The effect of dietary fibres on constituents of complex carbohydrates in various tissues of streptozotocin induced diabetic rats is presented by analysing different constituents of complex carbohydrates in presence and absence of dietary fibre. Wheat bran was effective in preventing the decrease (14%) in total sugars in spleen and an increase in total sugars in stomach (33%) during diabetes. Decrease in uronic acid content during diabetes in spleen was prevented to the extent of 25% by the presence of wheat bran in the diet. The other parameters which were affected by the presence of wheat bran in the diet during diabetes are amino sugar (brain and stomach), sulphates (liver) and protein (lungs and stomach). Guar gum was effective in preventing the decrease in total sugar content in spleen by 28% and sulphate content in liver by 14% during diabetes. Variation in protein content in lungs was observed in diabetes. The results indicated beneficial role of dietary fibres like wheat bran and guar gum on complex carbohydrates to varying extents in different tissues. 相似文献

Apoptosis, or programmed cell death, is a well-ordered process that allows damaged or diseased cells to be removed from an organism without severe inflammatory reactions. Multiple factors, including microbial infection, can induce programmed death and trigger reactions in both host and microbial cellular pathways. Whereas an ultimate outcome is host cell death, these apoptotic triggering mechanisms may also facilitate microbial spread and prolong infection. To gain a better understanding of the complex events of host cell response to microbial infection, we investigated the molecular role of the microorganism Enteropathogenic Escherichia coli (EPEC) in programmed cell death. We report that wild type strain of EPEC, E2348/69, induced apoptosis in cultured PtK2 and Caco-2 cells, and in contrast, infections by the intracellularly localized Listeria monocytogenes did not. Fractionation and concentration of EPEC-secreted proteins demonstrated that soluble protein factors expressed by the bacteria were capable of inducing the apoptotic events in the absence of organism attachment, suggesting adherence is not required to induce host cell death. Among the known EPEC proteins secreted via the Type III secretion (TTS) system, we identified the translocated intimin receptor (Tir) in the apoptosis-inducing protein sample. In addition, host cell ectopic expression of an EPEC GFP-Tir showed mitochondrial localization of the protein and produced apoptotic effects in transfected cells. Taken together, these results suggest a potential EPEC Tirmediated role in the apoptotic signaling cascade of infected host cells. 相似文献

The two major cortices of the brain--the cerebral and cerebellar cortex--are massively connected through intercalated nuclei (pontine, cerebellar and thalamic nuclei). We suggest that the two cortices co-operate by generating precise temporal patterns in the cerebral cortex that are detected in the cerebellar cortex as temporal patterns assembled spatially in the mossy fibers. We will begin by showing that the tidal-wave mechanism works in the cerebellar cortex as a read-out mechanism for such spatio-temporal patterns due to the synchronous activity they generate in the parallel fiber system which drives the Purkinje cells--the output neurons of the cerebellar cortex--to fire action potentials. We will review the anatomy of the mossy fibers and show that within a "beam", or "row" of cerebellar cortex the mossy fibers in principle could embed a vast number of tidal-wave generating sequences. Based on anatomical data we will argue that the cerebellar mossy fiber-granule cell-Purkinje cell system can potentially detect and--through learning--select from an enormous number of spatio-temporal patterns. 相似文献

The present study describes the effects of several high-fat low-cholesterol antiatherogenic diets on the hepatic lipid peroxidation and hepatic antioxidant systems in apolipoprotein E-deficient mice. Eighty mice were distributed into five groups and fed with regular mouse chow or chow supplemented with coconut, palm, olive and sunflower seed oils. After ten weeks, they were sacrificed and the livers were removed so that lipid peroxidation and -tocopherol concentrations, and superoxide dismutase, glutathione peroxidase and glutathione reductase activities could be measured. The size of the atherosclerotic lesions in the aortas was also measured. Results showed that the diets supplemented with olive oil, palm oil or sunflower seed oil significantly decreased the size of the lesion. However, there was an association between those mice that were on diets supplemented with palm or coconut oils and a significant increase in hepatic lipid peroxidation. This association was not found in animals fed with olive or sunflower seed oils, the diets with the highest content of vitamin E. The dietary content of vitamin E was significantly correlated (r = 0.98; p < 0.05) with the hepatic concentration of this compound. Our study suggests that the high content of vitamin E in olive oil or sunflower seed oil may protect from the undesirable hepatotoxic effects of high-fat diets in apo E-deficient mice and that this should be taken into account when these diets are used to prevent atherosclerosis. 相似文献

A method was developed for the determination of selenium concentration in serum by flow injection-hydride generation-atomic
absorption spectrometry (FI-HG-AAS) following microwave digestion of serum samples and reduction of selenate to selenite.
The detection limit of the method was 0.3 μg Se/L and the characteristic concentration, corresponding to the 0.0044 absorbance
signal, was 0.12 μg Se/L. The results from the analysis of two Seronorm standard reference materials showed good agreement
with the certified values. The method was then used to analyze selenium in sera of Austrian and Slovenian people for the calculation
of dietary intakes. The selenium concentrations in sera of mothers at delivery, their neonates, and the male and female adults
were 71 ± 14, 42 ± 6, 75 ± 21, and 65 ± 16 μg/L for the Austrians and 62 ± 15, 34 ± 7, 70 ± 12, and 66 ± 15 μg/L for the Slovenians.
The dietary intakes of selenium of the mothers and the male and the female adults were calculated as 52, 37, and 46 μg/d for
the Austrians and 45, 38, and 32 μg/d for the Slovenians. 相似文献

