快速、准确鉴定出病原体是临床感染性疾病诊断和传染病预防控制的基础。高通量测序基因检测技术突破了传统检测手段的时效性、灵敏度等的局限,为病原体检测和研究提供了便捷、高效的途径。本综述以高通量测序技术发展过程为基础,回顾纳米孔三代测序技术,及其在病毒性传染病检测鉴定及研究中的应用,并对该技术的应用前景及可能存在的问题进行阐述,期望它能在病毒性传染病的防控方面发挥更大的作用。  相似文献

Summary Genetically defined alkaline phosphatase (ALP) isozymes from the larval midgut tissues ofBombyx mori were purified and characterized. The membrane-bound form (m-ALP) was solubilized with 1% Triton X-100, then purified by DEAE-Sephacel, Con A-Sepharose 4B and Ultrogel AcA 34 column chromatography. The soluble form (s-ALP) was purified by DEAE-Sephacel, epoxy Toyopearl coupled with phosphonic acid and Ultrogel AcA 34 column chromatography. About 840- and 650-fold purification were achieved for m-ALP and s-ALP, respectively, and both ALPs were homogeneous as judged by poly-acrylamide gel electrophoresis. Both forms were found to be similar (MW=68 000 in gel permeation chromatography, and a single subunit as a monomer in denaturing SDS-polyacrylamide gels with MW=58 000 for m-ALP and MW=61 000 for s-ALP). The pH optima of ALPs were shown to lie at 10.9 (m-ALP) and 9.8 (s-ALP), the former being extremely stable even in pH 10–12 which accords with the physiological milieu inBombyx midgut lumen. The Km values of the m-ALP and s-ALP forp-nitrophenyl phosphate were 1.99 and 1.49 mM, respectively. Both ALPs had a similar substrate specificity.l-Cysteine strongly inhibited both ALPs, but inhibitory effects ofl-phenylalanine,l-homoarginine andl-leucine were undetectable for s-ALP and very weak for m-ALP. In this study, the amounts of trace elements (Fe (II) and Cu (II)) and pigments (chlorophyll and carotenoid) in soybean oil were evaluated under high voltage electric field (HVEF) bleaching method at different voltage (10 and 20 kV), temperature (35–65 °C), time (0−30 min) and clay percentage (0.5–2 %) and then were compared to the industrial bleaching method (IBM). The kinetic data of ions and pigments adsorbed on activated bentonite clay under IBM and HVEF at two voltages of 10 and 20 kV followed the mechanism of the pseudo-first-order model (PFOM). The carotenoid and chlorophyll equilibrium data followed a Freundlich isotherm type model, which demonstrated multilayer adsorption under HVEF. The thermodynamic parameters (ΔG°, ΔH°, and ΔS°) displayed that the adsorption of trace metal ions and pigments on bentonite clay under IBM and HVEF were feasible, endothermic and spontaneous between 35 and 65 °C. The results indicated that the HVEF, especially at higher voltage, has a high remarkable capability to remove metal ions and pigments from soybean oil than the IBM. The highest removal capacity for metal and pigments of soybean oil bleaching were obtained in the order of HVEF-20 kV > HVEF-10 kV > IBM.  相似文献

