A role for nitrogen reserves in forage regrowth and stress tolerance 总被引:20,自引:0,他引:20
Carbohydrate accumulation and utilization during shoot regrowth after defoliation and winter has been studied extensively in most species used as forage. However, recent work suggests that N reserves found in vegetative tissues also are important for defoliation tolerance and winter hardiness. Results suggest that these N reserves constitute an alternative N source used when N2 fixation and/or mineral N uptake are reduced. 15 N labelling experiments indicate that a large proportion of herbage N is derived from N reserves mobilized from stem bases or roots to developing leaves and shoots. Amino acids and specific proteins (i.e. vegetative storage proteins, VSPs) are deposited in roots and stem bases and, in the case of VSPs, are degraded rapidly after defoliation. Identification and characterization of VSPs will increase our understanding of the role N reserves play in stress tolerance and may lead to innovative approaches for improving forage persistence and productivity. 相似文献

Time courses of formation of inositol 1,4,5-trisphosphate (IP3 ) were followed in the leaves of non-acclimated and cold (2°C)-acclimated winter oilseed rape ( Brassica napus L. var. oleifera ) plants, subjected to different freezing temperatures or to polyethylene glycol 8000 (PEG) and abscisic acid (ABA) treatments. Changes in water potential (Ψw ) and in ABA level in the frost- and PEG-treated tissues were also determined. Results obtained indicate that temperatures sligthly higher than LT50 induced a transient and substantial increase in IP3 level, both in non-acclimated and cold-acclimated tissues. At comparable freezing temperature (–5°C) the response of cold-acclimated leaves was lower than that of non-acclimated ones. The PEG-depedent decrease in Ψw to –0.9 MPa or ABA (0.1 m M ) treatment gave rise to a transient increase in IP3 content in non-acclimated tissues only. Collectively, the data indicate that cold acclimation of plants may lead to lower cell responsiveness to the factors studied in terms of induction of IP3 formation. Changes in the IP3 content, observed in the present experiments, support our previous suggestion that non-killing freezing temperatures may induce the phosphoinositide pathway, both in non-acclimated and cold-acclimated tissues. Lowering of tissue water potential to some threshold value or a high exogenous ABA supply may mimic the freezing-dependent reaction in the non-acclimated leaves. 相似文献

The differentiation of phloem anastomoses linking the longitudinal vascular bundles has been studied in stem internodes of Cucurbita maxima Duchesne, C. pepo L. and Dahlia pinnata Cav. These anastomoses comprise naturally occurring regenerative sieve tubes which redifferentiate from interfascicular parenchyma cells in the young internodes. In all three species, severing a vascular bundle in a young internode resulted in regeneration of xylem to form a curved by-pass immediately around the wound. The numerous phloem anastomoses in these young internodes were not involved in this process, the regenerated vessels originating from interfascicular parenchyma alone. Conversely, in mature internodes of Dahlia, the regenerated vessels originated from initials of the interfascicular cambia, and their phloem anastomoses did not influence the pattern of xylogenesis. On the other hand, in old internodes of Cucurbita, in which an interfascicular cambium was not yet developed, the parenchyma cells between the bundles had lost the ability to redifferentiate into vessel elements, and instead, regenerated vessels were produced in the phloem anastomoses. Thus, the wounded region of the vascular bundle was not bypassed via the shortest, curved pathway, but by more circuitous routes further away from the wound. Some of the regenerated vessels produced in the phloem anastomoses were extremely wide, and presumably efficient conductors of water. Variation in base water potential (Ψb, a daily maximum level of plant water potential, which is presumed to correspond to the condition of equilibrium between
the soil and plant water potentials) was examined in shoots of Norway spruce trees growing in well-drained and waterlogged
soils. The influence of soil water content, air temperature, and vapour pressure deficit of the atmosphere on Ψb was studied using the pressure chamber technique. Maximum daily water potentials were not always observable before dawn;
some were registered up to two hours later. This tendency being characteristic of trees growing under stress (shade, waterlogging)
conditions, increased with declining soil water availability. In trees growing in well-drained soil, Ψb depended asymptotically on the available soil water storage (R2=0.73), while the values were slightly influenced by vapour pressure deficit of the atmosphere as well. In trees growing in

