Mixed-rod cone bipolar (Mb) cells of goldfish retina have large synaptic terminals (10 mum in diameter) that make 60-90 ribbon synapses mostly onto amacrine cells and rarely onto ganglion cells and, in return, receive 300-400 synapses from gamma-aminobutyric acid (GABA)-ergic amacrine cells. Tissue viewed by electron microscopy revealed the presence of double-membrane-bound processes deep within Mb terminals. No membrane specializations were apparent on these invaginating processes, although rare vesicular fusion was observed. These invaginating dendrites were termed "InDents". Mb bipolar cells were identified by their immunoreactivity for protein kinase C. Double-label immunofluorescence with other cell-type-specific labels eliminated Müller cells, efferent fibers, other Mb bipolar cells, dopaminergic interplexiform cells, and somatostatin amacrine cells as a source of the InDents. Confocal analysis of double-labeled tissue clearly showed dendrites of GABA amacrine cells, backfilled ganglion cells, and dendrites containing PanNa immunoreactivity extending into and passing through Mb terminals. Nearly all Mb terminals showed evidence for the presence of InDents, indicating their common presence in goldfish retina. No PanNa immunoreactivity was found on GABA or ganglion cell InDents, suggesting that a subtype of glycine amacrine cell contained voltage-gated Na channels. Thus, potassium and calcium voltage-gated channels might be present on the InDents and on the Mb terminal membrane opposed to the InDents. In addition to synaptic signaling at ribbon and conventional synapses, Mb bipolar cells may exchange information with InDents by an alternative signaling mechanism.  相似文献

Electrical stimulation of the central nervous system creates both orthodromically propagating action potentials, by stimulation of local cells and passing axons, and antidromically propagating action potentials, by stimulation of presynaptic axons and terminals. Our aim was to understand how antidromic action potentials navigate through complex arborizations, such as those of thalamic and basal ganglia afferents-sites of electrical activation during deep brain stimulation. We developed computational models to study the propagation of antidromic action potentials past the bifurcation in branched axons. In both unmyelinated and myelinated branched axons, when the diameters of each axon branch remained under a specific threshold (set by the antidromic geometric ratio), antidromic propagation occurred robustly; action potentials traveled both antidromically into the primary segment as well as "re-orthodromically" into the terminal secondary segment. Propagation occurred across a broad range of stimulation frequencies, axon segment geometries, and concentrations of extracellular potassium, but was strongly dependent on the geometry of the node of Ranvier at the axonal bifurcation. Thus, antidromic activation of axon terminals can, through axon collaterals, lead to widespread activation or inhibition of targets remote from the site of stimulation. These effects should be included when interpreting the results of functional imaging or evoked potential studies on the mechanisms of action of DBS.  相似文献

The Carboniferous and Permian of the Baoshan block consist of three major depositional sequences: a Lower Carboniferous carbonate sequence, a Lower Permian siliciclastic sequence, and a Middle Permian carbonate sequence. These three sequences were interrupted by two major regressive events: first, the Namurian Uplift ranging in age from Serpukovian to Gzhelian, and second, the Post-Sakmarian Regression occurring probably at Artinskian time in the Baoshan block, although the precise time interval of the latter event is still unclear. The Baoshan block is characterized by warm-water, highly diverse and abundant faunas during the Early Carboniferous, by cold-water and low diversity faunas during the Early Permian, and by possibly warm-water but low diversity faunas during the Middle Permian. The Sweetognathus bucaramangus conodont fauna constrains the upper boundary of the diamictite-bearing siliciclastic deposits (Dingjiazhai Formation) to the Sakmarian to early Artinskian, as well as the eruption of the rifting basalts (Woniusi Formation) to, at least, the post-early Artinskian. Paleozoogeographically, affiliation of the faunas in the Baoshan block changed from Eurasian in the Early Carboniferous, to Peri-Gondwanan in the Early Permian, and to Marginal Cathaysian/Cimmerian in the Middle Permian. Cimmerian blocks have more or less comparable geohistory to one another in the Carboniferous and Permian. During the Middle Permian, the eastern Cimmerian blocks such as Sibumasu (s.s), Baoshan, and Tengchong are not far from the palaeoequator, but apparently more distant than the western Cimmerian blocks based on the presence or absence of some index taxa such as the fusulinaceans Eopolydiexodina and Neoschwagerina, and the corals Thomasiphyllum and Wentzellophyllum persicum.  相似文献

We present the idea of using multiresponse incomplete block designs when not all responses can be observed in all experimental units. For a special class of such designs, in which partial designs are PBB designs, a method for estimating natural treatment contrast is given. We also consider the problem of testing the hypotheses concerning the natural and any estimable treatment contrasts. For testing this hypothesis the Wald statistics, being asymptotically chi-square distributed, is proposed.  相似文献

Analysis of sample survey data on the species composition of waterfowl kill for resident and nonresident hunters for provinces of Canada during 1974 and 1975 showed that a successful game bird hunter who hunted in more than one area (degree block) had both a higher average season kill and more successful days of hunting in which one or more birds were bagged, than one who hunted in the same area during the season. Adjustment for days hunted resulted in a higher daily hunting for those who hunted in more than one degree block. However, the higher kill of the successful hunter is attributable more to a larger number of days hunted than to a larger kill per successful day. Also, hunters hunting in more than one degree block had their successful hunting days more evenly spread across the season. The estimates of kill per successful hunter and days hunted for stratified sampling by areas of hunting were considerably more efficient than simple random sampling.  相似文献

Structural features of elastic laminae within arteries can provide vital information for both the mechanobiology and the biomechanics of the wall. In this paper, we propose, test and illustrate a new computer-based scheme for automated analysis of regional distributions of elastic laminae thickness, inter-lamellar distances and fragmentation furcation points (FPs) from standard histological images. Our scheme eliminates potential artefacts produced by tissue cutting, automatically aligns tissue according to physiologic orientations and performs cross-sectional measurements along radial directions. A statistical randomised complete block design and F test were used to assess the potential (non)-uniformity of lamellar thicknesses and separations along both radial and circumferential directions. Illustrative results for both normotensive and hypertensive thoracic porcine aorta revealed marked heterogeneity along the radial direction in nearly stress-free samples. Clearly, regional measurements can provide more detailed information about morphologic changes that cannot be gained by globally averaged evaluations alone. We also found that quantifying FP densities offers new information about potential elastin fragmentation, particularly in response to increased loading due to hypertension.  相似文献
