Questions: What is the best grassland management regime for the threatened plant species Gladiolus imbricatus; is the stage structure of local populations a feasible indicator of the effect of changed management. Location: Coastal meadow system in southwestern Estonia. Methods : The effect of five management regimes was studied in a long‐term (three‐year) field experiment: (1) mowing in late July, (2) grazing by cattle, (3) grazing by sheep, (4) sheep grazing during the first year and mowing during subsequent years, (5) no management (control). Results: The population density increased significantly in response to the mowing treatment and to the mowing after sheep grazing treatment. The proportion of grazed plant individuals was higher in the sheep‐grazed than in the cattle‐grazed treatment. Generative and vegetative adult individuals of G. imbricatus were significantly more damaged by cattle herbivory than juveniles. All management regimes shifted the population structure towards a dynamic state where juvenile stages dominate, while the not managed control retained a regressive population structure. Conclusions: Population stage structure was a useful indicator of different management conditions, even in the case where population density did not differ. As indicated by population stage structure, the best management regime for G. imbricatus was either mowing in late July only, or alternation of grazing and mowing in different years. 相似文献

Biosensors for signaling molecules allow the study of physiological processes by bringing together the fields of protein engineering, fluorescence imaging, and cell biology. Construction of genetically encoded biosensors generally relies on the availability of a binding “core” that is both specific and stable, which can then be combined with fluorescent molecules to create a sensor. However, binding proteins with the desired properties are often not available in nature and substantial improvement to sensors can be required, particularly with regard to their durability. Ancestral protein reconstruction is a powerful protein-engineering tool able to generate highly stable and functional proteins. In this work, we sought to establish the utility of ancestral protein reconstruction to biosensor development, beginning with the construction of an l-arginine biosensor. l-arginine, as the immediate precursor to nitric oxide, is an important molecule in many physiological contexts including brain function. Using a combination of ancestral reconstruction and circular permutation, we constructed a Förster resonance energy transfer (FRET) biosensor for l-arginine (cpFLIPR). cpFLIPR displays high sensitivity and specificity, with a Kd of ∼14 µM and a maximal dynamic range of 35%. Importantly, cpFLIPR was highly robust, enabling accurate l-arginine measurement at physiological temperatures. We established that cpFLIPR is compatible with two-photon excitation fluorescence microscopy and report l-arginine concentrations in brain tissue. 相似文献

为探讨真菌诱导子以及硝酸银等非生物诱导子对雷公藤不定根生长及次生代谢产物含量的影响。以雷公藤不定根为材料,通过向培养基中添加不同种类的真菌诱导子以及硝酸银等非生物诱导子,采用高效液相色谱检测不定根中雷公藤甲素和生物碱含量。结果表明:各种真菌诱导子不影响不定根的生长,但对次生代谢产物含量有显著影响,其中,苹果炭疽和柿子炭疽诱导子的加入不仅使不定根中雷公藤甲素的含量分别提高了2.24和1.93倍,生物碱的含量也各提高了2.02和2.07倍。苹果炭疽诱导子浓度为50μg/m L时比较适合雷公藤不定根生长及雷公藤甲素和生物碱的积累。硝酸银抑制不定根的生长和生物碱的积累,但促进雷公藤甲素的积累。硝酸银浓度为25μmol/L时雷公藤甲素含量为对照的1.71倍。茉莉酸甲酯浓度为50μmol/L时,不定根增长量为对照的1.04倍,雷公藤甲素和生物碱含量分别为对照的1.64倍和2.12倍。酵母提取物浓度为2 g/L时,雷公藤甲素含量为对照的1.48倍。表明培养基中添加硝酸银和酵母提取物对不定根中雷公藤甲素的合成具有明显的促进作用,苹果炭疽和茉莉酸甲酯的协同作用既能促进雷公藤甲素的合成又能促进雷公藤生物碱的合成。 相似文献

A new species of Noblella is described from the humid montane forest of the Región Cusco in Peru. Specimens were collected at 2330–2370 m elevation in Madre Selva, near Santa Ana, in the province of La Convención. The new species is readily distinguished from all other species of Noblella by having a broad, irregularly shaped, white mark on black background on chest and belly. The new species further differs from known Peruvian species of Noblella by the combination of the following characters: tympanic membrane absent, small tubercles on the upper eyelid and on dorsum, tarsal tubercles or folds absent, tips of digits not expanded, no circumferential grooves on digits, dark brown facial mask and lateral band extending from the tip of the snout to the inguinal region. The new species has a snout-to-vent length of 15.6 mm in one adult male and 17.6 mm in one adult female. Like other recently described species in the genus, this new Noblella inhabits high-elevation forests in the Andes and likely has a restricted geographic distribution. 相似文献