Physiological and biochemical changes in relation to inorganic nitrogen availability were studied for tank-cultivated Ulva
rigida grown under nitrogen- enriched and nitrogen-depleted seawater. U. rigida was initially cultivated in nitrogen-enriched
seawater (daily concentrations of NH4+ and NO3- + NO2- ranged between 0.5–1.7 and 0.06–0.15 mg L-1, respectively), then transferred
to nitrogen-depleted seawater where photosynthetic capacity decreased to zero after 23 d. At the time (14 d) when photosynthetic
rates were lower than 2.0 μmol O2 g-1 FW min-1 and strong bleaching had occurred, some algae were returned to the initial
nitrogen-enriched seawater to study recovery from N-limited growth. Data on biochemical composition (chlorophylls, ash, caloric
content, fatty acids and dietary fibres) and colouration varied significantly depending on the nitrogen conditions. C:N ratios
correlated significantly with biochemical parameters. Fatty acid (FA) synthesis continued during the N-starvation period;
saturated and mono-unsaturated FA increased to a maximun of 72.2%, while poly-unsaturated fatty acids (PUFA) decreased to
27.7%. During the N-enriched recovery period, the reverse was found. C:N ratios above 10 correlated with carbohydrate synthesis
as shown by the dietary fibre level. Under nitrogen enriched conditions, C:N ratios decreased along with a decrease in fibre
level. Under controlled conditions, nitrogen represents a major influence on the development of intensive tank cultivation
of Ulva rigida, not only by affecting parameters closely related to nitrogen metabolism but also some clearly influenced by
carbon uptake.

A compartmental model of myelinated nerve fiber was used to show that primary afferent depolarization (PAD), as elicited by axo-axonic synapses, reduces the amplitude of propagating action potentials primarily by interfering with ionic current responsible for the spike regeneration. This reduction adds to the effect of the synaptic shunt, increases with the PAD amplitude, and occurs at significant distances from the synaptic zone. PAD transiently enhances the sodium current activation, which partly accounts for the PAD-induced fiber hyperexcitability, and enhances sodium inactivation on a slower time course, thus reducing the amplitude of action potentials. In vivo, intra-axonal recordings from the intraspinal portion of group I afferent fibers were carried out to verify that depolarizations reduced the amplitude of propagating action potentials as predicted by the model. Because a biochemical function has not been defined for boron (B), its nutritional essentiality has not been firmly established.
Nonetheless, dietary guidance should be formulated for B, because it has demonstrated beneficial, if not essential, effects
in both animals and humans. Intakes of B commonly found with diets abundant in fruits, vegetables, legumes, pulses, and nuts
have effects construed to be beneficial in macromineral, energy, nitrogen, and reactive oxygen metabolism, in addition to
enhancing the response to estrogen therapy and improving psychomotor skills and cognitive processes of attention and memory.
Perhaps the best-documented beneficial effect of B is on calcium (Ca) metabolism or utilization, and thus, bone calcification
and maintenance. The paradigm emerging for the provision of dietary guidance that includes consideration of the total health
effects of a nutrient, not just the prevention of a deficiency disease, has resulted in dietary guidance for chromium (Cr)
and fluoride; both of these elements have beneficial effects in humans, but neither has a defined biochemical function. Knowledge
of B nutritional effects in humans equals or is superior to that of Cr and fluoride; thus, establishing a dietary reference
intake for B is justified. An analysis of both human and animal data suggests that an acceptable safe range of population
mean intakes of B for adults could well be 1–13 mg/d. Recent findings indicate that a significant number of people do not
consistently consume more than 1 mg B/d; this suggests that B could be a practical nutritional or clinical concern. 相似文献

Knowledge of daily boron (B) intakes will assist researchers in establishing B requirements and elucidating the metabolic
role of B in humans. B concentrations in commonly consumed foods were utilized to approximate the B intake of selected US
populations. Triplicate food samples were digested at low temperatures with 16M HNO3 and 30% H2O2, and analyzed by inductively coupled plasma spectroscopy (ICP). Milk and dairy products, juices, and beverages were the largest
contributors to dietary B. Total B content of average daily diets ranged from 548 μg for toddlers (2 yr old) to 883 μg for
mature males (60–65 yr old), excluding tap water contributions. Toddlers consumed 3.7 times more B than mature males when
adjusted for body weight (body wt) and consumed the diet with the highest boron density (1.8 μg/kJ or 0.43 μg/kcal). Adolescent
females consumed a diet with the lowest B density (1.12 μg/kJ or 0.26 μg/kcal). Food B concentrations applied to diet records
(1020 μg/d) and ICP analysis of the corresponding food composites (1170 μg/d) were comparable (p < 0.05). Current estimations of B in US diets are consistent with reports in the literature stating that normal adult daily
B intakes are approx 1 mg.

We report the development of a new method of alkali‐catalyzed low temperature wet crosslinking of plant proteins to improve their breaking tenacity without using high temperatures or phosphorus‐containing catalysts used in conventional poly(carboxylic acid) crosslinking of cellulose and proteins. Carboxylic acids are preferred over aldehyde‐containing crosslinkers for crosslinking proteins and cellulose because of their low toxicity and cost and ability to improve the desired properties of the materials. However, current knowledge in carboxylic acid crosslinking of proteins and cellulose requires the use of carboxylic acids with at least three carboxylic groups, toxic phosphorous‐containing catalysts and curing at high temperatures (150–185°C). The use of high temperatures and low pH in conventional carboxylic acid crosslinking has been reported to cause substantial strength loss and/or undesired changes in the properties of the crosslinked materials. In this research, gliadin, soyprotein, and zein fibers have been crosslinked with malic acid, citric acid, and butanetetracarboxylic acid to improve the tenacity of the fibers without using high temperatures and phosphorus‐containing catalysts. The new method of wet crosslinking using carboxylic acids containing two or more carboxylic groups will be useful to crosslink proteins for various industrial applications. © 2009 American Institute of Chemical Engineers Biotechnol.
