Imazaquin, imazethapyr and pendimethalin showed high toxicity to sorghum plants grown in a greenhouse soil mix. However, mycorrhizal sorghum plants were less affected by herbicide toxicity than non-mycorrhizal ones, at low to moderate herbicide concentrations. VAM herbicide safening effects were more evident on imazaquin-treated plants, than for those treated with the other two herbicides. Applications of imazethapyr and pendimethalin at the two highest concentrations, but not imazaquin, reduced VAM colonization rates in sorghum. Applications of the VAM stimulating isoflavonoids, biochanin A and formononetin, at 5 ppm solutions to a field soil sample containing toxic levels of imazaquin (13 ppb) and indigenous VAM fungi, reduced herbicide-induced injury in corn and sorghum under growth chamber conditions. The benefits of isoflavonoids were reduced when additional propagules of Glomus intraradix were added into field-soil samples, and were eliminated when VAM fungi were inactivated by autoclaving. This indicates that herbicide safening effects of biochanin A, and formononetin are VAM-mediated and also suggests the potential use of these isoflavonoids as herbicide safeners.  相似文献

The time courses of some Photosystem II (PS II) parameters have been monitored during in-vivo and in-vitro photoinhibition of spinach chloroplasts, at room temperature and at 10 °C or 0 °C. Exposing leaf discs of low-light grown spinach at 25 °C to high light led to photoinhibition of chloroplasts in-vivo as manifested by a parallel decrease in the number of functional PS II centres, the variable chlorophyll fluorescence at 77K (F v /F m ), and the number of atrazine-binding sites. When the photoinhibitory treatment was given at 10 °C, the former two parameters declined in parallel but the loss of atrazine-binding sites occurred more slowly and to a lesser extent. During in-vitro photoinhibition of chloroplast thylakoids at 25 °C, the loss of functional PS II centres proceeded slightly more rapidly than the loss of atrazine-binding sites, and this difference in rate was further increased when the thylakoids were photoinhibited at 0 °C. During the recovery phase of leaf discs (up to 9 h) the increases in F v /F m preceded that of the number of functional PS II centres, while only a further decline in the number of atrazine-binding sites was observed. The recovery of variable chlorophyll fluorescence and the concentration of functional PS II centres occurred more rapidly at 25 °C than at 10 °C. Summary A urea and NPK-mixture at concentration of 5, 10 and 15 mg g–1 air dry litter stimulated microbial populations, microbial activity and rate of decomposition of the litter. The stimulation was more pronounced as the concentration of the fertilizers was increased. However, this trend was reverse after two months in case of urea except for bacterial population. Fewer fungal species were isolated from the fertilizer-treated litter, together with a certain degree of alteration in the composition of mycoflora.  相似文献

(Benzamidooxy)acetic acid (common name benzadox) which has herbicidal properties was evaluated as a potential inhibitor of photosynthesis in C4 plants. Among enzymes of the C4 pathway, it was a relatively strong inhibitor of alanine aminotransferase in in vitro experiments at concentrations of 5mM. In benzadox treated leaves of Panicum miliaceum, a NAD-malic enzyme type C4 species, there was strong inhibition of both alanine and aspartate aminotransferase and of photosynthetic O2 evolution within one hour. Consistent with the inhibition of these enzymes of the C4 cycle, the pool sizes of metabolites of the cycle was altered: the aspartate level was increased two fold, while the levels of other metabolites such as pyruvate, alanine, oxalacetate and malate were decreased. Kinetic studies with partially purified alanine aminotransferase showed that benzadox is a competitive inhibitor with respect to alanine and a noncompetitive inhibitor with respect to 2-oxoglutarate. Comparisons between the structures and inhibitory actions of benzadox and (aminooxy)acetic acid, the latter a potent inhibitor of alanine and aspartate aminotransferases, suggest that in vivo, benzadox may exert its effect through metabolism to (aminooxy)acetic acid.Abbreviations benzadox (benzamidooxy)acetic acid - DTE dithioerythritol This research was supported in part by gift funds from Monsanto Agricultural Products Company. The uptake of the auxin type herbicide 2,4-D into rice seedlings ( Oryza sativa L. cv. Dunghan Shali) and its effects on the K+, NH+4 and NO3 ion uptake and the K+ content were investigated at different pH values. A short incubation of the roots in 0.01 m M 2,4-D caused a marked ion uptake inhibition only at low pH. The non-auxin type herbicide benthiocarb did not produce such an inhibitory effect. Lowering of the pH in the external medium led to an increased 2,4-D uptake by the roots. These results can be explained by the increased H+ permeability of the membranes, allowing a more rapid entrance of 2,4-D into the root cells, thereby inhibiting the active ion uptake. Rice roots not subjected to 2,4-D treatment responded to H+ stress with an increased anomalous K+ uptake and a decreased K+ content. With reference to the effects of pH changes on the ion and 2,4-D uptake, possible transport mechanism of NH+4 and 2,4-D are briefly discussed.  相似文献

