Nitrogen (N) mineralization is a spatially variable and difficult component of the N cycle to quantify accurately under field conditions. Net N-mineralization was compared by direct measurement, indirect estimate, and laboratory incubation for a restored tallgrass prairie and for deficiently and optimally N-fertilized, no-tillage (NT) and chisel-plowed (CP) maize (Zea mays L.) agroecosystems on Plano silt loam soil (fine-silty, mixed, superactive, mesic Typic Argiudoll) in Wisconsin, USA. Four years of in-situ field measurements using an incubated-soil-core/ion-exchange-resin-bag technique showed that land use significantly affected net N-mineralization. Net N-mineralization was consistently smaller in the restored prairie than in the maize agroecosystems and typically larger in the CP than in the NT maize agroecosystems. Three independent methods for indirectly estimating annual net N-mineralization (i.e., N budget residual, deficiently N-fertilized plant N uptake, and profile-scaled in-situ field measurements) were relatively consistent at capturing land-use and tillage effects on net N-mineralization. Laboratory incubation and periodic leaching of Fall-sampled soils demonstrated that both mineralized N and labile C were co-limiting factors influencing N-mineralization in agricultural soils and generally supported field measurements by showing a significant difference in net N-mineralization with and without added fertilizer-N.  相似文献

Organ preservation solutions have been designed to protect grafts against the injury inflicted by cold ischemia. However, toxicity of University of Wisconsin (UW) solution during rewarming has been reported. Therefore, we here assessed the toxicity of UW, histidine-tryptophan-ketoglutarate (HTK), Euro-Collins, histidine-lactobionate (HL), sodium-lactobionate-sucrose and Celsior solutions in cultured hepatocytes under hypothermic (4 °C), intermediate (21 °C) and physiological (37 °C) conditions. Marked toxicity of UW, HTK, HL and Euro-Collins solutions was observed at both 37 and 21 °C. With the exception of UW solution, these solutions also increased cell injury during cold incubation (LDH release after 18 h at 4 °C: HTK 76 ± 2%, Euro-Collins 78 ± 17%, HL 81 ± 15%; control: Krebs-Henseleit buffer 20 ± 6%). Testing of individual components using modified Krebs-Henseleit buffers suggested that histidine and phosphate are responsible for (part of) this toxicity. These potential toxicities should be taken into account in the development of future preservation solutions.  相似文献

Abstract. We evaluated variability in cover estimation data obtained by (1) two sampling teams who double sampled plots and (2) one team that used two methods (line intercepts and visual estimation of cover classes) to characterize vegetation of herbaceous wetlands. Species richness and cover estimates were similar among teams and among methods, but one sampling team scored cover higher than the other. The line intercept technique yielded higher cover estimates but lower species richness estimates than the cover class method. Cluster analyses of plots revealed that 36% and 11% of plots sampled consecutively by two teams or using two methods, respectively, were similar enough in species composition and abundance to be paired together in the resulting clustering tree. Simplifying cover estimate data to presence/absence increased the similarity among both teams and methods at the plot scale. Teams were very similar in their overall characterization of sites when cover estimation data were used, as assessed by cluster analysis, but methods agreed best on their overall characterization of sites when only presence/absence data were considered. Differences in abundance estimates as well as pseudoturnover contribute to variability. For double sampled plots, pseudoturnover was 19.1%, but 57.7% of pseudo‐turnover cases involved taxa with ≤ 0.5% cover while only 3.4% involved taxa with > 8% cover. We suggest that vegetation scientists incorporate quality control, calibrate observers and publish their results.  相似文献

We evaluated several factors influencing the taxonomic richness of macrophytes, benthic invertebrates, snails, and fish in a series of northern Wisconsin lakes. We chose the study lakes to decouple the potential effects of ionic strength of lake water and stream connection, two factors that are usually highly correlated and therefore have been confounded in previous studies. In addition, our study lakes covered a wide range in a variety of characteristics, including residential development, abundance of exotic species, nutrient concentrations, predator abundance, and lake size. Species richness within each of the four taxonomic groups was significantly positively related to ionic strength (as measured by specific conductance); we also found secondary associations with other variables, depending on the specific group of organisms. The relationship between richness and lake area was dependent on the specific conductance of the lake and the vagility of the organisms; less vagile groups of organisms showed stronger and steeper species–area relationships in low-conductivity lakes. Further, after variance owing to specific conductance was removed, the presence of stream connections was positively related to species richness for fish, snails, and macrophytes as well as familial richness in benthic invertebrates. Our results indicate that lakes with relatively more groundwater input have lower extinction rates for all four groups of taxa and that lakes with stream inlets and outlets have enhanced immigration rates for fish, snails, benthic invertebrate families, and macrophytes. These findings link processes of immigration and extinction of four groups of organisms of varying vagility to landscape-level hydrologic characteristics related to the glacial history of the region.  相似文献

