The effect of mechanical agitation on the microalgae Phaeodactylum tricornutum and Porphyridium cruentum was investigated in aerated continuous cultures with and without the added shear protectant Pluronic F68. Damage to cells was quantified through a decrease in the steady state concentration of the biomass in the photobioreactor. For a given aeration rate, the steady state biomass concentration rose with increasing rate of mechanical agitation until an upper limit on agitation speed was reached. This maximum tolerable agitation speed depended on the microalgal species. Further increase in agitation speed caused a decline in the steady state concentration of the biomass. An impeller tip speed of >1.56 m s–1 damaged P. tricornutum in aerated culture. In contrast, the damage threshold tip speed for P. cruentum was between 2.45 and 2.89 m s–1. Mechanical agitation was not the direct cause of cell damage. Damage occurred because of the rupture of small gas bubbles at the surface of the culture, but mechanical agitation was instrumental in generating the bubbles that ultimately damaged the cells. Pluronic F68 protected the cells against damage and increased the steady state concentration of the biomass relative to operation without the additive. The protective effect of Pluronic was concentration-dependent over the concentration range of 0.01–0.10% w/v.  相似文献

Light supply is one of the most important factors affecting autotrophic growth of microalgae. This study investigated the effect of the type and light intensity of artificial light sources on the cell growth of an indigenous microalga Chlorella vulgaris ESP‐31 obtained from southern Taiwan. In addition, a dissolved inorganic carbon source (i.e. sodium bicarbonate) was used to improve the biomass production of strain ESP‐31. The results show that a new fluorescent light source (TL5) was effective in indoor cultivation of microalgae. Better overall productivity of 0.029 g dry cell weight/L‐d was obtained when using TL5 lamps as the light source with a light intensity of 9 W/m2. A carbon source (sodium bicarbonate) concentration of 1000 mg/L was found to be optimal for the growth of strain ESP‐31 in terms of both biomass production and carbon source utilization. Under the optimal growth conditions, the resulting microalgal biomass consisted of 25–30% protein, 6–10% carbohydrate, and 30–40% lipid.  相似文献

Ethanol use as a fuel additive or directly as a fuel source has grown in popularity due to governmental regulations and in some cases economic incentives based on environmental concerns as well as a desire to reduce oil dependency. As a consequence, several countries are interested in developing their internal market for use of this biofuel. Currently, almost all bio-ethanol is produced from grain or sugarcane. However, as this kind of feedstock is essentially food, other efficient and economically viable technologies for ethanol production have been evaluated. This article reviews some current and promising technologies for ethanol production considering aspects related to the raw materials, processes, and engineered strains development. The main producer and consumer nations and future perspectives for the ethanol market are also presented. Finally, technological trends to expand this market are discussed focusing on promising strategies like the use of microalgae and continuous systems with immobilized cells.  相似文献

The growth characteristics of microalgae under different light conditions (continuous or intermittent) are essential information for photobioreactor design and operation. In this study, we constructed a thin-layer (10 mm) flat plate photobioreactor device with a light/dark (L/D) alternation system to investigate the growth of Spirulina platensis under two different light regimes: (1) continuous illumination in a wide range of light intensities (1.00-77.16 mW cm−2); (2) intermittent illumination in medium frequency (0.01-20 Hz). Specific growth rate and light efficiency based on biomass production were determined for each round of experiment. Four regions (light limited region, intermediate region, light saturated region and light inhibition region) were recognized according to the results under continuous illumination. Under intermittent illumination, when L/D frequency increased from 0.01 Hz to 20 Hz, specific growth rate and light efficiency were enhanced. However, the enhancement was different, depending on the applied light intensity and light fraction. The higher the light intensity, the greater the enhancement would be when L/D frequency increased from 0.01 Hz to 20 Hz; and the higher the light intensity, the lower the light fractions is needed to maintain light efficiency as high as that under continuous illumination in light limited region.  相似文献

Algae are currently used for production of niche products and are becoming increasingly interesting for the production of bulk commodities, such as biodiesel. For the production of these goods to become economically feasible, production costs will have to be lowered by one order of magnitude. The application of two-phase systems could be used to lower production costs. These systems circumvent the costly step of cell harvesting, whilst the product is extracted and prepared for downstream processing. The mechanism of extraction is a fundamental aspect of the practical question whether two-phase systems can be applied for in situ extraction, viz, simultaneous growth, product formation and extraction, or as a separate downstream processing step. Three possible mechanisms are discussed; 1) product excretion 2) cell permeabilization, and 3) cell death. It was shown that in the case of product excretion, the application of two-phase systems for in situ extraction can be very valuable. With permeabilization and cell death, in situ extraction is not ideal, but the application of two-phase systems as downstream extraction steps can be part of a well-designed biorefinery process. In this way, processing costs can be decreased while the product is mildly and selectively extracted.Thus far none of the algal strains used in two-phase systems have been shown to excrete their product; the output has always been the result of cell death. Two-phase systems can be a good approach as a downstream processing step for these species. For future applications of two-phase in situ extraction in algal production processes, either new species that show product excretion should be discovered, or existing species should be modified to induce product excretion.  相似文献

