目的探讨不同分娩方式对婴儿出生后1年内肠道菌群定植的影响。方法选取45例新生儿为研究对象,根据分娩方式分为自然分娩组(n=27)和剖宫产组(n=18)。收集婴儿出生后0(胎粪)、3、6和12个月的粪便标本,应用高通量测序技术分析肠道菌群多样性及组成。结果与自然分娩组比较,在0个月时剖宫产组婴儿粪便标本拟杆菌门的相对丰度显著降低(Z=-2.374 1,P=0.017 6)。2组研究对象中,除自然分娩组和剖宫产组0个月时婴儿粪便标本分别以埃希菌-志贺菌属和克雷伯菌属为优势菌属外,余下均以双歧杆菌属为优势菌属。相比于自然分娩组,在0个月时剖宫产组婴儿粪便标本埃希菌-志贺菌属和肠杆菌属所占比例显著降低(Z=-2.136 4,P=0.032 7;Z=-2.940 8,P=0.003 3),克雷伯菌属和罗氏菌属所占比例显著升高(Z=-2.642 4,P=0.008 2;Z=-2.299 4,P=0.021 5);6个月时罗氏菌属所占比例显著降低(Z=-2.045 0,P=0.040 9),肠球菌属所占比例显著升高(Z=-2.109 2,P=0.034 9)。结论不同分娩方式下的婴儿肠道菌群的构成存在显著差异。  相似文献

目的探究中部和西部地区幼儿肠道菌群的结构差异与膳食的关系,为幼儿营养健康状况监测和营养改善工作提供有效的营养干预。方法选择“贫困地区儿童营养改善项目”河南汝阳和贵州福泉,随机抽取107名汉族健康幼儿为调查对象,食物摄入采用24 h消费调查方法。应用高通量测序技术对肠道菌群进行测序和生物信息分析,研究两县幼儿肠道菌群差异。结果两县幼儿肠道菌群组成结构较为一致,但Alpha多样性分析表明,贵州福泉县幼儿肠道物种丰富度和多样性高于河南汝阳县。细菌属水平分析中,两县幼儿肠道内均以拟杆菌属、普雷沃菌、柔嫩梭菌和双歧杆菌属为主导的菌群结构,但河南汝阳县幼儿肠道双歧杆菌属和乳杆菌属丰度显著高于贵州福泉县(12.36% vs. 7.44%;0.19% vs. 0.03%)。结论中西部地区幼儿肠道菌群结构差异不大,但有益菌含量存在显著差异,这为研究肠道菌群与膳食及营养健康状况之间的关系提供了依据。  相似文献

目的探讨去卵巢对小鼠肠道菌群和血脂的影响。方法 12只10周龄C57BL/6小鼠随机分为2组:假手术组(SHAM组)和去卵巢组(OVX组),每组6只,进行12周的喂养。每2周测定小鼠体质量,12周后测肝脏指数、血清三酰甘油水平和游离脂肪酸水平,小肠进行病理学检查,收集小鼠粪便并在Illumina MiSeq测序平台进行16S rRNA基因测序检测。结果与SHAM组相比,OVX组小鼠体质量、肝脏指数、血清三酰甘油水平和游离脂肪酸水平明显增加(t=4.745,t=15.090,t=11.140,t=4.038,均P0.01)。与SHAM组相比,OVX组小鼠肠道Shannon Wiener指数明显降低(t=4.520,P0.01);在SMB53属(P=0.001)、拟杆菌属(P=0.017)、优杆菌属(P=0.018)、粪球菌属(P=0.019)、赖氨酸芽胞杆菌属(P=0.020)的丰度上差异有统计学意义。结论去卵巢小鼠血脂升高和肠道菌群失衡,提示肠道菌群可能是预防和治疗雌激素缺乏后脂质代谢异常的潜在靶点。  相似文献

This short paper focuses on some technical aspects concerning plant sample preparation for LM technology, with particular attention to the application of gene expression and proteomics studies. Transport of calcium (Ca) and cadmium (Cd) was examined along the gastro-intestinal tract (GIT) of freshwater and seawater Oncorhynchus mykiss irideus (FWT and SWTies respectively) using in vitro and in vivo experiments. Based on known physiological differences between FWT and SWT which aid in regulating ion levels and osmolarity, we hypothesized that SWT would have lower rates of Ca uptake. Also, we predicted that Cd rates would also be lower because Cd is known to share a common transport mechanism with Ca. Kinetics of Ca and Cd transport were determined using mucosal salines of varying concentrations [1, 10, 30, 60, and 100 (mmol L− 1 for Ca, μmol L− 1 for Cd)]. Linear and saturating relationships were found for Ca for FWT and SWT, but overall SWT had lower rates. Linear and/or saturating relationships were also found for Cd uptake, but rates varied little between fish types. Elevated Ca had no inhibitory effect on Cd transport, and Ca channel blockers nifedipine and verapamil had little effect on Ca or Cd uptake. However, lanthanum reduced Ca transport into some compartments. A 21 day in vivo feeding experiment was also performed where FWT and SWT were exposed to control diets or Cd-spiked diets (552 μg Cd g− 1 food). Whole body Cd uptake between fish types was similar, but the majority of Cd in SWT remained in the posterior intestine tissue, while FWT transported more Cd through their gut wall. Overall it appears that large differences in Ca and Cd uptake between FWT and SWT exist, with SWT generally having lower rates.  相似文献