Post-transplantation thrombosis may occur in donor segments of iliac arteries and livers following surgical removal and storage in University of Wisconsin (UW) solution for transplantation. We have previously suggested that purine receptors are vulnerable to denaturation after UW storage. The aims of the present study were to determine what particular subtypes of purine P2Y receptors in rabbit thoracic aorta deteriorate after 8 days of UW storage by studying vascular reactivity to acetylcholine, ATP, 2MeSATP and UTP. Ring segments of aortae from male New Zealand White rabbits were mounted upon fine-wire myographs and vasodilatation to the above agents tested on fresh tissue, and after 8 days of UW storage. Vasodilatation to ATP was attenuated by 100 microM L-NAME in fresh tissue suggesting that the relaxant response was, in part, due to nitric oxide (NO). P2Y-mediated relaxation to ATP was significantly attenuated by UW storage and cholinergic responses were not. This attenuated relaxation to ATP was not further attenuated by L-NAME, suggesting a loss of the NO-dependent mechanism. De-endothelialisation indicated that UTP-mediated vasorelaxation, via P2Y(2) receptors, was endothelium-dependent. Any residual endothelium-independent relaxation to UTP was abolished by UW storage and endothelium-dependent UTP relaxation was reduced to the same level as that seen in fresh, de-endothelialised tissue. In contrast responses to 2MeSATP, via P2Y(1) receptors, were predominantly endothelium-independent and were only partially attenuated by UW storage. Responses to pyridoxalphosphate-6-azophenyl-2('),4(')-disulphonic acid (PPADS) and L-NAME suggested that vasorelaxation to 2MeSATP and UTP was mediated by P2Y(1) and P2Y(2) receptors, respectively. It is therefore concluded that UW storage predominantly decreases P2Y(2) receptor-mediated vascular reactivity.  相似文献

In this work we address key phenomena observed with classical set shifting tasks as the “Wisconsin Card Sorting Test” or the “Stroop” task: Different types of errors and increased response times reflecting decreased attention. A component of major importance in these tasks is referred to as the “attentional control” thought to be implemented by the prefrontal cortex which acts primarily by an amplification of task relevant information. This mode of operation is illustrated by a neurodynamical model developed for a new kind of set shifting experiment: The Wisconsin-Delayed-Match-to-Sample task combines uninstructed shifts as investigated in Wisconsin-like tasks with a Delayed-Match-to-Sample paradigm. These newly developed WDMS experiments in conjunction with the neurodynamical simulations are able to explain the reason for decreased attention in set shifting experiments as well the different consequences of decreased attention in tasks requiring bivalent yes/no responses compared to tasks requiring multivalent responses. We previously described the entity of cold-induced apoptosis to rat hepatocytes and characterized its major, iron-dependent pathway. However, after cold incubation in some solutions, e.g. cell culture medium, hepatocytes show an additional, yet uncharacterized component of cold-induced injury. We here assessed the effects of organ preservation solutions on both components of cold-induced injury and tried to further characterize the iron-independent component. None of the preservation solutions (University of Wisconsin, histidine-tryptophan-ketoglutarate, Euro-Collins, histidine-lactobionate, sodium-lactobionate-sucrose and Celsior solutions) provided significant protection against cold-induced cell injury (LDH release after 24-h cold incubation/3h rewarming >65% for all solutions); three solutions even enhanced cold-induced injury. However, when the predominant iron-dependent mechanism was eliminated by the addition of iron chelators, all preservation solutions yielded hepatocyte protection that was clearly superior to the one obtainable in cell culture medium or Krebs-Henseleit buffer with iron chelators (LDH release after 24-h cold incubation/3h rewarming 相似文献

