Cadmium effects on leaf transpiration of sugar beet (Beta vulgaris)   总被引:1,自引:0,他引:1  
Seedlings of sugar beet ( Beta vulgaris L. cv Monohill) were cultivated for 4 weeks in nutrient solution containing different concentrations of CdCl2 (0 to 10 μ M ). The effects of Cd on appearance and function of stomata and leaf cuticle were investigated by water loss measurements and microscopy. The leaf transpiration rate increased with increasing Cd concentrations while the sum total of stomatal aperture area per unit leaf area decreased. Already at low Cd levels. an increase of defective and undeveloped stomata was found in Cd treated plants. These stomata are closed or have small apertures and probably lack a functional closing mechanism. The number of intact stomata per unit leaf area was lower in leaves of Cd treated plants than in controls, and Cd induced closure of intact stomata. The total number of stomata per leaf area slightly increases with increasing Cd concentration. as does the percentage of small stomata. Furthermore. specific leaf area increased, while the density of leaf structure was decreased by Cd. From this observation we conclude that the increase in transpiration rate caused by Cd is primarily due to effects on the permeability of the leaf cuticle to water.  相似文献

A leucine aminopeptidase (EC 3,4,11.1) was purified from cotyledons of resting kidney beans ( Phaseolus vulgaris L. cv. Processor) by acidic extraction, ammonium sulfate fractionation and chromatography on DEAE-Sephacel, Sephacryl S-300, Mono Q HPLC and Superose HPLC columns. The yield of the 317-fold purified enzyme was 9%. On gel filtrations on Sephacryl S-300 and Superose HPLC the elution volumes of the enzyme corresponded to an M, of 360 000. The enzyme gave one band on native gel electrophoresis and an electrophoretic titration in an immobilized pH gradient gave a single curve with a pI of 4.8. Two bands were observed in an SDS-gel electrophoresis with Mr values of 58 000 and 60 000 both with and without reduction by 2-mercaptoethanol, indicating that subunits of the enzyme are not linked by disulphide bridges. The purified enzyme most rapidly liberated Leu and Ala of the N-termini of di-and oligopeptides, optimally at pH 9.0 ± 0.5. The enzyme was stable in the presence of glycerol, dithiothreitol and Mg2+, while the latter also had an activating effect. Bestatin inhibited the enzyme competitively with Leu-Gly-Gly with a Ki-value of 1.5 nM . These observations indicate that the purified aminopeptidase from the cotyledons of resting kidney beans corresponds to the cytosolic leucine aminopeptidase of mammalian tissues (EC 3.4, 11.1). The high enzyme activity observed suggests that this aminopeptidase has an important role in the production of free amino acids during germination.  相似文献

Germinating seedlings of mung bean ( Phaseolus vulgaris L. cv. K-16) were treated with different concentrations of cadmium acetate (10, 50 and 100 μ M ). Cd2+ lowered the chlorophyll and heme levels. The level of lipid peroxides were higher on day 3 than on day 6. However, Cd2+ treatment significantly enhanced the level of lipid peroxides. Similarly, a dose-dependent induction of lipoxygenase (EC 1.13.11.12) activity was observed with Cd2+ treatment. Further, the activities of antioxidant enzymes such as superoxide dismutase (EC 1.15.1.1) and catalase (EC 1.11.1.6) were decreased. Our results suggest that lipoxygenase-mediated accumulation of lipid peroxides on the one hand and inhibition of free radical scavenging enzymes like superoxide dismutase and catalase on the other caused a pronounced reduction in the chlorophyll and heme levels of the seedlings. The experiments conducted on the effect of Cd2+ on dark-grown seedlings did not conform with the result of light-grown seedlings. Though chlorophyll and heme levels decreased in a dose-dependent manner, no accumulation of lipid peroxides was observed, suggesting that the inhibition of chlorophyll synthesis by Cd2+ is achieved both by reaction with constituent biosynthetic enzymes as well as peroxide-mediated degradation.  相似文献

