Xanthomonas campestris pv. vitians, a pathogen of lettuce, elicits a hypersensitive response within 12 hours of inoculation into Brassica leaves, characterized by tissue collapse, loss of membrane integrity, vein blockage and melanin production. In contrast, the compatible pathogen, X. c. pv. campestris, has no visible effects on leaves for 48 hours, after which inoculated areas show chlorosis which eventually spreads, followed by rotting.mRNA was prepared from leaves inoculated with suspensions of both pathovars or with sterile medium up to 24 hours following inoculation. In vitro translation of total and poly A+ RNA in rabbit reticulocyte lysate in the presence of 35S methionine followed by separation of the polypeptide products by 2D-PAGE, allowed comparison of the effects of these treatments on plant gene expression. Major changes in gene expression were observed as a consequence of the inoculation technique. In addition, after inoculation with X. c. vitians, up to fifteen additional major polypeptides appeared or greatly increased by four hours. Some of these had disappeared by nine hours and several more had appeared. No major polypeptides disappeared or decreased greatly in intensity following inoculation with X. c. vitians.  相似文献

The effect of constituents of guinea pig platelets on neutrophil adherence was examined. The platelet sonicate supernatant contained adherence-inhibiting activity which strongly inhibited neutrophil adherence to glass. When the platelet sonicate supernatant was treated with neuraminidase or trypsin, the adherence-inhibiting activity was significantly inhibited, suggesting that the adherence-inhibiting factor (AIF) is a glycoprotein. The subcellular fractionation experiments indicated that the AIF activity was present at about 40% in both the cytosol and granule fractions. From the Sephadex G-200 gel filtration analysis, AIF of cytosol fraction and granule fraction proved to be different molecules, with molecular masses of about 230 and 12 kDa, respectively. When platelets were stimulated with thrombin, about 20% of total AIF was released extracellularly without the release of the cytoplasmic enzyme lactate dehydrogenase. These results suggest the possibility that a biologically active substance, AIF, is released from platelets in response to stimuli and regulates neutrophil functions through interference with neutrophil adherence.  相似文献

Summary The hills of the Negev highlands, west of Sede Boqer, are typically covered by various half-shrub communities, including the deciduous species of Helianthemum vesicarium Boiss., an Irano-Turanian element that grows only on north facing slopes, and H. ventosum Boiss., a Saharo-Arabian element which grows on both slopes but mainly on those facing south. Upon irrigation, plants of H. vesicarium preserve their natural rhythm of activity during winter and remain deciduous throughout the summer. On the other hand, under a similar irrigation treatment, the growth and flowering season of H. ventosum is modified and is extended well into the summer. Thus, H. ventosum shows a high phenological plasticity, while H. vesicarium seems to be very conservative. The different response of the two species to an improved water regime may partly explain the differences in their natural habitats: H. vesicarium on the more humid north facing slopes and H. ventosum mostly on the more arid and extreme south facing ones. Plants of both species which grow near boulders attain larger sizes and denser stands than those on the slopes.Age analysis based on xylem ring counts of 859 plants of the two natural populations showed that the plants did not exceed 14 years and most of them reached the age of some 5 years only. In general more seedlings are established during rainy year-clusters than during dry ones. However, no direct correlation between seedling establishment and the annual rainfall of specific years could be found.The rapid turnover of plants in the stands of both species of Helianthemum resembles the behavior of desert annuals rather than that of most desert shrubs.  相似文献

Cultures of the unicellular green alga Chlamydomonas reinhardii can be synchronized by light/dark cycling not only under photoautotrophic but also under mixotrophic growth conditions. We observed that cultures synchronized in the presence of acetate continue to divide synchronously for one cell-cycle period when transferred to heterotrophic growth conditions. This finding enabled us to investigate the differential effects of light on cell growth and cell division. When cells were exposed to continuous light at the beginning of the growth period they entered the division phase earlier than dark-grown cells as a consequence of an increased growth rate. Illumination at the end of the growth period, however, caused a considerable delay in cell division and an extended growth period. The light-induced delay in cell division was also observed in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), an inhibitor of photosystem II. This finding demonstrates that cell division is directly influenced by a light/dard-responsive cell-cycle switch rather than by light/dark-dependent changes in energy metabolism. The importance of this light/dark control to the regulation of the Chlamydomonas cell cycle was investigated in comparison with other control mechanisms (size control, time control). We found that the light/dard-responsive cell-cycle switch regulates the transition from G1-to S-phase. Shells of Bosminidae and Chydoridae are quantitatively preserved in lake sediments. The chronological deposition of these remains provides the means for longterm observation of these Cladocera, both in terms of species and communities. Chydorid analysis, as based on subfossil assemblages, is an analysis of community and provides direct observation of community dynamics over extended periods of time. It has proved to be a valuable method to obtain information on the influence of environmental factors and time on community characteristics. Morphological variation inBosmina (Eubosmina) has been followed for some thousand years. This is of special interest for the evaluation of taxonomic rank (species, forms) if closely related taxa have co-existed. Bosmina successions, as well as shifts in the chydorid fauna, are related to environmental change. Thus, cladoceran analysis of lake sediments provides information on the developmental history of lakes and allows observation of the effects of longterm environmental changes, such as climatic changes and eutrophication.  相似文献

