The generation of a species-rich DNA barcode database in combination with rapid and affordable sequencing techniques will dramatically change specimen identification in ecological, biogeographical and taxonomic applications. Though cytochrome c oxidase 1 has been shown to be a useful tool for differentiating some groups of marine algae, its wide application in the Phaeophyceae has yet to be studied. The presence of polymerase chain reaction (PCR) inhibiting compounds in members of the Fucales, Laminariales and Tilopteridales, that are often co-extracted with DNA, has hampered the rapid processing associated with barcode projects. Polyphenolics and polysaccharides are present in concentrations such that DNA extraction methods typically include extensive series of washes, organelle extractions and/or cesium columns. In this paper we examine the utility of cytochrome c oxidase 1 for barcoding the Phaeophyceae and present a method for extracting PCR friendly DNA from brown macroalgae in about 2 h, dramatically reducing the time required from previous methods, some of which take days. This method is easily adapted to a 96 well, high-throughput format and may have applications in other organisms where the presence of similar PCR inhibiting compounds hinders molecular analyses. We extracted DNA from 106 isolates representing 29 species from 20 genera in nine families from five orders of Phaeophyceae. We were able to amplify the barcode marker (cytochrome c oxidase 1) from all samples and a nuclear marker (internal transcribed spacer region) from 54 selected samples. Cytochrome c oxidase 1 was able to differentiate clearly among species, showing within species divergence of 0.00–0.46%, with the exception of one previously studied genus, and between species divergences of greater than 3%.  相似文献

The taxonomy and biogeography of a genus with species that occur in geographically isolated regions is interesting. The brown algal genus Ishige Yendo is a good example, with species that apparently inhabit warm regions of both the northwestern and northeastern Pacific Ocean. We determined the sequences of mitochondrial cox3 and plastid rbcL genes from specimens of the genus collected over its distributional range. Analyses of the 86 cox3 and 97 rbcL sequences resulted in congruent trees in which Ishige sinicola (Setch. et N. L. Gardner) Chihara consisted of two distinct clades: one comprising samples from Korea and Japan, and the other comprising samples from the Gulf of California. Additional observations of the morphology and anatomy of the specimens agree with the molecular data. On the basis of results, we reinstated Ishige foliacea S. Okamura (considered a synonym of I. sinicola from the Gulf of California) for plants from the northwest Pacific region and designated a specimen in the Yendo Herbarium (SAP) as the lectotype. I. foliacea is distinguished by large (up to 20 cm) and wide (up to 20 mm) thalli, with a cortex of 4–7 cells, and a medulla composed of long, tangled hyphal cells. Both cox3 and rbcL sequence data strongly support the sister‐area relationship between the northwest Pacific region and the Gulf of California. A likely explanation for this pattern would be the presence of a species ancestral to contemporary species of Ishige in both regions during the paleogeological period, with descendants later isolated by distance.  相似文献

A new species, Sargassum boreale Yoshida et Horiguchi is described. It belongs to the subgenus Bactrophycus section Teretia, with cylindrical receptacles and is distinct from Sargassum confusum C. Agardh, S. pallidum (Turner) C. Agardh and Sargassum microceratium (Turner) C. Agardh in having a rather elongated stem with smooth surface and distantly issuing main branches, with narrow leaves. The distinction between S. boreale and these species is also revealed by a difference in internal transcribed spacer 2 (ITS‐2) sequences. In addition to the base substitutions, the existence of a large gap in S. boreale distinguishes this species from others. Sargassum boreale is distributed around Hokkaido and Saghalien to 50°N latitude. A key to the species of section Teretia is provided.  相似文献

The kelp Laminaria digitata growing in the low intertidal region along energetic coastlines are exposed to a range of hydrodynamic environments. Macroalgae in the intertidal zone can experience both waves and currents independently, but it is unknown how they influence growth rate. Relative growth rate of the meristematic region and the entire blade of L. digitata were measured to assess the influence of wave and current motion from three hydrodynamic environments: low current and low wave (LCLW), high current and low wave (HCLW), and high wave and low current (HWLC). Alongside hydrodynamic data, seawater nutrient concentrations and temperature were also collected and analyzed. Results suggest that differences in L. digitata relative growth rates were not attributed to the seawater nutrient concentrations or temperature, but attributed to the hydrodynamic environments. At the high current condition, kelp growth rate of the meristematic region was enhanced by 45% compared to the high wave condition. When including the entire blade growth rate, an average increase of 25% was observed between the high current and high wave condition. Potentially, the division in growth rate observed between the wave and current motion is related to the frequency and magnitude at which the hydrodynamic forces act. These findings highlight the complexity of the hydrodynamic environment and that forces associated with waves and currents may have a significant role on the productivity of kelp.  相似文献

The life cycle of an organism is one of its most elemental features, underpinning a broad range of phenomena including developmental processes, reproductive fitness, mode of dispersal and adaptation to the local environment. Life cycle modification may have played an important role during the evolution of several eukaryotic groups, including the terrestrial plants. Brown algae are potentially interesting models to study life cycle evolution because this group exhibits a broad range of different life cycles. Currently, life cycle studies are focused on the emerging brown algal model Ectocarpus. Two life cycle mutants have been described in this species, both of which cause the sporophyte generation to exhibit gametophyte characteristics. The ouroboros mutation is particularly interesting because it induces complete conversion of the sporophyte generation into a functional, gamete-producing gametophyte, a class of mutation that has not been described so far in other systems. Analysis of Ectocarpus life cycle mutants is providing insights into several life-cycle-related processes including parthenogenesis, symmetric/asymmetric initial cell divisions and sex determination.  相似文献

sporophytes, and both male and female gametophytes of Dictyopteris undulata Holmes were collected from the Pacific coast and the Japan Sea coast of Japan. Female gametophytes are reported for the first time in the present study. All reproductive structures are aggregated in sori that form on both sides of the midrib in the upper and the middle parts of a blade. Tetraspor-angia are spherical and project from the cortical layer with a 2–4-celled stalk at the base. They are sometimes divided tetrahedrally to form four spores, but often remain undivided. Oogoniat sori also project from the cortical layer. Each oogonium is ovoid, sometimes borne on a single stalk cell at the base. Antheridial sori appear swollen above the cortex and each antheridium has a basal stalk cell. Fertile female gametophytes and te-trasporophytes are difficult to distinguish from each other in surface view because tetrasporangial and oogonial sori are similar in size and undivided tetrasporangia resemble oogonia. Oogonia are not so closely packed in small groups, unlike in Dictyota species, and they can be distinguished by the cell sizes (i.e. tetrasporangia are bigger in diameter than oogonia).  相似文献

The distribution of sterols in 14 Chilean Rhodophyceae and in Lessonia sp. (Phaeophyceae) has been examined. Cholesterol is present in all the red algae examined. Golidium filicinum contains, in addition desmosterol and 24-methylene-cholesterol. Cholest-7-en-3-β-ol was found in Cryptonemia angustate and Neoagardhiella gaudichaudii. Fucosterol and 24-methylenecholesterol were found in Lessonia sp.  相似文献
