Rabbit antisera were produced against a lymphokine-activated human macrophage cell line, U937 (αU937), and human peritoneal macrophages (αPEMØ). After absorption with AB erythrocytes, pooled platelets, and B-lymphoblastoid cell lines, both antisera reacted by microcytotoxicity, indirect immunofluorescence (IF), and radioimmunoassay (RIA) with adherence-purified human peripheral blood monocytes, splenic and peritoneal macrophages, and leukemic myelomonoblasts. A panel of normal human T lymphocytes, B lymphocytes, and erythroid-myeloid or lymphoblastoid cell lines failed to react with both αU937 and αPEMØ. Although both heteroantisera reacted against polymorphonuclear leukocytes (PMNs), after absorption with PMNs specific reactivity against mononuclear phagocytes remained. Absorption of αU937 and αPEMØ with myelomonoblastic leukemia cells (AMML) removed IF and RIA activity against both PMNs and monocytes but not against splenic and peritoneal macrophages. In contrast, absorptions of both heteroantisera preparations with splenic macrophages abolished their IF and RIA reactivity not only to splenic and peritoneal macrophages but also to peripheral blood monocytes and leukemic myelomonoblasts. These results are consistent with (1) both antisera defining specific monocyte/macrophage-associated antigens(s) which are distinct from MHC-coded HLA-A,B,C, and DR antigens, and (2) expression of common monocyte/macrophage-associated antigen(s) and uniquely associated antigen(s) selectively expressed on tissue macrophages. These reagents will be useful in delineating human monocyte/macrophage differentiation as well as the immunological functions of mononuclear phagocytes.  相似文献

This paper considers the patterns of shell shape variation shown by Nucella canalicuata, N. emarginata and N. lamellosa from two areas of the Pacific Northwest: the shores near Friday Harbour on San Juan Island and near Bamfield on the west coast of Vancouver Island. No clear pattern of variation in association with changes in exposure was seen in either N. canaliculata or N. lamellosa . It appears that genetic influences are more important controls of shell shape than environmental selection in both these species. Nucella emarginata shows the nearest approximation to the pattern shown by the Atlantic species, N. lapillus , but only at the exposed end of the wave-action gradient. On those shores, enclaves from the most surf-washed open coast headlands have shells with proportionally larger apertures (and thus a shorter, squatter form) than their equivalents in local shelter. But, unlike in N. lapillus , the trend does not continue onto genuinely sheltered shores. Under these circumstances the species is generally rare and, where enclaves do occur, their shells are of much the same shape (although of a much larger size) as in more exposed situations.  相似文献

ABSTRACT Little is known about the relationship between seasonal food availability and the foraging strategies of insectivorous Neotropical birds. We studied a population of Tropical Kingbirds (Tyrannus melancholicus), a primarily insectivorous species, in eastern Bolivia to examine relationships between rainfall, food availability, and foraging strategies throughout the year. Our study site in the southern Amazon Basin was characterized by strong seasonal variation in the abundance of the kingbird's main insect prey (coleopterans and hymenopterans), with reduced abundance during the nonbreeding season which largely overlaps the dry season. Overall, mean search times for insect prey by Tropical Kingbirds during the breeding (96.9 ± 85.6 [SD] sec) and nonbreeding (83.7 ± 91.2 sec) seasons did not differ (P= 0.23). However, during the nonbreeding season, kingbird search times were negatively, but nonsignificantly, correlated with coleopteran abundance (r2= 0.43, P= 0.16) and significantly and negatively correlated with hymenopteran abundance (r2= 0.72, P= 0.03). Although insect abundance differed seasonally, kingbird search times did not, perhaps because kingbirds forage on a greater variety of insects during the nonbreeding season or, during the breeding season, kingbird search times may be influenced by the need to monitor and defend nests as well as constraints on the types of prey that can be fed to nestlings. However, the reduced abundance of their primary insect prey and negative relationships between the abundance of those prey and search times during the dry, nonbreeding season suggest that Tropical Kingbirds in southern Amazonia may be food limited, potentially explaining why some migrate and spend that season elsewhere.  相似文献

