A new approach to the development of a vaccine against meningococci of serogroups A and B was proposed. It involves the synthesis of conjugates of high-molecular capsule polysaccharides of the serogroup A meningococcus (PsA) with earlier synthesized protective fragments of membrane proteins from serogroup B meningococci. The conjugates were synthesized using a method that consists of the generation of aldehyde groups by oxidizing free vicinal hydroxyl groups of PsA and subsequent reaction of these groups with amino groups of the peptide. The reaction proceeds with the intermediate formation of the Schiff base, which is reduced to the stable secondary amine. The main parameters of the reaction were optimized in the synthesis of a PsA conjugate with a model peptide and methods of their characterization were developed. The reproducibility and efficiency of the synthetic procedure were demonstrated by the example of synthesis of PsA conjugates with fragments of protein PorA from the outer membrane of the serogroup B meningococcus. It was shown that, when administered without adjuvant, a conjugate of PsA with a protective peptide, which represents an exposed conserved fragment 306–332 of protein PorA, stimulates the formation of antibodies to the peptide and polysaccharide moieties of the molecule and is also capable of decreasing the degree of bacteremia in animals infected with serogroup A and serogroup B meningococci. The approach can be applied to the development of a complex vaccine for serogroup A and serogroup B meningococci.  相似文献

Sunfish rodlet cells were examined in vitro using a novel tissue explant system. Outgrowth of epidermal cell layers from explanted fish scales enabled both live cell videomicroscopy and immunocytochemical analysis of rodlet cells within the cell layer. Cells stained with fluorescent phallotoxin and antibody to tubulin showed that F‐actin is a component of the fibrous capsule that envelopes the cell and a microtubule network extends from the basal to apical ends of the cell interior. The fibrous capsule is also enriched for phosphotyrosine suggesting a potential signal‐transducing capability is present in this structure. Videomicroscopy analysis of live explant cultures demonstrated that rodlet cells are immobile and that interior structures are highly dynamic. Rodlet sacs can undergo extension and retraction, while intracellular particles can move rapidly within these cells. Fish scale tissue explants provide a useful system for analyzing the molecular composition and dynamic behavior of rodlet cells.  相似文献

目的

采用meta分析评价双歧杆菌三联活菌散/胶囊治疗儿童呼吸道感染继发腹泻的临床疗效。

方法

计算机检索中国知网、维普、万方、PubMed、Web of Science和Embase数据库,收集全球范围内在治疗儿童呼吸道感染继发腹泻中使用双歧杆菌三联活菌制剂(散/胶囊)的随机对照研究和前瞻性非随机对照研究,检索时间截至2022年7月,采用Jadad评分量表对纳入文献进行质量评价,采用Cochrane协作网提供的RevMan 5.4分析软件进行数据处理和meta分析。

结果

纳入19篇公开发表的文献进行meta分析,共3 086例患者,治疗组(对照组常规治疗措施+双歧杆菌三联活菌散/胶囊)1 578例,对照组1 508例,meta分析结果显示,治疗组的总体有效率显著高于对照组[OR = 6.54,95% CI(4.03,10.62),P<0.000 01],同时,治疗组的显效率优于对照组[OR=4.21,95% CI(2.97,5.97),P<0.000 01],并且,相比于对照组,治疗组的腹泻发病率降低[OR = 0.20,95% CI(0.12,0.34),P<0.000 01]),差异均有统计学意义。

结论

双歧杆菌三联活菌散/胶囊与常规治疗方法相比,可以提高儿童呼吸道感染继发腹泻的治疗总体有效率、治疗显效率,同时有效降低患儿的腹泻发病率。

The structure of the coating surrounding the metacercaria of Cryptocotyle lingua in the skin and the metacercaria of Liliatrema skrjabini in the muscles of the stone cockscomb was studied. Metacercariae of these trematodes were surrounded by a cyst adjacent to the helminth and by a capsule. In both species, the cyst consisted of electron- dense homogeneous material, which was probably synthesized by the parasite. The capsules were formed from host tissues; in C. lingua, they mostly consisted of ordered layers of collagen fibers and in L. skrjabini they were formed from fibroblasts and, presumably, phagocytes. The differences in capsule structure of the studied metacercariae may be related to the physiological features of surrounding tissues or to the taxonomic attribution of the trematodes.  相似文献

The sorption of cadmium (II), copper (II), lead (II),manganese (II), and zinc (II) by purified capsularpolysaccharide from the cyanobacterium Microcystis aeruginosafo. flos-aquae strainC3-40 was examined by four methods: equilibriumdialysis, metal removal from solution as detected byvoltammetry, metal accumulation by capsule-containingalginate beads, and calorimetry. The polysaccharide'ssaturation binding capacities for these metals rangedfrom 1.2 to 4 mmol of metal g-1 of capsule, whichcorresponds to 1 metal equivalent per 2 to 4saccharide subunits of the polymer. Competitionbetween paired metals was tested with simultaneous andsequential additions of metal. Cadmium (II) andlead (II), as well as lead (II) and zinc (II), competedrelatively equally and reciprocally for polymerbinding sites. In contrast, manganese (II) stronglyinhibited the binding of cadmium (II) and lead (II), butitself was not substantially inhibited by either theprior or simultaneous adsorption of cadmium (II) or lead (II).The data are interpreted with respect to overlap ofbinding sites and possibilities of altered polymerconformation or solvation. Calorimetric studies oflead (II) and cadmium (II) association reactions withthe polysaccharide suggest that the enthalpies aresmall and that the reactions may be driven by entropy.  相似文献

