An N2 strip gas was used in a packed bed sulfate-reducing bioreactor to recover the dissolved sulfide product and improve sulfate conversion. The highest volumetric productivity obtained was 261 mol H2S m−3 d−1. Lowering the initial pH of the medium from 7 to 6 increased the H2S content of the strip gas from 3.6 to 5.8 mol%. The ratio of strip gas to liquid flow rates (G/L) was found be to a suitable basis for scaling the process. Calculations indicated that modest G/L values (<102) were required to recover the residual dissolved sulfide in a downstream stripping column.  相似文献

This study identifies patterns of elephant Loxodonta africana africana impacts upon tree species and woody plant communities in Hluhluwe-Imfolozi Park, a South African savannahs/woodlands area. Elephants were reintroduced there from 1981, following more than 80 years of absence. Data were collected in 2003 on elephant impact on woodland in the Park. Different vegetation types were susceptible to different types and levels of damage by elephants, suggesting that elephants will not homogenize the vegetation. Elephants targeted larger stems for all types of damage, with a strong preference for some of the less abundant species such as Albizia versicolor (breaking and toppling) and Cordia caffra and Schotia brachypetala (debarking). Elephant impacts tended to be distributed evenly across the park landscape, irrespective of stem density or proximity to permanent water. Overall, elephants have little impact on slowing or reversing the spread of undesirable woody species, but are having a marked impact on certain less common tree species and larger tree size-classes in the Hluhluwe-Imfolozi Park.  相似文献

Bark stripping of hinoki cypress (Chamaecyparis obtusa (Sieb. et Zucc.) Endl.) by sika deer (Cervus nippon Temminck) was monitored for 4years on approximately 300 trees at three sites on Mt Takahara, central Japan. We investigated the effects of snow cover and food availability on seasonal and yearly changes in bark stripping. Bark stripping occurred during snowy periods (January and February) when less food was available because of the snow cover. The most serious bark stripping was observed in 1996 when the snow was the deepest and food resources were the poorest among the 4years of the study. In contrast, bark stripping did not occur in 1999 when the snow was thin and, consequently, more food was available. These results suggest that snow cover and food avail-ability influence the occurrence and intensity of bark stripping both seasonally and annually.  相似文献

Butanol is a precursor of many industrial chemicals, and a fuel that is more energetic, safer and easier to handle than ethanol. Fermentative biobutanol can be produced using renewable carbon sources such as agro-industrial residues and lignocellulosic biomass. Solventogenic clostridia are known as the most preeminent biobutanol producers. However, until now, solvent production through the fermentative routes is still not economically competitive compared to the petrochemical approaches, because the butanol is toxic to their own producer bacteria, and thus, the production capability is limited by the butanol tolerance of producing cells. In order to relieve butanol toxicity to the cells and improve the butanol production, many recovery strategies (either in situ or downstream of the fermentation) have been attempted by many researchers and varied success has been achieved. In this article, we summarize in situ recovery techniques that have been applied to butanol production through Clostridium fermentation, including liquid–liquid extraction, perstraction, reactive extraction, adsorption, pervaporation, vacuum fermentation, flash fermentation and gas stripping. We offer a prospective and an opinion about the past, present and the future of these techniques, such as the application of advanced membrane technology and use of recent extractants, including polymer solutions and ionic liquids, as well as the application of these techniques to assist the in situ synthesis of butanol derivatives.  相似文献

Feasibility of elemental sulfur reduction by Desulfovibrio desulfuricans in anaerobic conditions in a stirred reactor was studied. Hydrogen was used as energy source, whereas the carbonated species were bicarbonate and yeast extract. Attention was paid to reactor engineering aspects, biofilm formation on the sulfur surface, hydrogen sulfide formation rate and kinetics limitations of the sulfur reduction. D. desulfuricans formed stable biofilms on the sulfur surface. It was found that active sulfur surface availability limits the reaction rate. The reaction rate was first order with respect to sulfur and hydrogen velocity had no effect in the reaction rate for the range 8.2 x 10(-2) to 4.1 x 10(-1) Nm(3) m(-2) min(-1). At a superficial gas velocity (u(G)) = 3.1 x 10(-2) Nm(3) m(-2) min(-1), H(2)S(g) production rate decreased due to a deficient H(2)S stripping. A maximum H(2)S(g) production rate of 2.1 g H(2)S L(-1) d(-1) was achieved during 5 days with an initial sulfur density of 4.7% (w/v).  相似文献

