Response of Aphanizomenon ovalisporum to certain environmental parameters was studied to gain a better understanding of the conditions which may have stimulated its autumnal bloom in Lake Kinneret. Optimal temperature for A. ovalisporum growth was 26–30?°C, resulting in growth rates of 0.2–0.3?day?1, similar to those observed in the lake. Maximal rate of CO2 fixation (assimilation numbers of 6–8?μg?C?μg?1?Chl?h?1) was obtained at low irradiances (I k of 40–100?μmol?photons?m?2?s?1), 200?μM Pi and low N:Pi ratios. Growth was strongly affected by phosphorus availability, reaching a maximum at Pi concentrations above 40?μM. The high demand for phosphorus was indicated by an increase in alkaline phosphatase activity. The relative abundance of Pi in the cells increased by 4-fold in Pi-rich compared with Pi-limited cultures. Uptake of Pi was faster in Pi-depleted compared with Pi-sufficient cells. Maximal photosynthetic rates and K1/2(HCO3 ?) were 140–220?μmol?O2?mg?1?Chl?h?1 and 10–24?μM, respectively. At pH 7.0 the K 1/2(CO2) was 2.2 and fell to 0.04?μM at pH 9.0. These data indicated that A. ovalisporum is a HCO3 ? user, and can explain its high photosynthetic rates during the bloom, under high pH and low dissolved CO2 conditions. Na+ concentrations of about 5?mM were essential for A. ovalisporum growth at high pH approaching values in the lake.  相似文献

Lectins from Lens culinaris and Arachis hypogaea immobilized on polyacrylamide beads were used for selective isolation of glycosylated surface membrane domains of adult Schistosoma mansoni worms, and the method was compared with the membrane isolation procedure developed with polycationic (Affi-Gel) beads. The lentil lectin proved to be suitable for interaction with surface membrane components: an increment in the specific activities of tegumental phosphohydrolases was observed in the bound fraction with respect to that observed in a total worm homogenate. A characteristic polypeptide pattern on gel electrophoresis was also seen, more restricted than that obtained with the bound Affi-Gel fraction. Immobilized peanut lectin was not successful as a method for isolating membrane material from the tegument of adult worms. Solubilization and dissociation of the lentil lectin-bound enzyme markers was achieved after addition of detergent and competing sugars. Glycosylation of the solubilized enzymes was further confirmed by affinity chromatography with fresh lentil lectin-coated beads. These results, together with histochemical evidences, suggest that the active sites of some of these enzymes are locted within or close to the cytoplasmic leaflet of the surface tegumental membranes, and allow us to propose a model for the double surface membrane complex where some proteins may be crossing the two bilayers.  相似文献

有性繁殖是植物种群形成与维持的主要方式。为探索退化湿地的快速恢复方法,为松花江下游退化湿地恢复提供科学依据,本研究开展了香蒲和芦苇种子快速发芽的有性繁殖实验。研究采用滤纸为发芽基质,通过变温培养试验,以未浸种处理为参照,分析了蒸馏水、双氧水(H2O2)、硝酸钾(KNO3)和高锰酸钾(KMnO4)溶液浸种的预处理方式对香蒲、芦苇种子发芽率和发芽速率的影响。结果表明:不同预处理方式对香蒲、芦苇种子的发芽率和发芽速率均具有显著影响。KMnO4溶液浸种再清洗处理条件下,香蒲种子发芽率和发芽速率均显著高于其他处理,平均发芽率可达未浸种处理条件下的3.1倍,发芽速率为16.17±0.80。芦苇种子的发芽率和发芽速率在经KNO3溶液浸泡再清洗处理后效果最佳,种子发芽率达96%-99%,发芽速率达28.43±0.71。因此,分别对香蒲、芦苇种子采用KMnO4和KNO3溶液浸泡再清洗的预处理方式可以缩短出苗时间,提高发芽率,从而加速湿地植被恢复进程。  相似文献

以棕榈残渣(Empty fruit bunch,EFB)为原料,通过预处理、酶解、发酵等过程制备纤维乙醇.首先对比了碱、碱/过氧化氢等预处理条件对棕榈残渣组成及酶解的影响,结果表明稀碱预处理效果较好.适宜的稀碱预处理条件为:NaOH浓度为1%,固液比为1∶10,在40℃浸泡24 h后于121℃下保温30 min,在该条件下,EFB的固体回收率为74.09%,纤维素、半纤维素和木质素的含量分别为44.08%、25.74%和13.89%.对该条件下预处理后的固体样品,以底物浓度5%、酶载量30 FPU/g底物酶解72 h,纤维素和半纤维素的酶解率分别达到84.44%和89.28%.进一步考察了酶载量和底物浓度对酶解的影响以及乙醇批式同步糖化发酵,当酶载量为30 FPU/g底物,底物浓度由5%增加至25%时,利用酿酒酵母Saccharomyces cerevisiae(接种量为5%,VIV)发酵72 h后乙醇的浓度分别为9.76 g/L和35.25 g/L,可分别达到理论得率的79.09%和56.96%.  相似文献

