Protective immunity and production of anti-sporozoite (CSP) antibody was studied in A/J mice injected with X-irradiated sporozoites using different immunization schedules and antigen doses. Data were also obtained on the immunogenicity of X-irradiated as compared to nonirradiated sporozoites. After a single immunization (1.5 × 105 or 7.5 × 104 X-irradiated sporozoites) a number of animals was completely protected when challenged, but the percentage of protected mice varied considerably from experiment to experiment. Maximal protection was obtained 7 days after the immunization. When the first injection of parasites was followed by a single booster administered 3, 4 or 5 days later, protection was considerably enhanced and the results more consistent. After a single injection of 1.5 × 105 or 7.5 × 104 sporozoites, CSP antibody was detectable from the 19th and 23rd day, respectively, i.e., at a time point when protection was diminishing. This antibody persisted only for a short period. When a single booster was given soon after the first injection, CSP antibody was present in the sera of all the mice from the ninth day on and persisted for greater than 80 days. A single dose of X-irradiated sporozoites injected into rats, induced antibody (CSP) formation which reached a peak after 2 weeks and persisted at this level for more than 3 months. However in rats injected with viable sporozoites, the antibody titers fell rapidly and became undetectable after 4 weeks.From these data we can conclude that (a) the immune response induced by attenuated X-irradiated sporozoites is considerably longer-lasting than that induced by viable sporozoites; (b) CSP antibodies are not detectable during the early stages of the immune response; and (c) protective immunity precedes the presence of detectable serum and antibodies.  相似文献

Bumblebees harbour a wide range of parasitic organisms that attack all stages of their life cycles (reviews in Postner, 1951; Pouvreau, 1973, 1974; Alford, 1975; Kistner, 1982). Among them, conopid flies (Conopidae, Diptera) are particularly interesting because they attack foraging bumblebees which are handling flowers, or even on the wing (Frison, 1926; Cumber, 1949; Postner, 1951; Howell, 1967; Askew, 1971). A single egg is attached to (Frison, 1926; Plath, 1934; Cumber, 1949) or inserted into (DeMeijre, 1904; Howell, 1967) the host's abdomen, where the larva hatches and feeds on haemolymph and internal organs. Within 6–10 days the larva passes through three recognizable stages (Pouvreau, 1974) before the fly pupates in situ within the abdomen. The host bee dies shortly before the parasite pupates (Postner, 1951; Smith, 1966) and the parasite overwinters in its puparium; the adult fly then emerges in early summer (Frison, 1926; Townsend, 1935; Cumber, 1949; Postner, 1951). Conopid flies as parasites of bumblebees are known from all major habitats where the hosts occur (e.g. Kröber, 1939; Smith, 1966). However, the effect of parasitism on distribution and abundance of bumblebees is not known. In this preliminary note we have estimated degrees of infestation and concomitant reduction of life span in affected workers. The results are compared with literature reports on infestation levels in Europe.  相似文献

The time scales of evolutionary and ecological studies tend to converge, as shown by evidences that contemporary evolution can occur as fast as ecological processes. This opens new questions regarding variation of characters usually considered to change mostly along an evolutionary time scale, such as morphometric traits, including osteological and dental features such as mandibles and teeth of mammals. Using two-dimensional geometric morphometric approach, we questioned whether such features can change on a seasonal and local basis, in relation to the ecological dynamics of the populations. Our model comprised populations of house mice (Mus musculus domesticus) in two contrasted situations in mainland Western Europe: a feral population vs. two close commensal populations. Mitochondrial DNA (D-loop) provided insight into the diversity and dynamics of the populations.The feral population appeared as genetically highly diversified, suggesting a possible functioning as a sink in relation to the surrounding commensal populations. In contrast, commensal populations were highly homogeneous from a genetic point of view, suggesting each population to be isolated. This triggered morphological differentiation between neighboring farms. Seasonal differences in morphometric traits (mandible size and shape and molar size and shape) were significant in both settings, although seasonal variations were greater in the feral than in the commensal population. Seasonal variations in molar size and shape could be attributed to differential wear in young or overwintered populations. Differences in mandible shape could be related to aging in overwintered animals, but also possibly to differing growth conditions depending on the season. The impact of these ecological processes on morphometric traits is moderate compared to divergence over a large biogeographic scale, but their significance nevertheless underlines that even morphological characters may trace populations dynamics at small scale in time and space.  相似文献

