Summary The fate of ascending projections of thoracic interneurons in the metamorphosing brain of Tenebrio molitor is described. Persistent brain neurons were identified and their fate is described during metamorphosis. The projection sites of ascending elements are invariable throughout metamorphosis both in quantitative and in qualitative terms. Some of these ascending neurons are serotonin-immunoreactive and this set of neurons maintains a constant projection site within the metamorphosing brain. The alterations in the projection sites of these and other ascending neurons in the ventral nerve cord were analysed experimentally. The central projection sites of these persistent ascending neurons are not important for the maintenance of their nerve cord projections throughout metamorphosis. Experimental deletion of ascending neurons which project into the suboesophageal ganglion varies the shape of persistent central neurons.  相似文献

Summary A single-gene recessive mutation, bcd (broadened cortical domains), of Tetrahymena thermophila is characterized by a variable broadening of the spatial domains within which cortical organelles, including both the contractile vacuole pores (CVP) and oral apparatus (OA), are formed. The phenotype is not temperature-sensitive. During the development of the organelles of the mutant prior to cell division, extra CVPs and extra oral primordia (OP) appear near ciliary rows adjacent to the rows at which these structures normally form. In the later stages of development, some, but not all, of these extra structures are resorbed, or in the case of the oral domain, multiple adjacent OPs may be completely or partially integrated into a single enlarged OA. When multiple OAs persist, one or more of these may display a reversed orientation reminiscent of those encountered in janus mutants. However, unlike janus, bcd cells do not express any sign of a mirror-image global organization.Our results can best be accounted for by postulating that the bcd mutation affects some common determinant of the widths of both CVP and OA domains. Studies are in progress which explore the relationship between this width-determining mechanism(s) and the mechanism(s) determining the location of cortical organelles around the cell circumference.  相似文献

The physiological relationships between Ostrinia nubilalis Hübner and its tachinid parasitoid Pseudoperichaeta nigrolineata Walker are described under abiotic conditions which induce development of the host without diapause. The parasitoid lowers the larval growth of the host: the maximal weight attained by the parasitized larvae represented only 78% of that of healthy ones. The duration of the last larval host instar increased to 10.4 days in parasitized O. nubilalis compared to 8.0 days in unparasitized ones. The influence of the host on the parasitoid development was studied experimentally after parasitization of O. nubilalis larvae of instars 2 to 5. When the second larval instar of the host is parasitized, the overall duration of parasitoid larval development lasts twice as long as when the fifth instar is parasitized. The best yield of parasitoid pupariae (50%) is obtained when parasitization occurs in instar 3. We show that good synchronisation exists between the larval development of the host and its parasitoid. There are four phases of parasitoid development which would appear to require a signal from the host: the start of the growth of newly hatched parasitoid larvae and the 3rd to 4th instar ecdysis of the host; the first moulting of the parasitoid and the 4th to 5th instar ecdysis of the host; the growth resumption of the parasitoid instar II (weight about 1 mg) and the small rise of the ecdysteroid level in the middle of host instar 5; and in all probability, the second parasitoid moulting and the larval-pupal apolysis of the host.
Résumé Les relations physiologiques entre O. nubilalis et le tachinidae P. nigrolineata ont été étudiées dans des conditions abiotiques telles que l'hôte présente un développement sans diapause. Le parasitoïde ralentit la prise de poids de l'hôte: le poids maximal des chenilles parasitées ne représente que 78% de celui des chenilles saines. Seule la durée du 5ème stade est significativement plus longue chez les chenilles parasitées que chez les saines (10,4 contre 8,0 jours). L'influence de l'hôte sur le développement du parasitoïde à été expérimentée en parasitant des stades 2 à 5 d'O. nubilalis. Lorsque l'infestation a lieu au stade 2, le développement larvaire complet du parasitoî de dure deux fois plus longtemps que lorsque l'infestation a lieu au stade 5. Le meilleur rendement en pupes (50%) est obtenu lorsque l'infestation a lieu au stade 3. Il a été montré qu'il existe une bonne synchronisation entre le développement de l'hôte et de son parasitoî de. Il y a 4 phases physiologiques du développement larvaire de P. nigrolineata qui semblent nécessiter un signal provenant de l'hôte pour être dépassées. Ainsi peuvent être mis en relation: — le début de la croissance de la larve néonate du parasitoî de et l'ecdysis 3/4 de l'hôte; — la première mue du parasitoïde et l'ecdysis 4/5 de l'hôte; — la reprise de la croissance du stade II du parasitoïde, vers un poids de 1 mg et la remontée des taux d'ecdystéroïdes au milieu du stade 5 de l'hôte. et probablement, — la seconde mue du parasitoïde et l'apolyse nymphale de l'hôte. Les expérimentations vont se poursuivre pour déterminer les facteurs en cause. Ces phénomènes de synchronisation seront aussi étudiés dans le cas de la diapause de l'hôte.

