Piscidin 1 (Pis-1) is a novel cytotoxic peptide with a cationic α-helical structure isolated from the mast cells of hybrid striped bass. In our previous study, we showed that Pis-1[PG] with a substitution of Pro8 for Gly8 in Pis-1 had higher bacterial cell selectivity than Pis-1. We designed peptoid residue-substituted peptide, Pis-1[NkG], in which Gly8 of Pis-1 was replaced with Nlys (Lys peptoid residue). Pis-1[NkG] had higher antibacterial activity and lower cytotoxicity against mammalian cells than Pis-1 and Pis-1[PG]. We determined the tertiary structure of Pis-1[PG] and Pis-1[NkG] in the presence of DPC micelles by NMR spectroscopy. Both peptides had a three-turn helix in the C-terminal region and a bent structure in the center. Pis-1[PG] has a rigid bent structure at Pro8 whereas Pis-1[NkG] existed as a dynamic equilibrium of two conformers with a flexible hinge structure at Nlys8. Depolarization of the membrane potential of Staphylococcus aureus and confocal laser-scanning microscopy study revealed that Pis-1[NkG] effectively penetrated the bacterial cell membrane and accumulated in the cytoplasm, whereas Pis-1[PG] did not penetrate the membrane but remained outside or on the cell surface. Introduction of a lysine peptoid at position 8 of Pis-1 provided conformational flexibility and increased the positive charge at the hinge region; both factors facilitated penetration of the bacterial cell membrane and conferred bacterial cell selectivity on Pis-1[NkG].  相似文献

Intimal hyperplasia plays an important role in various types of vascular remodeling. Mechanical forces derived from blood flow are associated with the proliferation of vascular smooth muscle cells (VSMC). This contributes to many vascular disorders such as hypertension, atherosclerosis and restenosis after percutaneous transluminal angioplasty (PTA). In this study, we show that static pressure induces the proliferation of VSMC and activates its related signal pathway. VSMC from a rat aorta were treated with different pressures (0, 60, 90, 120, 150 and 180 mm Hg) in a custom-made pressure incubator for 24 h. The most active proliferation of VSMC was detected at a pressure of 120 mm Hg. VSMC was also incubated under a static pressure of 120 mm Hg for different time intervals (0, 2, 4, 8, 12 and 24 h). We found that static pressure significantly stimulates VSMC proliferation. Extracellular signal-regulated kinases 1/2 (ERK1/2) activation showed a peak at the pressure of 120 mm Hg at 4-h time point. Moreover, caveolin-1 expression was significantly inhibited by rising static pressure. Downregulation of VSMC proliferation could be found after PD98059 (ERK1/2 phosphorylation inhibitor) treatment. Our data also showed that a siRNA-mediated caveolin-1 knock down increased ERK1/2 phosphorylation and VSMC proliferation. These results demonstrate that static pressure promotes VSMC proliferation via the Caveolin-1/ERK1/2 pathway.  相似文献

Here, the leaf photoacclimatory plasticity and efficiency of the tropical seagrass Thalassia testudinum were examined. Mesocosms were used to compare the variability induced by three light conditions, two leaf sections and the variability observed at the collection site. The study revealed an efficient photosynthetic light use at low irradiances, but limited photoacclimatory plasticity to increase maximum photosynthetic rates (P(max)) and saturation (E(k)) and compensation (E(c)) irradiances under high light irradiance. A strong, positive and linear association between the percentage of daylight hours above saturation and the relative maximum photochemical efficiency (F(V)/F(M)) reduction observed between basal and apical leaf sections was also found. The results indicate that T. testudinum leaves have a shade-adapted physiology. However, the large amount of heterotrophic biomass that this seagrass maintains may considerably increase plant respiratory demands and their minimum quantum requirements for growth (MQR). Although the MQR still needs to be quantified, it is hypothesized that the ecological success of this climax species in the oligotrophic and highly illuminated waters of the Caribbean may rely on the ability of the canopy to regulate the optimal leaf light environment and the morphological plasticity of the whole plant to enhance total leaf area and to reduce carbon respiratory losses.  相似文献

