【目的】明确马铃薯Y病毒(potato virus Y, PVY)侵染后诱导的烟草营养成分的变化及其对烟蚜Myzus persicae生命特性的影响,旨在进一步解析PVY-烟草-烟蚜三者间的互作机制。【方法】通过蒽酮比色法和氨基酸自动分析仪测定了PVY不同侵染时期烟株体内的可溶性糖和游离氨基酸含量的变化;测定和比较了感病与健康烟草植株上烟蚜种群生长发育、成虫寿命、繁殖力和有翅蚜产生量的差异性。【结果】PVY侵染前、中、后期(分别为侵染后5, 12和20 d)的烟草叶片中游离氨基酸的总量均显著高于健康烟草叶片。相较于健康烟草叶片,在PVY侵染前期的烟草叶片中,谷氨酸、脯氨酸、天冬氨酸、色氨酸、缬氨酸、赖氨酸和组氨酸的含量显著增加;PVY侵染中期,感病叶片中丝氨酸含量显著下降,谷氨酸、天冬氨酸、色氨酸、缬氨酸、亮氨酸、苯丙氨酸、精氨酸和组氨酸含量显著提高;PVY侵染后期,感病叶片中甘氨酸含量显著下降,谷氨酸、脯氨酸、天冬氨酸、苏氨酸、缬氨酸、亮氨酸、丙氨酸、苯丙氨酸、组氨酸、酪氨酸和精氨酸含量显著提高。在PVY侵染的前期和中期,感病叶片中的可溶性糖含量显著高于健康烟叶,而在侵染后期感病叶片... 相似文献

In young systemically infected leaves of Datura stramonium L., a severe strain of Potato virus X (PVX) accumulated to a lower degree than a mild strain. Infected leaves had increased protease and RNase activities in comparison with those of healthy controls. The highest hydrolase activities were found in leaves infected with the severe strain. Negative‐staining electron microscopy of dips from the infected leaves indicated that PVX virions underwent destructive changes, which resulted in the appearance of abnormal (swollen and ‘thin’) particles. Immuno‐electron microscopic assays showed that thin PVX particles, in contrast to those of normal diameter, lost the ability to bind with specific antiserum. The relative number of thin virions in leaves infected with the severe PVX strain was considerably higher than in leaves infected with the mild strain. This shows that a correlation exists between increased protease activity and intracellular destruction of virions. In abnormal virions, the viral RNA appears to be available for RNase attack. Therefore, it seems that high RNase activity together with increased generation of abnormal virions in the leaves infected with the severe strain promote inactivation of the viral RNA with RNase. We suppose that the enhanced hydrolase activities in the leaves infected with severe PVX strain, on the one hand, limit viral accumulation and thus play a defensive role and, on the other hand, cause considerable intracellular pathological changes resulting in severe symptoms. 相似文献

A potential novel method of producing high-quality potato (Solanum tuberosum L.) nuclear seeds is through the process of somatic embryogenesis (SE). Somatic embryo formation has been successfully reported in many plant species, but in potato, reliable SE systems are still at the experimental stage. A key factor in the success of any SE system is the ability to discriminate SE-specific cellular structures from those emerging through an organogenic route. In the investigation reported here we attempted to discriminate the progression of specific stages of potato SE by histological means. Internodal segment (INS) explants from 4- to 6-week-old cv. Desiree in vitro cultures were successively cultured on SE induction (for 2 weeks) and expression/regeneration media (for 3 weeks) with and without 2,4-dichlorophenoxyacetic acid (5 M). Microscopic examination of histological slides prepared using INS explants at different stages revealed the presence of characteristic globular, heart and torpedo stages in the potato SE system along with other associated unique features such as protoderm development and discrete vascular connections. The level of genetic diversity within populations of introduced species has received increasing attention as an important
factor influencing their survival and adaptive potential. We examined this issue with the Guatemalan potato tuber moth Tecia solanivora, an agricultural pest which has successfully invaded South America and the Canary Islands within the last 20 years. To analyse
changes in T. solanivora genetic diversity, the mitochondrial marker cytochrome b was sequenced from individuals collected across its known distribution area. High haplotypic diversity was observed in Guatemala,
whereas only three haplotypes have been found in Venezuela and a single one in the remaining invaded South American countries
and the Canary Islands. Invasive haplotypes were not observed in our samples from Guatemala but are closely related to Guatemalan
haplotypes. These results are consistent with the hypotheses that (i) either a few individuals were introduced into Venezuela leading to a strong initial genetic bottleneck, or selection pressure
may have lead to the disparition of all but a reduced number of introduced haplotypes, (ii) a second bottleneck occured between Venezuela and Colombia, and (iii) the invasion of the Canary Islands originated from South America. We further reviewed the recent literature to compare this
change in genetic diversity with those reported for other invasive species. We quantified the changes in genetic diversity
between native and introduced ranges for 57 biological invasions. We found that the genetic homogenization in T. solanivora was among the strongest reported and discuss factors that can explain the success of invasive populations with low genetic

