The purpose of the study was to investigate the effects of mild hyperthermia on cell viability, release of lactate dehydrogenase (LDH), superoxide dismutase (SOD) activity, malondialdehyde (MDA) formation, total antioxidant capacity (T-AOC), and the relative mRNA levels of heat shock protein (HSP60, 70, and 90) in hepatic cells of grass carp (Ctenopharyngodon idellus) before and after temperature stress. Cultured cells were exposed to thermal stress (32 °C) for 0.5, 1, 2, 4, and 8 h. The results showed that hyperthermia stress significantly reduced cell viability (P<0.01) and increased LDH release at 0.5 and 1 h (P<0.05). Additionally, hyperthermia stress led to oxidative stress as evidenced by significantly decreased T-AOC after treating cells for 0.5 and 8 h (P<0.05). SOD activity also significantly decreased after 1 h of stress (P<0.05), but MDA formation increased after 8 h of stress (P<0.05). This may be partly responsible for the lower cell viability and higher LDH release we observed. The differences between SOD activity, MDA formation, and T-AOC between the 2 h treatment group and the control were smaller than that of other groups. This indicated that cellular antioxidant enzyme systems play an important role in the defense against oxidative stress. Further tests showed that the expression of HSP60 at 1, 2, and 4 h (P<0.05), HSP70 at 0.5 and 1 h (P<0.01), and HSP90 at all time points after stress were higher (P<0.01) than pre-stress levels. This suggested that HSPs possess the ability to modulate cellular anti-stress responses and play key roles in protecting organisms from heat stress. In conclusion, hyperthermia inhibits cell proliferation, induces cell oxidative stress, and enhances HSP expression in hepatic cells of grass carp.  相似文献

If the technosphere and the biosphere are divided into cells, the presence and turnover of a substance in a study area can be summarized in a vector of stocks and a matrix of flows between different pairs of cells. Likewise the stocks and flows of several substances or materials in one or more time periods can be summarized in multidimensional data cubes. In this article, we provide a theoretical framework for handling physical flow data, and we demonstrate how a set of matrix operations can facilitate exploratory analysis and quality assessment of such data regardless of the number of substances, materials, and time periods considered. In particular, we show how matrices and cubes of flow data can be recalculated when the collection of cells is modified by joining cells, and also what information is required to recalculate flows when cells are split. Furthermore, we illustrate how and under what circumstances substance-flow data originating from different studies with different collections of cells can be compared or merged. The generic character of the given formulae facilitates the development of software for physical flow data.  相似文献

Contemporary cycles for copper and zinc are coanalyzed with the tools of exploratory data analysis. One-year analyses (circa 1994) are performed at three discrete spatial levels-country (52 countries that comprise essentially all anthropogenic stocks and flows of the two metals), eight world regions, and the planet as a whole-and are completed both in absolute magnitude and in per capita terms. This work constitutes, to our knowledge, the first multiscale, multilevel analysis of anthropogenic resources throughout their life cycles. The results demonstrate that (1) A high degree of correlation exists between country-level copper and country-level zinc rates of fabrication and manufacturing, entry into use, net addition to in-use stocks, discard, and landfilling; (2) Regional-level rates for copper and zinc cycle parameters show the same correlations as exist at country level; (3) On a per capita basis, countries add to in-use stock almost 50% more copper than zinc; (4) The predominant discard streams for copper and zinc at the global level are different for the two metals, and relative rates of different loss processes differ geographically, so that resource recovery policies must be designed from metalspecific and location-specific perspectives; (5)When absolute magnitudes of life-cycle flows are considered, the standard deviations of the data sets decrease from country level to regional level for both copper and zinc, which is not the case for the per capita data sets, where the statistical properties of the data sets for both metals approach being independent of spatial level, thus providing a basis for predicting unmeasured per capita metal flow behavior.  相似文献

Hepatic blood vessels consist of the hepatic artery and three types of venous channels (the portal veins, the sinusoids, and the hepatic veins). This study was undertaken to analyze, by immunohistochemistry, connexin expression throughout the vascular development of the fetal mouse liver with special attention being given to portal vein development. In the adult liver, connexin37 and connexin40 were expressed in the endothelium of the portal vein and hepatic artery, but not in those of the hepatic vein and sinusoids. Connexin43 was expressed in mesothelial cells and smooth muscle cells of the portal veins. The preferential expression of connexin37 and connexin40 in portal veins was seen throughout liver development, including its primordium formation stage (10.5-day or 11.5-day stage), although connexin37 expression was transiently seen in free nonparenchymal cells in fetal stages. The differentiation of each blood vessel in the hepatic vascular system may occur in early developmental stages, soon after hepatic primordium formation. This study was carried out to evaluate the antifibrotic effect of ethanol extract of the fruits of Indian herb Piper longum Linn. Liver fibrosis was induced in rats by CCl(4) administration. The extent of liver fibrosis was assessed by measuring the level of liver hydroxy proline (HP) and serum enzyme levels. Following CCl(4) administration HP was significantly increased and serum enzyme levels were elevated. Treatment with the ethanol extract of Piper longum Linn. reduced the HP and also the serum enzymes. The liver weight that increased following CCl(4) administration due to the deposition of collagen was reduced by the ethanol extract. Hence, it is concluded that this extract inhibits liver fibrosis induced by CCl(4).  相似文献

Aplyronine A, isolated from the sea hare Aplysia kurodai, possesses an exceedingly potent antitumor effect in vivo and it is one of the promising candidates as an anticancer drug. This macrolide is known to depolymerize F-actin and inhibit the polymerization of actin by forming a 1:1 complex with monomeric actin. The first complex structure of actin-aplyronine A was determined via a synchrotron X-ray analysis at a 1.45 A resolution. As expected, aplyronine A binds to a hydrophobic cleft composed of subdomains 1 and 3 of actin by intercalating its aliphatic tail part into the actin molecule as do the other reported F-actin depolymerizing agents. Unexpectedly, this complex structure shows the specific structural features around the trimethylserine moiety, revealed as an important moiety of aplyronine A for cytotoxicity against HeLa cells. Combining this result and our previous one, the moiety should strongly relate to the specific biological activity of aplyronine A; i.e. a potent antitumor effect.  相似文献

ATP has recently emerged as a key molecule mediating pathological pain. The aim of this study was to examine whether spinal cord astrocytes could be a source of ATP in response to the nociceptive neurotransmitters glutamate and substance P. Glutamate stimulated ATP release from these astrocytes and this release was greatly potentiated by substance P, even though substance P alone did not elicit ATP release. Substance P also potentiated glutamate-induced inward currents, but did not cause such currents alone. When glutamate was applied alone it acted exclusively through alpha-amino-3-hydroxy-5-methylisoxazole-4-proprionate receptors to stimulate Ca(2+) influx-dependent ATP release. However, when substance P was co-applied with glutamate, ATP release could be elicited by activation of NMDA and metabotropic glutamate receptors. Activation of neurokinin receptor subtypes, protein kinase C and phospholipases A(2), C and D were needed for substance P to bring about its effects. These results suggest that astrocytes may be a major source of ATP in the spinal cord on activation of nerve fibres that release substance P and glutamate.  相似文献
