应用胶银技术,对软组织肿瘤特别是纤维源性肿瘤核仁组成区蛋白进行了定量研究。在软组织一些恶性肿瘤中,以血管肉瘤、横纹肌肉瘤核仁组成区蛋白均值最高,并以此排列了它们的恶性度顺序。在纤维源性肿瘤中,纤维瘤、瘤样纤维组织增生、纤维肉瘤及其亚类核仁组成区蛋白均值有显著与高度显著性差异(P<0.05、P<0.001),其中随访的34例纤维肉瘤患者,具有高核仁组成区蛋白均值(≥5)与低核仁组成区蛋白均值(<5)的5年生存率分别是20%和53%(P<0.05)。因此,作者认为核仁组成区蛋白定量在判定软组织肿瘤的恶性度、鉴别其良恶性及预测患者预后中均有一定实用价值。 相似文献

对48例Ⅲ°烧伤病人的创面,定量植入枯草杆菌BS224菌后,分别在24h、48h、72h及96h做痂下组织细菌定量检测。结果显示:枯草杆菌对痂下组织的致病菌有明显的拮抗作用。感染创面的BS224菌体数量24—48小时显著增加,72—96小时而下降。与清洁创面的BS224菌动态变化上相同,呈常态曲线的规律变化。 相似文献

Summary Embryogenic callus was formed from several cultivars of cotton (Gossypium hirsutum L.) when sections of hypocotyl and cotyledon were cultured on medium supplemented with 5 mg/liter 6-(γ, γ-dimethylallyl-amino)-purine
(2iP) and 0.1 mg/liter α-naphthaleneacetic acid (NAA) for callus initiation and proliferation, and subcultured on medium supplemented
with 5 mg/liter NAA and 0.1 to 1 mg/liter 2iP for embryogenic callus induction. It seems that a high 2iP:auxin ratio is preferred
for callus initiation and proliferation, but should be exchanged with a higher NAA:cytokinin ratio before differentiation
will occur. Embryogenic calluses were recovered at a frequency of 2 to 85% depending on the cultivar used. Coker cultivars
produced embryogenic callus faster and at higher frequencies than other cultivars. Embryogenic callus produced somatic embryos
on phytohormone-free medium. This medium was used to maintain and proliferate embryogenic callus for a perid of 18 to 24 mo.
Somatic embryos were converted to plants on a lower ionic strength medium supplemented with 0.1 mg/liter gibberellic acid
(GA3) and 0.01 mg/liter NAA. Glucose was the only carbohydrate used through all phases of tissue culture and was much better than
sucrose, on which phenolic production was very high. High temperature (30° C) and low light intensity (9 μE · m−2 · s−1) were optimal conditions for callus initiation, embryogenic callus induction, and maintenance, whereas lower temperature
(25° C) and high light intensity (90 μE · m−2 s−1) were the optimal conditions for somatic embryo maturation, germination, and plantlet development. Plants could be regenerated

Summary DNA fingerprints generated by the Jeffreys' probes, 33.6 and 33.15, indicated the presence of minisatellite-like sequences
in the red clover genome. The fingerprints generated by probe 33.6 gave less background and fewer but better defined bands
than those obtained with probe 33.15. Assay of a regenerative somaclonal variant (F49R) by DNA fingerprinting with probe 33.6
detected mutation that was unlinked to the regenerative trait. The fingerprints obtained under the applied conditions also
demonstrated genetic stability of consecutive generations of the regenerants in tissue culture. DNA fingerprints of F1 plants revealed that each polymorphic band was inherited from either one or the other parent. Both probes distinguished individual-specific

Summary Carcinogenesis is a lengthy process which eventually culminates in the transformed phenotype, cancer. However, much remains
to be defined about the process of transformation. In vivo models for the study of the carcinogenic process present limitations
because it is not possible to detect the premalignant stages in the animals. An in vitro model, on the other hand, facilitates
the study of the carcinogenic process because it enables one to dissect out the crucial events required for carcinogenesis
to occur. As carcinogenesis is believed to be a multistep process; initiation, promotion, and progression, a multistep, in
vitro system has been devised in our laboratory to mimic each of these stages. We have previously shown the formation of “microtumors”
in collagen gels, induced by 7,12-dimethylbenz(a)anthracene. In the present study the direct acting water soluble, mammary
carcinogen,N-nitroso-N-methylurea (NMU) was used for tumorigenesis of mammary epithelial cells in culture. Mammary epithelial cells from virgin
Sprague-Dawley rats were propagated and exposed to single or multiple doses of NMU while growing as a monolayer in glass petri
dishes (initiation). Initiated cells were then plated into a collagen gel matrix culture. Prolonged growth in the collagen
gels afforded for the progression of the transformed cells into discernable microtumors in the three-dimensional matrix of
the collagen. The morphology of these “tumors” was determined by histologic sections of the gels. Fewer, if any, such structures

