山黧豆属(又名香豌豆属)(Lathyrus Linn.)全世界共约140种,已做过染色体计数者72种。我国约16种,大部分产西南、西北和东北各省区。近年来皖南农学院朱玉简等同志在安徽宣城发现一新种,命名为安徽山黧豆(Lathyrus anhuiensis Y.J.Zhu et R.X.Meng)。鉴于该新种分布区非常局限,个体数也不多,为了及时掌握其细胞学情况,特通过该院叶如欣同志取得部分种子进行了染色体的计数和核型的描述。  相似文献

近年在金沙江干热河谷及其邻近地区陆续发现的苏铁天然植物群落,为本属植物在欧亚大陆内陆分布的最北边缘。其中,面积最大和保存较好的是渡口市攀枝花苏铁(Cycas panzhihuaensis)植物群落。本文研究了这个群落的分布与生境、生长状况、外貌、种类组成以及群落的综合特征等。认为具有稀树草原的特点,而这里的稀树草原是次生性质的,苏铁群落是其中幸存的残遗植被类型。这对研究我国植物地理有着重要意义。  相似文献

云南兔Lepus comus G.Allen是云贵高原和滇西山地目前发现的唯一的一种野兔,初由G.M.Allen(1927)根据采自云南西部腾冲的标本命名。其后,国内外学者(包括G.M.Allen本人在内)对云南兔究竟是一独立种或为高原兔Lepus oiostulus(Hodgson)的一个地理亚种意见纷纭(罗泽珣,1981;高耀亭等,1964;Allen,1938;Ellerman et al.,1951;Angermann,1967;Corbet,1978。我们曾对国内有关单位所收藏的这两个类群的100余号标本进行较详细的对比研究。发现云南兔在形态上与高原兔确有明显区别,在分布上又有同域现象,因而主张仍应将云南兔保持其原定种的种级地位;同时,又查对了采于各地的云南兔所有标本,发现云南北部(丽江)至中部(景东、景谷)标本以及云南东部、南部和贵州西部标本都有异于滇西地模标本,且占有不同的地理分布区。经研究,应属两个新的亚种。其中,云南东部,南部和贵州西部的新亚种,为纪念先师彭鸿绶先生毕生献身于云、贵、川的科学考察事业和他的指导与培养,特以彭先生之姓氏命名,以资纪念。  相似文献

The lysosomal enzyme alpha-mannosidase-1 is one of the earliest developmentally controlled gene products in Dictyostelium discoideum. Although this enzyme is synthesized throughout the first 20 h of development, it is not required for complete morphogenesis, since structural gene (manA) mutants lacking activity develop normally. We isolated six strains deficient in alpha-mannosidase-1 activity which, unlike structural gene mutants, fail to aggregate. Fruiting revertants of these strains accumulate wild-type levels of alpha-mannosidase-1 activity, suggesting that both the enzymatic and morphological defects are caused by single mutations in nonstructural genes essential for early development. Direct genetic evidence for mutations outside of the structural gene was obtained by complementation analysis. We used alpha-mannosidase-1-specific monoclonal antibodies to analyze the biochemical defects in these mad (alpha-mannosidase-1-deficient) mutants. All mad mutants show a significantly reduced relative rate of enzyme precursor biosynthesis. The mad-404 mutation results in a complete lack of precursor biosynthesis, as well as a lack of functional alpha-mannosidase-1 mRNA. In some cases, however, the enzymatic defect results from improper post-translational modification which affects precursor processing. We conclude that a small number of aggregation-essential genes are involved in regulating the synthesis, modification, and processing of alpha-mannosidase-1 during development.  相似文献

Borrelia hermsii, an agent of relapsing fever, undergoes antigenic variation in its host. Surface-exposed proteins with differing primary structures determine the serotype of each organism. Using amino acid sequence data from two of these variable proteins, we synthesized two mixed-sequence oligonucleotides and then used the oligonucleotides to probe mRNA and DNA of three isogenic serotypes of B. hermsii. In Northern blots the probes were specific for the mRNA of the homologous serotype. Southern blots revealed two classes of hybridizing fragments: those common to the three serotypes and those specific for a particular serotype. A serotype-specific DNA fragment, which had hybridized to both oligonucleotide probes, was cloned. Subsequent use of the cloned fragment as a probe provided further evidence that antigenic variation in B. hermsii is associated with DNA rearrangements and with occurrence of expression-linked copies of all, or part, of an antigen-specifying gene.  相似文献

The distance geometry approach for computing the tertiary structure of globular proteins emphasized in this series of papers (Goelet al., J. theor. Biol. 99, 705–757, 1982) is developed further. This development includes incorporation of some secondary structure information—the location of alpha helices in the primary sequence—in the algorithm to compute the tertiary structure of alpha helical globular proteins. An algorithm is developed which estimates the interresidue distances between chain-proximate helices. These distances, in conjunction with the global statistical average distances obtainable from a database of real proteins and determined by the primary sequence of the protein under study, are used to determine the tertiary structure. Five proteins, parvalbumin, hemerythrin, human hemoglobin, lamprey hemoglobin, and sperm whale myoglobin, are investigated. The root mean square (RMS) errors between the calculated structures and those determined by X-ray diffraction range from 4.78 to 7.56 Å. These RMSs are 0.21–2.76 Å lower than those estimated without the secondary structure information. Contact maps and three-dimensional backbone representations also show considerable improvements with the introduction of secondary structure information.  相似文献