Inconsistent results obtained with published methods for the elution of antibodies from tissue sections prompted the assessment of both old and new methods in combination with monoclonal rabbit antibodies of known, increased affinity (above 1×10-9 KD). We tested an acidic (pH 2) glycine buffer, a 6 M urea hot buffer and a 2-Mercaptoethanol, SDS buffer (2-ME/SDS). Some antibodies were not removed by the glycine pH 2 or 6 M urea hot buffers, indicating that antibodies survive much harsher conditions than previously believed. We found that the elution is dependent upon the antibody affinity and is reduced by species-specific crosslinking via a dimeric or Fab fragments of a secondary antibody. The high affinity bond of exogenous streptavidin with the endogenous biotin can be removed by 6 M urea but not by the other buffers. 2-ME/SDS buffer is superior to glycine pH 2 and 6 M urea hot elution buffers for all antibodies because of its irreversible effect on the structure of the antibodies. It also has a mild retrieving effect on some antigens present on routinely treated sections and no detrimental effect on the immunoreactivity of the tissue. Therefore, 2-ME/SDS buffer is the method of choice to perform multiple rounds of immunostaining on a single routine section.  相似文献

目的:探究改良曲张静脉点式剥除术在治疗中老年下肢静脉曲张的临床疗效。方法:收集我院已确诊为下肢静脉曲张的中老年患者37例,分成实验组与对照组。对照组18例行传统曲张静脉点式剥除术,实验组19例行改良曲张静脉点式剥除术。对比两组患者手术后的下肢静脉曲张的治疗效果。结果:实验组有效率(94.7%)显著高于对照组(72.2%),差异具有统计学意义(P0.05);与对照组相比,实验组患者手术时间较短、术中出血量较少、下床活动时间较早,术后并发症总治愈率较高,复发率、术后并发症发生率较低,其差异均有统计学意义(P0.05)。结论:采用改良曲张静脉点式剥除术治疗中老年下肢静脉曲张的患者能够更彻底的剥除曲张额静脉,有效的改善患肢症状,明显降低复发率。  相似文献

Drying of the tissue section, partial or total, during immunostaining negatively affects both the staining of tissue antigens and the ability to remove previously deposited antibody layers, particularly during sequential rounds of de-staining and re-staining for multiple antigens. The cause is a progressive loss of the protein-associated water up to the removal of the non-freezable water, a step which abolishes the immunoavailability of the epitope. In order to describe and prevent these adverse effects, we tested, among other substances, sugars, which are known to protect unicellular organisms from freezing and dehydration, and stabilize drugs and reagents in solid state form in medical devices. Disaccharides (lactose, sucrose) prevented the air drying-induced antigen masking and protected tissue-bound antigens and antibodies from air drying-induced damage. Complete removal of the bound antibody layers by chemical stripping was permitted if lactose was present during air drying. Lactose, sucrose and other disaccharides prevent air drying artifacts, allow homogeneous, consistent staining and the reuse of formalin-fixed, paraffin-embedded tissue sections for repeated immunostaining rounds by guaranteeing constant staining quality in suboptimal hydration conditions.  相似文献

Chronic neurodegenerative diseases of the CNS (central nervous system) are characterized by the loss of neurons. There is, however, growing evidence to show that an early stage of this process involves degeneration of presynaptic terminals prior to the loss of the cell body. Synaptic plasticity in CNS pathology has been associated with microglia and the phenomenon of synaptic stripping. We review here the evidence for the involvement of microglia in synaptic stripping and synapse degeneration and we conclude that this is a case of guilt by association. In disease models of chronic neurodegeneration, there is no evidence that microglia play an active role in either synaptic stripping or synapse degeneration, but the degeneration of the synapse and the envelopment of a degenerating terminal appears to be a neuron autonomous event. We highlight here some of the gaps in our understanding of synapse degeneration in chronic neurodegenerative disease.  相似文献

Summary The effects of several factors on the rate and the capacity of lead adsorption by river-mud were studies. These factors included the concentration of mud, the concentration of lead, the organic matter content of mud, pH, time of adsorption, temperature and agitation. The reaction order of the adsorption process with respect to lead and the type of adsorption were determined. Also, indications of the rate-limiting step were discussed and an empirical equation describing the adsorption of lead on river-mud was developed.  相似文献