Enhanced exercise capacity is not only a feature of healthful aging, but also a therapy for aging patients and patients with cardiovascular disease. Disruption of the Regulator of G Protein Signaling 14 (RGS14) in mice extends healthful lifespan, mediated by increased brown adipose tissue (BAT). Accordingly, we determined whether RGS14 knockout (KO) mice exhibit enhanced exercise capacity and the role of BAT in mediating exercise capacity. Exercise was performed on a treadmill and exercise capacity was assessed by maximal running distance and work to exhaustion. Exercise capacity was measured in RGS14 KO mice and their wild types (WT), and also in WT mice with BAT transplantation from RGS14 KO mice or from other WT mice. RGS14 KO mice demonstrated 160 ± 9% increased maximal running distance and 154 ± 6% increased work to exhaustion, compared to WT mice. RGS14 KO BAT transplantation to WT mice, resulted in a reversal of phenotype, with the WT mice receiving the BAT transplant from RGS14 KO mice demonstrating 151 ± 5% increased maximal running distance and 158 ± 7% increased work to exhaustion, at three days after BAT transplantation, compared to RGS14 KO donors. BAT transplantation from WT to WT mice also resulted in increased exercise performance, but not at 3 days, but only at 8 weeks after transplantation. The BAT induced enhanced exercise capacity was mediated by (1) mitochondrial biogenesis and SIRT3; (2) antioxidant defense and the MEK/ERK pathway, and increased hindlimb perfusion. Thus, BAT mediates enhanced exercise capacity, a mechanism more powerful with RGS14 disruption. 相似文献

用组织培养及逐步增加,NaCl浓度的方法,筛选出烟草耐盐愈伤组织变异体。能耐盐(2% NaCl)的愈伤组织在2% NaCl中继代29次,再移入无盐培养基中培养11和20代后不能保持提高的耐盐性,分别退化到只耐1.5%与1.0% NaCl的水平。耐2% NaCl愈伤组织产生的再生植株自交后代,其萌发种子、幼苗及成长植株均未能表现出耐盐性,说明用选择胁迫方法所筛选出的耐盐细胞系,其耐盐性的提高属于生理适应性。 相似文献

BackgroundPregnant women are among the key groups in iodine nutrition evaluation. The purpose of the present study was to summarize the evidence supporting the relationship between mild iodine deficiency (UIC: 100–150 μg/L) in pregnant women and levels of thyroid function tests.MethodsThis review follows the guidelines for systematic reviews (PRISMA 2020). Three electronic databases (PubMed, Medline, and Embase) were searched for relevant publications in English on the association between mild iodine deficiency in pregnant women and thyroid function. Articles published in Chinese were searched in China’s electronic databases (CNKI, WanFang, CBM, and WeiPu). Pooled effects were presented as standardized mean differences (SMDs) and odds ratios (ORs) with 95% confidence intervals (CIs) using fixed or random effect models, respectively. This meta-analysis was registered at www.crd.york.ac.uk/prospero as CRD42019128120.ResultsWe summarized the results from 7 articles with 8261 participants. The overall pooled results showed that the levels of FT3, FT4, and abnormal TgAb (the antibody levels exceeded the upper limit of the reference range) were significantly increased in pregnant women with mild iodine deficiency compared to pregnant women with adequate iodine status (FT3: SMD=0.854, 95% CI: 0.188, 1.520; FT4: SMD=0.550, 95% CI: 0.050, 1.051; TgAb: OR=1.292, 95% CI: 1.095; 1.524). Subgroup analysis was carried out on the sample size, ethnicity, country, and gestation of FT3, FT4, and TSH, but no plausible factor was found. Egger’s tests indicated no publication bias.The increase in FT3 and FT4, as well as TgAb levels, in pregnant women is associated with mild iodine deficiency.ConclusionMild iodine deficiency is associated with an increase in FT3,FT4 and TgAb levels in pregnant women. Mild iodine deficiency may increase the risk of thyroid dysfunction in pregnant women. 相似文献
