甜菜夜蛾促前胸腺激素cDNA的克隆和表达   总被引:1,自引:0,他引:1  
利用兼并引物扩增出甜菜夜蛾(Spodoptera exigua, Spe)的促前胸腺激素(prothoracicotropic hormone, PTTH) cDNA, 克隆和测序结果显示出PTTH cDNA编码226个氨基酸的开放阅读框: 包括信号肽、1个功能未知的肽和PTTH. 和其他已知物种的PTTH比较, 甜菜夜蛾和夜蛾科昆虫有较高的同源性, 和蚕蛾科和大蚕蛾科昆虫的相似性稍低, 但是PTTH分子中的7个半胱氨酸残基的位置是非常保守的. 整体免疫组织化学检测发现PTTH表达在甜菜夜蛾脑中的2对神经分泌细胞中. Northern杂交表明在脑中有高丰度的PTTH mRNA存在, 分子大小约为1.2 kb. 半定量RT-PCR检测了PTTH在幼虫期和蛹期的发育变化, 显示出PTTH表达和蜕皮变态有密切的关系, 暗示PTTH在甜菜夜蛾行使其生物学功能很可能是通过刺激前胸腺合成和分泌蜕皮激素来完成的.  相似文献

Genes up-regulated during red coloration in UV-B irradiated lettuce leaves   总被引:4,自引:0,他引:4  
Molecular analysis of gene expression differences between green and red lettuce leaves was performed using the SSH method. BlastX comparisons of subtractive expressed sequence tags (ESTs) indicated that 7.6% of clones encoded enzymes involved in secondary metabolism. Such clones had a particularly high abundance of flavonoid-metabolism proteins (6.5%). Following SSH, 566 clones were rescreened for differential gene expression using dot-blot hybridization. Of these, 53 were found to overexpressed during red coloration. The up-regulated expression of six genes was confirmed by Northern blot analyses. The expression of chalcone synthase (CHS), flavanone 3-hydroxylase (F3H), and dihydroflavonol 4-reductase (DFR) genes showed a positive correlation with anthocyanin accumulation in UV-B-irradiated lettuce leaves; flavonoid 3′,5′-hydroxylase (F3′,5′H) and anthocyanidin synthase (ANS) were expressed continuously in both samples. These results indicated that the genes CHS, F3H, and DFR coincided with increases in anthocyanin accumulation during the red coloration of lettuce leaves. This study show a relationship between red coloration and the expression of up-regulated genes in lettuce. The subtractive cDNA library and EST database described in this study represent a valuable resource for further research for secondary metabolism in the vegetable crops.  相似文献

This article reflects on the relations between xenophobic parties and immigration policy in Italy. It argues that the salience of the Northern League (Lega Nord), the instability of the Italian party system and the recent transformation of Italy from an emigration country to an immigration country all contribute to Italy's difficulty in achieving a coherent immigration policy. Since the first comprehensive immigration policy emerged in 1998 with the Turco-Napolitano law, there have been six changes of government, with frequent oscillation between centre-right and centre-left coalitions. While the effects of these oscillations, particularly of the centre-right's effort to restrict immigrants’ rights and entry possibilities, have been dampened by the Italian courts and the European Union, it is still problematic to speak of an Italian model. Drawing on Carvalho's book, this article will address the effects of the Lega on creating and maintaining this policy imbalance.  相似文献

The investigation of the lithic assemblages from three archaeological structures of the Upper Palaeolithic Mezhyrich site, mammoth bone dwelling no. 1, nearby located pit no. 7 and workshop with a portion of cultural layer situated on the border of the pit, exhibits a sufficient diversity of the main indexes of flint assemblages. According to the latter, the main flint resources, cores and large pieces of flint, as well as blades and several tools (scrapers), were kept just in the dwelling. At the same time, the remains of “debitage” and the modification products of several tools (burin spalls) are more numerous in the pit and the workshop. The higher percentages of backed microliths, included the forms with diagnostic projectile impact fracture, and burins with several working edges are also observed in the assemblages of the last two archaeological structures.  相似文献