The general aim of this study is to update the inventory of the fish species and to specify distribution patterns in the Nero River ichthyofauna in order to establish some basis for the conservation of these fish communities and their habitat. From February 2009 to January 2010, thirty-three sites were sampled monthly with gill nets and a backpack electrofisher, and environmental variables were recorded. Overall, 46 species included in 33 genuses, 24 families and 9 orders were collected. Eleven families and 30 species were the first records for the Nero River. Including all species previously listed in the literature, the number of species presently known in the Nero River and its tributaries is revised to 59. Four families, Alestidae (21%), Schilbeidae (19%), Cyprinidae (17%) and Cichlidae (16%) that made up 73% of the total number of the catches, were the most dominant. The most dominant numerical species were Schilbe mandibularis and Brycinus longipinnis. Fish species and sampling sites along with eight environmental variables were ordinated with canonical correspondence analysis (CCA) coupled to the Monte Carlo test. Ecological status based on fish assemblage according to environmental variables and anthropogenic pressures showed that miss dead wood leaves and roots, electrical conductivity, total dissolved solids, mud, nitrite, basin width, dissolved oxygen and pH, were the primary factors influencing fish distribution. The environmental tolerance index (ETI), ecological tolerance (tk) and optima (uk) values of 10 species to 8 different environmental variables were analyzed. Six species (Hemichromis fasciatus, Epiplatys chaperi, Barbus ablabes, B. longipinnis, Hemichromis bimaculatus and Chromidotilapia guntheri) have high ETI and a cosmopolitan distribution in the Nero River. In the tributaries of the middle course, high concentrations of nitrite in the water, added to the presence of a lot of tolerant species in the ichthyofauna are indications of disturbance of these areas. In greenhouse and field experiments, an invert emulsion (MSG 8.25) was tested with dried, formulated spores of the bioherbicidal fungus Colletotrichum gloeosporioides f. sp. aeschynomene, a highly virulent pathogen of the leguminous weed Aeschynomene virginica (northern jointvetch), but considered ‘immune’ against another more serious leguminous weed, Sesbania exaltata (hemp sesbania). A 1:1 (v/v) fungus/invert emulsion mixture resulted in 100% infection and mortality of inoculated hemp sesbania seedlings over a 21-day period under greenhouse conditions. In replicated field tests of the fungus/invert formulation conducted in Stuttgart, AR, and Stoneville, MS, hemp sesbania was controlled 85 and 90%, respectively. These results suggest that this invert emulsion expands the host range of C. gloeosporioides f. sp. aeschynomene, with a concomitant improvement of the bioherbicidal potential of this pathogen. 相似文献

Wild and tissue cultured plants of Portulaca grandiflora Hook. have shown to be able to decolorize a sulfonated diazo dye Navy Blue HE2R (NBHE2R) up to 98% in 40 h. A significant induction in the activities of lignin peroxidase, tyrosinase and DCIP reductase was observed in the roots during dye decolorization. The wild plants and tissue cultures could independently decolorize and degrade NBHE2R into metabolites viz. N-benzylacetamide and 6-diazenyl-4-hydroxynaphthalene-2-sulfonic acid. A dye mixture and a textile effluent were also decolorized efficiently by P. grandiflora. The phytotoxicity study revealed reduction in the toxicity due to metabolites formed after dye degradation. 相似文献

As an energy carrier, hydrogen gas is a promising substitute to carbonaceous fuels owing to its superb conversion efficiency, non-polluting nature, and high energy content. At present, hydrogen is predominately synthesized via chemical reformation of fossil fuels. While various biological methods have been extensively explored, none of them is justified as economically feasible. A sustainable platform for biological production of hydrogen will certainly impact the biofuel market. Among a selection of biological systems, algae and cyanobacteria have garnered major interests as potential cell factories for hydrogen production. In conjunction with photosynthesis, these organisms utilize inexpensive inorganic substrates and solar energy for simultaneous biosynthesis and hydrogen evolution. However, the hydrogen yield associated with these organisms remains far too low to compete with the existing chemical systems. This article reviews recent advances of biochemical, bioprocess, and genetic engineering strategies in circumventing technological limitations to hopefully improve the applicative potential of these photosynthetic hydrogen production systems. 相似文献