British populations of Senecio vulgaris frequently contain two common capitulum morphs (radiate and non-radiate) and one rare intermediate morph. The radiate morph shows a higher maternal rate of intermorph outcrossing than the non-radiate morph and due to the ‘cost of outcrossing’ should decline in frequency, ultimately to be lost from a population. To determine whether the radiate morph exhibits some inherent advantage in fitness to offset the ‘cost of outcrossing’, a comparison was made of the survivorship and fecundity of the radiate and non-radiate morphs raised in pure stands and 1 : 1 mixture at three planting dates (autumn 1983, and spring and autumn 1984). Plants in stands established in spring 1984 were harvested in late August 1984, while plants in stands established in autumn overwintered before being harvested the following summer. In spring planted stands, the two morphs exhibited equivalent survivorships, while the fecundity of the non-radiate morph tended to be greater than the radiate morph. In autumn planted stands, survivorship and Net Reproductive Output (survivorship × fecundity) of the non-radiate morph was greater than that of the radiate morph in mixture, and also in pure stands established in 1983. In no instance was the Net Reproductive Output of either morph significantly greater in mixture than in pure stand. Density had a contrasting effect on morph survivorship and fecundity in the spring and autumn 1984 planted stands. Whereas, in spring stands, fecundity was subject to compensating density dependent regulation while survivorship was density independent, the opposite trend was observed in autumn planted stands. It is concluded that under the conditions of the experiments, the radiate morph exhibited no fitness advantage which might offset the inherent disadvantage it suffers in natural polymorphic populations due to the ‘cost of outcrossing’.  相似文献

Seeds of Phaseolus vulgaris L. cv. White Kidney were germinated and grown either in a nitrogen-free or in an ammonia-supplied medium. The changes in the soluble protein concentration and in the levels of glutamine synthetase (GS, EC 6.3.1.2), NADH–glutamate synthase (NADH-GOGAT, EC 1.4.1.14), ferredoxin-glutamate synthase (Fd-GOGAT, EC 1.4.7.1) and glutamate dehydrogenase (GDH, EC 1.4.1.2), both NADH- and NAD+-dependent, were examined in cotyledons and roots during the first 10 days after sowing. Soluble protein declined rapidly in the cotyledons and increased slightly in the roots. GS activity was initially high both in cotyledons and roots but subsequently decreased during seedling growth. Exogenous ammonia hardly affected GS activity. High levels of NADH-GOGAT were present both in cotyledons and roots during the first days of germination. The activity then gradually declined in both organs. In contrast, Fd-GOGAT in cotyledons was initially low and progressively increased with seedling development. In roots, the levels of Fd-GOGAT were higher in young than in old seedlings. Supply of ammonia to the seedlings increased the levels of NADH-GOGAT and Fd-GOGAT both in cotyledons and roots. NADH-GDH (aminating) activity gradually increased during germination. In contrast, the levels of NAD+-GDH (deaminating) activity were highest during the first days of germination. Exogenous ammonia did not significantly affect the activities of GDH.  相似文献

Phaseolus vulgaris L. cv. Kinghorn Wax seedlings, supplied with nutrient solution containing either 0 or 5 mM nitrate as sole N source, were exposed to 0.25 μl/l NO2 for 6 hr each day for 10 days at continuous photosynthetic photon flux (PPF) of 100, 300, 500 or 700 μmol m−2 sec−1. There was a significant interaction of PPF and nitrate. Shoot and root dry weights increased with increasing PPFs only when nitrate was supplied. The main effects of NO2 on plant growth were significant; none of the interactions involving NO2 were significant. Exposure to NO2 decreased shoot and root dry weight in both the presence and absence of nutrient N and at all PPF levels. All interactions were significant for in vitro leaf nitrate reductase activity (NRA), which increased markedly at PPFs above 100 μmol m−2 sec−1 when nitrate was supplied. Treatment with NO2 strongly inhibited enzyme activity in the presence of nitrate, particularly at the 300 μmol m−2 sec−1 PPF level. These experiments demonstrated that PPF level does not modify the effect of NO2 on growth but does have a major effect on NRA and on NO2 effects on NRA in the presence of nutrient nitrate.  相似文献

