The effect of a chilling stress, at a moderate photon flux density for a few hours, on the peroxidation of membrane lipids and on superoxide dismutase (SOD) activity was compared in leaf slices of chilling-sensitive and chilling-insensitive plants. The aim was to determine if susceptibility to chill-temperature photoinhibition could be related to either damage to membrane lipids by superoxide and-or a decrease in activity of chloroplast SOD. Plants used were Nerium oleander L., grown at 45° C, and Cucumis sativus L., both susceptible to chill-temperature photoinhibition, and N. oleander, grown at 20° C and Spinacia oleracea L., both insensitive to chill-temperature photoinhibition. Lipid peroxidation was assessed by measuring the concentration of malondialdehyde (MDA). Leaf slices from all plants showed a basal level of MDA which decreased by about 15% when the leaf slices were chilled in the light. The level of MDA was not increased by the addition of either KHCO3 or methyl viologen during chilling but it was increased, up to threefold, by the addition of Rose Bengal, which produces singlet oxygen. Chloroplast SOD activity was assessed in leaf extracts as the cyanide-sensitive production of H2O2 in a system which produced superoxide. Activity of SOD was similar in all the plants and was altered little by chilling. The results show that for the plants tested, chilling at a moderate photon flux density for 5 h does not increase the susceptibility of cell membranes to peroxidative damage nor does it decrease the activity of SOD. Photoinhibition of photosynthesis in vivo is shown to be considerably promoted by O2 under circumstances where energy turnover by photorespiration and photosynthetic carbon metabolism are low. Intact protoplasts of Valerianella locusta L. were photoinhibited by 30 min irradiation with 3000 mol photons · m–2 · s–1 at 4° C in saturating [CO2] at different oxygen concentrations, corresponding to 2–40% O2 in air. The photoinhibition of light-limited CO2-dependent photosynthetic O2 evolution increased with increasing oxygen concentration. The uncoupled photochemical activity of photosystem II, measured in the presence of the electron acceptor 1,4-benzoquinone, and maximum variable fluorescence, Fv, were strongly affected and this inhibition was closely correlated to the O2 concentration. The effect of O2 did not saturate at the highest concentrations applied. An increase in photoinhibitory fluorescence quenching with [O2], although less pronounced than in protoplasts, was also observed with intact leaves irradiated at 4° C in air. Initial fluorescence, Fo, was slightly (about 10%) increased by the inhibitory treatments but not influenced by [O2]. A long-term cold acclimation of the plants did not substantially alter the O2-sensitivity of the protoplasts under the high-light treatment. Summary The ionizing radiation resistances of sixSalmonella species were examined. The experimental variables were the suspending medium, the presence or absence of air, and the temperature during the irradiation process.S. typhimurium ATCC 14028,S. enteritidis ATCC 9186,S. newport ATCC 6962,S. dublin ATCC 15480,S. anatum ATCC 9270, andS. arizonae ATCC 29933 were suspended in phosphate buffer (0.1 M, pH 7.0), brain heart infusion broth (BHI) or mechanically deboned chicken and exposed to gamma radiation from cesium-137 at 0.12 kGy per min. The radiation resistance of theSalmonella increased approximately two-fold when assayed in sterile mechanically deboned chicken rather than in buffer or BHI. The average radiation (0.30 to 1.20 kGy) D-value for all sixSalmonella strains was 0.56 kGy in mechanically deboned chicken.S. enteritidis was significantly more resistant to ionizing radiation than the other five strains ofSalmonella tested on mechanically deboned chicken. The temperature of irradiation but not the presence or absence of air significantly influenced the survival ofS. typhimurium andS. enteritidis in mechanically deboned chicken. Treatment of chicken meat with ionizing radiation would be an effective means for control ofSalmonella contamination.  相似文献

Summary The environmental factors influencing methanogenesis in a shallow anoxic aquifer were probed in a combined field and laboratory study. Field data collected over a year revealed that in situ rates of methane production were depressed in winter and elevated in summer. Over the same period, ground water pH values ranged from 6.0 to 7.8 while temperatures varied from 7–22°C. In situ methanogenesis was severely inhibited at temperatures < 13°C or by pH values < 7. The influence of these factors on microbial methane formation from both endogenous and exogenous substrates were tested in aquifer slurries adjusted to pH 5–9 and incubated at temperatures ranging from 5–45°C. Temperature optima for methane production from endogenous substrates varied as a function of pH, but the pH optimum was 8 at all temperatures. Optimal conditions for acetoclastic methanogenesis were found at pH 8 and 35°C. An analysis of variance revealed that pH, temperature, and a pH-temperature interaction are all significant variables influencing aquifer methanogenesis. In addition transient sulfate accumulations were also found to limit methane production in some areas. A comparison of field and laboratory methane production patterns suggest that pH, temperature, and sulfate accumultations are important, but not the only environmental variables influencing the mineralization of organic matter in shallow aquifers.  相似文献

