Aims: To determine if pretreatment with oxidizing agents sensitizes Bacillus subtilis spores to dry heat or desiccation. Methods: Bacillus subtilis spores were killed approx. 90% by oxidizing agents, and the sensitivity of treated and untreated spores to dry heat and desiccation was determined. The effects of pyruvate on spore recovery after oxidizing agent pretreatment and then dry heat or desiccation were also determined. Conclusions: Spores pretreated with Oxone? or hypochlorite were not sensitized to dry heat or freeze‐drying. However, hydrogen peroxide or t‐butylhydroperoxide pretreatment sensitized spores to dry heat or desiccation, and the desiccation caused mutagenesis in the survivors. Pyruvate increased recovery of spores treated with hydrogen peroxide alone or plus dry heat or desiccation, and with t‐butylhydroperoxide and desiccation, but not with t‐butylhydroperoxide alone or plus dry heat. Significance and Impact of the Study: Pretreatment with peroxides sensitizes bacterial spores to subsequent stress. This finding may suggest improved regimens for spore inactivation. 相似文献

Comparative analyses of bacterial community successions in the composting materials were done for a conventional windrow post-treatment
(WPOT) process with the hyperthermophilic pre-treatment (HTPRT) and simple windrow composting (SWC; without the HTPRT). Multidimensional
scaling profiles based on data of terminal restriction fragment length polymorphisms of the bacterial population in the samples
of every 7 days composting material and analyses of the 16S rRNA gene-based clone library of the 7 and 21 days composting
materials suggested that bacterial communities of the composting materials differed much between these two processes until
the 35 days of composting, whereas that they were closely related to each other at the final composting stage (42 days of
composting). Detailed phylogenetic analysis clarified that all WPOT clone libraries contained many clones of the lineages
of aerobic bacteria (for example, bacilli). However, the most abundant clones retrieved from all SWC materials were affiliated
with a clone cluster closely related to identified and classified members of the phylum Firmicutes that have strictly anaerobic metabolism pathways. From these results, we conclude that the HTPRT process contributed to easily
establish an aerobic ecosystem from the early stage to the final stage of WPOT composting with plowing the materials only

In order to reduce the risks of Legionnaires' disease, caused by the bacterium Legionella pneumophila, disinfection of tap water systems contaminated with this bacterium is a necessity. This study investigates if electrochemical disinfection is able to eliminate such contamination. Hereto, water spiked with bacteria (10(4)CFU Escherichia coli or L. pneumophila/ml) was passed through an electrolysis cell (direct effect) or bacteria were added to tap water after passage through such disinfection unit (residual effect). The spiked tap water was completely disinfected, during passage through the electrolysis cell, even when only a residual free oxidant concentration of 0.07 mg/l is left (L. pneumophila). The residual effect leads to a complete eradication of cultivable E. coli, if after reaction time at least a free oxidant concentration of 0.08 mg/l is still present. Similar conditions reduce substantially L. pneumophila, but a complete killing is not realised. 相似文献

This report presents data from experiments on seed dispersal by wind for ten species of the family Apiaceae. Seed shadows were obtained in the field under natural conditions, using wind speeds between four and ten m/s. The flight of individual seeds was followed by eye, and seed shadows were acquired, with median distances varying from 0.7 to 3.1 m between species. Multiple regression models of wind speed and seed weight on dispersal distance were significant for six out of ten species; wind speed had significant effects in seven cases, but seed weight only once. A good correlation between mean terminal falling velocity of the seeds of a species and median dispersal distance, indicates the promising explanatory power that individual terminal velocity data might have on dispersal distance, together with wind speed and turbulence. The theory that seeds that seem to be adapted to wind dispersal travel much longer distances than seeds that have no adaptation was tested. Flattened and winged seeds were indeed found to be transported further by wind, but not much further. Moreover, the species with wind-adapted seeds were also taller, being an alternative explanation since their seeds experienced higher wind speeds at these greater heights. Furthermore, flattened and winged seeds were disseminated from ripe umbels at lower wind speeds in the laboratory. This means that the observed difference in dispersal distance would have been smaller when species specific thresholds for wind speed were incorporated in the field experiments. Abstract Legionella pneumophila occurring in drinking water was subjected to environmental stress through holding tests at ambient and elevated temperatures and by chemical disinfection. The bacterium in its native environment was more resistant to adverse conditions, as compared with laboratory-grown organisms. Of the several chemical disinfectants acceptable to drinking water treatment and tested for Legionella inactivation, ozonation was the most efficient method.