The ability of follicle-stimulating hormone (FSH), the estrogens, estradiol-17β and diethylstilbestrol, and the estrogen antagonists, clomiphene and enclomiphene citrate to affect the growth and internalization of hypophysectomized rat granulosa cell gap junction membranes was compared in ovarian follicles assigned to one of four follicle size classes (60–149, 150–249, 250–319, and 320–450 μm diameter). In the absence of exogenous hormone stimulation, atresia prevents follicle growth beyond 320 μm in diameter but surface gap junction membrane increases throughout this early follicle growth. Internalization of gap junction membrane is first detected at the 150- to 249-μm follicle stage and also increases with follicle size. Therefore, growth and turnover of gap junction membrane occur at a basal rate in the absence of gonadotropin or steroid hormone stimulation. Estrogen and estrogen antagonist injections result in no significant differences in the amount of surface or internalized junction membrane in the three smallest follicle size classes when compared to the untreated hypophysectomized animals. However, estrogen but not estrogen antagonists rescues growing follicles from atresia and permits their further growth into the 320- to 450-μm follicle size class. As a result of the additional follicle growth, both surface and internalized junction membrane increase beyond that seen in the largest follicles from hypophysectomized animals. In contrast to other treatments, FSH stimulation promotes amplification of gap junction membrane in all size classes and, like estrogen, rescues follicles from atresia and promotes their entry into the 320- to 450-μm follicle size class. Surface gap junction membrane is amplified two- to fourfold over other treatments in the first three follicle size classes, but reaches maximal levels in the 250- to 319-μm follicles. The internalized junction membrane which first appears in the 150- to 249-μm size class is dramatically increased over other treatments in the 250- to 319- and 320- to 450-μm size classes. These studies indicate that exogenous estrogen stimulation promotes gap junction growth indirectly by sustaining the basal rate of junction synthesis in follicles rescued from atresia. In contrast, exogenous FSH stimulation directly amplifies the developmental sequence of gap junction growth and turnover. During early follicle growth, FSH stimulation preferentially promotes increases in surface gap junctions while internalization of surface junctions is increased during later follicle growth.  相似文献

We investigated the site occupancy dynamics of greater prairie-chickens at Konza Prairie Biological Station, a protected site in northeastern Kansas that is managed for ecological research. We surveyed the site during mid-Mar to mid-May, 1981–2008, and recorded detections of birds in a grid of 6.3 ha survey plots (n = 187 plots). We used multiseason occupancy models to estimate the probabilities of occupancy (ψ) and detection (p), and tested whether land cover in woody vegetation, and land use with prescribed fire or grazing management influenced the dynamic processes of site colonization and local extinction. Probability of detection per site was consistently <1 and varied among years (p = 0.12–0.82). Site occupancy of prairie-chickens declined 40% over the study period from a high of ψ = 0.19 ± 0.02 SE in 1981 to a low of 0.11 ± 0.03 in 2008, despite protection from disturbance at leks and losses to harvest. We found that different sets of environmental factors impacted the probabilities of colonization and local extinction. Probability of colonization for an unoccupied site was negatively associated with the proportion of site occupied by woodland cover (β = −1.25), and was lower for grazed sites (β = −0.62). In contrast, probability of local extinction was affected by a weak interaction between grazing and average frequency of prescribed fire (β = −1.01), but model-averaged slope coefficients were not statistically different than 0. To conserve prairie-chickens, we recommend prairies be managed with combinations of prescribed fire and grazing that maintain a heterogeneous mosaic of prairie habitats, while preventing woody encroachment. To assess biotic responses to land management practices, field sampling should be based on occupancy models or similar techniques that account for imperfect detection. © 2011 The Wildlife Society.  相似文献

Plants frequently display fruit characteristics that support multiple seed‐dispersal syndromes. These ambiguous characteristics may reflect the fact that seed dispersal is usually a complex process involving multiple dispersers. This is the case for the Neotropical ginger Renealmia alpinia (Zingiberaceae). It was originally suggested that the aromatic fruits of R. alpinia located at the base of the plant are adapted for terrestrial mammal seed dispersal. However, the dark‐purple coloration of the fruits and bright orange aril surrounding the seeds suggest that birds may play a role in R. alpinia seed dispersal. At La Selva Biological Station, Costa Rica, we used camera traps to record vertebrate visits to infructescences of R. alpinia. Most visitors were toucans and aracaris (Ramphastidae). However fruits were also removed by terrestrial mammals (coatis and armadillos). In addition to vertebrate fruit removal, some of the fruits dehisce and the seeds that fall on the ground are dispersed by ants. Fruitfall traps showed that 77 percent of fruits are removed by vertebrates. However, 15 percent of fruits fall to the base of parent plants to be potentially dispersed by ants. Experiments using a laboratory ant colony showed that ants are effective seed dispersers of R. alpinia. Ant seed manipulation increased germination success and reduced time to germination. In conclusion, primary seed dispersal in the Neotropical ginger R. alpinia is mostly performed by birds, additionally ants are effective dispersers at short distances. Seed dispersal in R. alpinia is a complex process involving a diverse array of dispersal agents.  相似文献

