The site of integration of phage Mu d (Ap lac) in mutant M9s which leads to deficiency of formic dehydrogenase (benzylviologen-linked) activity was determined. It was shown that the phage had inserted into the gene for the seleno-polypeptide of the enzyme (80 kd) leading to the formation of a truncated peptide (60 kd) still able to incorporate Se. Synthesis of the truncated polypeptide is subject to the same regulatory signals as that of the wild-type enzyme. The formation of the 110 kd seleno-polypeptide, which is a constituent component of the formic dehydrogenase from the formate-nitrate respiratory pathway, is unimpaired in mutant M9s. The location of the gene for the 80 kd seleno-polypeptide was mapped at 92.4 min of the Escherichia coli chromosome.Abbreviation PMSF phenylmethylsulfonyl fluoride Dedicated to Professor G. Summary The Cy transposable element system is composed of two genetically defined elements: an rcy receptor element inserted at the Bronze-1 locus; and an independently segregating regulatory element, Cy. The Cy system is not functionally homologous to any of the non-Mutator transposable element systems. Evidence is presented that supports a relationship between the Cy system and the family of Mu1-homologous transposable elements that are responsible for the Mutator phenomenon. Although related, Cy elements and the Mu1-homologous elements are not identical; Cy is inherited in a near-Mendelian fashion, in contrast to the non-Mendelian inheritance of the Mu1-homologous elements.  相似文献

The 663 amino acid Mu transposase protein is absolutely required for Mu DNA transposition. Mutant proteins were constructed in vitro in order to locate regions of transposase that may be important for the catalysis of DNA transposition. Deletions in the A gene, which encodes the transposase, yielded two stable mutant proteins that aid in defining the end-specific DNA-binding domain. Linker insertion mutagenesis at eight sites in the Mu A gene generated two proteins, FF6 and FF14 (resulting from two and four amino acid insertions, respectively, at position 408), which were thermolabile for DNA binding in vitro at 43°C. However, transposition activity in vivo was severely reduced for all mutant proteins at 37°C, except those with insertions at positions 328 and 624. In addition, site-specific mutagenesis was performed to alter tyrosine 414, which is situated in a region that displays amino acid homology to the active sites of a number of nicking/closing enzymes. Tyrosine 414 may reside within an important, yet non-essential, site of transposase, as an aspartate-substituted protein had a drastically reduced frequency of transposition, while the remaining mutants yielded reduced, but substantial, frequencies of Mu transposition in vivo.  相似文献

Abstract In nature, bacteria encounter a variety of environmental conditions, among the most frequent of these is the limitation or starvation of one or more essential nutrients. It is reasonable to assume, therefore, that bacteria have evolved mechanisms to enhance their survival over prolonged periods of nutrient starvation. We have identified eight genetic loci in the enteropathogen Salmonella typhimurium , using Mu d -directed lacZ operon fusion technology, that were induced in response to two or more different starvation conditions (sti) . In simultaneous studies, we also identified genetic loci, using Mu d-lac fusions, which respond to only phosphate-starvation conditions (psi) . We further characterized these loci as to their induction-characteristics, kinetics of induction, expression during growth on different carbon sources, and approximate location on the S. typhimurium genetic map. In concurrent studies, we analyzed whole cell extracts of S. typhimurium grown under a variety of nutrients oxydation conditions as well as under nonlimiting conditions, using two-dimensional polyacrylamide gel electrophoresis. Results from these studies correlated well with our gene fusion studies. In more recent studies, we have demonstrated a complex genetic regulation of a number of these starvation-inducible loci, and have implicated at least four of these loci in the long term starvation-survival of S. typhimurium .  相似文献

Abstract The ability of P-type plasmids, containing bacteriophage Mu, to survive in Rhizobia was examined under conditions in which the plasmids were not directly selected. The plasmid pRP4::Mu::Tn7 survived in all tested strains of Rhizobia and Agrobacteria . The surviving plasmids were analysed for expression of plasmid coded properties and for presence of both bacteriophage Mu and Tn7 DNA. A large variation in plasmid size and coding capacity was observed. In the parent plasmid Tn7 is inserted into the Mu DNA. The majority of surviving plasmids had lost Mu DNA but the Tn7 DNA was retained. Analysis of surviving plasmids which contained Mu DNA indicated that a small deletion of Mu is sufficient to allow plasmid survival. Deleted derivatives of pJB4J1 were also isolated in Rhizobium under similar conditions. The potential of these suicide vectors as potential vectors for transposon mutagenesis in Rhizobium is discussed.  相似文献

 本文旨在将毛乌素沙地植被建设的水分平衡与半固定沙丘持续发展原则应用于治沙造林的实践中。毛乌素沙地是一个灌木“王国”,然而沙地灌丛植被的发育常常受到水分亏缺的严重制约。为此,根据水分平衡的原则与方法确立适宜的植物种植密度,对沙地植被的经营管理具有重要的指导意义。在水分平衡研究中,蒸散是最难估计的一项。本文提供了一种根据叶面积指数的季节变化与蒸腾速率的观测资料计算蒸腾耗水量的方法,并根据沙地水分平衡的要求估算丁几种优势灌木的适宜种植密度。结果表明,毛乌素沙地灌丛生态系统的蒸发散主要来自植物蒸腾作用;在所研究的植物当中,除沙地柏(Sabina vulgaris)可以形成很大的密度外(因其强的蒸腾控制能力),其它灌木的适宜种植密度应控制在使沙丘处于半固定状态的水平上。  相似文献

