The present study was undertaken in an effort to elucidate the mechanism(s) responsible for desensitization, making use of both cutaneous and peritoneal manifestations of delayed hypersensitivity as parallel assay systems in the guinea pig model. Particular emphasis was placed on investigating the possibility of passively transferring desensitization with serum factors and attempting to characterize these. Our data support the hypothesis of an inhibitory environment in the anergic animal: The desensitized state was successfully transferred to immune recipients by the systemic administration of desensitized serum as measured by both skin testing and macrophage disappearance reaction (MDR). Transfer of the desensitized state is not due to residual antigen in donor sera and is nonspecific. Partial characterization of the serum inhibitor indicated that it is nondialyzable, heatstable at 56 °C for 30 min, but destroyed by heating at 80 °C for 60 min. Sephadex G-100 chromatography located the inhibitory activity in the size range of 10,000–45,000 daltons. Larger molecular weight fractions were devoid of inhibitory activity, suggesting that antibodies or immune complexes are not responsible for the effect observed and the inhibitory serum component is not associated with macroglobulins. The physicochemical properties of the desensitizing factor, and our previous finding that intravenous injection of lymphokines leads to transient cutaneous anergy, suggest that the desensitizing substances may represent endogenous lymphokines in the general circulation.  相似文献

A study has been made of the distribution of copper in the kidneys of growing rats. Renal copper concentrations increased steadily with age and were greater in female than in male animals. Most of the copper was present as (copper, zinc)-metallothionein and two forms of this protein were isolated and characterized from the kidneys of mature female rats. That copper metabolism in kidneys is subject to hormonal influence was indicated by a reduction in the concentrations of copper and (copper, zinc)-metallothionein in ovariectomized rats and by an increase in their concentrations after the administration of progesterone. Concentrations of renal (copper, zinc)-metallothionein were less in zinc-deficient than in zinc-adequate rats during pregnancy and after progesterone administration.  相似文献

It is shown that serum from mice heavily infected with BCG contains antibodies which block the cell transfer of delayed-type hypersensitivity (DTK) to purified protein derivative (PPD) when BCG-immune cells were preincubated in it. This suppressive activity is antigen specific in that the serum does not block the cell transfer of contact sensitivity to oxazolone. However, the suppressive activity is not antigen directed in that it is absorbed neither by PPD-coupled Sepharose beads nor by PPD-pulsed normal peritoneal exudate cells. On the other hand, the activity can be absorbed to BCG-immune T cells and eluted from a Sepharose column conjugated with affinity-purified mouse anti-PPD antibodies. The possibility that antireceptor antibodies arise during the BCG infection and regulate DTK reaction is discussed.  相似文献

The effect of agents stimulating the oxidative burst (OB) in oil-elicited guinea pig peritoneal macrophages (MPs) on cyclic adenosine 3′,5′-monophosphate (cAMP) levels was examined. We found that: (i) Phorbol myristate acetate (PMA), the Ca2+ ionophore A23187, concanavalin A (Con A), wheat germ agglutinin (WGA), N-formyl-l-methionyl-l-leucyl-l-phenylalanine (FMLP) and opsonized zymosan, elevated cAMP levels two- to fivefold; (ii) the biologically inactive PMA analog, 4-O-methyl-PMA, was proportionally less effective than PMA in stimulating cAMP accumulation; (iii) increased levels of cAMP were evident after 10 min of incubation with the stimulants, in the presence of the phosphodiesterase inhibitor 3-isobutyl methylxanthine (IBX); (iv) basal cAMP levels in MPs increased proportionally with the extracellular Ca2+ concentration; (v) the cAMP-elevating effect of all stimulants (with the exception of A23187) was more pronounced in low Ca2+ media, associated with lower basal cAMP levels. A23187 did not elevate cAMP levels in the absence of extracellular Ca2+; (vi) short-term incubation of MPs with arachidonic acid and with the arachidonic acid precursor, linoleic acid, induced an increase in the level of cAMP; (vii) the elevations in cAMP levels induced by OB stimulants were enhanced, not blocked, by mepacrine, 5,8,11,14-eicosatetraynoic acid (ETYA), indomethacin or aspirin, demonstrating that prostaglandin (PG) synthesis was not involved; (viii) the cAMP-elevating effect of arachidonic and linoleic acids was blocked by ETYA and indomethacin, indicating that it was mediated by PGs. The mechanism by which OB stimulants elevate cAMP levels could not be determined but changes in the cellular level of Ca2+ seem to play a pivotal role.  相似文献

Parathyroid hormone (PTH) and calcitonin exert well known effects on the renal tubule which are thought to involve specific hormone receptors and adenyl cyclase. In the intestine, it is not clear whether the action of PTH and calcitonin is only indirect or also direct, and their mechanisms of action are much less well established. In the present study, possibly direct effects of PTH and calcitonin on Na+ transport in isolated intestinal epithelial cells of rats were investigated. In the presence of bovine PTH (1.2 I.U./ml) in the incubation medium, the Na+ efflux rate constant (oKNa) of isolated enterocytes was significantly reduced when compared to that in control experiments with the hormone vehicle only. The mean depression of oKNa induced by bovine PTH was 26% as compared to the control (100%) and to that induced by ouabain (4.0mM) which was 44%. No depressant effect of bovine PTH on oKNa was observed when the isolated enterocytes were incubated with ouabain (4.0 mM). Thus, bovine PTH appeared to inhibit the ouabain-sensitive Na+ pump. When incubating the isolated epithelial cells in an EGTA-containing Ca2+-free medium, bovine PTH lost its capacity to inhibit (oKNa). Thus, the presence of extracellular Ca2+ appeared necessary for the inhibitory effect of bovine PTH. In contrast to its effect on oKNa, bovine PTH induced no change in net Na+ uptake by isolated enterocytes. Moreover, no significant effect on enterocyte Na+ transport could be demostrated for salmon or porcine calcitonin at two different concentrations in the incubation medium. Neither bovine PTH nor salmon calcitonin induced significant changes in enterocyte cyclic AMP or cyclic GMP concentrations. It was concluded that bovine PTH, but not calcitonin, exerted a direct inhibitory effect on the ouabain-sensitive oKNa of isolated rat enterocytes. The effect of bovine PTH occured without measurable activation of the cyclic nucleotide system but needed the presence of Ca2+ in the incubation medium to be operative.  相似文献

