Various factors (pollen diameter, in vitro germination and tube length, in vivo growth rate in selfed and nonselfed styles) which could possibly contribute to the competitive ability of pollen were investigated on 30 Zea mays L. inbred lines. The only factor with which pollen diameter was positively correlated was in vitro pollen-tube growth. Traits related to the early stages of growth (in vitro germination, in vitro tube length, early in vivo pollen growth rate) were all positively correlated with each other, and these early characteristics were negatively correlated with late in vivo tube growth rate, which is largely influenced by the stylar genotype.  相似文献

Summary The influence of different concentrations of the heavy metals cadmium (Cd2+), cobalt (Co2+), copper (Cu2+), iron (Fe2+ and Fe3+), mercury (Hg2+), manganese (Mn2+), and zinc (Zn2+), plus aluminium (Al3+) (a toxic metal in polluted areas), on pollen germination and tube growth ofLilium longiflorum was investigated using light microscopy. Effects could be observed with 3 M and 100 M of heavy metal, added as chloride salts to the medium. Cd2+, Cu2+, and Hg2+, showed the greatest toxicity, whereas germination and growth rate was less affected by Mn2+. Affected tubes showed swelling of the tip region. Tubes treated with Cd2+, Co2+, Fe2+, Fe3+, Hg2+, and Mn2+ were also prepared for ultrastructural studies. In all cases, the main effect was abnormal cell wall organization, mostly at the tip, where round, fibrillar aggregates, the shape and size of secretory Golgi vesicles were formed. They built up a loose network which could be up to 10 m thick compared to untreated tubes where the cell wall was composed of thin layers of long fibrils and about 100 nm thick. Cd2+ was the only metal which produced effects at the intracellular level: organelle distribution within the tip region appeared disorganized. A general mechanism of heavy metal action on pollen tube growth is discussed.  相似文献

Genetic parameters of size through sexual maturity have been relatively unexplored for Pacific salmon. In this study, individually tagged coho salmon were raised in freshwater, and the heritabilities of size and growth rate were estimated at several intervals between 13 and 24 months of age (spawning). Heritability estimates for size were moderate to high from 13 to 19 months of age, ranging from 0.36 to 0.50, and lower from 21 months to spawning at 24 months, ranging from 0.17 to 0.32. Heritabilities of specific growth rates estimated over 3-month intervals were moderate from 16 to 21 months of age, ranging from 0.21 to 0.34. Genetic and phenotypic correlations between sizes measured at different ages were moderate to high, ranging from about 0.7 to 1.0. Correlations between growth rate and size indicated that the larger fish were the fastest growing between 16 and 19 months of age and were slower growing between 19 and 21 months of age.  相似文献

Investigations were performed on growth phase-dependent EcoRII site-specific DNA methylation of the carrot genome during primary culture to elucidate physiological aspects of genome DNA variability in tissue culture. While DNA methylation of the root cambium and the secondary phloem and petioles of carrot leaves were strikingly different, the methylation level of the secondary phloem seemed to be independent of cultivar origin, the age of the plants and the extent of secondary root growth. As was shown earlier a change in the differentiated state of the secondary phloem by tissue culture leads to changes in genome modification. Whereas de novo methylation was observed during the first 2 weeks of growth initiation, the results presented demonstrate genome de-methylation during the transition to stationary growth indicating differential nome methylation during different phases of culture. The presence of kinetin in the nutrient medium of the primary culture was found to be antagonistic to changes in genome modification in general. De novo methylation and subsequent de-methylation of the carrot genome are discussed as gross changes obviously essential to molecular genome differentiation during tissue culture.  相似文献

An oil palm experiment was set up in the Ivory Coast to compare the effects of crossing and selfing within two origins, Deli and La Mé, on the mean and the variability of Deli x between-origin hybrids. The originality of the experiment lay in the crossing plan, which provided access to genetic parameters related to additivity, dominance and different components of epistasis. This first part covers the analysis of the components of the mean. The parents used were obtained from four palms, two from each origin. Those of La Mé origin were half-sibs. The common parent came from a wild stand in the Ivory Coast. Those of Deli origin were from two different populations bred in Southeast Asia for several generations from a narrow genetic base. These four parents gave rise to nine Deli x La Mé hybrid populations with double-cousintype links. The additive component is more important within the Deli origin than within the La Mé origin. This may be explained by the large genetic divergence between the two Deli parent palms. On the other hand, the additive* additive epistasis is more substantial within the La Mé origin, probably because of inbreeding. The discussion concentrates on how this information should be used when choosing parents to be crossed and tested and to produce improved populations. The crossing plan proposed can be of general use and is suitable for other species in a reciprocal recurrent selection programme.  相似文献

Four dicotyledonous Astcraceac and four monocotyledonous Iridaceac species were grown from seed under three different effective ultraviolet-B (UV-B, 280–320 nm) fluenccs, which approximated those received daily over the natural growing periods of these species at their southerly and northerly distribution limits under ambient stratospheric ozone, and at the northerly limit assuming a 20% ozone depletion. Diminished photochemical efficiencies, delayed flowering, decreased flower production, pollen germination and tube growth, and reduced numbers of seeds set, accompanied by corresponding increases in mass per seed, were observed mainly in dicotyledons under elevated UV-B radiation, despite their higher foliar and pollen polyphenolic contents, their possession of trinucleatc pollen and their natural occurrence at lower latitudes than monocotyledons with binucleate pollen. The results demonstrate the difficulties in determining plant sensitivity to UV-B radiation from polyphenolic content, pollen type and natural geographic distribution, and suggest that monocotyledons possess other attributes that confer greater resilience to UV-B radiation.  相似文献

