Cadmium is among the toxic and hazardous metal widely dispersed in the environment in high levels. Current studies have provided new insights into antioxidant properties of bioflavonoid which have emerged as probable therapeutic and nutraceutical agents. The present study is geared to investigate the possible role of Cymbopogon schoenanthus (L.) Spreng. (or Ethkher) on heavy metal cadmium (Cd) induced oxidative stress in mice. Mice were randomly divided into four groups and treated for 15 days as follows: group 1: normal control-treated (saline); group 2: Ethkher leaves extract-treated (100 mg/kg); group 3: cadmium chloride (CdCl2) treated; group 4: CdCl2 plus Ethkher leaves extract. The results showed a significant reduction in hemoglobin, RBC and hematocrit in cadmium-treated mice as compared to control. Exposure to Cd caused a significant increase in the number of white blood cells (P < 0.05) indicating the occurrence of systemic inflammation. The results of this study also revealed that the mice intoxicated with Cd showed a significant increase in bilirubin, aspartate aminotransferase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP) and gamma-glutamyl transpeptidase (GGTP) activities. Cd intoxication leads to suppression in humoral immunity. However, pretreatment with Ethkher extract reversed almost all the abnormalities in the blood parameters showing noteworthy protection against cadmium induced toxicity in mice. The outcome of the present study revealed that the Ethkher possessed significant immunomodulatory activity and had a preventive effect on the hematological alterations in Cd intoxicated mice.  相似文献

This study was carried out to determine the LC50 of zinc oxide nanoparticles (ZnONPs) on Oreochromis niloticus and to investigate the effect of vitamin E and C on hematological and biochemical alterations induced by two sublethal concentrations (1 and 2 mg/L) of ZnONPs. One hundred and eighty fish were used for studying the lethal concentrations of ZnONPs. For sublethal study two hundred and twenty-five males of O. niloticus were equally divided into five groups, control, the second and the third were treated with 1 and 2 mg/L ZnONPs respectively. The fourth and fifth were exposed to the same concentrations of ZnONPs plus vitamins E and C. The results revealed that the 96 h LC50 of ZnONPs was 3.1 ± 0.4 mg/L. The sublethal study revealed the presence of normocytic normochromic anemia in groups (2, 3 and 5) along the experiment period. The 4th group showed normocytic normochromic anemia at the 7th day and microcytic hypochromic anemia at the 15th day. Leukocytosis, heterophilia, lymphopenia and monocytopenia were recorded at the 7th day in all treated groups compared with the normal control. At the 15th day heteropenia, lymphopenia and monocytopenia were reported in all treated groups. A significant increase in the serum levels of alkaline phosphatase, aminotransferases, urea, creatinine and erythrocytic nuclear and morphological abnormalities along the experimental periods in all treated groups compared with the normal control. Serum total protein and albumin levels were significantly decreased at the same period in the same groups. Addition of vitamin E and C to the diet (groups 4 and 5) significantly improved all measured parameters compared with groups (2 and 3) which treated with ZnONPs only.  相似文献

The aim of the present study was to obtain a basic knowledge of the hematology in order to determine changes in blood parameters of the spotted rose snapper Lutjanus guttatus. The morphological features of blood cells were described according to the observations made by light microscopy of Wright-Giemsa-stained blood films. The reference intervals and the mean value were determined for each hematological parameter evaluated in healthy fish and data were compared to those of naturally infected, with dactylogyrid monogeneans fish.Infected fish showed a prevalence of 100% and a mean intensity of 246.6 parasites per fish. Mean values of HCT, WBC, thrombocytes percentage and eosinophils percentage were significantly higher (P < 0.05) in the infected fish. In addition, lymphocytes percentage and total protein were significantly lower (P < 0.05) in the infected fish compared to healthy fish. Only total WBC count, lymphocytes percentage and eosinophils percentage in infected fish were outside reference interval. The hematology of the spotted rose snapper of this study might serve as a basis for future studies and diagnosis. Changes observed in blood parameters in infected fish suggest that the immune system of L. guttatus was affected by the presence of the parasites.  相似文献

本文对从长江中获得的江豚进行了血液学、血液化学特征的测定和血红蛋白、血浆蛋白电泳图谱的研究。发现江豚的嗜中性粒细胞所占百分比较高,淋巴细胞的百分比较低,具有嗜碱性粒细胞。江豚血红蛋白的电泳类型为Ⅰ-Ⅱ,即电泳相对迁移率在人类Hb S和Hb F之间,并有4条非血红素蛋白带(NHP)。用醋酸纤维薄膜作支持物进行电泳,血浆的总蛋白(TP)带可分成8条主要的区带,从阳极到阴极依次是:前白蛋白(ζ)、白蛋白(A)和6条后白蛋白带(PA);然而用聚丙烯酰胺凝胶作支持物共可分成16条区带,其中14条后白蛋白带。脂蛋白(Lp)在聚丙烯酰胺凝胶上可分成4条区带:α_1-脂蛋白、α_2-脂蛋白和β-脂蛋白,分别相当于聚丙烯酰胺凝胶上总蛋白带电泳图谱中的PA9、PA10和PA11,而前-β-脂蛋白和乳糜微粒在起点处。运铁蛋白(Tf)的电泳图为一条区带,相当于PA6。  相似文献