This review is aimed to summarize the experimental researches in the influences of static magnetic field on laboratory rodent models, reported by laboratory scientists, experimental technicians, clinical surgeons, animal veterinarians, and other researchers. Past studies suggested that static magnetic field-singly applied or used combined with other physical or chemical substances-significantly relieved some pains and ameliorated certain diseases in different organ systems, e.g. hypertension, osteoporosis, neuralgia, diabetes and leukemia etc. But on the other hand, some harmful events have also been observed in a number of investigations, from cellular level to fetal development. So exposure to static magnetic field might have dual effects on experimental rodent in various environments, viz. there are potentially therapeutic benefits, as well as adverse effects from it. The positive effect may relate to moderate intensities, while negative influence seems to be in connection with acute strong static magnetic fields. In addition, different orientations of static magnetic field exert different degrees of impact. Thus, the bioeffects of static magnetic field exposure on mice/rats depend on magnetic field intensities, durations and directions, though the exactly relationship between them is still vague. Further researches need to perform with appropriate methodologies, ingenious designs repeatedly and systemically, not only in animal models, but also in human volunteers and patients.  相似文献

This study assessed the impact of introduced black rats (Rattus rattus) on Cory's shearwater (Calonectris diomedea borealis) in a multi-invaded insular ecosystem where rats are mesopredators. We hypothesized that black rats should have little impact on Cory's shearwaters in the presence of cats as superpredators. Stomach contents and stable isotope analysis (SIA) in tissues of black rats were analyzed to assess the trophic ecology and the importance of Cory's shearwater in their diet. We also studied the isotopic signature in tissues of house mouse (Mus domesticus) to confirm previous data showing no predation of this species on Cory's shearwaters. For both rodent species, temporal variation in diet composition in response to the availability of seabird prey was evaluated, and short- and long-term consistency in diet was tested using different tissues from the same individual. For black rats a Bayesian isotope mixing model (SIAR) was applied to determine the relative contribution of each prey to the individual diet. SIA of mouse tissues varied between the Cory's shearwater breeding and non-breeding periods. However, no significant differences were found in diet and SIA for black rats. In contrast, individuals of both species showed a strong consistency in diet which apparently benefited their body condition index. Although black rats supplement their diet with Cory's shearwater eggs and chicks (8.3% in stomach contents and 10.6% in the SIAR model), their current impact on the Cory's shearwater population appears to be small, probably due to several factors including the small size of the rat population and a high level of rat predation by cats.  相似文献

Fruit production of Castanea sativa Miller (Fagaceae) in southern Spain is carried out in traditional orchards using a large number of local varieties. The main areas of production are located in the Natural Park of Sierra de Aracena y Picos de Aroche (Huelva) and the Genal river Valley (Malaga). These agroforestry systems have shown a high genetic diversity and represent an example of in situ conservation. In this region, chestnut mainly suffers from attacks by Cydia splendana (Hübner) (Lepidoptera: Tortricidae), whose larvae feed on the fruits, thereby reducing their quality and economic value. The aim of this study was to evaluate the differential response between the chestnut local varieties to the attack of C. splendana. For that purpose, trees from the most representative varieties were evaluated on various farms. In cases where it was necessary, the trees were identified with a set of eight microsatellite markers. The infestation rate of C. splendana (percentage of infested fruits of the total fruits sampled per tree) differed among varieties, indicating a high genetic component in this effect. The results obtained in this study open the possibility of using this differential response in the control of C. splendana.  相似文献

The horn fly Haematobia irritans L. (Diptera: Muscidae) has recently spread to Argentina and Uruguay and is believed to cause damage to cattle hides. Four groups of ten Holstein steers each were maintained for 58 weeks under different infestation levels with H. irritans to determine if it was the cause of this problem. Hides (chrome tanned) from steers maintained under minimum infestation level had 4.7 +/- 3.8% of the area damaged. Maintaining the steers under low H. irritans level for the last 44 days of the trial using insecticidal ear-tags, resulted in 29.5 +/- 15.8% of hide area being damaged. Steers that were treated with 5% cypermethrin pour-on, when the H. irritans population was close to 50 flies, showed that 31.3 +/- 16.6% of hide area was injured, and 46.6 +/- 12.8% of damaged hide area was found in hides from non-treated steers. Significant differences were found between mean hide damage from steers maintained continuously under low H. irritans infestation levels and all other groups. Hyperaemia was significantly lower in the skin of steers under low H. irritans infestation level than in the skins of non-treated steers and steers maintained under low-level infestations for the final 44 days. Eosinophil and mononuclear cell infiltration was significantly lower when the population of H. irritans was less than six per steer than when the population was more than 100 flies per steer. Low numbers of Stomoxys calcitrans were found in all groups, but most hide damage was presumed due to H. irritans.  相似文献

The results of bioassay and colony evaluation demonstrated that British and Texas Buckfast honey bee stocks had one-third to one-half the mean prevalence and abundance of tracheal mites as Canadian standard stock, indicating that Buckfast stocks are less susceptible to tracheal mites than Canadian standard stock. Hybrid Canadian and Buckfast stocks exhibited resistance characteristics similar to only one of their parental stocks, suggesting the colony has an unknown effect on the expression of a bee's resistance to mites. A high correlation (r s=0.66) between abundance values from the bioassay and colony evaluations indicates that the bioassay can be used to screen bees for mite resistance.  相似文献