The rate constants for the reversible addition of protons and sulfite to the 5,6 double bond of cytidine and 3-methylcytidine have been spectrophotometrically measured under conditions (25°C, μ = 1.0 ) where the deamination of 5,6-dihydrocytidine-6-sulfonate is minimal. Both the addition and the elimination of sulfite from the ring system are subject to general catalysis of proton transfer. For the reaction in either direction, plots of the pseudo-firstorder rate constants against increasing buffer concentration are biphasic and indicative of at least a two-step reaction pathway with both steps being subject to general acid-base catalysis. Kinetic hydrogen-deuterium isotope effects were measured for both buffer-catalyzed steps of sulfite elimination from 3-methyl-5,6-dihydrocytidine-6-sulfonate and sulfite addition to 3-methylcytidine. Both H2O and D2O were used as solvent. For both the addition and the elimination of SO32− values of k2H/k2D were 6.3–7.1 and 2.3–2.6 at low and high imidazole buffer concentration, respectively. The large isotope effects values in the range of 6–7 can be attributed to rate-determining proton transfer to carbon-5 of the cytidine ring system. The smaller values are more likely caused by proton transfer to a electronegative atom such as the oxygen on carbon-2 of the cytidine ring. The equilibrium constants for bisulfite buffer addition to 3-methylcytidine and cytidine at 25°C, μ = 1.0 , pH 7.2, are 10.2 and 1.3 −1, respectively.  相似文献

Abstract. In controlled environments, the interactive effects of warm (16: 8°C, day: night) and cool (12: 4°C, day: night) temperatures and long (13.5 h) and short (10 h) photoperiods on the dehardening of seedlings of Pinus radiata D. Don were investigated. In another experiment, the effect of four photoperiods from 9 to 14 h was examined. In a third, dehardening at constant temperatures from 5 to 17°C was followed. There was no evidence for an interaction between photoperiod and temperature. Dehardening was temporarily delayed by photoperiods below about 10 h, but there was no other quantitative effect of photoperiod. At constant temperatures, the rate of dehardening was initially constant but declined as the minimum summer frost hardiness was reached. In the initial phase the rate of dehardening was a linear function of temperature, increasing from 0.05°C day−1 at 8°C to 0.30 °C day−1 at 17°C. Temperature controlled the loss of frost hardiness by regulating the rate of dehardening.  相似文献

Abstract. Coleus blumei Benth. (PI No. 354190), a green-leafed cultivar, was exposed to 5°C for 48 or 72 h after pretreatment for 48 h at two levels of photosynthetically active radiation (PAR) (8 and 320 μmol s−1 m−2), two temperatures (13 and 20°C), and two abscisic acid (ABA) levels (0 and 200 g m−3 of the racemic mixture). Plants given low PAR for only 48 h prior to chilling treatment (48 or 72 h at 5°C) showed increased protection against chilling injury while those given high PAR were severely injured. The former plants were darker green, contained greater concentrations of chlorophyll- a , chlorophyll- b , total chlorophyll and anthocyanin and generally had a lower abscission rate than the latter plants. There were no differences, however, in chlorophyll- a/b ratio among plants grown at the two PAR levels, two temperatures or two ABA concentrations. Temperature and ABA pretreatment and number of hours at 5°C had no significant effect on chilling injury as measured by leaf chlorosis, but generally had a significant effect on leaf abcission, especially at 3 and 7 d after returning the plants to the greenhouse. Enclosing intact plants or excised shoots in plastic bags to maintain 100% relative humidity during 72 h chilling treatment failed to provide protection against chilling injury. These findings indicate that the protective effects of low PAR applied prior to chilling treatment may be as important or more important than that applied during chilling. They also indicate the importance of making careful measurements of PAR levels when conducting studies on chilling injury.  相似文献

Abstract Chenopodium album L. plants, grown under controlled environmental conditions on different levels of soil nitrate, produced seeds with proportionately different NO?3 contents. Regardless of the endogenous NO?3 content, few seeds germinated in water or upon treatment with KNO3. Ethylene promoted germination, and the extent of germination was positively correlated with the endogenous seed NO?3 content. Combined application of ethylene and KNO3 in the dark had a synergistic effect on NO?3 -deficient seed. The synergism between ethylene and KNO3 was attributable to the NO?3 moiety of the nitrate salt. Ethylene and light showed moderate synergism in seeds with low or high endogenous nitrate. Addition of nitrate, however, masked the interaction between ethylene and light. Gibberellic acid4+7 (GA4+7) or red light, each alone or combined with KNO3, had little effect on germination. When applied together in the dark, ethylene and GA4+7 synergistically enhanced the germination of NO?3-deficient seed. The combined effects of the two hormones on this seed were further enhanced by the addition of KNO3. There was no synergism between ethylene and GA4+7 in NO?3-rich seed. These interactions among GA4+7, ethylene and KNO3 were not affected by light. The results confirm and further elaborate our earlier finding that the sensitivity of C. album seeds to ethylene may depend on nitrate availability.  相似文献
