Plasma membrane fractions were isolated from maize (Zea mays L.) endosperms and etiolated kernels to investigate the possible membrane location of the sucrose synthase (SS) protein. Endosperms from seedlings at both 12 and 21 days after pollination (DAP), representing early and mid-developmental stages, were used, in addition to etiolated leaf and elongation zones from seedlings. Plasma membrane fractions were isolated from this material using differential centrifugation and aqueous two-phase partitioning. The plasma membrane-enriched fraction obtained was then analyzed for the presence of sucrose synthase using protein blots and activity measurements. Both isozymes SS1 and SS2, encoded by the lociSh1 andSus1, respectively, were detected in the plasma membrane-enriched fraction using polyclonal and monoclonal antisera to SS1 and SS2 isozymes. In addition, measurements of sucrose synthase activity in plasma membrane fractions of endosperm revealed high levels of specific activity. The sucrose synthase enzyme is tightly associated with the membrane, as shown by Triton X-100 treatment of the plasma membrane-enriched fraction. The effects of environmental conditions on ear-shoot initiation have been investigated in three inbred genotypes of Zea mays L. which are used for seed production. Scanning electron microscopy (SEM) and binocular examination during the vegetative phase showed that axillary meristems are initiated at the same rate as the leaf primordia on the apical meristem, but with a delay of 5.6–7.0 plastochrons, depending on the genotype. Furthermore, the topmost axillary meristem is initiated on the same day as the tassel, whatever the genotype. One of the inbreds (B22) used in this study has been reported to exhibit, in field conditions, a reproductive failure affecting car initiation, causing the topmost car to be replaced at maturity by a sterile, leaf-like, structure. Scanning electron microscopic study of the formation of the abnormal axillary buds indicated that ear failure resulted from the early collapse of the axillary meristem followed by elongation of the prophyll or of the meristem itself. Using controlled environments, ear abortion was mimicked by a chilling treatment (10°C), given just before tassel initiation. Other factors, such as high irradiance and flooding, enhanced the abortive response. The critical stage for the main car was just before the initiation of the topmost axillary meristem which also corresponded to tassel initiation. Chilling the plants before or after tassel initiation either induced an acclimation response or had no effect. The three inbreds showed differential responses to the stress treatment, indicating that a genetic factor is implicated as well. It is suggested that chilling causes a perturbation of apical dominance which, in the responsive genotypes, represses axillary meristem development. The use of a stress-sensitive inbred such as B22 as a model system could yield some interesting clues to the mechanism of endogenous control of ear initiation in maize. 相似文献

Gravitropism of maize and rice coleoptiles was investigated with respect to its dependence on the angle of displacement or the initial stimulation angle (ISA). Close examination of curvature kinetics and the response to a drop in stimulation angle (SA) indicated that the gravtropic response during an early but substantial part of the curvature development is directly related to the ISA, there being no effect of the reduction of SA resulting from the curvature response itself. On the basis of this finding, the relationship between the steady SA and the curvature rate was determined. In maize, the curvature rate increased linearly with the sines of SAs up to an SA of 90 degrees. Rice coleoptiles, however, showed a saturation curve in the same range of SAs. The saturation profile was nearly identical between coleoptiles grown in air and those submerged in water, although the latter elongated much faster. Rice coleoptiles appeared to be far more sensitive to gravity than maize coleoptiles. It is concluded that the sensitivity to gravity, assessed through dependence on ISA, is a property inherent to a given gravitropic organ. Long-term measurements of curvature indicated that the coleoptiles bend back past the vertical. This overshooting was marked in submerged rice coleoptiles. 相似文献

The localization of the auxin receptor relevant to the control of elongation growth is still a matter of controversy. Auxin-induced elongation of maize coleoptile segments was measured by means of a high resolution auxanometer. When indole-3-acetic acid (IAA) was removed from the bathing solution, a rapid cessation of auxin-induced elongation was detected. This decline was delayed when the auxin efflux carrier was blocked by the phytotropins naphthylphthalamic acid (NPA) and pyrenoylbenzoic acid (PBA) or by triiodobenzoic acid (TIBA). The IAA concentration in NPA-pretreated segments was 2–3 times higher than in NPA-free controls 35 min after the removal of IAA in the bathing medium.
A similar rapid drop of growth after removal of auxin was observed for the rapidly-transported synthetic auxin, naphthaleneacetic acid (NAA). When the auxin efflux was blocked, growth induced by NAA was sustained much longer than IAA-stimulated elongation.

We conclude from these results that the presence of intracellular auxin is a necessary and sufficient condition for sustained auxin-induced elongation growth, at least for the phases during the 2 h after its application. Flooding effects on membrane permeability, lipid peroxidation and activated oxygen metabolism in corn (Zea mays L.) leaves were investigated to determine if activated oxygens are involved in corn flooding-injury. Potted corn plants were flooded at the 4-leaf stage in a controlled environment. A 7-day flooding treatment resulted in a significant increase in chlorophyll breakdown, lipid peroxidation (malondialdehye content), membrane permeability, and the production of superoxide (O
2
-
) and hydrogen peroxide (H2O2) in corn leaves. The effects were much greater in older leaves than in younger ones. Spraying leaves with 8-hydroxyquinoline (an O
2
-
scavenger) and sodium benzoate (an .OH scavenger) reduced the oxidative damage and enhanced superoxide dismutase (SOD) activity. A short duration flooding treatment elevated the activities of SOD, catalase, ascorbate peroxidase (AP), and glutathione reductase (GR), while further flooding significantly reduced the enzyme activities but enhanced the concentrations of ascorbic acid and reduced form glutathione (GSH). Summary The seed storage proteins of maize (Zea mays L.) are synthesized during endosperm development on membrane-bound polyribosomes. Protein body formation in normal genotypes occurs via a sequential deposition of the various types of zeins, and leads to the formation of spherical structures with a diameter of about l m. In the endosperm mutantopaque-2 the level of one zein class is reduced; these kernels exhibit an opaque phenotype instead of the vitreous phenotype displayed in normal genotypes, presumably due to the decrease in total zein protein at the time of desiccation. Previous microscopic examination ofopaque-2 protein bodies at 22 DAP (days after pollination) showed that the protein bodies were morphologically similar to those of normal genotypes. However, the endosperm ofopaque-2 maize at 14 DAP contains tubular arrays within the rough endoplasmic reticulum. These tubular arrays are tightly associated with the developing protein bodies. Long strands of tubules, sometimes 10 m in length, are observed in the endosperm, and partially formed protein bodies often seem to be forming directly from these tubular arrays. Maize seedlings were studied for their expression patterns of ABP1-mRNA and ABP1. In situ hybridization did not reveal hot spots of ABP1-mRNA accumulation. This result was supported by northern hybridization. In coleoptiles the ABP1-mRNA remains constant during day 1 to day 5 and is of low abundance (1.3 pg/μg total RNA). Northern blots indicated that in primary roots the mRNA level is even 10 times lower. Neither ABP1-mRNA nor ABP1 was found to be concentrated within the outer epidermis of the coleoptile. Analysis of immunostained western blots did not reveal pronounced differences in ABP1 content on the basis of equal amounts of fresh weight or total protein. We therefore assume ABP1 to be more or less equally distributed among the cells of the shoot tissues of maize seedlings. 相似文献