Certain mutations in S12, a ribosomal protein involved in translation elongation rate and translation accuracy, confer resistance to the aminoglycoside streptomycin. Previously we showed in Salmonella typhimurium that the fitness cost, i.e. reduced growth rate, due to the amino acid substitution K42N in S12 could be compensated by at least 35 different mutations located in the ribosomal proteins S4, S5 and L19. Here, we have characterized in vivo the fitness, translation speed and translation accuracy of four different L19 mutants. When separated from the resistance mutation located in S12, the three different compensatory amino acid substitutions in L19 at position 40 (Q40H, Q40L and Q40R) caused a decrease in fitness while the G104A change had no effect on bacterial growth. The rate of protein synthesis was unaffected or increased by the mutations at position 40 and the level of read-through of a UGA nonsense codon was increased in vivo, indicating a loss of translational accuracy. The mutations in L19 increased sensitivity to aminoglycosides active at the A-site, further indicating a perturbation of the decoding step. These phenotypes are similar to those of the classical S4 and S5 ram (ribosomal ambiguity) mutants. By evolving low-fitness L19 mutants by serial passage, we showed that the fitness cost conferred by the L19 mutations could be compensated by additional mutations in the ribosomal protein L19 itself, in S12 and in L14, a protein located close to L19. Our results reveal a novel functional role for the 50 S ribosomal protein L19 during protein synthesis, supporting published structural data suggesting that the interaction of L14 and L19 with 16 S rRNA could influence function of the 30 S subunit. Moreover, our study demonstrates how compensatory fitness-evolution can be used to discover new molecular functions of ribosomal proteins.  相似文献

The growing threat from the emergence of multidrug resistant pathogens highlights a critical need to expand our currently available arsenal of broad-spectrum antibiotics. In this connection, new antibiotics must be developed that exhibit the abilities to circumvent known resistance pathways. An important step toward achieving this goal is to define the key molecular interactions that govern antibiotic resistance. Here, we use site-specific mutagenesis, coupled with calorimetric, NMR, and enzymological techniques, to define the key interactions that govern the binding of the aminoglycoside antibiotics neomycin and kanamycin B to APH(3')-IIIa (an antibiotic phosphorylating enzyme that confers resistance). Our mutational analyses identify the D261, E262, and C-terminal F264 residues of the enzyme as being critical for recognition of the two drugs as well as for the manifestation of the resistance phenotype. In addition, the E160 residue is more important for recognition of kanamycin B than neomycin, with mutation of this residue partially restoring sensitivity to kanamycin B but not to neomycin. By contrast, the D193 residue partially restores sensitivity to neomycin but not to kanamycin B, with the origins of this differential effect being due to the importance of D193 for catalyzing the phosphorylation of neomycin. These collective mutational results, coupled with (15)N NMR-derived pK(a) and calorimetrically derived binding-linked drug protonation data, identify the 1-, 3-, and 2'-amino groups of both neomycin and kanamycin B as being critical functionalities for binding to APH(3')-IIIa. These drug amino functionalities represent potential sites of modification in the design of next-generation compounds that can overcome APH(3')-IIIa-induced resistance.  相似文献

The presence of cancer stem cells, in both hematopoietic and solid malignancies, has been recently linked to their pathogenesis. We aimed to identify the characteristics and stem-like properties of sphere-colony forming cells in rat osteosarcoma and malignant fibrous histiocytoma cell lines. The results showed that both cell lines possessed an ability to form spherical, clonally expanding colonies in anchorage-independent, serum-starved conditions in N2/1% methylcellulose medium. The sphere cells showed stem-like properties with the ability to self-renew, and expressed the stem cell-related STAT3 and Bmi1 genes. Interestingly, spheres from both sarcomas remarkably decreased the expression of INK4a/ARF locus genes, p16(INK4a) and p19(ARF), which could be related to the resistance against cell senescence and apoptosis. Spheres showed strong tumorigenicity with metastatic potential in vivo via the inoculation into syngeneic rats, suggesting the presence of these populations might contribute to the tumor development such as metastasis via the resistance to apoptotic stimuli.  相似文献

采用抑制剂加入法估算了中肋骨条藻、棕囊藻和东海原甲藻在不同生长期内二甲基硫化物的产生与消耗速率.结果表明:颗粒态二甲基巯基丙酸(DMSPp)和颗粒态二甲亚砜(DMSOp)在3种藻类的不同生长期内均为净消耗,溶解态二甲基巯基丙酸(DMSPd)和溶解态二甲亚砜(DMSOd)的含量受藻类产生与细菌病毒消耗控制,在藻类不同生长期内存在不同的产生与消耗速率,而二甲基硫(DMS)在3种藻不同生长期内均为净产生.同一种藻在不同生长期内以及不同藻在相同生长期内二甲基硫化物的产生与消耗速率均存在较大差异,表明藻类的生理状态和种间差异均对二甲基硫化物的产生与消耗速率产生影响.  相似文献