A broad spectrum of fungal antagonists was evaluated as potential biocontrol agents (BCAs) against the soil-borne pathogen Rhizoctonia solani using a new combination of in vitro and in vivo assays. The in vitro characterisation of diverse parameters including the ability to parasitise mycelium and to inhibit the germination of Rhizoctonia sclerotia at different temperatures resulted in the selection of six potential fungal antagonists. These were genotypically characterised by their BOX-PCR fingerprints, and identified as Trichoderma reesei and T. viride by partial 18S rDNA sequencing. When potato sprouts were treated with Trichoderma, all isolates significantly reduced the incidence of Rhizoctonia symptoms. Evaluated under growth chamber conditions, the selected Trichoderma isolates either partly or completely controlled the dry mass loss of lettuce caused by R. solani. Furthermore, the antagonistic Trichoderma strains were active under field conditions. To analyse the effect of Trichoderma treatment on indigenous root-associated microbial communities, we performed a DNA-dependent SSCP (Single-Strand Conformation Polymorphism) analysis of 16S rDNA/ITS sequences. In this first assessment study for Trichoderma it was shown that the pathogen and the vegetation time had much more influence on the composition of the microbiota than the BCA treatment. The main components (90%) in recovered vegetable oil (RVO) are esters of higher carboxylic acids and glycerol (triglycerides) which consist of three fatty acids: oleic (C18:1, Z), palmitic (C16:0) and linoleic (C18:2, Z) in the ratio 2.8:1.4:1. RVO may provide a low cost new supply of unsaturated fatty acid chlorides. Modification of potato starch using the mixture of fatty acid chlorides derived from RVO and using acid chlorides of the two major pure component acids has been performed. Films were prepared from the starch esters and mechanical properties tested. Commercially available potato starch, modified to DS-value 1.5 with RVO-derived acyl chlorides, was thermoplastic with a maximum tensile strength of 1.4 MPa and an elongation at break of 54%. 相似文献

Among the multiple environmental signals and hormonal factors regulatingpotato plant morphogenesis and controlling tuber induction, jasmonates (JAs)andgibberellins (GAs) are important components of the signalling pathways in theseprocesses. In the present study, with Solanum tuberosum L.cv. Spunta, we followed the endogenous changes of JAs and GAs during thedevelopmental stages of soil-grown potato plants. Foliage at initial growthshowed the highest jasmonic acid (JA) concentration, while in roots the highestcontent was observed in the stage of tuber set. In stolons at the developmentalstage of tuber set an important increase of JA was found; however, in tubersthere was no change in this compound during tuber set and subsequent growth.Methyl jasmonate (Me-JA) in foliage did not show the same pattern as JA; Me-JAdecreased during the developmental stages in which it was monitored, meanwhileJA increased during those stages. The highest total amount of JAs expressed asJA+Me-JA was found at tuber set. A very important peak ofJA in roots was coincident with that observed in stolons at tuber set. Also, aprogressive increase of this compound in roots was shown during the transitionof stolons to tubers. Of the two GAs monitored, gibberellic acid(GA3) was the most abundant in all the organs. While GA1and GA3 were also found in stolons at the time of tuber set, noothermeasurements of GAs were obtained for stolons at previous stages of plantdevelopment. Summary A mathematical model of the species-area relation is described for a group of limited species. This model is a modification
of that proposed earlier (Kobayashi, 1975), being assumed that the limited species are expected to occur in a habitat under consideration. The model equation
gives a sigmoid species-log area curve implying that few rare species are found in a group of species. The good agreement
between observation and this model is exemplified with the data of plant and arthropod communities. The implication of parameters
involved are examined in connection with those of the preceding model, and the underlying ecology of the model is discussed.

Over 6 yr the effectiveness of imazalil, prochloraz and fenpiclonil, applied either alone or in a mixture, in controlling gangrene, dry rot, skin spot and silver scurf on potato tubers in store was compared with that of 2-aminobutane and thiabendazole. An assessment was also made of their efficiency in controlling the development of these diseases on the daughter tubers of seed tubers treated at harvest or before planting. Overall, 2-aminobutane was more effective in controlling gangrene (Phoma foveata) in store than the spray-applied fungicides. Deposits of imazalil, thiabendazole and fenpiclonil were greater when sprays were applied with an electrostatic sprayer than with a hydraulic sprayer. The opposite was found with the mixture of prochloraz Mn and tolclofos-methyl. More effective gangrene control was associated with the highest deposits. Fenpiclonil and the mixture of thiabendazole and imazalil were more effective in controlling dry rot (Fusarium solani var. coeruleum) than imazalil alone. The development of dry rot was, however, increased by 2-aminobutane treatment on eight out of 14 stocks. 2-aminobutane gave the greatest reduction (83%) in the severity of skin spot during storage whereas thiabendazole alone, and the mixture of thiabendazole and imazalil, gave mean reductions of 70% and 65% respectively. This mixture and fenpiclonil gave the greatest reduction in the severity of silver scurf although, in general, reductions in silver scurf with fungicide treatment were less than with skin spot. The type of sprayer used to apply a fungicide did not affect the effectiveness of the fungicides in controlling either skin spot or silver scurf on tubers in store, or on the daughter tubers. The incidence of gangrene and dry rot on daughter tubers was not reduced consistently by fungicide treatment of seed tubers of the six stocks tested. However, the severity of skin spot and silver scurf was reduced by fungicide treatments of all eight stocks but the reduction in disease was greater for skin spot than for silver scurf. All fungicides gave reductions in the severity of skin spot, and fenpiclonil and the mixture of thiabendazole and imazalil were the most effective for silver scurf. The effectiveness of the fungicides in controlling these diseases was similar for seed treated at harvest and that treated before planting. 相似文献
