用硫酸铵分段盐析及DEAE-Sephadex A-50、羟磷灰石和CM纤维素等多种柱层析方法,从正常小鼠肝浸液中分离纯化出一种免疫抑制蛋白质(LISP)。在体外用微量该蛋白质就能强烈抑制小鼠T、B淋巴细胞对促有丝分裂原和同种异型抗原的增生反应。纯化的蛋白质在聚丙烯酰胺凝胶电泳(PACE)和等电聚焦(IEF)鉴定时均显示为一条区带,其等电点(pI)值在7.5—7.8范围。沉降系数利S_(20),w为5.39。Sephadex G-100凝胶层析测得LISP的分子量为78,000道尔顿。十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)提示LISP是由二个相同的亚基组成,亚基分子量为38,500道尔顿。LISP是一种既非糖蛋白又非脂蛋白的碱性蛋白质,对它的氨基酸组成也作了分析。  相似文献

Seedlings of barley (Hordeum vulgare L. cvs Salka and Zita), buckwheat (Fagopyrum esculentum Moench) and rape (Brassica napus L. ssp. napus cv. Line) were raised at 8 or 10 different extenral P concentrations in the range 0–2000 μM. Apart from P, the nutrient solutions were complete. Phosphate influx in roots of different P status was determined by use of a nutrient solution containing 0.1 mM32P-labelled phosphate. A double labelling technique was used for simultaneous determination of influx (33P) and efflux (32P) of phosphorus by roots of barley and rape with three selected P levels. Flux determinations were also done in presence of a metabolic uncoupler (2,4-dinitrophenol) and a protein synthesis inhibitor (cycloheximide). Influx of phosphate was maximal at a certin optimal P level of the roots and decreased at both lower and higher P levels. Maximum phosphate influex [μmol (g root)-?1 h?1] were: rape 4,4, buckwheat 2.2, barley cv. Salka 1.6, barley cv. Zita 1.5. Both Hill plots and plots of the untransformed decreasing phosphate influx vs root P concentrations above the optimal were linear and had high correlation coefficients. The Hill coefficient varied between -3.1 and -4.2. The decrease of phosphate influx from the maximum to the lowest value at the highest P concentration of the root was 60–70%. Hence, phosphate influex appeared to be regulated through negative feedback by the internal level of phosphorous in the roots. The regulation mechanism seems bascially similar for the three species and may be of an allosteric type. P efflux from roots of low and optimal (with regard to P influx) P status was 15–20% of the simultaneous P influx. Contary to P influx, P efflux increased at high P status and almost eliminated (barley) or halved (rape) net P uptake. 2,4-Dinitrophenol reduced both P influx and P efflux by low P roots and gave linearly increasing P efflux with increasing root P status. This indicates that P efflux partly occurred by counter transport and ion exchange at the uptake sites, partly by passive P efflux along an electrochemical potential gradient. Phosphate influx was not affected by inhibition of barley root growth with cycloheximide, but P efflux increased considerably.  相似文献

When the conditions for detecting proteins by ammoniacal silver staining (B. R. Oakley, D. R. Kirsch, and N. R. Morris (1980) Anal. Biochem. 105, 361-363.) following gel electrophoresis were varied, it was noted that glutaraldehyde pretreatment was necessary for maximal staining, which could not be explained simply as the result of "fixation." Further studies indicated that glutaraldehyde enhancement of protein staining with this silver reagent was probably due to oxidation of the aldehyde groups by silver ions, resulting in metallic silver depositions within the gel which act as nucleation sites for additional metallic silver localization in the protein bands upon the addition of formaldehyde developer. This proposed mechanism is consistent with the Tollen's reaction, as well as some aspects of the photographic process. Consistent with this notion, silver-staining intensities are directly related to mole percentage lysine of various standard proteins.  相似文献

本文运用申农(Shannon)提供的最大熵原理,提出一种构成单一综合性状的新方法,并以此与数量遗传学中的多元统计法作了比较。在作多元遗传分析吋,常用多元统计法求出多个数量性状的综合性状,再对这些相互关联的基本性状作主成份分析或用典范相关进行遗传分析。本文提出了不同于多元统计学的另一种新的方法——最大熵法求出多个数量性状的单一综合性状值。它具有数学结构简单,过程明晰,结果简明等优点。  相似文献

Light scattering studies were performed on bovine alpha-crystallin measuring the scattering intensities as a function of scattering angle, concentration, and temperature. The data yielded the molecular weight, radius of gyration, and second virial coefficient of alpha-crystallin at different temperatures. The second virial coefficient increased with increasing temperature. Both the enthalpy and entropy of solution of alpha-crystallin are positive. The Flory theta temperature was found to be 271 K.  相似文献

