Question: Is the stimulation of germination by thermal shock (resulting from the passage of fire) commonly observed for Mediterranean‐climate Fabaceae also apparent for NW European Genisteae (Fabaceae) species? Location: Southern England and NW France. Methods: The germination of Cytisus scopiarius, Genista anglica, Ulex europaeus, Ulex gallii and Ulex minor was examined following exposure to a range of temperatures (50°C, 65°C, 80°C, 95°C and 110°C) applied to seeds for 5 min. A sixth Mediterranean‐origin species (Spartium junceum) was also included since it is a common invasive in NW Europe and North America. Results: All five native NW European species displayed increased germination following thermal shock, even when seeds were heated to 110°C. However, there was some variation depending on provenance: in contrast to seeds collected from southern England, germination of French C. scopiarius seeds was unaffected by temperature. Spartium junceum germinated most at 95°C, but was the only species to show reduced germination when seeds were heated to 110°C. Conclusions: The NW European Genisteae appear to be pre‐adapted to the high temperatures associated with fire; a response attributable to their evolutionary origins in the fire‐prone ecosystems of the Mediterranean Basin. Consequently, projected increases in fire frequency linked to climate change may stimulate their regeneration in NW European heathlands, potentially altering the species composition of these ecosystems. Additionally, a clearer understanding of the interaction between thermal shock and germination may explain why fire has so frequently been ineffective in controlling invasive Genisteae populations worldwide.  相似文献

Question: The recovery of forest plant communities in post‐agricultural landscapes is largely determined by dispersal constraints, but can environmental legacies of former land use additionally limit the recolonization of recent forests by forest herbs? Location: Ancient forest and recent forest on former heavily fertilized agricultural land (Muizenbos, northern Belgium). Methods: Seeds and adults of two forest herbs with similar life‐history traits, but contrasting colonization capacity – the fast‐colonizing Geum urbanum and the slow‐colonizing Primula elatior– were introduced into both ancient and recent forest sites. Soil conditions and plant tissue nutrient concentrations were measured to characterize habitat quality. To determine whether the introduced species could successfully establish and persist, we monitored recruitment, longevity and adult performance during 8 years in permanently marked plots. Results: Phosphorus availability was ten times higher in recent forest soils and was also reflected in the plant tissue samples. Species longevity was clearly lower in recent forest sites indicating higher turnover. The fast‐colonizing G. urbanum counterbalanced this lower longevity by new establishment, while the slow‐colonizing P. elatior dropped below the number of originally introduced individuals. Additionally, G. urbanum performed better in recent forest sites in contrast to P. elatior. Conclusions: Even when dispersal constraints of the slow‐colonizing forest herb P. elatior are eliminated through introduction, environmental conditions in recent forest sites additionally restrict its recruitment, longevity and performance. These experimental results suggest that environmental constraints may strengthen the differences in colonization capacity among forest herbs if slow dispersers also tend to be less likely to establish.  相似文献

