The population genetic structure of sea bass (Dicentrarchus labrax) along a transect from the Atlantic Ocean (AO) to the Eastern Mediterranean (EM) Sea differs from that of most other marine taxa in this area. Three populations (AO, Western Mediterranean [WM], EM) are recognized today, which were originally two allopatric populations. How two ancestral genetic units have evolved into three distinct units has not been addressed yet. Therefore, to investigate mechanisms that lead to the emergence of the central WM population, its current status, and its connectivity with the two parental populations, we applied 20 nuclear loci that were either gene associated or gene independent. Results confirmed the existence of three distinct gene pools, with higher differentiation at two transitional areas, the Almeria‐Oran Front (AOF) and of the Siculo‐Tunisian Strait (STS), than within any population. Significant linkage disequilibrium and heterozygote excess indicated that the STS is probably another tension zone, as already described for the AOF. Neutrality tests fail to reveal marker loci that could be driven by selection within or among metapopulations, except for locus DLA0068. Collectively, results support that the central WM population arose by trapping two tensions zones at distinct geographic locations of limited connectivity. Population assignment further revealed that WM individuals were more introgressed than individuals from the other two metapopulations. This suggests that this population might result from hybrid swarming, and was or is still seeded by genes received through the filter of each tension zone.  相似文献

A trypsin was purified from the hepatopancreas of snakehead (Channa argus) by ammonium sulfate fractionation and a series of column chromatographies including DEAE-Sepharose, Sephacryl S-200 HR and Hi-Trap Capto-Q. The molecular mass of the purified trypsin was about 22 kDa, as estimated by SDS-PAGE. The optimum pH and temperature of the purified trypsin were 9.0 and 40 °C, respectively. The trypsin was stable in the pH range of 7.5-9.5 and below 45 °C. The enzymatic activity was strongly inhibited by serine proteinase inhibitors, such as MBTI, Pefabloc SC, PMSF, LBTI and benzamidine. Peptide mass fingerprinting (PMF) of the purified protein obtained 2 peptide fragments with 25 amino acid residues and were 100% identical to the trypsinogen from pufferfish (Takifugu rubripes). The activation energy (Ea) of this enzyme was 24.65 kJ·M− 1. Apparent Km was 1.02 μM and kcat was 148 S− 1 for fluorogenic substrate Boc-Phe-Ser-Arg-MCA. A trypsinogen gene encoding 247 amino acid residues was further cloned on the basis of the sequence obtained from PMF and the conserved site peptide of trypsinogen together with 5′-RACE and 3′-RACE. The deduced amino acid sequence contains a signal peptide of 15 residues and an activation peptide of 9 amino acid residues with a mature protein of 223 residues. The catalytic triad His-64, Asp-107, Ser-201 and 12 Cys residues which may form 6 disulfide bonds were conserved. Compared with the PMF data, only 2 amino acid residues difference were identified, suggesting the cloned trypsinogen is quite possibly the precursor of the purified trypsin.  相似文献

研究了木兰科(Magaoliaceae)3个杂交组合的亲本和杂交后代的核型。结果表明,云南含笑(Michelia yunnanensis)、灰岩含笑(Michelia calcicola)及其杂交组合A的核型分别为2n=2x=38=36m+2sm、2n=2x=38=34m+4sm和2n=2x=38=26m+12sm;紫花含笑(Michelia crassipes)及其与云南含笑杂交组合C的核型分别为2n=2x=38=32m+6sm和2n=2x=38=24m+12sm+2st;山玉兰(Mognolia delavayi)、广玉兰(Mognolia grondiflora)及其杂交组合U的核型分别为2n=2x=38=28m+10sm、2n=6x=114=88m+lOsm+16st和2n=4x=76=57m+15sm+4st。杂交组合的核型与理论核型存在明显的差异,可能是在杂交组合的形成过程中,来自亲本的染色体发生了结构变异。  相似文献

为建立鉴定大豆杂种的方法,采用亲本间有多态性的3对SSR引物,对148个耐盐与盐敏感大豆品种正反交F1植株进行分子鉴定,结果表明,有81.8%的F1为真杂种,且3对多态性SSR引物检测结果一致;在亲本基因型纯合的情况下,采用1对在亲本间有多态性的SSR引物即可对F1真伪进行准确判断;亲本间分子量差异大的SSR位点可用高浓度琼脂糖电泳进行快速鉴定。利用该方法对2007年参加国家大豆区试的大豆杂交种品系H02—286的50粒种子进行纯度鉴定,进一步验证了SSR标记检测杂种真伪的可行性。  相似文献