Synthetic glycoconjugates prepared by the direct reductive amination of di-N-acetylchitobiose and tetra-N-acetyl-chitotetraose to poly-l-lysine with sodium cyanoborohydride have been used to explore the binding specificities of the lectins wheat germ agglutinin and Bandeiraea simplicifolia II. These conjugates are effective precipitating antigens for these lectins, and hapten inhibition experiments, employing the per-N-acetylated oligomers of chitin as inhibitors, demonstrate that wheat germ agglutinin and Bandeiraea simplicifolia II lectin have binding sites complementary to three and two contiguous β 1,4-linked N-acetyl-d-glucosamine residues, respectively, in agreement with conclusions reached using other methods. Conjugates prepared by this technique should be useful for examining the binding specificities of other lectins, and the results of a study of the effect of chain length of the hapten on the affinity of the lectin for these conjugates should provide guidance in selection of the hapten most appropriate for these studies.  相似文献

Twenty-five newly bred improved cassava cultivars, twenty-three improved from the International Institute of Tropical Agriculture and two local cultivars were evaluated for their relative resistance to African cassava mosaic begomovirus disease (ACMD) at Ibadan, in an area of high disease pressure representative of the forest/savanna transition zone of Nigeria. These cultivars were exposed to natural infection by the viruliferous whiteflies ( Bemisia tabaci Gennadius) and the disease incidence (DI) and index of symptom severity (ISS) were assessed for all clones. Results for the Rank-sum (i.e., sum of ranks for DI and ISS for each cultivar) and the area under the disease progress curve (AUDPC) were used to assess the relative resistance of the cassava clones. Those that showed low Rank-sum and AUDPC values were rated 'moderately resistant (MR)', 'resistant ( R )', and 'highly resistant (HR)' to ACMD depending on their respective values and deviation from the mean distribution curve. Clones M94/0121 and 94/0239 were rated HR under the two methods. Clone M94/0583 was rated HR under the AUDPC with a deviation from the mean distribution curve of m 2.00 while it was rated R under the Rank-sum method with a deviation from the mean distribution of m 1.99. Also plants of clones ISU and TMS 30572 were rated highly susceptible (HS) under both methods. Clone TME-1 was intermediate between Moderately resistant (MR) and Moderately susceptible (MS) under the AUDPC method with a deviation from the mean distribution of 0.00 but observed to be MS under the Rank-sum method with a deviation of + 0.2. The two methods of evaluation gave similar results as revealed by Spearman rank correlation ( r equals; 0.99, P <0.01). However, the AUDPC method is less cumbersome compared to the Rank-sum method. None of the clones was observed to be immune to the disease.  相似文献

The Steller's sea cow – Hydrodamalis gigas (Dugongidae: Sirenia) – is an extinct herbivorous marine mammal which inhabited the North Pacific Ocean during the Pleistocene and Holocene. H. gigas was the largest member of the Sirenia order and disappeared in the middle of the 18th century. Here, we present the complete sequence of the mitochondrial genome of this extinct animal. The Steller's sea cow mitochondrial DNA (mtDNA) is 16,872 base pairs (bp) in length and contains a set of mitochondrial genes typical for mammals. Phylogenetic analysis based on complete mitochondrial genomes of the sirenian species allows accurate assessment of the degree of their mitogenomic diversification during millions of years of evolution.  相似文献

A major issue in shift-work research is to understand the possible ways in which shift work can impact performance and health. Nearly all body functions, from those of the cellular level to those of the entire body, are circadian rhythmic. Disturbances of these rhythms as well as the social consequences of odd work hours are of importance for the health and well-being of shift workers. This article reviews a number of common methodological issues which are of relevance to epidemiological studies in this area of research. It discusses conceptual problems regarding the use of the term “shift work,” and it underscores the need to develop models that explain the mechanisms of disease in shift workers.  相似文献

We demonstrated that interferon delays the maturation of human monocytes to macrophages in vitro as assessed by morphologic, histochemical, and biochemical parameters. After exposure to interferon, monocytes were slightly smaller, less stretched out, and had a delayed loss of granules with peroxidase positive reactivity, as compared with control, noninterferon-treated cells. Also, interferon prevented the increase of the specific activity of three lysosomal enzymes (β-galactosidase, β-glucuronidase, and β-N-acetylglucosaminidase) in the monocytes, and enzyme activities were 30–40% of that observed in untreated cells. Both human leukocyte and human fibroblast interferons were effective in delaying the maturation process. The effects of the interferon were species specific and reversible and were neutralized by antiinterferon serum. These studies describe a new nonantiviral effect of interferon, unique in that it involves the delay of maturation of cells in a system which is not associated with cell proliferation. Thus interferon could potentially effect host defense mechanisms and aspects of the immune response which are dependent on the maturation of monocytes to macrophages.  相似文献