以枣(Jujube)皮为原料,经酶解和微波超声波处理后提取红色素,研究酰基化剂(Acylating agent)对红色素理化性质的影响.结果显示,枣皮红色素分别与阿魏酸和水杨酸形成的复合物对光和部分金属如Na+、Ca2+和Fe3+稳定;在pH 1~7范围内不稳定,pH 9~14范围内稳定.结果表明,阿魏酸和水杨酸与枣皮红色素复合后产生的酰基化产物有助于红色素分子的稳定.  相似文献

目的:建立质粒pVAX1-PENK的大规模制备2--艺。方法:对大肠杆菌工程菌DH5α-pVAX1-PENK进行补料发酵,利用自行发明的连续碱裂解过程对菌体进行裂解,经超滤浓缩后,用Sepharnse 6 Fast Flow层析柱分离DNA与RNA,再经Plasmidselect Xtra层析柱分离超螺旋质粒DNA与开环或线性质粒DNA,最后经Source 15Q层析柱精制质粒DNA。结果:发酵获得质粒pVAX1-PENK的产率为182mg/L,经碱裂解及层析分离后,最终制备的质粒DNA超螺旋比例大于98%,总回收率为60.5%,纯度(D260nm/D280nm)为1.8~2.0。结论:建立的质粒DNA生产工艺可以制备大量高纯度的质粒DNA,并避免了使用动物源性的酶及有毒试剂。  相似文献

Nucleotide pyrophosphatases/phosphodiesterases (NPPs) release nucleoside 5′-monophosphates from nucleotides and their derivatives. They exist both as membrane proteins, with an extracellular active site, and as soluble proteins in body fluids. The only well-characterized NPPs are the mammalian ecto-enzymes NPP1 (PC-1), NPP2 (autotaxin) and NPP3 (B10; gp130RB13-6). These are modular proteins consisting of a short N-terminal intracellular domain, a single transmembrane domain, two somatomedin-B-like domains, a catalytic domain, and a C-terminal nuclease-like domain. The catalytic domain of NPPs is conserved from prokaryotes to mammals and shows remarkable structural and catalytic similarities with the catalytic domain of other phospho-/sulfo-coordinating enzymes such as alkaline phosphatases. Hydrolysis of pyrophosphate/phosphodiester bonds by NPPs occurs via a nucleotidylated threonine. NPPs are also known to auto(de)phosphorylate this active-site threonine, a process accounted for by an intrinsic phosphatase activity, with the phosphorylated enzyme representing the catalytic intermediate of the phosphatase reaction. NPP1-3 have been implicated in various processes, including bone mineralization, signaling by insulin and by nucleotides, and the differentiation and motility of cells. While it has been established that most of these biological effects of NPPs require a functional catalytic site, their physiological substrates remain to be identified.  相似文献

Here, to determine the effects of transport stress on blood parameters in dogs, we investigated the changes in hematologic and serum chemical parameters in healthy beagle dogs transported from Beijing, China, to Osaka, Japan, to obtain the background data. Only the activity of serum alkaline phosphatase increased clearly upon arrival, a change attributed to transport stress, but the activity gradually reduced afterward. No marked changes in levels of other blood parameters were noted. Our findings here suggest that alkaline phosphatase is a useful tool for studying transport stress.  相似文献

New molecular peroxotungstate(VI) complexes with dipeptides as ancillary ligands of the type, [WO(O2)2(dipeptide)(H2O)].3H2O, dipeptide = glycyl-glycine or glycyl-leucine, have been synthesized and characterized by elemental analysis, spectral and physico-chemical methods including thermal analysis. The complexes contain side-on bound peroxo groups and a peptide zwitterion bonded to the metal centre unidentately through an O(carboxylate) atom. Investigations on certain biologically important key properties of these compounds and a set of dimeric compounds in analogous co-ligand environment, Na2[W2O3(O2)4(dipeptide)2].3H2O, dipeptide = glycyl-glycine and glycyl-leucine, reported previously by us revealed interesting features of the compounds. Each of the compounds despite having a 7 co-ordinated metal centre exerts a strong inhibitory effect on alkaline phosphatase activity with a potency higher than that of the free dipeptide, tungstate or peroxotungstate. The compounds exhibit remarkable stability in solutions of acidic as well as physiological pH and are weaker as substrate to the enzyme catalase, compared to H2O2. The mononuclear and dinuclear peroxotungsten compounds are efficient oxidants of reduced glutathione (GSH), a reaction in which only one of the peroxo groups of a diperoxotungsten moiety of the complexes was found to be active.  相似文献

Abstract

A solution of 2-methyl-5-tert-butylthiophenol and triethylamine in acetonitrile efficiently removed the methyl protecting groups from phosphate and phosphorothioate triesters and yielded oligonucleotides of high quality. This inexpensive odorless liquid is not toxic and is a suitable replacement for hazardous thiophenol and other reagents often used for this purpose.   相似文献
