Soybean cyst nematode (SCN) is a major soybean yield-limiting pest. The present study was conducted to map broad-based SCN resistance loci from the cultivar Hartwig. Two-hundred F23 lines derived from the cross Williams 82 x Hartwig were screened with a fourth-generation SCN inbred and 56 polymorphic molecular markers. Allele states and phenotypes were analyzed using stepwise regression and the model selection was made at P 0.01. Four unlinked RFLP markers (A006, A567, A487, A112) were associated with SCN resistance and the partial coefficient of determinations (R2) were 91%, 1%, 1%, and 1%. We have mapped a new, major SCN resistance locus (A006) and three minor loci (A567, A487, A112). This complete mapping will accelerate the transfer of broad-based resistance without linkage drag and aid in the determination of relationships among various SCN-resistant germplasm sources. 相似文献

The hatching responses of Globodera rostochiensis (golden potato cyst nematode) to purified and partially-purified preparations of natural (including the potato glycoalkaloids solanine and α-chaconine) and artificial hatching factors (HFs) were bimodal. At least 10 HFs, mostly anionic, were resolved from potato root leachate by a combination of gel permeation and ion-exchange chromatography. Whereas potato roots were the principal source of HFs, haulm leachate also contained such chemicals. Root leachate from aseptically-grown potato plants lacked several HFs which were present in conventionally-produced leachate. 相似文献

本文采用脐血浆治疗实验性肝衰竭大鼠模型,并与成人血浆的疗效进行了比较。结果表明,脐血浆降低大鼠死亡率、血谷丙转氨酶水平,总胆红素浓度及使甲胎蛋白水平升高均明显高于成人血浆的作用(P<0.05).肝脏病理组织学也显示脐血浆具有减轻病理损害和较快修复坏死组织的作用。提示脐血浆中含有促进肝细胞再生的因子,具有显著提高实验性大鼠肝衰竭的存活率。 相似文献

Evaluation of enzyme activities in combination with taxonomic analyses may help define the mechanisms involved in microbial decomposition of orgaic amendments and biological control of soilborne pathogens. In this study, powdered pine bark was added to nematode-infested soil at rates of 0, 5, 10, 15, 20, 25, 30, 35, 40, 45, and 50 g kg–1. Total fungal populations did not differ among treatments immediately after application of pine bark. After 7 days, fungal populations were positively correlated with increasing levels of pine bark. This increase was sustained through 14 and 21 days.Penicillium chrysogenum andPaecilomves variotii were the predominant fungal species isolated from soil amended with pine bark. Total bacterial populations did not change with addition of pine bark at 0, 7, and 14 days after treatment. At 21 and 63 days, total bacterial populations declined in soil receiving the highest rates of pine bark. Addition of pine bark powder to soil caused a shift in predominant bacterial genera fromBacillus spp. in nonamended soil, toPseudomonas spp. in amended soil. Soil enzyme activities were positively correlated with pine bark rate at all sampling times. Trehalase activity was positively correlated with total fungal populations and with predominant fungal species, but was not related to bacterial populations. The number of non-parasitic (non-stylet bearing) nematodes andMeloidogyne arenaria in soil and roots were not correlated with pine bark rate. Lines of wheat with the 6Mv chromosome from Aegilops ventricosa display partial resistance to both pathotypes Hal2 and Ha41 of Heterodera avenae. With either pathotype, the effect of this alien chromosome on cyst production, size, and fecundity was expressed in resistance tests. Partial resistance of five 6Mv(6D) substitution lines varied according to the intrinsic cyst-forming capacity of the nematode pathotypes and the recipient germplasms. Using standard hybridoma technology and hierarchical screening, monoclonal antibodies (MAbs) were obtained with specific reactivity against two developmental stages of Globodera pallida. The procedure was based on enzyme-linked immunosorbent assay (ELISA) with homogenates prepared from second-stage juveniles, young adult females, and potato roots. Hybridomas were formed by fusing myelomas with splenocytes derived from mice immunized with either infective juveniles or females of G. pallida. About 600 hybridoma lines were screened from the fusion involving the mouse immunized with juveniles. Two MAbs (LJMAbl &2) were identified with high reactivity toward second-stage juveniles but no reactivity with either potato roots or females of G. pallida. A total of 630 cell lines was screened from the corresponding fusion involving the spleen of a mouse receiving immunogens from adult female nematodes. One MAb (LFMAbl) was obtained with the required specificity against only adult female G. pallida. This work extends the application of monoclonal antibodies in nematology from valuable probes for research and species identification to recognition of developmental stages. While investigating dinoflagellate cyst assemblages in surface sediments of the Gulfs of Naples and Salerno (Mediterranean Sea), we found a new calcareous resting cyst. This cyst has a round to oval body surrounded by a thick mineral layer, which gives it the shape of a Napoleon hat, with a flat, oval face bearing the archeopyle and a convex keel on the opposite side. The cyst shape is variable in both natural samples and clonal cultures. The organic membrane underlying the calcareous covering is resistant to acetolysis, thus demonstrating the presence of sporopolleninlike material. The cyst germinated into a motile stage having the same morphological features and thecal plate pattern as Peridinium tyrrhenicum Balech. We believe the validity of the genus Pentapharsodinium Indelicato & Loeblich should be accepted. Based on the comparative examination of the species we collected and of a similar species, Pentapharsodinium trachodium Indelicato & Loeblich, we propose Pentapharsodinium tyrrhenicum as a new combination for Peridinium tyrrhenicum. The genus Pentapharsodinium also includes P. dalei Indelicato & Loeblich (= Peridinium faeroense Dale), which produces spiny, organic-walled cysts. The presence of species forming calcareous cysts and species producing noncalcareous cysts in the same genus raises questions about maintaining the family Calciodinellaceae. This family should only include calcareous cyst-forming peridinioids, in order to maintain a unified system of classification of fossil and recent dinoflagellates. 相似文献

Two monoclonal antibodies, which differentially recognise the two species of potato cyst nematodes (PCN), Globodera pallida and G. rostochiensis, are described. They have been shown to have potential for quantification of these two species, recognising proteins of the same molecular weight (34 kD) in both species. Further investigation showed these proteins to have isoelectric points at pH values of 5.7 in G. pallida and 5.9 in G. rostochiensis, in common with the proteins used by Fleming & Marks (1983) to differentiate the species of PCN. They are likely to be structurally very similar, with the same physiological function (and therefore similar concentrations) in the two species. In cross-reactivity tests with a wide range of soil nematode species, the antibodies reacted strongly only with species of the genus Globodera, and thereby confirmed their potential as the basis of a quantitative immunoassay likely to be useful in management of PCN populations. 相似文献
