Summary Proton chemical shifts of a series of disordered linear peptides (H-Gly-Gly-X-Gly-Gly-OH, with X being one of the 20 naturally occurring amino acids) have been obtained using 1D and 2D 1H NMR at pH 5.0 as a function of temperature and solvent composition. The use of 2D methods has allowed some ambiguities in side-chain assignments in previous studies to be resolved. An additional benefit of the temperature data is that they can be used to obtain ‘random coil’ amide proton chemical shifts at any temperature between 278 and 318 K by interpolation. Changes of chemical shift as a function of trifluoroethanol concentration have also been determined at a variety of temperatures for a subset of peptides. Significant changes are found in backbone and side-chain amide proton chemical shifts in these ‘random coil’ peptides with increasing amounts of trifluoroethanol, suggesting that caution is required when interpreting chemical shift changes as a measure of helix formation in peptides in the presence of this solvent. Comparison of the proton chemical shifts obtained here for H-Gly-Gly-X-Gly-Gly-OH with those for H-Gly-Gly-X-Ala-OH [Bundi, A. and Wüthrich, K. (1979) Biopolymers, 18, 285–297] and for Ac-Gly-Gly-X-Ala-Gly-Gly-NH2 [Wishart, D.S., Bigam, C.G., Holm, A., Hodges, R.S. and Sykes, B.D. (1995) J. Biomol. NMR, 5, 67–81] generally shows good agreement for CH protons, but reveals significant variability for NH protons. Amide proton chemical shifts appear to be highly sensitive to local sequence variations and probably also to solution conditions. Caution must therefore be exercised in any structural interpretation based on amide proton chemical shifts.  相似文献

Generating developmental gradients by temperature gradients established within a developing organism is an easy, non-invasive technique to study physiological interdependencies between locally separated subsystems. A linear temperature gradient of about 10° C/mm was maintained up to 5 h in either direction along the long axis of a long-germ-type hymenopteran egg, which was simultaneously filmed by the 16 mm timelapse technique. The result was a dramatic desynchronization of development, which between the egg poles could reach up to 9.3 h relative to normal development. Within the same egg, up to seven mitotic waves (i.e. eight different nuclear generations) were observed at the same time, and the subsequent cellularization process was extremely asynchronous. The initial regions of the mitotic waves, the fountain flow of the ooplasm, and the gastrulation process were shifted towards the egg pole kept at higher temperatures. Developmental processes occurring successively in normal development now took place simultaneously, with either direction of the temperature gradient. For instance, while gastrulation had started in the warm region, midblastula transition and cellularization were in progress in the middle of the egg, and intravitelline nuclear multiplication occurred at the cold pole, by rapid and still biphasic cell cycles. In some respects, development resembled that of a short-germ-type insect egg. Nevertheless, the developmental processes were resynchronized after the temperature gradient was switched off. Surprisingly, the extreme desynchronization during early development did not affect the segment pattern of the resulting embryos. The technique of inducing well-defined developmental asynchronies might be applied in Drosophila to analyse the subtle interplay between maternal and zygotic gene activities described in this species.  相似文献

A procedure was developed to encapsulate mycelia of an atoxigenic strain of Aspergillus flavus in alginate pellets for seeding into agricultural fields in order to reduce aflatoxin contamination via competitive exclusion. Kaolin, a clay filler commonly employed in alginate formulations, was detrimental to pellet performance as measured by spore yield. Corn cob grits, a by-product of the corn industry, was found to be an excellent replacement for kaolin. Of nine nutritive adjuvants tested, wheat gluten improved pellet performance the most, although gluten concentrations above 5% were difficult to process. The best formulation tested consisted of 1% sodium alginate, 5% corn cob grits and 5% wheat gluten. On a 'per gram' basis, this alginate formulation yielded more spores than either A. flavus sclerotia or colonized wheat seed. Pesticides were also tested as adjuvants with potential use for protecting pellets under field conditions. Only one (chloramphenicol) of four tested pesticides (the others were dichloran, rose Bengal and cyfluthrin) reduced pellet sporulation. Formulations with or without pesticide adjuvants retained similar spore yield potential during a 2-year storage at 8 C. However, spore production in stored products lagged behind that of fresh products. At 75% relative humidity (RH), pellet storage stability decreased with increasing temperature from 27 to 42 C. Pellet spore yield at 32 C decreased as RH decreased from 100 to 90%. Sporulation occurred at 90% RH but not at 88% RH. Spore yield varied widely in four field tests, and the cumulative spore yield was inversely correlated (r2= -0.798, P 0.01) with rainfall. The results suggest that alginate pellets may be effective formulations for delivery of atoxigenic A. flavus strains to furrow-irrigated cotton in desert environments, where aflatoxin contamination of cottonseed is most severe.  相似文献