Photosystem II particles which retained high rates of herbicide-sensitive activity were used to examine the site(s) of action of various herbicides. A polypeptide of 32–34 kdaltons was identified as the triazine-herbicide binding site based upon: (a) parallel loss of atrazine activity and the polypeptide during either trypsin treatment or selective detergent depletion of protein in the Photosystem II complex, and (b) covalent labeling of the polypeptide by a 14C-labeled photoaffinity triazine.In Photosystem II particles depleted of the 32–34-kdalton polypeptide, electron transport was still active and was slightly sensitive to DCMU and largely sensitive to dinoseb (urea and nitrophenol herbicides, respectively). On the basis of this result it is proposed that the general herbicide binding site common to atrazine, DCMU and dinoseb is formed by a minimum of two polypeptides which determine affinity and/or mediate herbicide-induced inhibition of electron transport on the acceptor side of Photosystem II.  相似文献

A DCMU* (diuron)-resistant algal mutant was selected and characterized. Chlorophyll content, growth, and photosystem-I activity are as in the wild-type. Growth in liquid medium with 3 M DCMU present is half of the control. Apparently only the herbicide-binding site is affected within the redox chain. In contrast to the wild-type, trypsin treatment of isolated chloroplast material completely abolishes photosynthetic electron transport inhibition by DCMU or atrazine.DCMU resistance of chloroplasts is accompanies by tolerance to triazinones and phenylpyridazinones, but not to symmetric triazines. Sensitivity to diphenylethers, DBMIB or o-phenanthroline is not altered.Data on this algal mutant combined with those from triazine-resistant mutants of higher plants give direct evidence of overlapping binding sites at a (hypothetical) binding protein located at the reducing side of photosytem II.  相似文献

A modified chlorophyll fluorescence technique was evaluated as a rapid diagnostic test of the susceptibility of wheat cultivars to chlorotoluron. Two winter wheat cultivars (Maris Huntsman and Mercia) exhibited differential response to the herbicide. All of the parameters of chlorophyll fluorescence examined were strongly influenced by herbicide concentration. Additionally, the procedure adopted here for the examination of winter wheat cultivar sensitivity to herbicide indicated that the area above the fluorescence induction curve and the ratio FV/FM are appropriate chlorophyll fluorescence parameters for detection of differential herbicide response between wheat cultivars. The potential use of this technique as an alternative to traditional methods of screening new winter wheat cultivars for their response to photosynthetic inhibitor herbicide is demonstrated here.  相似文献

The binding characteristics and the inhibitory power of atrazine and DCMU towards uncoupled electron flow activity were studied in acyl lipid-depleted thylakoid membranes from atrazine-susceptible and-resistant biotypes of Solanum nigrum L. For this purpose, phospholipase A2 from Vipera russelli and the lipase from Rhizopus arrhizus were used to obtain a selective lipid class (phospholipids or galactolipids) depletion which was restricted to the outer monolayer. Neither phospholipid nor galactolipid removal affected the dissociation constant and the number of binding sites of atrazine. In contrast, the dissociation constant of DCMU was increased in phospholipid-depleted thylakoid membranes but remained unchanged after galactolipid depletion. The number of DCMU binding sites decreased significantly after both lipase treatments, but only in the resistant biotype. The inhibitory effectiveness of the herbicide was either decreased or increased (to different extents) depending on the lipid class which was removed from the membrane and on the biotype considered.