Question: To what extent can aerial photography be used for taxonomic identification of Amazonian tree crowns? Objective: To investigate whether a combination of dichotomous keys and a web‐based interface is a suitable approach to identify tree crowns. Location: The fieldwork was conducted at Tiputini Biodiversity Station located in the Amazon, eastern Ecuador. Methods: High‐resolution imagery was taken from an airplane flying at a low altitude (600 m) above the ground. Imagery of the observable upper layer of the tree crowns was used for the analysis. Dichotomous identification keys for different types of crowns were produced and tested. The identification keys were designed to be web‐based interactive, using Google Earth as the main online platform. The taxa analysed were Iriartea, Astrocaryum, Inga, Parkia, Cecropia, Pourouma, Guarea, Otoba, Lauraceae and Pouteria. Results: This paper demonstrates that a combination of photo‐imagery, dichotomous keys and a web‐based interface can be useful for the taxonomic identification of Amazonian trees based on their crown characteristics. The keys tested with an overall identification accuracy of over 50% for five of the ten taxa with three of them showing accuracy greater than 70% (Iriartea, Astrocaryum and Cecropia). Conclusions: The application of dichotomous keys and a web‐based interface provides a new methodological approach for taxonomic identification of various Amazonian tree crowns. Overall, the study showed that crowns with a medium‐rough texture are less reliably identified than crowns with smoother or well‐defined surfaces.  相似文献

In vitro generation of free radicals by xanthine oxidase acting on xanthine as substrate depressed steady-state calcium uptake by canine cardiac sarcoplasmic reticulum vesicles. The effect of free radicals on the calcium-dependent ATPase activity of the SR vesicles was pH dependent. At pH 7.0, ATPase activity was decreased, but at pH 6.4 it was unchanged. Exposure to free radicals increased the passive permeability of the vesicles to calcium. This increase was inhibited by superoxide dismutase (SOD) at pH 7.0, and SOD plus mannitol at pH 6.4. The increased permeability per se was insufficient to explain the effects of free radicals on ATPase activity, since the calcium ionophore A23187 was unable to mimick these effects. Direct measurement of the number and turnover of the pump units indicated that the number of units was unchanged but turnover was decreased by free radicals at pH 7.0. The overall data suggest at least two mechanisms of free radical damage, one associated with an increase in passive permeability and another associated with an as yet undefined change in some specific steps of the ATPase reaction.  相似文献

Whole pituitary glands, as well as equivalent-sized pieces of kidney, liver, and adrenal tissue, were implanted into the anterior chamber of the adult newt eye. Secondary lens formation was present in 16 out of 17 cases of the pituitary implantation experiments in which the pituitary tissues were exposed to the anterior chamber fluid. In only one case out of 23, did another implanted tissue (liver) stimulated secondary lens formation. The extent of secondary lens formation was correlated with the position (closeness) of the implant in relationship to the dorsal iris. In all cases of secondary lens formation from the dorsal iris stimulated by tissue implants, the polarity of the developing lens was reversed so that the pole of the fiber axis was directed toward the implant rather than the retina. It appears that the ectopic pituitary produces a diffusible substance capable of initiating lens formation from the dorsal margin of the iris and capable of determining the orientation of the fiber axis pole of the developing lens.  相似文献

Luteinizing hormone levels were measured in blood samples collected at 5 minute (min) intervals for 3 hours (hr) during the a.m. and p.m. of 3 consecutive days from long-term ovariectomized cows. Levels of LH fluctuated in a pulsatile manner in all animals. During the pulses, LH levels increased rapidly (2.5 to 6.0 ng/ml). Following the rapid increase, a more gradual exponential decline was observed. The interval between pulses was consistent both within and between days of blood sample collection within cows. From the results we suggest that each cow may have an inherent consistent rhythmic pattern of LH release in the absence of an ovarian source of hormones.  相似文献
