Isolation and purification of mesenchymal stem cells (MSCs) from mouse via plastic adherent cultures is arduous because of
the unwanted growth of hematopoietic cells and non-MSCs. In this work, homogenous populations of CD34+ MSCs from mouse bone marrow were isolated via positive selection. For this purpose, C57Bl/6 mice were killed and bone marrow
cells were aspirated before incubation with magnetic bead conjugated to anti-CD34 antibody. A sample of positively selected
CD34+ cells were prepared for flow cytometry to examine the expression of CD34 antigen and others were subcultured in a 25-cm2 culture flask. To investigate the mesenchymal nature, the plastic adherent cultivated cells were induced to differentiate
along osteoblastic and adipogenic lineages. Furthermore, the expression of some surface markers was investigated by flow cytometry.
According to the result, purified populations of fibroblast-like CD34+ cells were achieved in the first passage (1 wk after culture initiation). The cells expressed CD34, CD44, Sca-1, and Vcam-1
antigens (markers) but not CD11b and CD45. They were capable of differentiating into osteocytes and adipocytes. This study
indicated that our protocol can result in the efficient isolation of homogenous populations of MSCs from C57BL/6 mouse bone
marrow. We have shown that murine bone marrow-derived CD34+ cells with plastic adherent properties and capability of differentiating into skeletal lineages in vitro are MSCs. 相似文献

Questions: 1. Is there any post‐dispersal positive effect of the exotic shrub Pyracantha angustifolia on the success of Ligustrum lucidum seedlings, as compared to the effect of the native Condalia montana or the open herbaceous patches between shrubs? 2. Is the possible facilitation by Pyracantha and/or Condalia related to differential emergence, growth, or survival of Ligustrum seedlings under their canopies? Location: Córdoba, central Argentina. Methods: We designed three treatments, in which ten mature individuals of Pyracantha, ten of the dominant native shrub Condalia montana, and ten patches without shrub cover were involved. In each treatment we planted seeds and saplings of Ligustrum collected from nearby natural populations. Seedlings emerging from the planted seeds were harvested after one year to measure growth. Survival of the transplanted saplings was recorded every two month during a year. Half of the planted seeds and transplanted saplings were cage‐protected from rodents. Results: Ligustrum seedling emergence did not differ among treatments while growth was significantly higher in the absence of shrub cover. Sapling survival was significantly higher under the canopy of Pyracantha, intermediate under Condalia, and lowest in the absence of shrub cover. Caging did not affect growth but enhanced seedling emergence and sapling survival. Conclusion: The differential sapling survival in the shrub canopy treatments is consistent with natural sapling distribution. Pyracantha and, less so, Condalia, has a nurse‐plant effect on Ligustrum. This results from contrasting effects of the shrubs on different stages of the life cycle of Ligustrum: no effect on seedling emergence, negative on seedling growth, and positive on sapling survival. This suggests that efforts to control the expansion of Ligustrum over the landscape should tackle Pyracantha as well. 相似文献

A mathematical model is presented for the emergence of perceptual-cognitive-behavioral modes in psychophysical experiments in which participants are confronted with two alternatives. The model is based on the theory of self-organization and, in particular, the order parameter concept such that the emergence of a mode is conceptualized as an instability leading to the emergence of an appropriately defined order parameter. The order parameter model is merged with a second model that describes adaptation in terms of a system parameter dynamics. It is shown that the two-component model predicts hysteretic mode-mode transitions when control parameters are increased or decreased beyond critical values. The two-component model can account for both positive and negative hysteresis effects due to the interaction between order parameter and system parameter dynamics. Moreover, the model-based analysis reveals that response time curves look rather flat when response times are relatively decoupled from the mode-mode transition phenomenon. In general, response time curves exhibit a peaked close to the mode-mode transition point. In this context, the possibility is discussed that such peaked response time curves belong to the class of critical phenomena of self-organizing systems. In order to illustrate the relevance of peaked response time curves for future research and research reported in the past, results from a perceptual judgment experiment are reported, in which participants judged their ability to stand on a tilted slope for various angles of inclination. Creating multiyear cycles in population density demands, in traditional models, causal factors that operate on local populations in a density-dependent way with time lags. However, cycles of the geometrid Epirrita autumnata in northern Europe may be regional, not local; i.e., successive outbreaks occur in different localities. We review possible causes of cycles of E. autumnata under both local and regional scenarios, including large-scale synchrony. Assuming cyclicity is a local phenomenon, individual populations of E. autumnata display peaks but populations all over the outbreak range fluctuate in synchrony. This concept assumes that the peaks at most localities are so low that they do not lead to visible defoliation and easily remain unnoticed. In this scenario, populations are able to start recovery a few years after the crash, i.e., at the time of the mitigation of detrimental delayed density-dependent factors, such as delayed inducible resistance of the host plant or parasitism. In that case, the same factors that lead to crashes also explain the periodicity of cyclic fluctuations. According to the regional cyclicity scenario, different factors can be important in different phases of the cycle. The key is to identify the factors that tend to produce outbreaks with a periodicity of about 10 years. Initiation of the increase phase seems to coincide with maxima in sunspot activity, but causal connections remain unclear. Climatic factor(s) associated with the solar cycle could contribute to the large-scale geographic synchrony. 相似文献