Hartbeespoort Dam is a hypertrophic man-made lake which is located in the Transvaal Province of South Africa. This region has recently experienced its most severe drought of the century. However, on three occasions in the summer rainy seasons of 1984 and 1985, major rainfalls (> 50 mm) occurred which caused large inflows to the lake. Inflowing river water entered as a density current causing marked silting of the water. Within the epilimnion (0–10 m) prior to these rainfalls there was usually no variation of bacterial numbers with depth, but heterotrophic bacterial activity (glucose uptake) decreased with depth concomitant with primary production. With the increased river inflow bacterial numbers did not increase but bacterial activity at the bottom of the epilimnion (10 m) increased to as high as 2.7 µg C l–1 h–1 in January 1985, reversing the depth profile of bacterial activity within the epilimnion. This resulted in decreased glucose concentrations (Kt + Sn) and turnover times. Heterotrophic activity per cell increased by between 2.5 and 5 times. These data demonstrate that storm events are important phenomena causing short-term changes in the metabolic activity of planktonic heterotrophic bacteria in lakes.  相似文献

Summary The physical localization of sequences homologous to three cloned genes was determined by in situ hybridization to metaphase chromosomes. Previous work had assigned the skeletal myosin heavy chain gene cluster (Myh), the functional locus for the cellular tumor antigen p53 (Trp53-1), and the cellular homologue of the viral erb-B oncogene (Erbb) toMus musculus chromosome 11 (MMU11). Our results provide regional assignments ofMyh andTrp53-1 to chromosome bands B2C, and ofErbb to bands A1A4. Taken together with in situ mapping of three other loci on MMU 11 (Hox-2 homeobox-containing gene cluster, theSparc protein, and theColla-1 collagen gene), which have been reported elsewhere, these data allowed us to construct a physical map of MMU11 and to compare it with the linkage map of this chromosome. The map positions of the homologous genes on human chromosomes suggest evolutionary relationships of distinct regions of MMU11 with six different human chromosome arms: 1p, 5q, 7p, 16p, 17p, and 17q. The delineation of conserved chromosome regions has important implications for the understanding of karyotype evolution in mammalian species and for the development of animal models of human genetic diseases.  相似文献

Purification of a Ciliary Neurotrophic Factor from Bovine Heart   总被引:4,自引:0,他引:4  
A neurotrophic factor that promotes the survival of cholinergic parasympathetic ciliary neurons has been purified approximately 20,000-fold from bovine cardiac tissue under nondenaturing conditions using heparin-affinity chromatography. Up to 22 micrograms of purified factor having a specific activity of 4 X 10(5) trophic units/mg can be obtained from 250 g of heart muscle. Sodium dodecyl sulfate (SDS)-polyacrylamide gels of the purified material show a broad band that is sometimes resolvable into a closely spaced pair of bands of 22 and 23 kilodaltons. Partially purified factor can be resolved into two peaks of activity (pI 5.6 and 5.0) by high-resolution anion-exchange chromatography and chromatofocusing, although these procedures have not proved useful as purification methods because of the large losses of activity incurred. It is likely that these two peaks represent the two bands seen on SDS-polyacrylamide gels. The bovine cardiac factor(s) differs from similar factors purified from chick optic tissues and pig brain in that it is irreversibly denatured by SDS.  相似文献

Chromaffin cells both recently isolated or in culture present a high-affinity adenosine transporter with a Km value of 1 microM. When cells were exposed to nerve growth factor (NGF; 10 ng/ml), the adenosine transporter affinity decreased to 3 microM. This value was maintained from 3 days after plating to the end of the culture period. A change in the transport capacity was observed, with a significant increase (approximately 200-260%) in NGF-cultured cells throughout the period studied.  相似文献

Platelet-derived growth factor (PDGF) increases the mitogenic activity of epidermal growth factor (EGF) in several cells lines, including BALB/C-3T3. PDGF-treated BALB/C-3T3 cells manifest a reduced capacity to bind 125I-labeled EGF due to a loss of high affinity EGF receptors. Cholera toxin potentiates the ability of PDGF to both decrease EGF binding and initiate mitogenesis. Whether PDGF increases EGF sensitivity via its effects on EGF receptors is not known and requires a more complete understanding of the mechanism by which PDGF decreases EGF binding. 12-O-tetradecanoylphorbol 13-acetate (TPA) also reduces EGF binding in BALB/C-3T3 and other cells, presumably by activating protein kinase C and, consequently, inducing the phosphorylation of EGF receptors at threonine-654. PDGF indirectly activates protein kinase C, and EGF receptors in PDGF-treated WI-38 cells are phosphorylated at threonine-654. Thus, the effects of PDGF on EGF binding may also be mediated by protein kinase C. We investigated this hypothesis by comparing the actions of PDGF and TPA on EGF binding in density-arrested BALB/C-3T3 cells. Both PDGF and TPA caused a rapid, transient, cycloheximide-independent loss of 125I-EGF binding capacity. The actions of both agents were potentiated by cholera toxin. However, whereas TPA allowed EGF binding to recover, PDGF induced a secondary and cycloheximide-dependent loss of binding capacity. Most importantly, PDGF effectively reduced binding in cells refractory to TPA and devoid of detectable protein kinase C activity. These findings indicate that PDGF decreases EGF binding by a mechanism that involves protein synthesis and is distinct from that of TPA.  相似文献