Binding activity of a putative central neurotransmitter, L-glutamic acid, was examined in the supernatant preparations solubilized from rat retinal membranes by Nonidet P-40. [3H]Glutamate binding activity increased linearly with increasing concentrations of the solubilized proteins up to 15 micrograms. The binding activity reached an equilibrium within 10 min at 2 degrees C, while increasing with incubation time up to 60 min at 30 degrees C. Addition of an excess of nonradioactive glutamate rapidly decreased the activity at 30 degrees C. Scatchard analysis revealed that the solubilized retinal binding activity consisted of a single component with a KD of 0.25 microM and a Bmax of 57.4 pmol/mg protein. The solubilized binding activity exhibited a stereospecificity and a structure selectivity to L-glutamate, and was abolished by quisqualate, L-glutamate diethyl ester, and DL-2-amino-3-phosphonopropionate. None of the other agonists and antagonists for the central excitatory amino acid receptors affected the binding activity. Reduction of incubation temperature from 30 degrees C to 2 degrees C resulted in a drastic attenuation of the binding activity due to decrement of the number of the apparent binding sites. Cation-exchange column chromatography revealed that unidentified radioactive material was in fact formed during the incubation of [3H]glutamate with the retinal preparations at 30 degrees C. These results suggest that retinal [3H]glutamate binding activity may be derived at least in part from the quisqualate-sensitive membranous enzyme with a stereospecific and structure-selective high affinity for the central neurotransmitter.  相似文献

Mortality from acute myocardial infarction (MI) over the 5 year period 1982–1987 in Brown County, Wisconsin, was analyzed to assess the relationship with environmental temperature. Deaths occurrring on the day of and the day following a significant snowfall as well as deaths occuring in health care facilities were eliminated from consideration because the focus was upon temperature, not snowfall or events within a hospital. These criteria resulted in the inclusion of 1,802 days and 926 cases of acute MI. The mean temperature on the day of death was obtained from climatological data and were grouped into six categories covering a range of temperatures from<–17.8°C (0°F) to 16.1°C (61°F). The number of deaths in each category was tabulated. The effect of temperature, sex, and age were analyzed by regression analysis. The results indicated a linear increase in mortality as mean daily temperature decreased over the temperature range. The inverse temperature effect was most pronounced in males over the age of 60. These results indicate that cold temperatures appear to be associated with an increased mortality from myocardial infarction.  相似文献

We investigated the time relationship between ambient temperature change and antigen stimulation on immune responses to sheep red blood cells (SRBC) and polyvinylpyrrolidone (PVP) in mice. In the case of a shift from comfortable (25°C) to cold (8°C) temperatures, suppression in the number of splenic plaque-forming cells (PFC) took place mainly when the shift was done between 1 day before and 2 to 4 days after immunization. The suppression of the PVP response lasted for up to a maximum of 6 days when mice were transferred 1 day before immunization. In the case of a temperature shift from 25° to 36.5°C, the suppressive effect was found when the temperature shift was done between 4 days before and 2 days after immunization. The effect lasted longer than that of the temperature shift to cold, i.e., at least 9 days after the temperature shift. Blood corticosterone levels after the temperature shifts corresponded to changes in the immune responses: elevation of the blood corticosterone levels was observed for only the first 3 days after a temperature shift to 8°C but for 10 days after a temperature shift to 36.5°C during the period time of the experiment. These result suggested that blood corticosterone level contributes to the duration of the effects of temperature shifts on immune responses of mice. Furthermore, it appeared that the early stage of the immune response is more susceptible to temperature shifts than the later stage. To explain these results, the terms effective period in the course of physiological adaptation to changed ambient temperature and susceptible period in the course of the immune response, were proposed.  相似文献

Summary To investigate the physiological, behavioral, and genetic contributions to growth rate, we studied the thermal sensitivity of growth rate in hatchlings of the iguanid lizards Sceloporus occidentalis and S. graciosus in the laboratory. We used a cycling thermal regime patterned after thermal environments found in nature. Growth rates increased with duration of access to radiant heat. Thus, variation in the thermal environment can cause phenotypic variation in growth rate and hence body size. The two species differed in both the magnitude and thermal sensitivity of growth rate, and these differences were associated with differences in behavioral thermoregulation. Thus, growth is determined interactively by both behavior and physiology. We found evidence of among-family variation in the growth rates of S. occidentalis, suggesting that growth rate has the genetic potential to evolve. In S. occidentalis, both growth rate and egg size affected body size of hatchlings at several weeks of age. In turn, hatchling size may affect fitness: for example, larger S. occidentalis hatchlings had higher sprint speeds and may therefore be more adept at capturing prey or evading predators. Our results demonstrate that growth rate has genetic, behavioral, and physiological components, and that the resulting effects on body size may have important consequences for ecological performance e.g., sprint speed.  相似文献

1. 1.|The effects of thermal stimulation of the preoptic region (POAH) and the spinal cord on non-shivering thermogenesis (NST) and shivering were studied in euthermic golden hamsters.

2. 2.|Shivering intensity is suppressed by heating the POAH but is independent of spinal cord temperature. Therefore, NST in the interscapular brown adipose tissue does not suppress shivering.

3. 3.|NST is inhibited by heating of the POAH as well as of spinal cord. It is discussed that the control of NST by two different central thermosensitive areas is significant for thermoregulation during exercise.

Summary The amount of copper extracted from soils by EDTA was found to increase with the temperature of extraction. Under controlled conditions of extraction, the amount of applied copper extracted from soils by EDTA decreased during incubation of soils under both moist and dry conditions. The recovery of applied copper by EDTA appeared to be related to the amounts of manganese and iron oxides in the soil. On the basis of comparisons with calcium chloride-extractable copper and isotopically-exchangeable copper it is suggested that EDTA extraction could over-estimate the availability of soil copper to plants.  相似文献