The effect of NH
4
+
on the regulation of NO
3
–
and NO

–
transport systems in roots of intact barley (Hordeum vulgareL.) seedlings grown in NO
3
–
or NO
2
–
was studied. Ammonium partially inhibited induction of both transport systems. The inhibition was less severe in NO
2
–
-fed than in NO
3
–
-fed seedlings, presumably due to lower uptake of NH
4
+
in the presence of NO
2
–
. In seedlings pretreated with NH
4
+
subsequent induction was inhibited only when NH
4
+
was also present during induction, even though pretreated roots accumulated high levels of NH
4
+
. This indicates that inhibition may be regulated by NH
4
+
concentration in the cytoplasm rather than its total accumulation in roots. L-Methionine sulfoximine did not relieve the inhibition by NH
4
+
, suggesting that inhibition is caused by NH
4
+
itself rather than by its assimilation product(s). Ammonium inhibited subsequent expression of NO
3
–
transport activity similarly in roots grown in 0.1, 1.0, or 10 mM NO
3
–
for 24 h (steady-state phase) or 4 d (decline phase), indicating that it has a direct, rather than general feedback effect. Induction of the NO
3
–
transport system was about twice as sensitive to NH
4
+
as compared to the NO
2
–
transport system. This may relate to higher turnover rates of membraneassociated NO
3

Hydraulic and osmotic properties were measured along developing maize (Zea mays L.) roots at distances between 15 and 465 mm from the root tip to quantify the effects of changes in root structure on the radial and longitudinal movement of water and solutes (ions). Root development generated regions of different hydraulic and osmotic properties. Close to the root tip, passive solute permeability (root permeability coefficient, Psr) was high and selectivity (root reflection coefficient, sr) low, indicative of an imperfect semipermeable root structure. Within the apical 100–150 mm, Psr decreased by an order of magnitude and sr increased significantly. Root hydraulic conductivity (Lpr) depended on the nature of the force (hydrostatic and osmotic). Osmotic Lpr was smaller by an order of magnitude than hydrostatic Lpr and decreased with increasing distance from the root tip. Throughout the root, responses in turgor of cortical cells and late metaxylem to step changes in xylem pressure applied to the base of excised roots were measured at high spatial resolution. The resulting profiles of radial and longitudinal propagation of pressure showed that the endodermis had become the major hydraulic barrier in older parts of the root, i.e. at distances from the apex ä 150 mm. Other than at the endodermis, no significant radial hydraulic resistance could be detected. The fate of most nonhuman primate species is intimately related to man. The increasing encroachment on the natural habitat
has resulted in the decimation and even near extinction of some species. Along with this development, the basic concept in
many modern zoos has changed from one of merely display to self-perpetuating units. Primate research facilities are orienting
their research programs towards reproductive physiology and behavior in an effort to provide basic knowledge of reproduction
in these species. This increased emphasis in the area of reproductive biology and the various efforts to improve breeding
of these mostly endangered primates in captivity has stimulated the author to write this review. It represents an attempt
to provide the reader with basic background information relating to the endocrinology and behavior of reproduction in the
clawed New World monkeys as it exists at the time of publication.
The intermediate evolutionary position ofCallimico goeldii between the clawed New World monkeys and the ‘true New World monkeys’ and our relatively poor knowledge about reproduction
and behavior in this particular species fully justifies the focus on Goeldi’s monkey in this essay. This review is an attempt
to provide a brief history of previous studies but also the basis for research in the future. The current status of knowledge
of the small-bodied clawed monkeys is also discussed in an evolutionary context, with an emphasis on the different reproductive
strategies in this dynamic group of primates.