Environmental changes that have occurred due to the use of fossil fuels have driven the search for alternative sources that have a lower environmental impact. First-generation biofuels were derived from crops such as sugar cane, corn and soybean, which contribute to water scarcity and deforestation. Second-generation biofuels originated from lignocellulose agriculture and forest residues, however these needed large areas of land that could be used for food production. Based on technology projections, the third generation of biofuels will be derived from microalgae. Microalgae are considered to be an alternative energy source without the drawbacks of the first- and second-generation biofuels. Depending upon the growing conditions, microalgae can produce biocompounds that are easily converted into biofuels. The biofuels from microalgae are an alternative that can keep the development of human activity in harmony with the environment. This investigation examines the possible results of policy support in developed and developing economies for developing algal biodiesel through to 2040. This investigation adopts the Taiwan General Equilibrium Model-Energy for Bio-fuels (TAIGEM-EB) to predict competition among the development of algal biodiesel, bioethanol and conventional crop-based biodiesel. Analytical results show that algal biodiesel will not be the major energy source in 2040 without strong support in developed economies. In contrast, bioethanol enjoys a development advantage relative to both forms of biodiesel. Finally, algal biodiesel will almost completely replace conventional biodiesel. CO2 reduction benefits the development of the bio-fuels industry.  相似文献

Population outburst together with increased motorization has led to an overwhelming increase in the demand for fuel. In the milieu of economical and environmental concern, algae capable of accumulating high starch/cellulose can serve as an excellent alternative to food crops for bioethanol production, a green fuel for sustainable future. Certain species of algae can produce ethanol during dark-anaerobic fermentation and thus serve as a direct source for ethanol production. Of late, oleaginous microalgae generate high starch/cellulose biomass waste after oil extraction, which can be hydrolyzed to generate sugary syrup to be used as substrate for ethanol production. Macroalgae are also harnessed as renewable source of biomass intended for ethanol production. Currently there are very few studies on this issue, and intense research is required in future in this area for efficient utilization of algal biomass and their industrial wastes to produce environmentally friendly fuel bioethanol.  相似文献

High production cost is a major obstacle to the extensive use of microalgae biodiesel. To cut the cost and achieve higher biomass productivity, Chlorella minutissima UTEX2341 was cultured under photoheterotrophic conditions. With the carbon, nitrogen and phosphorus concentration of 26.37, 2.61 and 0.03 g L?1 d?1 respectively, a maximum biomass productivity of 1.78 g L?1 d?1 was obtained, which was 59 times more than that cultured under autotrophic condition. The lipid productivity reached 0.29 g L?1 d?1, which was 11.9 times higher than the highest value reported by Oh et al. (2010). The conversion rate of microalgae lipids to FAME was found to be elevated from 45.65% to 62.97% and the FAME productivity increased from 1.16 to 180.68 mg L?1 d?1 after the optimization. 94% of the fatty acid of C. minutissima UTEX2341 was found to be composed of palmitic, oleic, linoleic and γ linoleic and the unsaturated fatty acids were the main parts (79.42%).  相似文献

Microalgae have the ability to grow rapidly, synthesize and accumulate large amounts (approximately 20-50% of dry weight) of lipids. A successful and economically viable algae based oil industry depends on the selection of appropriate algal strains. In this study ten species of microalgae were prospected to determine their suitability for oil production: Chaetoceros gracilis, Chaetoceros mulleri, Chlorella vulgaris, Dunaliella sp., Isochrysis sp., Nannochloropsis oculata, Tetraselmis sp., Tetraselmis chui, Tetraselmis tetrathele and Thalassiosira weissflogii. The study was carried out in 3 L glass flasks subjected to constant aeration and controlled artificial illumination and temperature at two different salinities. After harvesting, the extraction of oil was carried out using the Bligh and Dyer method assisted by ultrasound. Results showed that C. gracilis presented the highest oil content and that C. vulgaris presented the highest oil production.  相似文献