Laboratory tests with eggs of Diabrotica virgifera virgifera LeConte showed that during a 10-day hatching period, hatch of male eggs predominated on the first and second days, eggs of mixed sex, with ca. 1:1 ratio, hatched on the third and fourth days, and eggs hatching from the fifth to the tenth days were nearly all female. Overall, female eggs hatched a mean of 2.9 days later than male eggs. Not only did female eggs hatch later, but the time for posthatch development to the adult stage was 1.8 days longer for females. The later egg hatch and longer posthatch development for females resulted in female adults emerging a mean of 4.7 days later than male adults. Total adult emergence lasted 14 days; of this, males predominated during the first 5 days, and females predominated during the last 9 days. Males of D. v. virgifera appear to have evolved protandry (the tendency for males to emerge before females) by developing both a postdiapause embryonic stage and a combined larval and pupal stage of shorter duration.
Résumé L'observation, au laboratoire, pendant une période d'éclosion de dix jours, des oeufs de D. v. virgifera LeConte, a montré que les oeufs mâles prédominent les deux premiers jours d'éclosion, que les oeufs des deux sexes, avec des fréquences 0,5/0,5, ont éclos les troisième et quatrième jours, et que les oeufs éclos du cinquième au dixième jour étaient presque tous femelles. Globalement, les oeufs femelles ont éclos en moyenne 2,9 jours plus tard que les oeufs mâles. De plus, la durée du développement post-embryonnaire des femelles a demandé 1,8 jour en plus. Une éclosion plus tardive et un développement post-embryonnaire plus long ont entrainé une émergence des femelles en moyenne 4,7 jours après les mâles. La période d'émergence des adultes s'est étalée sur 14 jours; les mâles ayant dominé pendant les 5 premiers jours et les femelles pendant les 9 derniers. Les mâles de D. v. virgifera semblent avoir évolué vers la protandrie en acquerant, tant une diapause post-embryonnaire que des stades de développements larvaire et nymphal plus brefs.

A calcium requirement was shown for both vesicle development and nitrogenase activity by Frankia strains EAN1pec and CpI1. Washing cells with EGTA or EDTA inhibited both vesicle development and nitrogenase activity. The inhibition of both was reversed by the addition of calcium. A variety of agents known to affect calcium-dependent biological processes, such as a Ca-ATPase inhibitor, Ca-channel blockers, Ca-ionophores, calmodulin antagonists and the local anaesthetics, tetracaine and dibucaine, inhibited nitrogenase activity. Respiratory studies showed that a CN-insensitive respiration process occurred only under nitrogen derepressing conditions. Respiration by NH4Cl-grown cells was completely inhibited by KCN while N2-grown cells were inhibited by only 70%. Removal of calcium ions by EGTA or by the addition of dibucaine or tetracaine blocked the CN-insensitive respiration. The relationship between nitrogen fixation and development of a specialized cell structure, called the vesicle, was studied using four Frankia isolates. Nitrogenase activity was repressed in all four strains during growth with ammonia. Strain CpI1 formed no vesicles during NH4 growth. Strains ACN1 ag , EAN1pec and EUN1f produced low numbers of vesicles in the presence of ammonia. Following transfer to nitrogen-free media, a parallel increase in nitrogenase activity and vesicle numbers occurred with all four isolates. Appearance of nitrogenase activity was more rapid in those strains that possessed some vesicles at the time of shift to N2 as a nitrogen source. The ratio of vesicle numbers to level of nitrogenase activity varied widely among the four strains and in response to different growth conditions and culture age of the individual strains. Optimum conditions of temperature, carbon and energy source, nitrogen source and availability of iron and molybdenum were different for each of the four strains. Those conditions that significantly reduced nitrogenase activity were always associated with decreased numbers of vesicles.  相似文献

Summary Little is known about the effects of enriched CO2 environments, which are anticipated to exist in the next century, on natural plant-insect herbivore interactions. To begin to understand such effects on insect growth and survival, I reared both early and penultimate instar larvae of the buckeye, Junonia coenia (Lepidoptera: Nymphalidae), on leaves from one of their major hostplants, plantain, Plantago lanceolata (Plantaginaceae), grown in either ambient (350 PPM) or high (700 PPM) CO2 atmospheres. Despite consuming more foliage, early instar larvae experienced reduced growth on high CO2-grown compared to ambient CO2-grown leaves. However, survivorship of early instar larvae was unaffected by the CO2 treatment. Larval weight gain was positively correlated with the nitrogen concentration of the plant material and consumption was negatively correlated with foliar nitrogen concentration, whereas neither larval weight gain nor consumption were significantly correlated with foliar water or allelochemical concentrations. In contrast, penultimate instar larvae had similar growth rates on ambient and high CO2-grown leaves. Significantly higher consumption rates on high CO2-grown plants enabled penultimate instar larvae to obtain similar amounts of nitrogen in both treatments. These larvae grew at similar rates on foliage from the two CO2 treatments, despite a reduced efficiency of conversion of ingested food (ECI) on the low nitrogen, high CO2-grown plants. However, nitrogen utilization efficiencies (NUE) were unaffected by CO2 treatment. Again, for late instar larvae, consumption rates were negatively correlated with foliar nitrogen concentrations, and ECI was also very highly correlated with leaf nitrogen; foliar water or allelochemical concentrations did not affect either of these parameters. Differences in growth responses of early and late instar larvae to lower nitrogen, high-CO2 grown foliage may be due to the inability of early instar larvae to efficiently process the increased flow of food through the gut caused by additional consumption of high CO2 foliage.  相似文献