2005年在长江口赤潮频发海域调查期间,分别于4月27日、5月4日和5月8日,在zzf1、zc18a和ra5站位利用现场船基培养的方法,研究了添加到赤潮密度10^6cells L^-1的东海原甲藻(Prorocentrum donghaiense)对原生动物群落结构的影响。结果发现,赤潮密度的东海原甲藻抑制了小型无壳纤毛虫的种群数量,而没有抑制中大型砂壳纤毛虫和夜光虫(Noctiluca scintillans)的种群数量,从而使得原生动物群落向中大型种类演替,这种影响的程度与原生动物本身的群落组成和浮游植物的组成密切相关。添加东海原甲藻72h后,在以小型无壳纤毛虫管游虫(Cyrtostrombidium sp.)为优势种的zzf1站位,演替为以大型原生动物夜光虫为优势种的群落;在以中大型原生动物百乐拟铃虫(Tintinnospsis beroidea)和夜光虫为优势种的zc18a站位,仍然是以此为优势种,且大型原生动物夜光虫在群落中的比例上升;在以小型无壳纤毛虫急游虫2(Strombidium sp.2)为优势种的ra5站位,演替为以中大型原生动物百乐拟铃虫和亚速岛网纹虫(Favella azorica)为优势种的群落。zzf1和ra5站位实验组中原生动物的总丰度都呈下降趋势,而zc18a站位变化不明显,这是由于前两个站位的最优势种管游虫(zzf1站位)和急游虫2(ra5站位)的丰度迅速下降,而zc18a站位的优势种百乐拟铃虫和夜光虫的丰度比较稳定造成的。在zzf1和zc18a站位,对照组和实验组中原生动物的总生物量在实验前后都没有大的变化,而在ra5站位却均呈下降的趋势。这可能与浮游植物的组成有关,zzf1和zc18a站位是以无毒的锥状斯氏藻(Scrippsiella trochoidea)和螺旋环沟藻(Gyrodinium spirale)为主,而ra5站位是以有毒的亚历山大藻(Alexandriumsp.)为主。综上可见,可预测当东海大规模赤潮爆发时,会使原生动物群落向中大型种类演替,进而可能影响海洋生态系统的结构和功能。  相似文献

The minimum sampling areas (MSAs) for the shrub communities in the arid valley in the upper reach of the Minjiang River, China, were studied by fitting community species-area relationships using 3 types of equations. The MSAs were determined at the proportional factor (ρ) 0.6, 0.7, 0.8 and 0.9. The proportional factors represent the proportion of the number of species within a sampling plot in the total number of species. The MSAs of the shrub communities at different elevations and on different slope faces for ρ = 0.6, 0.7 and 0.8 were all around 100 m2. Hence, the MSAs could be set to be 100 m2 (10 m × 10 m) at 60%–80% precision levels. For ρ = 0.9, that is, for a 90% precision level, the MSAs were less than 200 m2 (10 m × 20 m). The MSAs and species richness increased gradually with the rising elevation. At the elevation below 2000 m, the MSAs and species richness on the north-facing slope were larger than those on the south-facing slope. However, at the elevation around 2200 m, there was no difference amongst different facing slopes. For the shrub communities in the arid valley in the upper reach of the Minjiang River, the species-area curves by fitting the first two equations are better than that by fitting the third equation.  相似文献

To assess soil quality, a minimum dataset (MDS) of soil properties has to be proposed commonly through calculating the total load of each candidate soil parameter on all of the qualified principal components by use of principal component analysis (PCA) and Norm-value computation. Considering intensive land-use changes, the method introduced in this study on MDS establishment integrates the quantified contributions of land-use type and land-use duration on each soil parameter by using multivariate analysis and mean multiple comparison. In this way, a MDS maximally representing all candidates with minimal loss of the soil quality information contained by those non-MDS soil parameters is established. The MDS proposed can not only well integrate the quantified influence of land-use changes and land-use duration on soil parameters, but is also quite flexible and extendable with the potential to be extrapolated to assess soil quality in other regions. Based on two sets of soil database obtained separately in 1985 and 2004, two MDSs established are compared with each other. It is found that only quite a small change in MDS components occurs during a 20-year period. For a better assessment of soil quality, it seems necessary to examine on what kind of temporal scale and how much MDS will change for a site-specific area with intensive land-use changes.  相似文献