Slow wave potentials (SWPs) are transient depolarizations which propagate substantial distances from their point of origin. They were induced in the epidermal cells of pea epicotyls by injurious methods such as root excision and heat treatment, as well as by externally applied defined steps in xylem pressure (Px) in the absence of wounding. The common principle of induction was a rapid increase in Px. Such a stimulus appeared under natural conditions after (i) bending of the epicotyl, (ii) wounding of the epidermis, (iii) rewatering of dehydrated roots, and (iv) embolism. The induced depolarization was not associated with a change in cell input resistance. This result and the ineffectiveness of ion channel blockers point to H(+)-pumps rather than ion channels as the ionic basis of the SWP. Stimuli such as excision, heat treatment and pressure steps, which generate SWPs, caused a transient increase in the fluorescence intensity of epicotyls loaded with the pH-indicator DM-NERF, a 2',7'-dimethyl derivative of rhodol, but not of those loaded with the pH indicator 2',7'bis(2-carboxyethyl)-5-(and-6)-carboxyfluorescein (BCECF). Matching kinetics of depolarization and pH response identify a transient inactivation of proton pumps in the plasma membrane as the causal mechanism of the SWP. Feeding pump inhibitors to the cut surface of excised epicotyls failed to chemically simulate a SWP; cyanide, azide and 2,4-dinitrophenol caused sustained, local depolarizations which did not propagate. Of all tested substances, only sodium cholate caused a transient and propagating depolarization whose arrival in the growing region of the epicotyl coincided with a transient growth rate reduction. 相似文献

The production of antimicrobial phytoalexins is one of the best-known inducible defence responses following microbial infection of plants or treatment with elicitors. In the legume soybean (Glycine max L.), 1,3-1,6--glucans derived from the fungal pathogen Phytophthora sojae have been identified as potent elicitors of the synthesis of the phytoalexin, glyceollin. Recently it has been reported that during symbiotic interaction between soybean and the nitrogen-fixing bacterium Bradyrhizobium japonicum USDA 110 the bacteria synthesize cyclic 1,3-1,6--glucans. Here we demonstrate that both the fungal and the bacterial -glucans are ligands of -glucan-binding sites which are putative receptors for the elicitor signal compounds in soybean roots. Whereas the fungal -glucans stimulate phytoalexin synthesis at low concentrations, the bacterial cyclic 1,3-1,6--glucans appear to be inactive even at relatively high concentrations. Competition studies indicate that increasing concentrations of the bacterial 1,3-1,6--glucans progressively inhibit stimulation of phytoalexin synthesis in a bioassay induced by the fungal 1,3-1,6--glucans. Another type of cyclic -glucan, a 1,2--glucan from Rhizobium meliloti, that does not nodulate on soybean, seems to be inactive as elicitor and as ligand of the -glucan-binding sites. These results may indicate a novel mechanism for a successful plant-symbiont interaction by suppressing the plant's defence response.Abbreviations HG-APEA

This study examined the effects of pituitary-interrenal hormones on catecholamine storage and release in the rainbow trout Oncorhynchus mykiss. An extract of trout pituitary elicited the release of adrenaline, but not noradrenaline, using an in situ perfusion preparation. A variety of doses of adrenocorticotropic hormone (2–2000 mU) caused the release of both catecholamines in situ which was unaffected by pre-treatment with the ganglion blocker, hexamethonium, or the serotonergic receptor antagonist, methysergide, but was abolished in calcium-free media. Intra-arterial injections of adrenocorticotrophic hormone in vivo caused an elevation of plasma adrenaline but not noradrenaline levels. Injections of cortisol in situ did not elicit catecholamine release. Trout given an intraperitoneal implant of cortisol (50 mg·kg-1 body weight) had significantly higher plasma cortisol concentrations when compared to controls after 7 days of implantation. Increases in the levels of stored catecholamines were observed in various regions of the kidney and posterior cardinal vein following 3 and 7 days of cortisol treatment. The ability of the chromaffin cells to release catecholamines in response to cholinergic stimulation was assessed in situ after 7 days of treatment. Basal (non-stimulated) adrenaline outflowing perfusate levels were greater in the cortisol-treated fish. Cortisol treatment increased the responsiveness of the catecholamine release process to low doses of the cholinoceptor agonist carbachol. Three or 7 days of cortisol treatment did not alter the in vitro activity of the enzyme phenylethanolamine-N-methyl-transferase. The results of this study demonstrate that interactions within the pituitary-adrenal axis can influence both catecholamine storage and release in the rainbow trout.Abbreviations