MassMatrix is a program that matches tandem mass spectra with theoretical peptide sequences derived from a protein database. The program uses a mass accuracy sensitive probabilistic score model to rank peptide matches. The MS/MS search software was evaluated by use of a high mass accuracy dataset and its results compared with those from MASCOT, SEQUEST, X!Tandem, and OMSSA. For the high mass accuracy data, MassMatrix provided better sensitivity than MASCOT, SEQUEST, X!Tandem, and OMSSA for a given specificity and the percentage of false positives was 2%. More importantly all manually validated true positives corresponded to a unique peptide/spectrum match. The presence of decoy sequence and additional variable PTMs did not significantly affect the results from the high mass accuracy search. MassMatrix performs well when compared with MASCOT, SEQUEST, X!Tandem, and OMSSA with regard to search time. MassMatrix was also run on a distributed memory clusters and achieved search speeds of ~100 000 spectra per hour when searching against a complete human database with eight variable modifications. The yeast (Saccharomyces cerevisiae) 26S proteasome consists of the 19S regulatory particle (19S RP) and 20S proteasome subunits. We detected comprehensively co‐ and post‐translational modifications of these subunits using proteomic techniques. First, using MS/MS, we investigated the N‐terminal modifications of three 19S RP subunits, Rpt1, Rpn13, and Rpn15, which had been unclear, and found that the N‐terminus of Rpt1 is not modified, whereas that of Rpn13 and Rpn15 is acetylated. Second, we identified a total of 33 Ser/Thr phosphorylation sites in 15 subunits of the proteasome. The data obtained by us and other groups reveal that the 26S proteasome contains at least 88 phospho‐amino acids including 63 pSer, 23 pThr, and 2 pTyr residues. Dephosphorylation treatment of the 19S RP with λ phosphatase resulted in a 30% decrease in ATPase activity, demonstrating that phosphorylation is involved in the regulation of ATPase activity in the proteasome. Third, we tried to detect glycosylated subunits of the 26S proteasome. However, we identified neither N‐ and O‐linked oligosaccharides nor O‐linked β‐N‐acetylglucosamine in the 19S RP and 20S proteasome subunits. To date, a total of 110 co‐ and post‐translational modifications, including Nα‐acetylation, Nα‐myristoylation, and phosphorylation, in the yeast 26S proteasome have been identified.  相似文献

Osteogenesis imperfecta (OI) is a skeletal disorder primarily caused by mutations in the type I collagen genes. However, recent investigations have revealed that mutations in the genes encoding for cartilage-associated protein (CRTAP) or prolyl 3-hydroxylase 1 (P3H1) can cause a severe, recessive form of OI. These reports show minimal 3-hydroxylation of key proline residues in type I collagen as a result of CRTAP or P3H1 deficiency and demonstrate the importance of P3H1 and CRTAP to bone structure and development. P3H1 and CRTAP have previously been shown to form a stable complex with cyclophilin B, and P3H1 was shown to catalyze the 3-hydroxylation of specific proline residues in procollagen I in vitro. Here we describe a mouse model in which the P3H1 gene has been inactivated. Our data demonstrate abnormalities in collagen fibril ultrastructure in tendons from P3H1 null mice by electron microscopy. Differences are also seen in skin architecture, as well as in developing limbs by histology. Additionally bone mass and strength were significantly lower in the P3H1 mice as compared with wild-type littermates. Altogether these investigations demonstrate disturbances of collagen fiber architecture in tissues rich in fibrillar collagen, including bone, tendon, and skin. This model system presents a good opportunity to study the underlying mechanisms of recessive OI and to better understand its effects in humans.  相似文献

摘要 目的:基于脑肠轴理论探讨健脾化痰方联合针刺治疗卒中后抑郁(PSD)的治疗效果。方法:选取石家庄市中医院2018年4月~2022年5月期间收治的113例PSD患者。按照双色球随机分组法将患者分为对照组(n=56,基于脑肠轴理论,常规治疗联合健脾化痰方治疗)和研究组(n=57,在对照组基础上联合针刺治疗)。对比两组疗效、中医证候积分、美国国立卫生研究院卒中量表(NIHSS)评分、汉密尔顿抑郁量表-17(HAMD-17)评分、日常生活活动能力(ADL)评分、P物质(SP)和神经肽Y(NPY),观察两组治疗期间不良反应发生情况。结果:研究组的临床总有效率高于对照组(P<0.05)。与对照组相比,研究组治疗4周后中医证候积分总分、NIHSS评分、HAMD-17评分和血清SP水平更低,ADL评分和血清NPY水平更高(P<0.05)。两组均未见明显的不良反应发生。结论:基于脑肠轴理论采用健脾化痰方联合针刺治疗PSD患者,可改善患者的临床症状、生活自理能力,同时还可减轻神经缺损状况,调节血清SP、NPY水平。  相似文献