We prepared capsules containingSaccharomyces cerevisiae andZoogloea ramigera cells for the removal of lead (II) and cadmium ions. Microbial cells were encapsulated and cultured in the growth medium. TheS. cerevisiae cells grown in the capsule did not leak through the capsule membrane. The dried cell density reached to 250 g/l on the basis of the inner volume of the 2.0 mm diameter capsule after 36 hour cultivation. The dry whole cell exopolymer density of encapsulatedZ. ramigera reached to 200 g/L. The capsule was crosslinked with triethylene tetramine and glutaric dialdehyde solutions. The cadmium uptake of encapsulated whole cell exopolymer ofZ. ramigera was 55 mg Cd/g biosorbent. The adsorption line followed well Langmuir isotherm. The lead uptake of the encapsulatedS. cerevisiae was about 30 mg Pb/g biomass. The optimum pH of the lead uptake using encapsulatedS. cerevisiae was found to be 6. Freundlich model showed a little better fit to the adsorption data than Langmuir model. 95 percent of the lead adsorbed on the encapsulated biosorbents was desorbed by the 1 M HCl solution. The capsule was reused 50 batches without loosing the metal uptake capacity. And the mechanical strength of the crosslinked capsule was retained after 50 trials.  相似文献

Experimental studies suggest that the magnitude of chondrocyte deformation is much smaller than expected based on the material properties of extracellular matrix (ECM) and cells, and that this result could be explained by a structural unit, the chondron, that is thought to protect chondrocytes from large deformations in situ. We extended an existing numerical model of chondrocyte, ECM and pericellular matrix (PCM) to include depth-dependent structural information. Our results suggest that superficial zone chondrocytes, which lack a pericellular capsule (PC), are relatively stiff, and therefore are protected from excessive deformations, whereas middle and deep zone chondrocytes are softer but are protected by the PC that limits cell deformations in these regions. We conclude that cell deformations sensitively depend on the immediate structural environment of the PCM in a depth-dependent manner, and that the functional stiffness of chondrocytes in situ is much larger than experiments on isolated cells would suggest.  相似文献

目的:探讨双胶润通胶囊的润肠通便作用的效果及剂量.方法:实验采用动物试验,将小白鼠(雌雄各半,体重18~22 g)分成两个实验组,每组50只,实验一组进行小肠推进试验,实验二组进行排便试验,时间均为7 d.按推荐剂量每人每日5 g设计,分为低、中、高3个剂量组,另设空白对照和模型对照组.结果:在不影响小鼠体重的情况下,双胶润通胶囊高剂量组小肠推进率与模型对照组比较差异均有显著性变化(P《0.05);3个剂量组小鼠的首便时间、粪便粒数均高于便秘模型对照组,差异具有极显著性差异(P《0.01);结论:双胶润通胶囊对小鼠小肠运动有增强作用,并能缩短小鼠首便时间、增加小鼠的粪便粒数与重量,即:具有润肠通便作用.  相似文献

In mammals, the Transforming Growth Factor-beta (TGF-beta) superfamily controls a variety of developmental processes. In Drosophila, by contrast, a single member of the superfamily, decapentaplegic (dpp) performs most TGF-beta developmental functions. The complexity of dpp functions is reflected in the complex cis-regulatory sequences that flank the gene. Dpp is divided into three regions: Hin, including the protein-coding exons; disk, including 3' cis-regulatory sequences; and shortvein (shv), including noncoding exons and 5' cis-regulatory sequences. We analyzed the cis-regulatory structure of the shortvein region using a nested series of rearrangement breakpoints and rescue constructs. We delimit the molecular regions responsible for three mutant phenotypes: larval lethality, wing venation defects, and head capsule defects. Multiple overlapping elements are responsible for larval lethality and wing venation defects. However, the area regulating head capsule formation is distinct, and resides 5' to these elements. We have demonstrated this by isolating and describing two novel dpp alleles, which affect only the adult head capsule.  相似文献

The diversity and dynamics of Neisseria meningitidis populations generate a requirement for high resolution, comprehensive, and portable typing schemes for meningococcal disease surveillance. Molecular approaches, specifically DNA amplification and sequencing, are the methods of choice for various reasons, including: their generic nature and portability, comprehensive coverage, and ready implementation to culture negative clinical specimens. The following target genes are recommended: (1) the variable regions of the antigen-encoding genes porA and fetA and, if additional resolution is required, the porB gene for rapid investigation of disease outbreaks and investigating the distribution of antigenic variants; (2) the seven multilocus sequence typing loci-these data are essential for the most effective national, and international management of meningococcal disease, as well as being invaluable in studies of meningococcal population biology and evolution. These targets have been employed extensively in reference laboratories throughout the world and validated protocols have been published. It is further recommended that a modified nomenclature be adopted of the form: serogroup: PorA type: FetA type: sequence type (clonal complex), thus: B: P1.19,15: F5-1: ST-33 (cc32).  相似文献
