23S,25-Dihydroxyvitamin D3 was isolated from the plasma of vitamin D3-toxic pigs. An ultraviolet absorbance spectrum confirmed its purity. The configuration of the 23-hydroxyl group was determined to be S by comparison of the natural product with synthetic 23R,25- and 23S,25-dihydroxyvitamin D3 by high-pressure liquid chromatography. The affinity of both 23S,25- and 23R,25-dihydroxyvitamin D3 for the plasma vitamin D binding protein was similar to vitamin D3. Thus, with respect to the plasma vitamin D binding protein, 23S,25-dihydroxyvitamin D3 is the least potent, naturally-occurring, dihydroxylated vitamin D3 metabolite known.  相似文献

1,25-Dihydroxyvitamin D3 [1,25-(OH)2D3] receptor was characterized after partial purification of thymus cytosol by ammonium sulfate fractionation. The 1,25-(OH)2D3 receptor sediments at 3.7S in 5–20% sucrose gradients. The binding of 1,25-(OH)2D3 in thymic cytosol was a saturable process with high affinity (Kd = 0.12?0.48 nM) at 4°C. Competition for 1,25-(OH)2[3H]D3 receptor by nonradioactive analogs demonstrated the affinities of these analogs to be in order; 1,25-(OH)2D3 = 1,24R,25-(OH)3D3 = 1,25S,26-(OH)3D3 = 1,25R,26-(OH)3D3 > 1,25-(OH)2D3-26,23 lactone > 25-OHD3 > 23R,25-(OH)2D3 > 24R,25-(OH)2D3 > 23S,25-(OH)2D3 ? 25-OHD3-26,23 lactone. The receptor bound to DNA cellulose columns in low salt buffer and eluted as a single peak at 0.21 M KCl. These findings provide evidence that the thymus possesses a 1,25-(OH)2D3 receptor with properties indistinguishable from 1,25-(OH)2D3 receptors in other tissues.  相似文献

本文对我国无锡分离的兔出血症病毒A_2R-3毒株核酸的某些理化性质进行了研究。采用孚尔根染色、二苯胺反应和核酸酶解实验证实病毒核酸为DNA类型。吖啶橙染色、甲醛反应、核酸酶S_1消化和核酸热变性实验表明病毒核酸为单链型。核酸电泳呈单一组分。电境观察显示核酸分子链呈线状,平均长度约为2.15μ。计算分子量约为2.1—2.5×10~6d。核酸碱基组盛为A25.34、T29.37、G23.85、C21.43、(G C)克分子百分比值为45.28。结合以前的报道、我们认为:兔出血症病毒可以归类于细小病毒科。  相似文献

Emphasis has increased on accuracy in predicting the effect that anthropogenic stress has on natural ecosystems. Although toxicity tests low in environmental realism, such as standardized single species procedures, have been useful in providing a certain degree of protection to human health and the environment, the accuracy of such tests for predicting the effects of anthropogenic activities on complex ecosystems is questionable. The use of indigenous communities of microorganisms to assess the hazard of toxicants in aquatic ecosystems has many advantages. Theoretical and practical aspects of microbial community tests are discussed, particularly in related to widely cited problems in the use of multispecies test systems for predicting hazard. Further standardization of testing protocols using microbial colonization dynamics is advocated on the basis of previous studies, which have shown these parameters to be useful in assessing risk and impact of hazardous substances in aquatic ecosystems.  相似文献

Mangrove ecosystems occupy different locations on Caribbean island coasts, ranging from open bays (fringe mangals) to totally enclosed salt ponds and salinas. On geomorphologically active coastlines, such as south Jamaica, systems are at varying degrees of maturity and productivity. Furthermore, because of system variability, the interactions between mangroves and associated marine systems, such as coral reefs and seagrass beds, are developed to different degrees.Community structure and productivity of a range of mangals on different islands of the Greater and Lesser Antilles are discussed. Forcing functions and levels of interaction with the marine environment are identified.The rational choice of management options must be based on the range of goods and services provided by the different systems; and a good understanding of their ecology is essential when choosing sites for protection, waste disposal, landfill, marina development and fisheries enhancement. Examples are given from current studies in Jamaica, St. Lucia, the British Virgin Islands and Trinidad, of a flexible management response to mangrove ecosystem diversity.  相似文献

