Tall fescue grass cultivars with or without endophytes were evaluated for their susceptibility to Meloidogyne incognita in the greenhouse. Tall fescue cultivars evaluated included, i) wild-type Jesup (E+, ergot-producing endophyte present), ii) endophyte-free Jesup (E-, no endophyte present), iii) Jesup (Max-Q, non-ergot producing endophyte) and iv) Georgia 5 (E+). Peach was included as the control. Peach supported greater (P ≤ 0.05) reproduction of M. incognita than all tall fescue cultivars. Differences in reproduction were not detected among the tall fescue cultivars and all cultivars were rated as either poor or nonhosts for M. incognita. Suppression of M. incognita reproduction was not influenced by endophyte status. In two other greenhouse experiments, host susceptibility of tall fescue grasses to two M. incognita isolates (BY-peach isolate and GA-peach isolate) did not appear to be related to fungal endophyte strain [i.e., Jesup (Max-Q; nontoxic endophyte strain) vs. Bulldog 51 (toxic endophyte strain)]. Host status of tall fescue varied with species of root-knot nematode. Jesup (Max-Q) was rated as a nonhost for M. incognita (BY-peach isolate and GA-peach isolate) and M. hapla, a poor host for M. javanica and a good host for M. arenaria. Bulldog 51 tall fescue was also a good host for M. arenaria and M. javanica, but not M. incognita. Jesup (Max-Q) tall fescue may have potential as a preplant control strategy for M. incognita and M. hapla in southeastern and northeastern United States, respectively.  相似文献

The melanin polyketide synthase (pks) gene of Nodulisporium sp. MF5954 (ATCC74245) was cloned by exploiting its homology to the Colletotrichum lagenarium pks1 gene. Sequence analysis demonstrated that this gene is 70% identical to the C. lagenarium pks1 gene. A gene disruption construct, designed to replace both the ketoacyl synthase and acyl transferase domains with a hygromycin resistance (Hyr) gene, was synthesized, and used to disrupt the Nodulisporium melanin pks1 gene via homologous recombination, resulting in a mel(−) phenotype. Sequence analyses of the gene and of cDNA segments generated by RT-PCR indicate that there are three introns in the 5′ half of the gene. The proposed 2159-amino acid product is 72% identical and 78% similar to the 2187-amino acid sequence deduced from the C. lagenarium pks1 gene. This similarity is notable, considering that C. lagenarium is a member of the order Phyllachoales or Sordariales, whereas Nodulisporium is generally believed to be member of the order Xylariales. However, despite the strong resemblance between the amino acid sequences in the acyl transferase domains of the two proteins, only one in five codons are conserved in the DNA sequences that encode this motif. The Nodulisporium sp. pks1 gene sequence and the amino acid sequence deduced from its coding region have been deposited in Genbank under Accession No. AF151533. 3- Hydroxypropionic acid was isolated by bioactivity-guided fractionation of extracts obtained from submerged cultures of several endophytic fungi isolated from above-ground plant organs. This compound showed selective nematicidal activity against the plant-parasitic nematode Meloidogyne incognita with LD50 values of 12.5-15 microg/ml. Activity against the saprophytic Caenorhabditis elegans was fivefold lower. No antimicrobial, cytotoxic or phytotoxic effects were observed. Propionic acid and D- and L-lactic acids were not active against either nematode species. Based on morphological features and ITS, 18S and 28S rDNA analyses, the producing strains were identified as Phomopsis phaseoli isolated from the leaf of a tropical tree, and four strains of Melanconium betulinum isolated from twigs of Betula pendula and B. pubescens in Germany. This is the first report of 3-hydroxypropionic acid in fungi, and of the nematicidal activity of this metabolite.  相似文献

Numerous species of Ardisia (shrubs in the Myrsinaceae) possess conspicuous bacterial nodules in their leaf margins. This is an obligate, life-cycle symbiosis: the bacteria are maintained in the bud, and re-infect each new leaf primordium, as well as flowers and seeds, and are transmitted vertically to the next generation. Previous studies have shown that treatments which kill the bacteria in the buds lead to death of the plant. This study is the first to test for a net cost or benefit of the nodules in seedling growth capacity. A net benefit of the symbiosis would be expected from the elaborate nodule structure, and also from evolutionary theory. Seedlings of two symbiotic species (A. crenata and A. virens) and two non-symbiotic species (A. elliptica and A. sieboldii) were grown comparatively. For the symbiotic species, performance was assessed for intact plants, for plants with nodules clipped off, and for control plants in which the lamina was clipped between the nodules. The nodules did not contribute to, or detract from, seedling performance in high resource supply. Although plants increased ca. 4- to 6-fold in dry mass, nodule removal had no significant impact on plant growth, gas exchange, biomass allocation, or on foliar concentrations of chlorophyll or of 11 nutrients. No significant advantage was observed for the two symbiotic species over the two non-symbiotic species. The nodules might contribute to growth capacity during other life stages, during resource shortage, or during exposure to specific herbivores or pathogens.  相似文献