Abstract. Chara vulgaris L. growing in an oligohaline lake was adapted to laboratory conditions and subjected to long-term salinity treatments ranging from 0 to 350 mol m 3 NaCl added to the lake water (40–680 mosmol kg 1). Osmotic potential and concentration of the main osmotically active solutes (K+, Na+, Mg2+, Cl and sucrose) in the vacuolar sap of the central internodal cells were estimated. C. vulgaris did regulate turgor but incompletely. Turgor decreased from 335 mosmol kg 1 under control conditions to 52–111 mosmol kg 1 at 350 mol m 3 NaCl. The enhancement of πi was achieved by increase in both ions and sucrose. Sterile and fertile plants differed in their response to osmotic stress. In sterile plants, the ions accounted for about 87% of the vacuolar osmotic potential. The increase of πi under osmotic stress was exclusively due to an accumulation of Na+ and Cl-. In fertile plants, sucrose accounted for about 35% of πi and ions for about 51% Under osmotic stress, sucrose content increased together with the ionic content of Na+ and Cl-.  相似文献

Female birds can be identified through the presence of a W-chromosome. We describe a procedure for amplifying a W-linked DNA marker in the starling (Sturnus vulgaris) by the polymerase chain reaction (PCR) so allowing the diagnosis of sex in this species. The technique is sensitive, allowing even the smallest chicks to be sexed from a blood sample. The method possesses a positive internal control to ensure accuracy. It is also applicable to the spotless starling (S. unicolor) but not to two bird species outside the genus. The nucleotide sequence of the female-specific PCR product is given.  相似文献

分别用丝瓜花粉、桔全爪螨、朱砂叶螨、丝瓜花粉+桔全爪螨、丝瓜花粉+朱砂叶螨作为食物,测定普通钝绥螨(Amblyseius vulgaris)在不同食物条件下的发育历期、繁殖力、死亡率及子代性比,并组建其实验种群生命表。结果表明,普通钝绥螨取食有丝瓜花粉存在的食物比单独取食桔全爪螨或朱砂叶螨发育快,产卵量大,子代雌性比高。在前者情况下,普通钝绥螨种群的内禀增长力(r_m)为0.209—0.259,净增率(R_0)为18.918—35.771,由于雌成螨产卵期长,其平均世代时间(T)较长,为15.295—19.529天;在后者情况下,种群内禀增长力为0.165—0.191。净增率为7.091—17.113。由于雌成螨产卵期短,其平均世代历期亦较短,为11.50—14.23天。本研究还分别用丝瓜、党参、七里香、养麦、紫云英、玉米、兰花、栎树、西瓜、油菜、向日葵和益母草等12种植物的花粉饲养普通钝绥螨。结果表明,只有兰花花粉不能使该螨发育至成螨;后5种花粉虽能使其发育至成螨,但不能产卵;其余6种花粉能使其完成发育并产卵,其中丝瓜、党参和七里香花粉饲养效果较好。  相似文献

Summary Each cnidocil complex of the hydrozoans Tubularia larynx and Hydra vulgaris consists of 9 or 7–10 large stereovilli (=stereocilia), respectively, and a modified cilium, the cnidocil. The cnidocils comprise the regular 9 microtubule doublets, up to 30 additional microtubules, as well as a central filament body. Adjacent stereovilli are linked together by intermembrane connectors forming the stereovillar cone. The distal tips of the stereovilli surround the cnidocil in a closed tubular arrangement measuring up to 0.7 m in length. Within this contact region the cnidocil is linked to the stereovillar tube by another set of intermembrane connectors, which seem to hold the cnidocil in a central position within the stereovillar cone. Stereovillar membrane and actin core are linked by 16-nm long cross bridges, which display a periodicity of 16 nm and emerge from the actin core. Within the cnidocils periodically arranged membrane-cytoskeleton bridges are uniformly restricted to the contact region. Here, 24-nm long cross bridges, which are spaced by a regular distance of 20 nm, interconnect the A-tubules of the microtubule doublets and the membrane. The cnidociliary membrane is differentiated into distinct domains as revealed by freeze-fracturing. Within the contact region of the nematocytes of Tubularia larynx, intramembrane particles are arranged in 9 rows of 700 nm length and 50 nm width, separated by particlefree areas. Intramembrane particles are irregularly distributed distal to the contact region. Considering recent physiological results we presume that the latter represent chemoreceptor units, while mechanical stimuli are transmitted via the intermembrane connectors and the microtubule-membrane bridges to mechanosensitive channels within the domain of the cnidociliary membrane in the contact region.  相似文献