Frequency spectra of the surface undulations (flickering) of erythrocyte plasma membranes are measured by direct spectral analysis of the intensity fluctuations of the light passing the cells in a phase contrast microscope. Spectra are taken as a function (1) of the temperature (2) of the viscosity and osmolarity of the outer medium (3) of the aging of cells and (4) of pathological transformations. The spectra are approximately superpositions of two Lorentzian lines. At large frequencies,f, the spectra follow f?2. This behaviour can be interpreted in terms of cell thickness fluctuations caused by thermally excited membrane undulations provided the range of wavelengths is small. The undulations are determined by the membrane curvature elasticity while the lateral tension is negligibly small for cells of discoid shape. The technique presented allows accurate measurements of relative curvature (bending) elastic constants. The spectra of freshly drawn cells are remarkably reproducible. Aging of the cells in the medium leads to an increase in the curvature elastic constant. A decrease in osmolarity causes a reduction in the intensity and line width of the spectra and the flickering vanishes if the cell approaches a spherical shape. The effect of temperature between 10 and 40°C is astonishingly small with the exception of a sudden increase in the amplitude with increasing temperature at 35°C. The flicker spectra of a large fraction of the cells from patients suffering from cronical alcoholism exhibit a reduced line width or an increase in the curvature elastic constant.  相似文献

Using dynamic light scattering we have been able to determine precisely the hydrodynamic radius of l-α-dimyristoylphosphatidylcholine (DMPC) vesicles as a function of temperature. We have detected a sharp, thermally reversible change in the vesicle radius at a phase transition temperature 24°C, corresponding to an approximate 11% increase in surface are. A short period (15–30 min) at 30° C promotes germination of seeds of Lactuca sativa L. cv. Repolhuda in darkness. Far-red light reverses this stimulation, and the escape curves for phytochrome and high-temperature action are quite similar, indicating that the two factors act at a common point in the chain of events leading to germination. Summary Effects of P fertilizers on growth of Douglas fir (Pseudotsuga menziesii var.menziesii (Mirb.) Franco.) seedlings were examined in pots and nursery beds. In pot experiments levels of P equivalent to 300 kg/ha were adequate for maximum growth over 14–18 weeks and resulted in available soil P levels of 80 ppm after 15 weeks' growth. Maximum growth in pots was obtained with shoot P concentrations of 0.18%–0.20%, with higher values at lower temperatures, but the optimum concentration for one-year old (1-0) nursery seedlings was 0.16% P. Growth of seedlings was greatly restricted at a soil temperature of 5°C and an air temperature of 12°C. At a soil temperature of 10°C and an air temperature of 14°C seedling P requirement was greater than at soil and air temperatures of 20°C.Comparison showed that monammonium phosphate was more effective than calcium superphosphate in stimulating growth in pots and nursery. Triple superphosphate was also effective in the nursery. Diammonium phosphate, potassium dihydrogen phosphate and phosphoric acid had no advantages as P sources in the nursery. Available P levels of 100–130 ppm, in the loamy sand and sandy loam nurseries studied, and needle P concentrations of 0.18%, when sampled in October, were associated with maximum growth of two-year old (2-0) seedlings.P fertilization decreased root/shoot ratio, but did not alter the allometric relationship of shoot to root. Improving P status from a low level increased root growth capacity in 2-0 seedlings and P fertilization of potted seedlings increased dry weight/height ratio. Uptakes per seed bed ha of 236 kg N, 31 kg P, 81 kg K and 73 kg Ca by 2-0 seedlings were comparable with, or greater than, uptake rates of agricultural crops. Recoveries of 6–11% of P from fertilizer were recorded in the nursery.  相似文献