Abstract: Lateral fluid-percussion brain injury in rats results in cognitive deficits, motor dysfunction, and selective hippocampal cell loss. Neurotrophic factors have been shown to have potential therapeutic applications in neurodegenerative diseases, and nerve growth factor (NGF) has been shown to be neuroprotective in models of excitotoxicity. This study evaluated the neuroprotective efficacy of intracerebral NGF infusion after traumatic brain injury. Male Sprague-Dawley rats received lateral fluid-percussion brain injury of moderate severity (2.1–2.3 atm). A miniosmotic pump was implanted 24 h after injury to infuse NGF (n = 34) or vehicle (n = 16) directly into the region of maximal cortical injury. Infusions of NGF continued until the animal was killed at 72 h, 1 week, or 2 weeks after injury. Animals were evaluated for cognitive dysfunction (Morris Water Maze) and regional neuronal cell loss (Nissl staining) at each of the three time points. Animals surviving for 1 or 2 weeks were also evaluated for neurobehavioral motor function. Although an improvement in memory scores was not observed at 72 h after injury, animals receiving NGF infusions showed significantly improved memory scores when tested at 1 or 2 weeks after injury compared with injured animals receiving vehicle infusions ( p < 0.05). Motor scores and CA3 hippocampal cell loss were not significantly different in any group of NGF-treated animals when compared with controls. These data suggest that NGF administration, in the acute, posttraumatic period following fluid-percussion brain injury, may have potential in improving post-traumatic cognitive deficits.  相似文献

Nerve Growth Factor as a Mitogen for a Pancreatic Carcinoid Cell Line   总被引:1,自引:0,他引:1  
Abstract: Carcinoid tumors are a group of neuroendocrine neoplasms distributed widely throughout the body but most commonly occurring in the gut. These tumors retain many characteristics of their neural crest origin, including secretion of neuroactive peptides and responsiveness to neurotrophic substances. Nerve growth factor (NGF), a neurotrophic protein involved in maintenance and differentiation of peripheral sympathetic and sensory neurons, regulates growth of several neural tumor cells by inducing a differentiated phenotype and subsequent inhibition of cell growth rate. We examined the actions of NGF in a functioning human pancreatic carcinoid cell line (termed BON). NGF has no effect on the cytoarchitecture or constitutive secretion of bioamines in this carcinoid cell line. NGF, however, stimulates the in vitro cellular proliferation of BON cells. BON cells possess mRNA for the NGF receptors (p75LNGFR and p140trkA) and membrane-associated tyrosine kinase activity is increased in response to NGF. Both the mitogenic activity of NGF, as well as the receptor-linked tyrosine kinase activity, can be abrogated in BON cells by the trkA inhibitor K-252a and specific anti-NGF antibody. Our studies demonstrate that NGF is a mitogen for this carcinoid cell line without effect on cellular phenotype or cytoarchitecture. NGF may play a role in the development and progression of human carcinoid tumors.  相似文献

Abstract: Nerve growth factor (NGF) induces the synthesis and the phosphorylation of the orphan nuclear receptor NGFI-B in PC12 cells. Previous work has shown that phosphorylation, by protein kinase A, of a specific serine in the DNA-binding domain inhibits its binding to the NGFI-B response element. Also, cytoplasmic extracts from PC12 cells phosphorylate this serine, and phosphorylation is greater in extracts from cells treated with NGF. The present work describes the induction, identification, and partial purification of a kinase (termed NGFI-B kinase I) from PC12 cell extracts that catalyzes this phosphorylation. Phosphorylation of the DNA-binding domain with this purified preparation inhibits its binding to the NGFI-B response element. The kinase is rapidly activated by treatment of the cells with NGF, and the activation lasts for at least several hours. It also is activated by fibroblast growth factor and epidermal growth factor (EGF), but the activation by EGF is quite transient. The kinase requires Mg2+ but will use Mn2+. The molecular mass of the kinase is 95–100 kDa, and it is different from protein kinase A, Fos kinase, or pp90 rsk . Comparison with a partially purified preparation of cyclic AMP response element-binding protein kinase, however, indicates that the two are either very similar or identical.  相似文献

Abstract: SR 57746A {1-[2-(naphth-2-yl)ethyl]-4-(3-trifluoromethylphenyl)-1,2,5,6-tetrahydropyridine hydrochloride} exhibits neurotrophic activities in vivo and in vitro. We used the rat pheochromocytoma PC12 cell line to investigate in vitro cellular changes induced by SR 57746A. A significant increase in the percentage of cells bearing neurite-like processes was obtained in cells treated by SR 57746A and nerve growth factor (NGF) compared with NGF treatment alone. SR 57746A added alone, however, had no effect on morphogenesis or on survival of cells in serum-free medium. In contrast, SR 57746A induced a "priming" effect on PC12 cells for neurite outgrowth within 6 h of addition of the protein tyrosine kinase inhibitor genistein. An increase in α-actinin content resulted from treatment with SR 57746A. Expression of NGF-mediated acetylcholinesterase and choline acetyltransferase was enhanced within 5 days by SR 57746A. The molecule also induced rapid F-actin redistribution. Within 2 min of incubation, outgrowth of F-actin-containing filopodia was clearly visible at the cell periphery, as previously shown with NGF. It is interesting that this effect of SR 57746A could be mimicked by protein tyrosine kinase inhibitors and abolished by preincubation with sodium orthovanadate, a protein tyrosine phosphatase inhibitor.  相似文献
