We report here the results of mapping a set of 92 leaf cDNA clones in maize. The ends of each of these cDNA clones have previously been partially sequenced, and the sequence comparison has revealed the putative function for 28 clones. It is expected that the RFLP map developed using these expressed sequence tags will be of great importance for future maize genome analysis, such as for PCR-based gene mapping or gene function identification.Contribution from the Missouri Agricultural Experiment Station. The objectives of this study were two fold: (1) to determine whether divergent selection for kernel protein concentration, which produced the Illinois high protein (IHP), Illinois low protein (ILP), reverse low protein (RLP), and reverse high protein (RHP) maize (Zea mays L.) strains, had generated coupling-phase linkages among genes controlling protein concentration or other traits and (2) to measure the effectiveness of random mating in reducing linkage disequilibrium in segregating generations from crosses between the strains. To achieve these objectives, design III progenies from the F2 and F6 (produced by random mating the F2) from the crosses of IHP × ILP, IHP × RHP, ILP × RLP, and RHP × RLP were evaluated. Estimates of additive variance for percent protein in the crosses of IHP × ILP and ILP × RLP were significantly less in the F6 than in the F2 indicating the presence of coupling-phase linkages in the parents and their breakup by random mating. In addition, a significant reduction in dominance variance for grain yield from the F2 to the F6 in IHP × ILP suggested the presence of repulsion-phase linkages. No other evidence of coupling or repulsion-phase linkages was found for any of the traits measured. These results demonstrate the effectiveness of long-term divergent selection in the development of coupling-phase linkages and of random mating to dissipate linkage disequilibrium.Research supported by the Illinois Agricultural Experiment Station  相似文献

Konieczny and Ausubel have described a technique whereby Arabidopsis thaliana loci can be rapidly mapped to one of the ten chromosome arms using a small number of F2 progeny from crosses between the ecotypes Landsberg erecta and Columbia. The technique involves the use of 18 co-dominant, cleaved amplified polymorphic sequence (CAPS) markers which are evenly distributed throughout the Arabidopsis genome. We have mapped these 18 markers using recombinant inbred (RI) lines generated in our laboratory. These data enable a better integration of loci mapped relative to the CAPS markers into the restriction fragment length polymorphism (RFLP) map generated using Arabidopsis RI lines.  相似文献

Two cDNA clones corresponding to anaerobically induced maize mRNAs were found to have homology to a previously identified maize pyruvate decarboxylase gene. DNA sequencing and RFLP mapping indicate that these cDNAs represent two additional maize pdc genes. Each of the clones is approximately 85% identical in predicted amino acid sequence to the other two. All three clones are induced by hypoxic stress, but with different levels and kinetics of induction.  相似文献

Four near-isonuclear polycytoplasmic versions of 81A and two of Pb 402A male-sterile lines of pearl millet (Pennisetum typhoides) were used in factorial matings with five inbred male testers in different combinations in three sets. The cytoplasmic differences were studied for several agronomic traits using mean values and general combining effects (gca) of male-sterile lines, and specific combining ability effects of hybrids. The fertility/ sterility behaviour of different male-sterile lines in crosses with common male parents was also studied. Significant differences among near-isonuclear polycytoplasmic lines were observed in mean values for a few traits such as plant height, leaf length and peduncle length, but the differences for combining ability were more pronounced. The A3 cytoplasm was a better general combiner than the A2 cytoplasm for grain yield and both A2 and A3 cytoplasms were better general combiners for leaf length and peduncle length. In addition, superiority of A3 cytoplasm for gca was observed for plant height and ear characters over the A2 cytoplasm in set II. A differential behaviour of cytoplasms, both in combination with a common pollinator and across pollinators, was observed for several traits. The results provide evidence for the distinctiveness of different cytoplasmic sources in pearl millet and for the influence of cytoplasmic factors on the phenotypic expression of nuclear genes. A diversification of male sterility sources in the breeding of pearl millet hybrids is suggested.  相似文献

Summary This study investigates the characteristics of two human cell lines—1PT and 1PT VARIANT A—both derived from the same histologically undifferentiated, neuroendocrine positive, non-small cell lung carcinoma (NSCLC) and capable of growth in unsupplemented serum-free minimum essential medium. In stationary culture, the cells of both lines grew both attached to a plastic substratum and in suspension; the 1PT VARIANT A line formed three-dimensional clusters of loosely adherent cells. The cell lines differed in their DNA content, the 1PT having 1.44 times and the 1PT VARIANT A having 2.39 times the normal human diploid DNA content. Chromosome counts supported this observation, the ploidy of the 1PT and VARIANT A lines being 1.11 and 1.64, respectively. On transmission electron microscopy the cells of both lines had dense core granules and immature desmosomes, whereas only the 1PT VARIANT A line had mucin granules. Both lines formed, in nude mice, tumors that, like the original tumor from which they were derived, were histologically undifferentiated and showed local invasion. The original tumor and both lines had demonstrable neuroendocrine markers. Cytokeratins were apparent in the tumor but not the cell lines, and neurofilaments were present in the cell lines only. Staining for epithelial membrane antigen, neural cell adhesion molecule, and desmoplakin differentiated between the two lines. These lines provide a useful model for the investigation of the biology of the neuroendocrine positive subgroup of NSCLC, which is clinically important because of the possible responsiveness of these tumors to chemotherapy.  相似文献

Summary From the central nervous system ofDrosophila melanogaster 3rd instar larvae, eight continuous cell lines have been established (named ML-DmBG1 to 8). Using ML-DmBG2, single colony isolation was carried out and six colonial clones were obtained. All reacted to the antibody to horseradish peroxidase, which is a neuronal marker in insects. Acetylcholine, a known neurotransmitter inDrosophila, was detected in three of the colonial clones by high performance liquid chromatography. Therefore, it is concluded that the established colonial clones are neural cells originating in the larval central nervous system. Among them, some variation was observed with respect to morphology, acetylcholine content, and reactivity to anti-HRP. The variation may reflect the heterogeneity of cells composing the central nervous system.  相似文献

Post-harvest losses for maize and beans are common storage challenges affecting Tanzania's smallholder and large-scale farmers' storage facilities. Consequently, this leads to a reduction of over 50% of harvested grain qualitatively and quantitively per year. Therefore, this experimental study was conducted at the entomological laboratory of the Sokoine University of Agriculture Tanzania, where the research was aimed to assess different common storage insect pests of beans and maize at Morogoro major markets (i.e., Kihonda, Mazimbu and Morogoro Central Market) with similar storage conditions in Morogoro municipality. The complete block design (CBD) with three experimental replications was used. Where locations (Kihonda, Mazimbu and Central market) markets were considered as experimental treatments. The results obtained show that there were no significant differences in number of insect pests (such as maize weevils and bean bruchids) identified between the Kihonda and Central markets (P > 0.05). However, there is a significant difference (P< 0.05) between Mazimbu and other markets regarding the number of insect pest multiplication resulting from the storage condition of those grains in a particular market. Nevertheless, continuous winnowing of stored grains was observed to significantly intensify the infestation of the storage pest in maize and common beans. Therefore, alternatively, to reduce insect pest infestation to maize and common beans in storage facilities, it is important to avoid constant winnowing of stored grains.  相似文献