To assess if pollinators’ behaviour could explain the maintenance of hybrid zones between different flower colour morphs, we analyzed flower colour variation in an Antirrhinum hybrid zone using spectrometry and a model of bee perception. Some colours generated by hybridization were not observed in any Antirrhinum species and even appeared to be rare among angiosperms. Variation in flower colours within the hybrid zone was continuous; the most similar colours were predicted not to be discriminated from one another in natural foraging situations. However, when compared at a scale corresponding to bees’ foraging range, some flower colours could be discriminated from all colours displayed by neighbouring plants. This could affect pollinator behaviour and explain lower visitation rates within the centre of the hybrid zone. Behavioural studies involving bumblebees and plant mixtures of parental and hybrid flower colours carefully characterized with appropriate visual models will be necessary to test this hypothesis. NovW, novU and novS genes have been characterized as dTDP-4-keto-6-deoxy-D-glucose 3-epimerase, C-5 methyltransferase and dTDP-glucose 4-ketoreductase, respectively involved in noviose biosynthetic pathway. We have cloned and expressed the Streptomyces spheroids novWUS genes in S. venezuelae YJ003-OTBP1. This established the function of novWUS and, at the same time, it also proved that the noviosyl derivative of 10-deoxymethynolide(2)/narbonolide(4) obtained from S. venezuelae YJ003-OTBP1 is a novel hybrid antibiotic.  相似文献

The effect of red- and blue-light-emitting diodes on shoot and root growth of Hybrid Franc grape, a rootstock cultivar, along with two other grape genotypes, Kadainou R-1 and Vitis ficifolia var. ganebu, were investigated in vitro. Plants cultured under red-light-emitting diodes produced the longest shoots with longer internodes for all genotypes. The chlorophyll content measured as SPAD value and leaf number per explant were highest on plants cultured under blue-light-emitting diodes in all the genotypes. Blue light was also responsible for a higher number of stomata in all the genotypes; however, there was no significant difference in size of stomata in all genotypes under the different light conditions tested. Different light-emitting diodes did not affect the rooting percentage of Hybrid Franc but red-light-emitting diodes gave a higher rooting percentage along with higher root numbers for the two other grape genotypes.  相似文献

Subunit heterogeneity within a particular subunit in hemoglobin A have been explored with electron paramagnetic resonance spectroscopy using the nitrosyl hemes in Ni-Fe hybrid Hb under various solution conditions. Our previous studies on the crystal structure of NiHb demonstrated the presence of subunit heterogeneity within alpha-subunit. To further cross check this hypothesis, we made a hybrid Hb in which either the alpha- or beta-subunit contains iron, which alone can bind to NO. By this way dynamic exchange between penta- and hexa-coordinated forms within a subunit was confirmed. Upon the addition of inositol hexa phosphate (IHP) to these hybrids, R to T state transition is observed for [alpha(2)(Fe-NO)beta(2)(Ni)] but such a direct transformation is less marked in [alpha(2)(Ni)beta(2)(Fe-NO)]. Hence the bond between N(epsilon) and Fe is fundamental to the structure-function relation in Hb, as the motion of this nitrogen triggers the vast transformation, which occurs in the whole molecule on attachment of NO.  相似文献

实验比较了丰鲤与奥尼罗非鱼摄食低质和高质两种等能饲料时的氮收支和能量收支。低质饲料的蛋白质含量为 34.2 5 %、蛋白质主要来源为豆粕 ,高质饲料的蛋白质含量为 4 5 .4 4 %、蛋白质主要来源为鱼粉。 5 3d的生长结果显示 :摄食低质饲料时 ,奥尼罗非鱼通过降低排泄氮、排泄能和代谢能的比例 ,使生长氮和生长能的比例显著高于丰鲤 ,摄食高质饲料时 ,则通过降低粪氮和粪能的比例取得快速生长的效果 ;丰鲤生长氮和生长能比例不受饲料质量的显著影响 ,但高质饲料使排泄氮和代谢能比例显著降低 ;奥尼罗非鱼的氮收支不受饲料质量的显著影响 ,但高质饲料使生长能和排泄能的比例显著增高 ,代谢能的比例显著降低。结果表明 ,在两种饲料条件下 ,丰鲤利用氮和能量的能力显著低于奥尼罗非鱼。  相似文献

Ursodeoxycholic acid (UDCA), a component of bile acid, which is abundant in the gall bladder of bears, has been used in clinical medicine for cholestatic liver diseases. Recently, it was demonstrated that UDCA and its derivative tauroursodeoxycholic acid block apoptotic cell death in both hepatic and non-hepatic cells. Cisplatin, an effective anti-cancer drug, is known to cause sensory neuropathy in patients receiving the drug. In the present study, whether UDCA is effective in blocking cisplatin-induced cell death in mouse hybrid sensory neurons was conducted. N18D3 mouse hybrid sensory neurons exposed to cisplatin were found to undergo apoptotic cell death. Preincubation with UDCA completely blocked cisplatin-induced apoptotic cell death in the sensory neurons, and cisplatin-induced p53 accumulation was suppressed by UDCA treatment. These results indicate that UDCA has a neuroprotective effect on the cisplatin-induced neuronal cell death of sensory neurons via the downregulation of the p53 signaling pathway.  相似文献