Two nuclear techniques, Energy-Dispersive X-Ray Fluorescence Analysis (EDXRF) and Instrumental Neutron Activation Analysis (INAA), were used to analyze aerosol samples collected in the city of São Paulo, Brazil. Na, Cl, Mn, V, Al, Sm, Mo, W, La, As, Br, Sb, K, Ba, Se, Th, Cr, Rb, Ca, Fe, Ce, and Sc were determined by INAA, and Al, Si, P, S, Cl, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, Ga, As, Se, Br, Rb, Sr, Hg, and Pb were determined by EDXRF. A preliminary identification of the main source of the atmospheric aerosol was performed based on enrichment factor and correlation coefficient calculations.

Matrix metalloproteinases (MMP) play a cardinal role in the breakdown of extracellular matrix involved in a variety of biological and pathological processes. Research on MMPs has classified and characterized these enzymes according to their matrix substrate specificity, gene and protein domain structure, and regulation of activity and expression. However, the discovery of new MMPs has introduced a need for a more comprehensive and systematic method of classification and quantitative comparison of known and newly discovered members. This study compiles a sequence alignment, constructs a dendrogram, and calculates physical data and homology percentage assignments in order to obtain further insight into MMP structure-function relationships. Thorough analysis of MMP primary sequence domains, physical data patterns, and statistical analysis of sequence homology yields higher resolution in the similarities and differences that group MMP members.  相似文献

Hydraulic and osmotic properties were measured along developing maize (Zea mays L.) roots at distances between 15 and 465 mm from the root tip to quantify the effects of changes in root structure on the radial and longitudinal movement of water and solutes (ions). Root development generated regions of different hydraulic and osmotic properties. Close to the root tip, passive solute permeability (root permeability coefficient, Psr) was high and selectivity (root reflection coefficient, sr) low, indicative of an imperfect semipermeable root structure. Within the apical 100–150 mm, Psr decreased by an order of magnitude and sr increased significantly. Root hydraulic conductivity (Lpr) depended on the nature of the force (hydrostatic and osmotic). Osmotic Lpr was smaller by an order of magnitude than hydrostatic Lpr and decreased with increasing distance from the root tip. Throughout the root, responses in turgor of cortical cells and late metaxylem to step changes in xylem pressure applied to the base of excised roots were measured at high spatial resolution. The resulting profiles of radial and longitudinal propagation of pressure showed that the endodermis had become the major hydraulic barrier in older parts of the root, i.e. at distances from the apex ä 150 mm. Other than at the endodermis, no significant radial hydraulic resistance could be detected. The dynamic pressure method (DPM) is used for measurement of k(L)a in a 1-m(3) pilot scale fermentor in coalescing (distilled water) and noncoalescing (0.3 M Na(2)SO(4) aqueous solution) batches. The method consists in recording oxygen concentration in a batch after a small pressure change (20 kPa) in the fermentor. The upward pressure change is brought about by temporary closing and subsequent throttling of outlet gas stream and the downward change by full reopening of the gas outlet. Absorption of pure oxygen yields the same k(L)a values as absorption of air. In noncoalescing batch, the downward k(L)a values are always higher than the upward values owing to spontaneous nucleation of bubbles. The experiments performed in a stirred cell confirm this behavior. Thus, only upward pressure change should be used for measurement. The correlation of k(L)a data measured in small (18-L) and large (1000-L) vessels based on power dissipated and superficial gas velocity are in a good agreement. Unlike the DPM, the classical dynamic methods yield, under the same conditions, excessively low values of k(L)a (the dynamic startup method) or fail to produce data at all (the dynamic method with interchange of air for N(2)). Annual sweetclover plants [Melilotus segetalis (Brot) Ser.] were grown for a complete life cycle with and without saline (NaCl treatment of CE=15 dS m−1). Growth and partitioning analyses were performed. Sequential harvests (every 15 d) during the life cycle, and separation of plant material into roots, stems, petioles, leaves and reproductive structures were carried out Salt treatment reduced growth during the early and middle stages of the life of the plant, but did not significantly affect RGR in the reproductive phase. The root–shoot allometric coefficient of salinized plants in the generative phase decreased more than in control plants. We suggest that salinity-induced growth reduction in M. segetalis was primarily a result of a lower unit leaf rate (ULR) despite an increased leaf area ratio (LAR). Earlier flowering, higher biomass allocation to shoot and greater reproductive investment, but similar relative growth rate (RGR), were some of the main characteristics of salt-stressed plants compared to controls during the reproductive phase, these apparently being associated with increased sink strength caused by developing flowers and fruits.  相似文献