The endophytic fungus Piriformospora indica colonizes the roots of many plant species including Arabidopsis and promotes their performance, biomass, and seed production as well as resistance against biotic and abiotic stress. Imbalances in the symbiotic interaction such as uncontrolled fungal growth result in the loss of benefits for the plants and activation of defense responses against the microbe. We exposed Arabidopsis seedlings to a dense hyphal lawn of P. indica. The seedlings continue to grow, accumulate normal amounts of chlorophyll, and the photosynthetic parameters demonstrate that they perform well. In spite of high fungal doses around the roots, the fungal material inside the roots was not significantly higher when compared with roots that live in a beneficial symbiosis with P. indica. Fifteen defense- and stress-related genes including PR2, PR3, PAL2, and ERF1 are only moderately upregulated in the roots on the fungal lawn, and the seedlings did not accumulate H2O2/radical oxygen species. However, accumulation of anthocyanin in P. indica-exposed seedlings indicates stress symptoms. Furthermore, the jasmonic acid (JA) and jasmonic acid-isoleucine (JA-Ile) levels were increased in the roots, and consequently PDF1.2 and a newly characterized gene for a 2-oxoglurate and Fe2+-dependent oxygenase were upregulated more than 7-fold on the dense fungal lawn, in a JAR1- and EIN3-dependent manner. We conclude that growth of A. thaliana seedlings on high fungal doses of P. indica has little effect on the overall performance of the plants although elevated JA and JA-Ile levels in the roots induce a mild stress or defense response.  相似文献

为探讨铜(Cu)胁迫条件下土壤微生物对海州香薷(Elsholtzia splendens)光合生理和叶绿素荧光参数的影响,实验设置添加Cu(Cu胁迫)、接种土壤微生物、添加Cu与接种土壤微生物等3个处理,以不添加Cu与不接种土壤微生物为对照(CK)。结果表明:接种土壤微生物处理的植株相对叶绿素含量、净光合速率(Pn)、水分利用效率(WUE)均显著高于CK;且对初始荧光(Fo)和最大光化学效率(Fv/Fm)均有显著性影响。与CK相比,添加Cu降低了海州香薷的Pn和气孔导度(Gs),但胞间CO2浓度(Ci)的变化与Pn相反,表明其对光合作用的影响主要是非气孔限制因素。添加Cu的植株相对叶绿素含量显著下降,但Cu胁迫下接种土壤微生物提高了植株相对叶绿素含量,差异显著。在Cu胁迫条件下,接种土壤微生物的植株具有较高的Fv/Fm及较低的Fo,显著提高了海州香薷的WUEPnGs。说明接种土壤微生物可通过提高相对叶绿素含量、改善叶绿素荧光和光合作用来减轻Cu胁迫对海州香薷植株造成的伤害,从而提高海州香薷耐受Cu胁迫的能力。  相似文献

Grassland desertification seriously threatens sustainable economic and social development. Much attention has been paid to the control of grassland desertification, and even to the restoration and reconstruction of the grassland. Vegetation restoration is considered to be a very effective solution. Soil sustains an immense diversity of microbes, and the characteristics of soil microbial communities are sensitive indicators of soil. It is important to understand the relationship between vegetation and soil microbial diversity during the restoration process. Soil microbial, which is the main index to evaluate soil quality, plays a significant role in ecosystem and soil microbial diversity is the important one of global diversity. Exploring the effects of different vegetation patterns on soil microbial diversity can provide scientific bases and technical support for systemic and impersonal assessment of the best vegetation restoration patterns, as well as the vegetation restoration and reconstruction of Hulunbeier sandy land. Based on PCR–DGGE technology, a case study was carried out to investigate the effects of five different vegetation restoration patterns on soil microbial functional diversity after 4 years in sandy land in Hulunbeier, China. The five vegetation restoration patterns included mono-cultivar planting of Agropyron cristatum (UA), mono-cultivar planting of Hedysarum fruticosum (UH), mono-cultivar planting of Caragana korshinskii (UC), mixed-cultivar planting of Agropyron cristatum and Hedysarum fruticosum (AC) and mixed-cultivar planting of Agropyron cristatum, Hedysarum fruticosum, Caragana korshinskii and Elymus nutans (ACHE). Completely degraded sandy land was used as control.The results indicated that the vegetation restoration increased the genetic diversity of soil bacterial community obviously, and the structure of soil bacterial community was changed. The results of phylogenetic analysis suggested that the bacterial community in Hulunbeier sandy land mainly attributed to Proteobacteria, Bacteroidetes, Firmicutes, Actinobacteria, and Acidobacteria. The dominant groups were Proteobacteria and Bacteroidetes. The effects of different vegetation type on soil bacterial community structures were different.  相似文献