The activities and kinetics of the enzymes G6PDH (glucose-6-phosphate dehydrogenase) and 6PGDH (6-phosphogluconate dehydrogenase) from the mesophilic cyanobacterium Synechococcus 6307 and the thermophilic cyanobacterium Synechococcus 6716 are studied in relation to temperature. In Synechococcus 6307 the apparent K m's are for G6PDH: 80M (substrate) and 20M (NADP+); for 6PGDH: 90M (substrate) and 25M (NADP+). In Synechococcus 6716 the apparent K m's are for G6PDH: 550M (substrate) and 30M (NADP+); for 6PGDH: 40M (substrate) and 10M (NADP+). None of the K m's is influenced by the growth temperature and only the K m's of G6PDH for G6P are influenced by the assay temperature in both organisms. The idea that, in general, thermophilic enzymes possess a lower affinity for their substrates and co-enzymes than mesophilic enzymes is challenged.Although ATP, ribulose-1,5-bisphosphate, NADPH and pH can all influence the activities of G6PDH and 6PGDH to a certain extent (without any difference between the mesophilic and the thermophilic strain), they cannot be responsible for the total deactivation of the enzyme activities observed in the light, thus blocking the pentose phosphate pathway.Abbreviations G6PDH glucose-6-phosphate, dehydrogenase - 6PGDH 6-phosphogluconate dehydrogenase - G6P glucose-6-phosphate - 6PG 6-phosphogluconate - RUDP ribulose-1,5-bisphosphate - Tricine N-Tris (hydroxymethyl)-methylglycine  相似文献

Thermophilic Humicola lanuginosa, Penicillium duponti, Sporotrichum thermophile and Mucor pusillus required succinate in addition to glucose for optimal growth. The requirement for succinate was concentration-dependent and the concentration needed for one half of the maximal growth was 6.14 mM. In the presence of succinate, glucose utilization from the medium was markedly increased and this was associated with increased levels of the enzymes of the glycolytic and Krebs cycle pathways. Addition of succinate to cultures growing in glucose at any stage of growth stimulated the growth with the resulting rate of growth remaining high if the addition was made within 3 days of inoculation. Cycloheximide (71.4 M) prevented the succinate-mediated derepression of the enzymes suggesting that succinate may remove the catabolite repression in the presence of glucose.A preliminary part of this work was presented at the 17th annual meeting of the Association of Microbiologists of India at Manipal (India) held from Dec. 13 to 15, 1976  相似文献

Summary With weeds as with many plant species the main or first level factor determining the area of distribution is a (complex) climatic one. As they have an artificially enlarged area of distribution, they have a huge border area (in an ecological sense), where the climate is not optimal for them, and where they have a narrow ecological and sociological amplitude and are especially sensitive to some measures of modern intensified agriculture. In their northern border areas species of southern origin are restricted to calcarcous substrates and to agrestal and finally ruderal communities, while in their optimal climate they are indifferent to that soil factor and able to compete with other species even in natural vegetation types. Species presumably of origin in atlantic areas are restricted with increasing continentality to very poor and acid soils, as they cannot compete with other species on better sites any more, because of their physiological properties. Thus weed distribution demonstrates the complicated reaction of plant species to the complexes of soil-climatic factors and to the competition of other species. As far as weeds are concerned, species may be only relatively calciphilous, but genuinely calcifuge species, the control being climatic in the former case and physiological in the second.The measures of modern agriculture bring about a gradual extinction of sensitive species from the limit of their range towards their centre of distribution, where they can find refuge habitats in the natural vegetation. The sensitivity of such species (also against herbicides) seems to increase towards their limits. Resistant species occur with increasing densities after the removal of their competitors. In addition, they are able to enlarge their area and to invade sites, where they had not been able to compete before, or sites where they could not previously bear the environmental conditions together with the competition of the rich weed flora.As the complex climatic gradients responsible for the ranges of weed species show smooth transitions, the alteration of species composition in weed communities is also a gradual one.