The ability of leaves to acclimate photosynthetically to low temperature was examined during leaf development in winter rye plants ( Secale cereale L. cv. Puma) grown at 20°C or at 6°C. All leaves grown at 6°C exhibit increased chlorophyll (Chl) levels per leaf area, higher rates of uncoupled, light-saturated photosystem I (PSI) electron transport, and slower increases in photosystem II (PSII) electron transport capacity, when compared with 20°C leaves. The stoiehiometry of PSI and PSII was estimated for each leaf age class by quantifying Chl in elcctrophorctic separations of Chl-protein complexes. The ratio of PSII/PSI electron transport in 20°C leaves is highly correlated with the ratio of core Chl a -proteins associated with PSII (CPa) to those associated with PSI (CP1). In contrast, PSII/PSI electron transport in 6°C leaves is not as well correlated with CPa/CP1 and is related, in part, to the amount and organization of light-harvesting Chl a/b -proteins associated with PSII. CPa/CP1 increases slowly in 6°C leaves, although the ratio of CPa/CP1 in mature 20°C and 6°C leaves is not different. The results suggest that increased PSI activity at low temperature is not related to an increase in the relative proportion of PSI and may reflect, instead, a regulatory change. Photosynthetic acclimation to low environmental temperature involves increased PSI activity in mature leaves shifted to 6°C. In leaves grown entirely at 6°C, however, acclimation includes both increased PSI activity and modifications in the rate of accumlation of PSII and in the organization of LHCII.  相似文献

The gas exchange properties of whole plant canopies are an integral part of crop productivity and have attracted much attention in recent years. However, insufficient information exists on the coordination of transpiration and CO2 uptake for individual leaves during the growing season. Single-leaf determinations of net photosynthesis (Pn), transpiration (E) and water use efficiency (WUE) for field-grown cotton (Gossypium hirsutum L.) leaves were recorded during a 2-year field study. Measurements were made at 3 to 4 day intervals on the main-stem and first three sympodial leaves at main-stem node 10 from their unfolding through senescence. Results indicated that all gas exchange parameters changed with individual main-stem and sympodial leaf age. Values of Pn, E and WUE followed a rise and fall pattern with maximum rates achieved at a leaf age of 18 to 20 days. While no significant position effects were observed for Pn, main-stem and sympodial leaves did differ in E and WUE particularly as leaves aged beyond 40 days. For a given leaf age, the main-stem leaf had a significantly lower WUE than the three sympodial leaves. WUE's for the main-stem and three sympodial leaves between the ages of 41 to 50 days were 0.85, 1.30, 1.36 and 1.95 μmol CO2 mmol−1 H2O, respectively. The mechanisms which mediated leaf positional differences for WUE were not strictly related to changes in stomatal conductance (gs·H2O) since decreases in gs·H2O with leaf age were similar for the four leaves. However, significantly different radiant environments with distance along the fruiting branch did indicate the possible involvement of mutual leaf shading in determining WUE. The significance of these findings are presented in relation to light competition within the plant canopy during development.  相似文献

When pea (Pisum sativum L.) embryos were cultured on low osmotica, with or without added abscisic acid (ABA), there was very little change in the total mRNA translation products resolved by one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). The only marked alteration was an increase in production of two low-molecular-weight proteins. The purification and partial characterisation of these two ABA-responsive seed proteins (ABR17 and ABR18) is described. Both proteins were purified to homoeneity, as judged by SDS-PAGE, from embryos cultured in the presence of ABA. Antisera were raised against both proteins. Each serum cross-reacted with the other protein, indicating that the proteins are closely related. Their apparent molecular masses (Mrs) were estimated to be 17200 (ABR17) and 18100 (ABR18) by SDS-PAGE, and 26000 by gel filtration. Both proteins were heterogeneous on isoelectric focusing. Neither protein was detected (by immunoblotting or immunoprecipitation of cell-free translation products) in embryos grown in vivo at early to mid-development stages but both were present in embryos late in development. These proteins appear to be produced late in seed development but are capable of being induced early in development by culturing embryos in vitro and are markedly enhanced by ABA.  相似文献