BackgroundThe optimal induction treatment in potentially-resectable stage IIIA-N2 NSCLC remains undefined.AimTo compare neoadjuvant high-dose chemoradiotherapy (CRT) to neoadjuvant chemotherapy (CHT) in patients with resectable, stage IIIA-N2 non-small-cell lung cancer (NSCLC).MethodsRetrospective, multicentre study of 99 patients diagnosed with stage cT1-T3N2M0 NSCLC who underwent neoadjuvant treatment (high-dose CRT or CHT) followed by surgery between January 2005 and December 2014.Results47 patients (47.5%) underwent CRT and 52 (52.5%) CHT, with a median follow-up of 41 months. Surgery consisted of lobectomy (87.2% and 82.7%, in the CRT and CHT groups, respectively) or pneumonectomy (12.8% vs. 17.3%). Nodal downstaging (to N1/N0) and Pathologic complete response (pCR; pT0pN0) rates were significantly higher in the CRT group (89.4% vs. 57.7% and 46.8% vs. 7.7%, respectively; p < 0.001)). Locoregional recurrence was significantly lower in the CRT group (8.5% vs. 13.5%; p = 0.047) but distant recurrence rates were similar in the two groups. Median PFS was 45 months (CHT) vs. “not reached” (CRT). Median OS was similar: 61 vs. 56 months (p = 0.803). No differences in grade ≥3 toxicity were observed. On the Cox regression analysis, advanced pT stage was associated with worse OS and PFS (p < 0.001) and persistent N2 disease (p = 0.002) was associated with worse PFS.ConclusionsCompared to neoadjuvant chemotherapy alone, a higher proportion of patients treated with preoperative CRT achieved nodal downstaging and pCR with better locoregional control. However, there were no differences in survival. More studies are needed to know the optimal treatment of these patients.  相似文献

Medical imaging using X-rays has been one of the most popular imaging modalities ever since the discovery of X-rays 125 years ago. With unquestionable benefits, concerns about radiation risks have frequently been raised. Computed tomography (CT) and fluoroscopic guided interventional procedures have the potential to impart higher radiation exposure to patients than radiographic examinations. Despite technological advances, there have been instances of increased doses per procedure mainly because of better diagnostic information in images. However, cumulative dose from multiple procedures is creating new concerns as effective doses >100 mSv are not uncommon. There is a need for action at all levels. Manufacturers must produce equipment that can provide a quality diagnostic image at substantially lesser dose and better implementation of optimization strategies by users. There is an urgent need for the industry to develop CT scanners with sub-mSv radiation dose, a goal that has been lingering. It appears that a new monochromatic X-ray source will lead to replacement of X-ray tubes all over the world in coming years and will lead to a drastic reduction in radiation doses. This innovation will impact all X-ray imaging and will help dose reduction. For interventional procedures, the likely employment of robotic systems in practice may drastically reduce radiation exposures to operators- but patient exposure will still remain an issue. Training needs always need to be emphasized and practiced.  相似文献

Extensive use of antibiotics has caused the microbial resistance to rise drastically within the last few decades, and new approaches are therefore needed to develop effective antibacterial substances. In this study, we identified peptide in the hemolymph of Hermetia illucens larvae using reverse-phase chromatography, HPLC and Nano-LC-ESI-MS/MS system. We investigated the antibacterial effect of HP/F9 peptides against Klebsiella pneumoniae in vitro and in vivo. The peptide effectively inhibited the growth of K. pneumoniae in vitro and completely removed K. pneumoniae from the lungs of mice. Importantly, peptides (22,000 Da, HP/F9) successfully reduced lung inflammation upon K. pneumoniae infection. These results indicate that the HP/F9 peptide from H. illucens larva can effectively protect the mouse from K. pneumoniae infection. HP/F9 could be a new candidate for the development of effective antibacterial substance.  相似文献

研究了大兴安岭北部奥克里堆山不同海拔(750~1420 m)天然林土壤胞外酶活性(EEA)和酶计量比的变化特征及影响机制。结果表明: 海拔、季节及其交互作用对β-葡萄糖苷酶(BG)、N-乙酰-β-氨基葡萄糖苷酶(NAG)、亮氨酸氨基肽酶(LAP)和酸性磷酸酶(AP)活性均存在显著影响。5月,BG和NAG活性随海拔升高呈逐渐增大趋势,AP活性随海拔升高呈先升高后降低的趋势。7月,NAG活性随海拔升高而增大,AP活性则先升高后降低。9月,不同海拔上NAG活性变化幅度较大,在1420 m处活性最高,为124.22 nmol·h-1·g-1。随着海拔的升高,酶化学计量比ln(BG)∶ln(NAG+LAP)呈降低的趋势。除海拔830 m外,7月化学计量比值最高。土壤C、N、P转化酶活性对数转换后的比值为1∶1.25∶0.82。海拔和土壤温度是影响土壤胞外酶的主要因素,土壤温度与BG、NAG和AP呈显著正相关。ln(BG)∶ln(NAG+LAP)和ln(NAG+LAP)∶ln(AP)与pH分别呈显著正相关和负相关,与DOC分别呈显著负相关和正相关,而ln(BG)∶ln(AP)受土壤容重的影响较大。  相似文献