We analyze members of the receptor-like kinase (RLK) gene family in Arabidopsis thaliana for positive selection. Likelihood analyses find evidence for positive selection in 12 of the 52 RLK family sequences groups. These 12 groups represent 97 of the 403 sequences analyzed. The majority of genes in groups subject to positive selection have not been functionally characterized, but sites under selection are predominantly located in the extracellular region. The pattern of selection in the extracellular leucine-rich repeat (LRR) motif of groups 14 and 51 is similar to previous studies where positively selected positions are located in a solvent exposed β-strand that may determine disease specificity, raising the possibility that some RLK genes function in a similar role.[Reviewing Editor: Rasmus Nielsen] 相似文献

Nonrandom patterns associated with adaptively evolving genes can shed light on how selection and mutation produce rapid changes
in sequences. I examine such patterns in two independent families of antimicrobial peptide genes: those in frogs, which are
known to have evolved under positive selection, and those in flatfishes, which I show have also evolved under positive selection.
I address two recently proposed hypotheses about the molecular evolution of antimicrobial peptide genes. The first is that
the mature peptide region is replicated by an error-prone polymerase that increases the mutation rate and the transversion/transition
ratio compared to the signal sequence of the same genes. The second is that mature peptides evolve in a coordinated fashion
with their propieces, such that a change in net charge in one molecular region prompts an opposite change in charge in the
other region. I test these hypotheses using alternative methods that minimize alignment errors, correct for phylogenetic nonindependence,
reduce sequence saturation, and account for differing selection pressures on different regions of the gene. In both gene families
I show that divergence at both synonymous and nonsynonymous sites within the mature peptide region is enhanced. However, in
neither gene family is there evidence of an increased mutational transversion/transition ratio or coordinated evolution. My
observations are consistent with either an elevated mutation rate in an adaptively evolving gene region or widespread selection
on “silent” sites. These hypotheses challenge the assumption that mutations are random and can be measured by the synonymous
substitution rate.

The vomeronasal receptor 1 (V1R) are believed to be pheromone receptors in rodents. Here we used computational methods to identify 95 and 62 new putative V1R genes from the draft rat and mouse genome sequence, respectively. The rat V1R repertoire consists of 11 subfamilies, 10 of which are shared with the mouse, while rat appears to lack the H and I subfamilies found in mouse and possesses one unique subfamily (M). The estimations of the relative divergence times suggest that many subfamilies originated after the split of rodents and primates. The analysis also reveals that these clusters underwent an expansion very close to the split of mouse and rat. In addition, maximum likelihood analysis showed that the nonsynonymous and synonymous rate ratio for most of these clusters was much higher than one, suggesting the role of positive selection in the diversification of these duplicated V1R genes. Because V1R are thought to mediate the process of signal transduction in response to pheromone detection, we speculate that the V1R genes have evolved under positive Darwinian selection to maintain the ability to discriminate between large and complex pheromonal mixtures.Reviewing Editor: Dr. Rasmus Nielsen 相似文献