PURPOSE: Performance of anatomical metrics of Response Evaluation Criteria in Solid Tumors (RECIST1.1) versus Positron Emission Tomography Response Criteria in Solid Tumors (PERCIST1.0) for neoadjuvant chemoradiation (nCR) of pancreatic adenocarcinoma was evaluated based on the pathological treatment response (PTR) data. METHODS AND MATERIALS: The pre- and post-nCR CT and PET data for 14 patients with resectable or borderline resectable pancreatic head adenocarcinoma treated with nCR followed by surgery were retrospectively analyzed. These data were compared with the PTR which were graded according to tumor cell destruction (cellularity), with Grade 0, 1, 2 or 3 (G0, G1, G2 or G3) for complete, moderate, minimal and poor responses, respectively. Maximum standardized uptake value (SUVmax) was defined using body-weight (SUVbw). PERCIST1.0 was defined using lean-body mass normalized SUV (SUVlb or SUL). RECIST1.1 was defined by contouring the whole pancreas head on the CT image. Pre- and post-SUL-peak and SUVmax, RECIST1.1 and PETRECIST1.0 were correlated with PTR using Pearson’s correlation coefficient test. RESULTS: The average mean and SD in SUL-peak for all patients analyzed were lower in post-nCR (3.63±1.06) compared to those at pre-nCR (4.29±0.89). Using PERCIST1.0, 62% of patients showed stable metabolic disease (SMD), 23% partial metabolic response (PMR), and 15% progressive metabolic disease (PMD). Using RECIST1.1, 85% of patients showed stable disease (SD), 8% partial response (PR), and 7% progressive diseases (PD). A poor insignificant correlation was established between PRT and PERECIST1.0 (r=0.121), whereas no correlation was seen with RECIST1.1. CONCLUSIONS: PERCIST1.0 appears to increase the chance of detecting patients with progressive disease compared to the conventional anatomical-based assessment of RECIST1.1. The integration of these additional radiographic metrics in assessing treatment response to nCR for pancreatic adenocarcinoma may provide a promising strategy to better select patients that are most suitable for therapeutic intensification.  相似文献

Both neurocognitive deficits and schizophrenia are highly heritable. Genetic overlap between neurocognitive deficits and schizophrenia has been observed in both the general population and in the clinical samples. This study aimed to examine if the polygenic architecture of susceptibility to schizophrenia modified neurocognitive performance in schizophrenia patients. Schizophrenia polygenic risk scores (PRSs) were first derived from the Psychiatric Genomics Consortium (PGC) on schizophrenia, and then the scores were calculated in our independent sample of 1130 schizophrenia trios, who had PsychChip data and were part of the Schizophrenia Families from Taiwan project. Pseudocontrols generated from the nontransmitted parental alleles of the parents in these trios were compared with alleles in schizophrenia patients in assessing the replicability of PGC‐derived susceptibility variants. Schizophrenia PRS at the P‐value threshold (PT) of 0.1 explained 0.2% in the variance of disease status in this Han‐Taiwanese samples, and the score itself had a P‐value 0.05 for the association test with the disorder. Each patient underwent neurocognitive evaluation on sustained attention using the continuous performance test and executive function using the Wisconsin Card Sorting Test. We applied a structural equation model to construct the neurocognitive latent variable estimated from multiple measured indices in these 2 tests, and then tested the association between the PRS and the neurocognitive latent variable. Higher schizophrenia PRS generated at the PT of 0.1 was significantly associated with poorer neurocognitive performance with explained variance 0.5%. Our findings indicated that schizophrenia susceptibility variants modify the neurocognitive performance in schizophrenia patients.  相似文献

Dermatoglyphic ridge counts of the prints of 59 rhesus monkeys (Macaca mulatta) whose mothers had been treated with injections of testosterone during their pregnancies were studied to determine the effect of the day the hormone began to be administered, the amount of hormone administered, and the number of days of hormone administration upon the dermatoglyphic variation of the offspring. Of the three hormone variables, only the day of beginning administration (STARTDAY) was significantly associated with dermatoglyphic variation, and its positive significance was demonstrated with the ridge counts of Area I on both the left and right hand, Area II of the left hand, and the total ridge counts of both hands. These results are discussed within the context of the timing of the dermatoglyphic window, and the differences in the findings between the monkey and earlier human studies are addressed. © 1994 Wiley-Liss, Inc.  相似文献
