Summary Streptomycin-resistant Pseudomonas and Arthrobacter were isolated from semi-arid grassland soil and their relative responses in the rhizosphere of blue grama (Bouteloua gracilis) subjected to herbage removal were evaluated. Using plants grown in normal soil, the two bacteria showed differential responses to herbage removal, which were most marked in the rhizoplane, where the Pseudomonas showed a two-log unit increase over a 60 hour period, while Arthrobacter, in contrast, exhibited a one-log unit decrease in viable counts for at least 48 hours after defoliation, responses which are similar to those observed in root exudate medium experiments by earlier workers. These results suggest that the rhizoplane may be a critical environment for interaction of these two types of microorganisms, and that sequential responses of the root-associated soil microorganisms may occur after herbage removal from this important rangeland plant. These responses are most likely associated with increased exudate release following herbage removal, which has been best documented using blue grama grown under sterile conditions. 相似文献

Urban and periurban forests, which are integrated within the concept of Green Infrastructure, provide important Ecosystem Services, including air purification. In this study, we quantified the Ecosystem Service of particulate matter (PM10) and Ozone (O3) removal from urban and periurban forests in ten metropolitan cities in Italy, and its total monetary value. In order to gain a better understanding of how Ecosystem Services can be regulated on a wider scale, the vegetation ecosystem types were grouped into Physiognomic-Structural Categories of Vegetation according to morphofunctional criteria. The pollution removal was mapped using a remote sensing and GIS approach, by applying a deposition model and a stomatal flux model. We estimated, for the ten metropolitan cities, an overall pollution abatement of 7150 Mg of PM10 and 30,014 Mg of O3 in the year 2003, which was an extremely hot year. Our findings indicate that structural characteristics (i.e. Leaf Area Index) and functional diversity, linked to stomatal conductance, exert a marked influence on the provision of the regulating Ecosystem Services, whose total monetary value was estimated to be equal to 47 and 297 million USD for PM10 and O3 removal, respectively. This study represent the first national-scale assessment of the Ecosystem Services of air pollution removal in Europe, thus providing information that may be useful to stakeholders to manage Green Infrastructure more efficiently. 相似文献

Abstract An instrument has been built which reproducibly exerts measured amounts of force in an up and down rubbing motion to bean plant stems (Phaseolus vulgaris L. cv. Cherokee Wax). The thigmostimulator has, for the first time, permitted an accurate determination of the relationship between the amount of mechanical stimulus or stress (MS) and the resulting elongation of the plant (E). Over the stress range used, the analyzed relationship is best expressed by the equation: E = m In MS + b, where m is the slope of the line and b is the E-intercept. The sensory function is saturated at very low forces (e.g. 2 rubs at 3.59 N). Reciprocity between force and the number of stimuli holds only at low forces and numbers of stimuli (e.g. 12 rubs at 0.97 N to four rubs at 2.92 N). Analysis using linear regression and differential calculus reveals that for five 1-cm rubs, the sensory system is saturated at 6 N and that there is a threshold of 0.3 N below which no response occurs. Using the thigmostimulator for calibration, two 1-cm rubs given by hand are equivalent to a force of 4.41 ± 0.60 N. The nitrate-removal activity of a biofilm attached to a perlite carrier from an aerobic bioreactor used for treating dairy farm wastewater was examined by batch experiments under continuous aeration conditions. Despite aeration, the biofilm removed nitrate at a rate of 114.4 mg-N/kg-perlite/h from wastewater containing cow milk and manure. A simple and efficient tool to isolate epithelial cells from bacteria-contaminated samples has been developed using two different microparticles functionalized with chemical molecules. The epithelial cells could be captured simply by biocompatible anchors for membranes (BAM), consisting of poly(ethylene glycol) functionalized with oleyl-chain-conjugated NHS (N-hydroxysuccinimide) on glass microparticles, whereas bacteria were adsorbed on 3-aminopropyltrimethoxysilane (ATPS)-functionalized magnetic microparticles. In the case of samples highly contaminated with bacteria, epithelial cells were not isolated successfully by both of the single BAM- and antibody-functionalized microparticles. Therefore, serial isolation steps of these two different chemical functionalized microparticles were introduced. The concentration of bacteria was decreased dramatically by using APTS-functionalized magnetic particles prior to the isolation of epithelial cells by BAM microparticles. With these serial processes, successful isolation of epithelial cells was achieved from bacteria-contaminated epithelial samples. High throughput covalent urease immobilization was performed through the amide bond formation between the urease and the amino-functional MNPs. The enzyme’s performances, including shelf-life, reusability, enzymatic kinetics, and the enzyme relative activity in organic media was improved. At optimal conditions, the immobilization efficiency was calculated about 95.0% with keeping 94.7% of the urease initial specific activity. The optimal pH for maximum activity of the free and immobilized urease was calculated as 7.0 at 37.0 °C and 8.0 at 60.0 °C, respectively. The kinetics studies showed the Km of 26.0 mM and 8.0 mM and the Vmax of 5.31 μmol mg−1 min−1 and 3.93 μmol mg−1 min−1 for the free and immobilized urease, respectively. The ratio Kcat/Km as a measure of catalytic efficiency and enzyme specificity was calculated as 0.09 mg mL−1 min−1 and 0.22 mg mL−1 min−1 for the free and immobilized urease, respectively, indicating an improvement in the enzymatic kinetics. The shelf-life and operational studies of immobilized urease indicated that approximately 97.7% and 88.5% of its initial activity was retained after 40 days and 17 operational cycles, respectively. The immobilized urease was utilized to urea removal from water samples with an efficiency between 91.5–95.0%. 相似文献
