The aim of the study was to examine whether the difference in lactate concentration in different blood fractions is of practical importance when using blood lactate as a test variable of aerobic endurance capacity. Ten male firefighters performed submaximally graded exercise on a cycle ergometer for 20-25 min. Venous and capillary blood samples were taken every 5 min for determination of haematocrit and lactate concentrations in plasma, venous and capillary blood. At the same time, expired air was collected in Douglas bags for determination of the oxygen consumption. A lactate concentration of 4.0 mmol.l-1 was used as the reference value to compare the oxygen consumption and exercise intensity when different types of blood specimen and sampling sites were used for lactate analysis. At this concentration the exercise intensity was 17% lower (P less than 0.01) when plasma lactate was compared to venous blood lactate, and 12% lower (P less than 0.05) when capillary blood lactate was used. Similar discrepancies were seen in oxygen consumption. The results illustrated the importance of standardizing sampling and handling of blood specimens for lactate determination to enable direct comparisons to be made among results obtained in different studies.  相似文献

The influence of sampling and sample treatment upon adenylic nucleotide (ATP, ADP, AMP) content of microplankton is studied. Changes in light conditions during nigh-sampling and extracting do not induce significant variations, in the adenylic nucleotide content of microplankton or in energy charge values.The contribution of zooplankton (size up to 200 µm) to microplankton adenosine values can be neglected for inshore surface waters and traditional sample volumes (about one liter). This result can been explained by the low density of zooplankton in such a small sample volume and by differences in efficiency of the extracting method used.

The abundances, biomass, and seasonal succession of rotifer and crustacean zooplankton were examined in a man-made, eutrophic lake, Lake Oglethorpe, over a 13 month period. There was an inverse correlation between the abundance of rotifers and crustaceans. Rotifers were most abundant and dominated (>69%) the rotifer-crustacean biomass during summer months (June–September) while crustacean zooplankton dominated during the remainder of the year (>89%). Peak biomasses of crustaceans were observed in the fall (151 µg dry wt l–1 in October) and spring (89.66 µg dry wt l–1 in May). Mean annual biomass levels were 46.99 µg dry wt l–1 for crustaceans and 19.26 µg dry wt l–1 for rotifers. Trichocerca rousseleti, Polyarthra sp., Keratella cochlearis and Kellicottia bostoniensis were the most abundant rotifers in the lake. Diaptomus siciloides and Daphnia parvula were the most abundant crustaceans. Lake Oglethorpe is distinct in having an unusually high abundance of rotifers (range 217–7980 l–1). These high densities can be attributed not only to the eutrophic conditions of the lake but also to the detailed sampling methods employed in this study.The research was supported by National Science Foundation grants DEB 7725354 and DEB 8005582 to Dr. K. Airborne particles and ammonia were monitored in horse stalls managed under four conditions. Two ventilation rates, high (27 air changes per h) and low (5 air changes per h) and two bedding types, paper and straw, were employed. At both ventilation rates, the number of airborne particles generated while the stalls were mucked out was higher with straw than with paper. Particles were more efficiently cleared at the higher ventilation rate in both the straw and paper stalls. Ammonia measurements reflected an accumulation over time. In the stalls with low ventilation, ammonia levels were significantly higher than in those stalls with high ventilation regardless of bedding type. Management decisions and their relationships to respiratory disorders are discussed.  相似文献

Abstract. The Institute of Terrestrial Ecology (ITE) has classified the 1 km squares in Great Britain (GB) into thirty-two environmental strata, termed land classes, as a basis for ecological survey. The classes have been used in biogeographical studies of the distribution of individual species and species assemblages. The concept behind the technique is that there is an association between the environmental character of land and ecological parameters. The initial classification was based on a sample of squares drawn from a regular grid. The data for the 12121 km squares classified were drawn from published maps; the number of squares was limited by the available computing power. Subsequently the availability of more powerful computers and the need to improve geographical definition have led to the allocation of every 1 km square to its appropriate class. This paper has been written to summarise the principles involved in the development of the system and indicate the range of projects for which it has been used. The extension of the classification from a sample to the complete coverage of GB revealed the importance of the structure and style of data used to produce the classification. The significance of these conclusions for future work is discussed, with particular reference to automated methods of data capture.  相似文献

The traditional sampling method for estimating frequency (the number of sub-quadrats containing a basal part of the organisms) is compared, using both computer simulations and direct comparison in the field, to two new methods that use a compound series of variable-sized concentric sub-quadrats. Both the new frequency-score and the new importance-score methods are closer approximations of density than is the standard frequency method, and the estimates produced by both of the new methods are less affected by the choice of sub-quadrat size and the spatial distribution (dispersion) of the organisms (i.e. clumping and regularity). Thus, the two nested-quadrat methods appear to ameliorate the usual frequency limitations associated with sub-quadrat size and organism dispersion, by the use of a range of different sub-quadrat sizes. This is important in community studies, where the component species may show a wide range of densities and dispersions. Both of the new methods are easily employed in the field. The importance-score method involves no more sampling effort than does standard qualitative (presence-absence) sampling, and it can therefore be used to sample a larger quadrat area than would normally be used for frequency sampling. This makes the method much more cost-effective as a means of estimating abundance, and it allows a greater number of the rarer species to be included in the sampling. The frequency-score method is more time-consuming, but it is capable of detecting more subtle community patterns. This means that it is particularly useful for the study of species-poor communities or where small variations in composition need to be detected.  相似文献