Summary We have constructed a yeast artificial chromosome (YAC) library of tomato for chromosome walking that contains the equivalent of three haploid genomes (22 000 clones). The source of high molecular weight DNA was leaf protoplasts from the tomato cultivars VFNT cherry and Rio Grande-PtoR, which together contain loci encoding resistance to six pathogens of tomato. Approximately 11 000 YACs have been screened with RFLP markers that cosegregate withTm-2a andPto — loci conferring resistance to tobacco mosaic virus andPseudomonas syringae pv.tomato, respectively. Five YACs were identified that hybridized to the markers and are therefore starting points for chromosome walks to these genes. A subset of the library was characterized for the presence of various repetitive sequences and YACs were identified that carried TGRI, a repeat clustered near the telomeres of most tomato chromosomes, TGRII, an interspersed repeat, and TGRIIl, a repeat that occurs primarily at centromeric sites. Evaluation of the library for organellar sequences revealed that approximately 10% of the clones contain chloroplast sequences. Many of these YAC clones appear to contain the entire 155 kb tomato chloroplast genome. The tomato cultivars used in the library construction, in addition to carrying various disease resistance genes, also contain the wild-type alleles corresponding to most recessive mutations that have been mapped by classical linkage analysis. Thus, in addition to its utility for physical mapping and genome studies, this library should be useful for chromosome walking to genes corresponding to virtually any phenotype that can be scored in a segregating population.  相似文献

Forty pregnant long-tailed macaques were treated daily for 30 d with 0, 25, 150 or 300 μg selenium as L-selenomethionine/kg body weight. Erythrocyte and plasma selenium and glutathione peroxidase specific activities, hair and fecal selenium, and urinary selenium excretion were increased by and were linearly related to L-selenomethionine dose. Hair selenium was most sensitive to L-selenomethionine dose, with an 84-fold increase in the 300 μg selenium/(kg-d) group relative to controls (r=0.917). Daily urinary selenium excretion (80-fold,r=0.958), plasma selenium (22-fold,r=0.885), erythrocyte selenium (24-fold,r=0.920), and fecal selenium (18-fold,r=0.911) also responded strongly to L-selenomethionine. Erythrocyte and plasma glutathione peroxidase specific activities increased 154% and 69% over controls, respectively. Toxicity was associated with erythrocyte selenium >2.3 μg/mL, plasma selenium >2.8 μg/mL, and hair selenium >27 μg/g. Plasma, erythrocyte, and hair selenium concentrations may be useful for monitoring and preventing the toxicity of L-selenomethionine administered to humans in cancer chemoprevention trials.  相似文献

The partitioning of trace metals and hydrophobic organic contaminants to phytoplankton determines their toxicity as well as their fate and transport in aquatic ecosystems. Accurate impact assessments, therefore, depend on a good understanding of the factors regulating the sorption of these compounds to biotic particles. The accumulation of chlorinated organic compounds in phytoplankton is generally considered as being due solely to physical sorption, described by reversible equilibrium models based on Langmuir or Freundlich isotherms. On the other hand, the uptake of trace metals is a two phase process: a fast sorption component viewed as an ionexchange or a covalent bonding process with cell surface ligands, followed by an intracellular transport phase that is dependent on cellular metabolic activity. The uptake of inorganic and hydrophobic organic pollutants and their bioaccumulation are influenced in a complex manner by duration of exposure and cell density, by environmental factors such as pH, the concentration of cations and of dissolved and colloidal organic matter, as well as by phytoplankton physiological condition. High concentrations of H+, Ca2+, and Mg2+ ions will reduce trace metal sorption by directly competing for uptake sites on the cell's surface, whereas the presence of dissolved organic carbon such as natural and synthetic chelators and phytoplankton exudates will reduce the bioavailability of both trace metals and hydrophobic organic contaminants. Thus, the impact of toxic contaminants on phytoplankton may be determined as much by the factors influencing uptake and partitioning as by the potency of the toxicants and interspecies differences in sensitivity. Recommendations for improving toxicity assessments are presented.  相似文献
