We developed a modified method enabling stable MALDI-MS analysis and fluorescent detection of sialyl-compounds. The modification involved the amidation of sialic acid (Neu5Ac) at the position of the carboxyl group using the fluorescent reagent, 2-(2-pyridilamino)ethylamine (PAEA). In this study the following sialyl-compounds were amidated, 3′-sialyllactose (3′-SL), 6′-sialyllactose (6′-SL), and ganglioside GM3. Yields of PAEA-3′-SL, PAEA-6′-SL, and PAEA-GM3 were 45%, 60%, and 30%, respectively. The PAEA-amidation enabled fluorescence detection of structural isomers using HPLC and TLC at sensitivity levels as low as pmol. In MALDI-TOF-MS and/or MS/MS analysis in positive ion mode, PAEA-amidation provided the following advantages: suppression of preferential cleavage of Neu5Ac; enhancement of molecular-related ion intensities; simplification of MS spectra; and finally, since PAEA-amidation did not cleave the linkage between sugar and aglycon of sialylglycoconjugate, MALDI-TOF-MS and MS/MS analyses revealed the complete structure of the molecule. 相似文献

A new Pliocene deposit with vegetal impressionswas discovered at Rasteau (near Vaison-la-Romaine, Vaucluse. The study of these impression and the sporopollinic analysis from many levels allows us to specify its stratigraphical position in the Pliocene of the Rhone valley. So, the above mentioned levels settled just before the Hautimagne deposit (Neogene zone MN 14 for the mammals). In Mediterranean climates, bimodal growth patterns, corresponding to two peaks in radial increment during favorable seasons, have been described in several tree species. However, we lack a better mechanistic understanding of bimodality and its potential responses to the predicted warming and aridification trends. Filling this research gap is important since growth duration affects the capacity of trees to form wood and uptake carbon. Here we used an 11-year (1994–2004) long record of dendrometer data of the Mediterranean Holm oak (Quercus ilex) and compared how climate related to radial increment in trees from the south- and the north-facing slopes. We also related climate variables to tree-ring width and the production of intra-annual density fluctuations (IADFs), which reflects bimodality. In this paper, we introduce a model called VS-Lite2 to simulate tree-growth dynamics, which is a modified version of the process-based Vaganov-Shashkin Lite model. The VS-Lite2 model adequately reproduced the bimodal intra‐annual pattern of radial growth, IADFs, and annual tree growth. Trees from the south-oriented slope grew more, produced more IADFs and showed a more marked bimodal pattern than trees from the north-facing slope. These differences agree with the observation that late-summer drought constrained growth. Therefore, radial-growth models should consider plastic bimodality and micro-environmental conditions in areas subjected to seasonal droughts. 相似文献

In this study, wood anatomy, tree-ring width and wood density of Pinus sylvestris at the northern timberline in Fennoscandia were used to identify relationships among the parameters and to screen them for their climatic signals. Furthermore we investigated the influence of the juvenile wood section for all parameters developed. The measurements of wood anatomy were conducted with confocal laser scanning microscopy (CLSM) while the density profiles were produced using an Itrax MultiScanner. We developed chronologies of ring width, wood density and anatomy for a period between 1940 and 2010. Correlations between wood density and wood anatomy were strong in the latewood part. For some wood anatomy and density chronologies youth trends were found in the juvenile part. Wood density decreased from the pith up to the 9th ring and stabilized afterwards, while cell lumen diameter and lumen area increased simultaneously up to the 15th ring. All chronologies contained strong summer temperature signals. The wood anatomical variables provided additional information about seasonal precipitation which could not be found in wood density and tree-ring widths. Our study confirmed previous results stating that the parameter maximum density contains the strongest climate signal, that is, summer temperatures at the northern timberline. Nevertheless, the intra-annual data on tracheid dimensions showed good potential to supply seasonal climatic information and improve our understanding of climatic effects on tree growth and wood formation. 相似文献

The fate of a memory, whether stored or forgotten, is determined by the ability of an active or tagged synapse to undergo changes in synaptic efficacy requiring protein synthesis of plasticity-related proteins. A synapse can be tagged, but without the “capture” of plasticity-related proteins, it will not undergo long lasting forms of plasticity (synaptic tagging and capture hypothesis). What the “tag” is and how plasticity-related proteins are captured at tagged synapses are unknown. Ca2+/calmodulin-dependent protein kinase II α (CaMKIIα) is critical in learning and memory and is synthesized locally in neuronal dendrites. The mechanistic (mammalian) target of rapamycin (mTOR) is a protein kinase that increases CaMKIIα protein expression; however, the mechanism and site of dendritic expression are unknown. Herein, we show that mTOR activity mediates the branch-specific expression of CaMKIIα, favoring one secondary, daughter branch over the other in a single neuron. mTOR inhibition decreased the dendritic levels of CaMKIIα protein and mRNA by shortening its poly(A) tail. Overexpression of the RNA-stabilizing protein HuD increased CaMKIIα protein levels and preserved its selective expression in one daughter branch over the other when mTOR was inhibited. Unexpectedly, deleting the third RNA recognition motif of HuD, the domain that binds the poly(A) tail, eliminated the branch-specific expression of CaMKIIα when mTOR was active. These results provide a model for one molecular mechanism that may underlie the synaptic tagging and capture hypothesis where mTOR is the tag, preventing deadenylation of CaMKIIα mRNA, whereas HuD captures and promotes its expression in a branch-specific manner. 相似文献

Different maximum latewood density and blue intensity measurements techniques reveal similar results
Annually resolved and absolutely dated Maximum Latewood Density (MXD) and Blue Intensity (BI) measurements are frequently used for reconstructing summer temperature variability over the last centuries to millennia. A direct comparison of the outcome of both methods using similar material is needed due to how quickly this method is being adopted. The application of slightly different measuring systems (hardware) and analysis tools (software) in tandem with different wood samples and preparation procedures further challenges any straightforward assessment. Here we process 26 Norway spruce samples from the upper timberline in the Polish Tatra Mountains with the six most frequently used MXD and BI applications. Although offset is found in the raw MXD and BI data (0.04–0.13 g/cm3 and 0.45–1.58 dimensionless blue intensity), interannual and longer-term fluctuations are significantly (p < 0.01) positively correlated between all MXD and BI time-series. Our results emphasize the potential of faster and cheaper, as well as overall more user-friendly techniques to generate reliable MXD surrogates for high-frequency dendroclimatological studies. Although the correlations between MXD and BI were lower than within MXD and BI, the results of growth-climate response performed for both proxies show only marginal differences. The obtained level-offset further questions the suitability of joining different density surrogates for developing long-term composite chronologies to reconstruct low-frequency climate variability. 相似文献

Spatial patterns in vessel diameter, vessel density and xylem conducting efficiency within a crown were examined in closed-canopy
trees of silver birch (Betula pendula). The variation in anatomical and hydraulic characteristics of branches was considered from three perspectives: vertically
within a crown (lower, middle and upper crown), radially along main branches (proximal, middle and distal part), and with
respect to branch orders (first-, second- and third-order branches). Hydraulically weighted mean diameter of vessels (D
h) and theoretical specific conductivity of the xylem (k
t) exhibited no vertical trend within the tree crown, whereas leaf-specific conductivity of the xylem (LSCt) decreased acropetally. Variation in LSCt was governed by sapwood area to leaf area ratio (Huber value) rather than by changes in xylem anatomy. The acropetal increase

T) within the birch canopy is attributable to longer path length within the lower-crown branches and higher hydraulic resistance
