The highly heterogeneous nature of neuronal cell types and their connections presents a major challenge to the characterization of neural circuits at the protein level. New approaches now enable an increasingly sophisticated dissection of cell type- and cellular compartment-specific proteomes, as well as the profiling of the protein composition of specific synaptic connections. Here, we provide an overview of these approaches and discuss how they hold considerable promise toward unravelling the molecular mechanisms of neural circuit formation and function. Finally, we provide an outlook of technological developments that may bring the characterization of synaptic proteomes at the single-synapse level within reach.  相似文献

The current work deals with downstream processing of lactoperoxidase using liquid emulsion membrane from the bovine milk whey, which is a by-product from dairy industry. It is an alternate separation technique that can be used for the selective extraction of lactoperoxidase. The extraction of lactoperoxidase in liquid emulsion membrane takes place due to the electrostatic interaction between the enzyme and polar head group of reverse micellar surfactant. The optimum conditions resulted in 2.86 factor purity and activity recovery of 75.21%. Downstream processing involving liquid emulsion membrane is a potential technique for the extraction of lactoperoxidase from bovine whey.  相似文献

Fruits of Euterpe oleracea (FEO) are currently known as elements that present a very high antioxidant activity (AAO), as measured by the Oxygen Radical Absorbance Capacity. They are particularly rich in total phenolics (TP) and total anthocyanins (TA). Response Surface Methodology was used to optimise the solvent extraction of phenolic antioxidants from FEO, using a second-order polynomial equation to describe the experimental data for TP, TA, and AAO. In order to determine the best solid-to-liquid ratio and time of extraction, some preliminary studies were conducted. A rotatable central composite design with three variables (ethanol proportion, hydrochloric acid concentration and temperature) was then used. The results showed a good fit to the proposed model (R2 > 0.89). TP and TA, as well as TA and AAO, showed significant correlations (P < 0.05). The optimised conditions that maximized the yields of phenolic compounds (TP and TA) and AAO from FEO were: ethanol proportion between 70% and 80%, hydrochloric acid concentration between 0.065 and 0.074 mol/L and a temperature of 58 °C.  相似文献

Algae are considered excellent candidates for renewable fuel sources due to their natural lipid storage capabilities. Robust monitoring of algal fermentation processes and screening for new oil-rich strains requires a fast and reliable protocol for determination of intracellular lipid content. Current practices rely largely on gravimetric methods to determine oil content, techniques developed decades ago that are time consuming and require large sample volumes. In this paper, Nile Red, a fluorescent dye that has been used to identify the presence of lipid bodies in numerous types of organisms, is incorporated into a simple, fast, and reliable protocol for measuring the neutral lipid content of Auxenochlorella protothecoides, a green alga. The method uses ethanol, a relatively mild solvent, to permeabilize the cell membrane before staining and a 96 well micro-plate to increase sample capacity during fluorescence intensity measurements. It has been designed with the specific application of monitoring bioprocess performance. Previously dried samples or live samples from a growing culture can be used in the assay.  相似文献

Summary A quick and simple semi-quantitative method of selecting single seeds of rye for their content of 5-alkyl-resorcinols is presented. The resorcinols are extracted from whole, intact caryopses with acetone. Under the conditions described the germination rate is hardly decreased. The single grain method offers especially striking advantages when selffertile inbred lines are used.  相似文献

In the present research effort, production of crude sericin extracts from Bombyx mori silk cocoons was attempted using two different approaches. Sericin was extracted from cocoons by high-temperature autoclaving followed either by lyophilization or freezing-thawing precipitation, to obtain a crude sericin powder. The physico-chemical and biological characteristics of the crude sericin extracts were evaluated in detail, via FTIR, XRD, XRF, XRT, UV–vis scanning, TGA and DSC, protein quantification, antimicrobial activity, free radical scavenging activity, cytotoxic activity, potential for inducing chromosomal aberrations via Allium cepa assays, and genotoxicity via Comet™ analyses. The molecular weight distribution of the crude sericin extracts was also investigated, via sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE), and the results duly compared to standard sericin. The results gathered clearly suggest that the crude sericin extracts had both obvious radical scavenging effects with the 2.2-diphenyl-1-picryl-hydrazil (DPPH) assay, and antibacterial activity, further suggesting that this protein might be a valuable addition for either food and biopharmaceutical applications.  相似文献

Four hemicelluloses and cellulose fractions were extracted with 10% KOH or 7.5% NaOH at 15°C for 16 h and with 24% KOH or 17.5% NaOH at 15°C for 2 h from defatted, protein and pectin free, lignified or delignified sugar beet pulp (SBP). There was no significant difference in the yield and sugar composition of isolated hemicelluloses and cellulose obtained from four different procedures. 7.5% NaOH extraction at 15°C for 16 h from lignified SBP gave a slightly higher yield of hemicelluloses (10.96%), while 24% KOH extraction at 15°C for 2 h from delignified SBP produced the highest yield of cellulose (18.35%). Molecular-average weights ranged from 88 850 to 91 330 Da for the hemicelluloses obtained from lignified SBP, and 21 620–21 990 Da for the hemicelluloses isolated from delignified SBP. The neutral sugar composition of the hemicelluloses consisted of glucose, arabinose, galactose, xylose, and minor quantities of rhamnose and mannose. The infrared spectra showed an absorption band at 900 cm−1, indicating some amounts of β-linked polysaccharides. Besides ferulic and p-coumaric acids, six other phenolics were also identified in the mixture of alkaline nitrobenzene oxidation of associated lignin in the isolated hemicelluloses and cellulose fractions.  相似文献

Extraction of methane-oxidizing bacteria from soil particles   总被引:7,自引:0,他引:7  
Abstract: We present a method for extraction of active methane (CH4)-oxidizing bacteria from soil samples. The method is based on physical dispersion of bacteria from the soil particles followed by separation of bacteria and soil particles by floatation in the density media Nycodenz or Percoll. Separation on Nycodenz produced very pure bacterial suspensions while separation on Percoll produced rather impure suspensions. However, more than 60% of the methane-oxidizing activity was irreversibly inhibited in the procedure using Nycodenz compared to less than 10% irreversible inhibition when Percoll was employed. The bacterial suspensions extracted from soil can be used to study the physiology and ecology of soil bacteria that oxidize methane at atmospheric concentrations. Our data indicated that these bacteria are extremely difficult to dislodge from particles compared to the majority of bacteria in soil. Tentatively, we interpret the strong attachment to long residence time (i.e. slow turnover) of the methane-oxidizing bacteria. A slow turnover/growth rate would explain why soil disturbances, like cultivation, have a long lasting effect on the oxidation of atmospheric methane in soil.  相似文献

We carried out reactive extractions at moderate temperatures using Rhizopus oryzae resting cells and various solid materials containing different proportions of vegetable oil. Our methodology allows for free fatty acids and either methyl or solketal fatty esters to be prepared in moderate to high yields. Moreover, the process can be carried out in either a solvent or a solvent-free system. In a solvent-free system, the final yield can be increased if easily ground solid material is used. The water present in each material and the solvents used has an influence on the results.  相似文献