选择毛乌素沙地优势灌丛柠条(Caragana intermedia)、紫穗槐(Amorpha fruticosa)和沙柳(Salix psammophila)作为保育植物,选择2年生油松(Pinus tabulaeformis)和樟子松(Pinus sylvestris)幼苗作为目标植物,将其分别种植在开阔地、柠条、紫穗槐和沙柳4种微生境下,连续3a监测油松和樟子松幼苗的存活和生长动态,从而确定毛乌素沙地优势灌丛是否对植物幼苗的定居有促进作用。结果表明:柠条、紫穗槐和沙柳冠层下光照强度、大气温度和土壤温度都显著低于开阔地,致使土壤湿度增高,油松幼苗总存活率均显著高于开阔地,而樟子松幼苗总存活率仅紫穗槐冠层下显著高于开阔地(P0.05)。紫穗槐冠层下油松和樟子松幼苗总存活率均最高(P0.05),外来种紫穗槐可选为保育植物。保育灌丛下松树幼苗的主枝生长良好,并未受到抑制,但第1年主枝生长长度最小。光照强度和土壤湿度是决定主枝生长长度的重要因素。保育灌丛高度和冠幅面积与油松和樟子松幼苗存活率呈显著正相关(P0.05)。固氮灌丛(紫穗槐和柠条)对松树幼苗存活有持续的促进作用,但柠条冠幅结构紧密,可能因为对光的竞争,减弱了促进作用。沙柳灌丛下松树幼苗的存活率较低。油松幼苗具有一定程度的耐阴性,樟子松幼苗喜光性强,保育灌丛下油松幼苗存活率显著高于樟子松幼苗(P0.05)。总的来说,保育灌丛对松树幼苗的定居有促进作用,保育植物技术可作为一种有效的恢复措施,应用在毛乌素沙地的植被恢复中来。但是,保育灌丛的选择要考虑是本土种或外来种,以及灌丛的高度、冠幅大小、冠幅结构和化学效应,恢复物种的选择要考虑其自身的生物学特性及其与保育植物的相互作用。  相似文献

Assembly of the functional tetrameric form of Mu transposase (MuA protein) at the two att ends of Mu depends on interaction of MuA with multiple att and enhancer sites on supercoiled DNA, and is stimulated by MuB protein. The N-terminal domain I of MuA harbours distinct regions for interaction with the att ends and enhancer; the C-terminal domain III contains separate regions essential for tetramer assembly and interaction with MuB protein (IIIα and IIIβ, respectively). Although the central domain II (the ‘DDE’ domain) of MuA harbours the known catalytic DDE residues, a 26 amino acid peptide within IIIα also has a non-specific DNA binding and nuclease activity which has been implicated in catalysis. One model proposes that active sites for Mu transposition are assembled by sharing structural/catalytic residues between domains II and III present on separate MuA monomers within the MuA tetramer. We have used substrates with altered att sites and mixtures of MuA proteins with either wild-type or altered att DNA binding specificities, to create tetrameric arrangements wherein specific MuA subunits are nonfunctional in II, IIIα or IIIβ domains. From the ability of these oriented tetramers to carry out DNA cleavage and strand transfer we conclude that domain IIIα or IIIβ function is not unique to a specific subunit within the tetramer, indicative of a structural rather than a catalytic function for domain III in Mu transposition.  相似文献

通过西北-东南和东北-西南方向两条宽5m×长3 km的样带和一块4m×4m的样方调查,研究了浑善达克沙地无芒雀麦、植被、土壤水分及土壤盐分的空间变异特点及其相互关系。结果表明,样带内植被盖度、无芒雀麦盖度、土壤水分和土壤盐分的空间变化格局相似,然而,在不同的空间方向上,它们具有不同的空间变化;样方内无芒雀麦地上部生物量、分株数和土壤盐分具有相似的空间变化格局,但却不同于植被地上部总生物量和土壤水分的空间变化格局。相关分析表明,样带内植被盖度、无芒雀麦盖度与土壤水分和土壤盐分之间均具有显著的正相关关系。土壤水分和土壤盐分也具有显著的正相关。样方内无芒雀麦地上部生物量、分株数及植被地上部总生物量与土壤水分均具有显著的正相关关系,但与土壤盐分没有显著的相关关系。无芒雀麦地上部生物量、分株数和植被地上部总生物量三者之间,以及土壤水分和土壤盐分之间也均具有显著的正相关关系。研究结果表明了植被与土壤之间的相互关系依赖于空间尺度。  相似文献

行为规癖 (古板行为 )是母猪一种常见的异常行为 ,表现为重复、不变化的行为模式 ,并且不带有明显的目的性。Mu阿片受体 (Muopioidreceptor,简称MOR)属于G蛋白偶联受体 ,分布在痛觉传导区以及与情绪和行为有关的区域 ,影响动物的神经反应和行为表现。本研究以Mu阿片受体基因作为候选基因 ,探讨影响母猪规癖性状的可能性。根据Mu阿片受体基因外显子Ⅲ的序列设计引物 ,用PCR -SSCP的方法对大白猪、长白猪和杜洛克猪进行单核苷酸多态性分析 ,发现该位点存在多态性。对两种纯合子片段克隆并测序表明 ,mRNA第 1169处存在C→T的单碱基突变 ,在第 12 2 6处存在C→A的单碱基突变 ,均为沉默突变。统计结果发现 3种基因型 (AA ,AB ,BB)在各品种中的分布不一致 ,χ2 独立性检验差异极显著 (P <0 0 1)。将大白猪 3种基因型同行为规癖性状进行最小二乘分析 ,结果表明 ,BB基因型与其他 2种基因型相比有较高的无食咀嚼表现频率 ,同AA型比较差异极显著 (P <0 0 1) ,咬栏和站立基因型间差异不显著。因此 ,推测Mu阿片受体基因可能是影响母猪无食咀嚼性状的主效基因或与控制该性状的主效基因连锁  相似文献