目的探讨宫颈病变患者阴道微生态与高危型HPV感染及宫颈癌相关增殖基因表达的相关性。方法选择2018年1月至2019年1月间在我院确诊为原发性宫颈癌的患者50例作为宫颈癌组,在我院诊断为宫颈糜烂的患者78例作为宫颈糜烂组,同期在我院进行体检的健康女性100例作为正常对照组。对比3组研究对象的阴道微生态失调率、高危型HPV感染率以及宫颈癌组、宫颈糜烂组患者宫颈病灶组织中宫颈癌相关增殖基因(Prdx4、Nek2、Fhit、BLCAP)mRNA表达量的差异。采用Pearson检验分析宫颈癌患者阴道微生态失调率与高危型HPV感染及宫颈癌相关增殖基因表达的相关性。结果宫颈癌组、宫颈糜烂组患者的阴道微生态失调率、高危型HPV感染率高于正常对照组,其中宫颈癌组患者这两项指标水平高于宫颈糜烂组(均P0.05)。宫颈癌组患者宫颈病灶组织中Prdx4、Nek2 mRNA表达量高于宫颈糜烂组,Fhit、BLCAP mRNA表达量低于宫颈糜烂组(均P0.05)。相关性分析发现,宫颈癌患者阴道微生态失调率与高危型HPV感染率呈正相关,与癌基因(Prdx4、Nek2)mRNA表达量呈正相关,与抑癌基因(Fhit、BLCAP)mRNA表达量呈负相关(均P0.05)。结论宫颈癌患者阴道微生态失调率较高,可能与高危型HPV感染及癌细胞增殖旺盛密切相关。  相似文献

目的探讨轻度支气管哮喘儿童的诱导痰菌群特征及临床意义。方法纳入年龄为6~12周岁于2018年11月至2019年1月在深圳市儿童医院呼吸科门诊定期复诊的轻度支气管哮喘患儿51例(哮喘组),留取诱导痰,匹配同年龄段97例健康无过敏儿童的口咽拭子作为对照。诱导痰及口咽拭子提取总DNA并扩增,对16S rRNA基因进行高通量测序并对测序结果进行生物信息学分析。结果NMDS分析结果显示哮喘组与健康对照组研究对象菌群结构存在差异;哮喘组的诱导痰菌群多样性指数(Shannon index)高于健康对照组(2.34±0.53 vs 1.87±0.50,P<0.05)。门水平分析显示,哮喘组与健康对照组的菌群均主要为厚壁菌门(38.34%vs 44.74%,P<0.05)、变形杆菌门(31.14%vs 19.78%,P<0.05)、拟杆菌门(14.59%vs 20.52%,P<0.05)、放线菌门(10.41%vs 7.85%,P<0.05)和梭杆菌门(2.82%vs 6.67%,P<0.05),但两组之间的构成比有明显差异。与健康对照组相比,在属水平上哮喘组韦荣球菌属(5.27%vs 8.96%)、普雷沃菌属(8.38%vs 17.35%)、罗斯菌属(1.50%vs 5.46%)、纤毛菌属(1.37%vs 4.39%)等非条件致病菌属比例明显下降(均P<0.05),而嗜血杆菌属(9.83%vs 6.17%)、卟啉单胞菌属(2.48%vs 1.41%)、莫拉菌属(5.66%vs 0.42%)、诺卡菌属(3.40%vs 0.00%)等条件致病菌属比例明显上升(均P<0.05)。结论尽管轻度支气管哮喘患儿已临床控制,但诱导痰内菌群仍存在结构紊乱。气道菌群紊乱可能是儿童支气管哮喘的发病机制之一。除了致病菌属外,非致病菌菌属的构成变化可能也是儿童哮喘的一个发生机制。  相似文献