Due to the increased production of polyacrylamide microplastics in the environment impacting the adverse effects on aquatic organisms have become a global concern. The present study aimed to evaluate the toxicity of different concentrations (0.018, 0.03 0.09 g/1) of polyacrylamide microplastics on Oreochromis niloticus. Polyacrylamide microplastics were characterized by Fourier transform infrared and Raman spectroscopy. The FTIR technique revealed the spherical morphology and size range of polyacrylamide (0.1–0.4 mm) with 99 % purity. Raman Spectroscopy results showed peaks around (1100 cm?1 and 1650 cm?1) in gills, these peaks confirmed the presence of polyacrylamide microplastics in the gills of Oreochromis niloticus. Polyacrylamide microplastics significantly shortened the antioxidant enzymes (Catalase and Reduced glutathione) proceedings and the increase was observed in Malondialdehyde and Lipid peroxide levels in high-dose treated groups. Moreover, total protein contents were expressively increased, while other blood parameters (AST, ALP, ALT) were significantly decreased. Hemoglobin (g/dl), Erythrocyte (106/µl) and TLC (1x109/l) levels were reduced significantly (p < 0.05) in high concentrations. The administration of polyacrylamide microplastics induced different histological changes in the gills, liver and intestine of O. niloticus. It was concluded that polyacrylamide microplastics are toxic agents having harmful effects on fish health.  相似文献

Vitamin E (VE), an important lipid-soluble antioxidant, has great influence on growth and maintenance in animal. The effects of VE supplemented diet on growth and feed usage in Nile tilapia (Oreochromis niloticus) was investigated in this study. Three formulated diets containing VE (0, 50 and 100 mg/kg) were fed to Nile tilapia (3.56 ± 0.16 g) in glass aquaria maintaining three replicate groups for 56 days (8 weeks). Survival, growth performance including weight gain, percent weight gain, and specific growth rate (WG, % WG, and SGR), and feed utilization comprising protein efficiency ratio and feed conversion ratio (PER and FCR) were calculated. Hemato-biochemical indices including hemoglobin level (Hb), white blood cell (WBC), red blood cell (RBC) and glucose level were analyzed. In addition, muscle morphology was examined after completion of the experiment. At the end of the trial, WG, %WG, SGR, FCR and PER increased significantly which had dietary VE supplimentation. However, no distinct changes were observed in Hb level, RBC count, WBC count and glucose level among these different dietary groups. Dietary VE treatments significantly upgraded the muscle fiber diameter and lowered the intra-muscle gap. Moreover, quantity of hyperplastic muscle fiber as well as nucleus also significantly enhanced by VE. Morphological structure of muscle characterized by a huge proportion of hyperplastic muscle that may be supposed to contribute the enhanced growth of Nile tilapia receiving VE supplemented diet. Therefore these results suggested that VE incorporation into the feed can be effective to improve the feed efficiency and maximize the growth of O. niloticus.  相似文献

Pneumocyte type II produces a phospholipid, dipalmitoyl lecithin, which is stored in and secreted from the cell's inclusion bodies and is indispensable for alveolar stability. Cloned rat lung type II cells were harvested at monolayer confluence and homogenized in swelling buffer. After sequential differential centrifugations, the crude membrane fraction was subjected to discontinuous sucrose density gradient centrifugation at 65,000 × g. Quality of the relevant fractions was monitored by enzyme activities and phase contrast and electron microscopy of two major bands at densities 1.16 and 1.18, respectively. The less dense band contained only small quantities of organelles, little cytochrome c oxidase, and some glucose 6-phosphatase, but had a significant (Na+, K+)-ATPase activity; this and ultrastructural evidence certified the product as a suitable plasma membrane preparation. Upon sodium dodecyl sulfate-poly acrylamide gel electrophoresis, the protein pattern consisted of 11 major protein bands between 13,000 and 68,000 Mr ranges, and several minor ones. The lipid pattern was studied by two-dimensional thin layer chromatograpy, followed by various group reactions (e.g., amine, unsaturation, phosphorus, sugars). In the two major phospholipids, phosphatidyl choline and phosphatidyl ethanolamine, palmitic acid was the least abundant of four major fatty acids, accounting for 14.20% in phosphatidyl choline and 5.70% in phosphatidyl ethanolamine, whereas the most abundant were stearic and palmitoleic with about 28% each in phosphatidyl choline, and palmitoleic (29.90%) and oleic (23.05%) in the ethanolamine phosphatide. Apparently, the palmitic acid containing phosphatidyl choline must be in the lamellar inclusion bodies of type II cells and not in their plasma membranes.  相似文献

This model is described by a first order partial differential equation with a time delay and a retardation of the maturation variable, both due to cell replication. Zebrafish (Danio rerio) have been extensively utilized for understanding mechanisms of development. These studies have led to a wealth of resources including genetic tools, informational databases, and husbandry methods. In spite of all these resources, zebrafish have been underutilized for exploring pathophysiology of disease and the aging process. Zebrafish offer several advantages over mammalian models for these studies, including the ability to perform saturation mutagenesis and the capability to contain thousands of animals in a small space. In this review, we will discuss the use of mature zebrafish as an animal model and provide specific examples to support this novel use of zebrafish. Examples include demonstrating that clinical pathology can be performed in mature zebrafish and that age-associated changes in heat shock response can be observed in zebrafish. These highlights demonstrate the utility of zebrafish as a model for disease and aging.  相似文献