Summary A spontaneous variant, obtained from aFrankia isolate fromAlnus rubra nodules, was compared with the parent strain with regard to infectivity, nitrogenase activity, and electrophoretic and immunological profiles. Both the parent and the variant strain were equally effective in inducing nodulation in seedlings ofA. rubra. All inoculated plants had an active nitrogenase system as measured by the acetylene reduction assay. Electrophoresis of whole cell homogenates on SDS-polyacrylamide slab gels showed similar electrophoretic profiles; however, the variant strain also exhibited striking differences in protein patterns that distinguish it from the parent strain. Immunological analysis of the originalFrankia strain and its variant revealed shared antigens as well as immunologically distinct antigenic determinants in the two strains. The variant strain exhibits a distinct morphology and growth patterns which remain stable after many passages through culture.  相似文献

Leaf tissue of tall fescue Festuca arundinacea Schreb., hard fescue Festuca ovina L., red fescue Festuca rubra L. and perennial ryegrass Lolium perenne L. was stained with rose Bengal or aniline blue to detect the presence of the fungal endophyte Acremonium sp., Specimens were cleared using methyl salicylate, an optical clearing agent, and viewed using bright field microscopy. Tissue was presenred as dried tissue or stored in 70% aqueous ethyl alcohol before staining and clearing. Tissue was observed at 2, 4 and 12 weeks following clearing to check for stain retention. Staining with rose Bengal was inferior to aniline blue when followed by the clearing agent methyl salicylate. Fungal mycelia stained lighter with rose Bengal and were more difficult to detect than mycelia stained with aniline blue. The results illustrate the usefulness of combining staining and methyl salicylate clearing for detecting fungal endophytes.  相似文献

【目的】分离产绿原酸的内生菌并对其绿原酸合成途径的一种关键酶基因进行克隆和功能研究。【方法】采用表面消毒法从金银花中分离内生菌,以高效液相色谱(HPLC)和液相色谱-质谱联用(LC-MS)筛选确定产绿原酸的内生菌,克隆、表达该内生菌的组氨酸解氨基酶(HAL)并进行酶活测定。【结果】从金银花根中分离到一株产绿原酸的内生细菌RP1,同时在该内生菌发酵液中检测到了绿原酸代谢的中间物肉桂酸。对RP1的分子鉴定表明该内生菌为枯草芽孢杆菌(Bacillus subtilis)。对RP1的HAL基因进行克隆和原核表达,酶活测定表明该酶具有HAL和PAL (苯丙氨酸解氨基酶)双功能,其PAL活性产生的肉桂酸与LC-MS检测的结果一致。【结论】推测该内生菌可能利用其HAL的苯丙氨酸解氨基活性将苯丙氨酸的催化产物肉桂酸导入苯丙烷途径,从而产生绿原酸。  相似文献

We provide the first analysis of the fungi associated with a very special habitat, the aeroponic roots found in caves and mines in New Brunswick, Canada. Fungal diversity was assessed by Illumina sequencing using three complementary primer sets targeting ribosomal RNA genes, and roots were identified using the non-coding trnH-psbA spacer. Early colonizing ectomycorrhizal fungi such as Agaricales, Helotiales, Pezizales, and Thelephorales were predominant. Saprotrophs, endophytes and plant pathogens were also present, but Glomeromycota (arbuscular mycorrhizal fungi) were not detected. Fungal root communities were generally most similar within sites. Fungal diversity was inversely correlated with winter dark zone temperatures and distance from the entrance. By using a combination of three primer sets, we detected more fungal taxa than with any one primer set. This study adds to the understanding of these subterranean ecosystems and suggests that future studies investigate factors limiting the presence of late-stage ectomycorrhizal fungi and Glomeromycota.  相似文献

In French subalpine grasslands, cessation of mowing promotes dominance of Festuca paniculata, which alters plant diversity and ecosystem functioning. One of the mechanisms underpinning such effects may be linked to simultaneous changes in the abundance of fungal symbionts such as endophytes and arbuscular mycorrhizal fungi. In field conditions, mowing reduced the abundance of the endophyte Neotyphodium sp. in leaves of F. paniculata by a factor of 6, and increased mycorrhizal densities by a factor of 15 in the soil. In greenhouse experiments, the mycorrhizal colonization of Trifolium pratense and Allium porrum increased 3- fold and 3.8- fold respectively in mown vs unmown grassland soil. Significantly reduced growth of the two host plants was also observed on soil from the unmown grassland. Such opposite effects of mowing on the two functional groups of fungal symbionts could suggest interactions between these two groups, which in turn could contribute to structuring plant communities in subalpine grasslands.  相似文献