The new, monotypic camaenid genus Setocallosa is described for the new species S. pathutchingsae from Arnhem Land, Top End of the Northern Territory. This camaenid is characterized by a combination of distinct morphological features, such as a small, weakly elevated shell with widely open umbilicus and thick callus on the parietal wall, a wide epiphallic flagellum, elongated penis and short bursa copulatrix with subglobose end. The relationships of Setocallosa with the other camaenid genera in the area are investigated by means of mtDNA, but its phylogenetic position remains ambiguous. S. pathutchingsae occurs in sympatry with Arnhemtrachia ramingining, but based on morphology the two camaenids can be readily differentiated.

Changes in vegetative growing seasons are dominant indicators of the dynamic response of ecosystems to climate change. Therefore, knowledge of growing seasons over the past decades is essential to predict ecosystem changes. In this study, the long‐term changes in the growing seasons of temperate vegetation over the Northern Hemisphere were examined by analyzing satellite‐measured normalized difference vegetation index and reanalysis temperature during 1982–2008. Results showed that the length of the growing season (LOS) increased over the analysis period; however, the role of changes at the start of the growing season (SOS) and at the end of the growing season (EOS) differed depending on the time period. On a hemispheric scale, SOS advanced by 5.2 days in the early period (1982–1999) but advanced by only 0.2 days in the later period (2000–2008). EOS was delayed by 4.3 days in the early period, and it was further delayed by another 2.3 days in the later period. The difference between SOS and EOS in the later period was due to less warming during the preseason (January–April) before SOS compared with the magnitude of warming in the preseason (June–September) before EOS. At a regional scale, delayed EOS in later periods was shown. In North America, EOS was delayed by 8.1 days in the early period and delayed by another 1.3 days in the later period. In Europe, the delayed EOS by 8.2 days was more significant than the advanced SOS by 3.2 days in the later period. However, in East Asia, the overall increase in LOS during the early period was weakened in the later period. Admitting regional heterogeneity, changes in hemispheric features suggest that the longer‐lasting vegetation growth in recent decades can be attributed to extended leaf senescence in autumn rather than earlier spring leaf‐out.  相似文献

We used the ecosystem process model Biome‐BGC to simulate the effects of harvest and residue removal management scenarios on soil carbon (C), available soil nitrogen (N), net primary production (NPP), and net ecosystem production (NEP) in jack pine (Pinus banksiana Lamb.) and sugar maple (Acer saccharum Marsh) ecosystems in northern Wisconsin, USA. To assess harvest effects, we simulated short (50‐year) and long (100‐year) harvest intervals, high (clear‐cut) and low (selective) harvest intensities, and three levels of residue retention (15%, 25%, and 35%) over a 500‐year period. The model simulation of NPP, soil C accumulation, and NEP agreed reasonably well with biometric and eddy‐covariance measurements of these two ecosystems. The more intensive (50‐year rotation clear‐cuts with low residue retention) harvest scenarios tended to have the greatest NEP (420 and 678 t C ha?1 for the 500‐year interval for jack pine and sugar maple, respectively). All the harvest scenarios decreased mineral soil C and available mineral soil N content relative to the no‐harvest scenario for jack pine and sugar maple. The rate of change in mineral soil C decreased the greatest in the most intensive biomass removal scenarios (?0.012 and ?0.072 t C ha?1 yr?1 relative to no‐harvest for jack pine and sugar maple, respectively) and the smallest decrease was observed in the least intensive biomass removal scenarios (?0.002 and ?0.009 t C ha?1 yr?1 relative to no‐harvest for jack pine and sugar maple, respectively). The more intensive biomass removal harvest scenarios in sugar maple significantly decreased peak productivity (NPP) in the simulation period.  相似文献
