We present a fluorescence‐lifetime based method for monitoring cell and tissue activity in situ, during cell culturing and in the presence of a strong autofluorescence background. The miniature fiber‐optic probes are easily incorporated in the tight space of a cell culture chamber or in an endoscope. As a first application we monitored the cytosolic calcium levels in porcine tracheal explant cultures using the Calcium Green‐5N (CG5N) indicator. Despite the simplicity of the optical setup we are able to detect changes of calcium concentration as small as 2.5 nM, with a monitoring time resolution of less than 1 s. Liver fibrosis occurs in most cases of chronic liver disease, which are somewhat common, but also a potentially deadly group of diseases. In vitro modeling of liver fibrosis relies primarily on the isolation of in vivo activated hepatic stellate cells (aHSCs) and studying them in standard tissue culture dishes (two-dimensional [2D]). In contrast, modeling of fibrosis in a biofabricated three-dimensional (3D) construct allows us to study changes to the environment, such as extracellular matrix (ECM) composition and structure, and tissue rigidity. In the current study, we used aHSCs produced through subcultures in 2D and encapsulated them in a 3D collagen gel to form spherical constructs. In parallel, and as a comparison, we used an established HSC line, LX-2, representing early and less severe fibrosis. Compared with LX-2 cells, the aHSCs created a stiffer environment and expressed higher levels of TIMP1 and LOXL2, all of which are indicative of advanced liver fibrosis. Collectively, this study presents a fibrosis model that could be incorporated with multi-cellular models to more accurately reflect the effects of a severe fibrotic environment on liver function. 相似文献

The role of endocellulases and endoxylanase during liquefaction and saccharification of hydrothermally pretreated wheat straw was studied. The use of a flow‐loop setup with in‐line magnetic resonance imaging enabled frequent measurements of viscosity at 55°C during saccharification at 6% total solids content. Viscosity data were complemented with off‐line measurements of fiber lengths and release of soluble sugars. A clear correlation between fiber attrition and a decrease in viscosity was found. Fiber lengths and viscosity dropped quickly within the first hour and then stagnated, while sugar yields increased substantially thereafter, illustrating that liquefaction and saccharification are separate mechanisms. Both endoglucanase and endoxylanase were shown to have a significant effect on viscosity during liquefaction while the addition of endoxylanase also increased sugar yield. © 2014 American Institute of Chemical Engineers Biotechnol. The need for the extensive use of sustainability assessment as a standalone tool to evaluate the environmental, economic and social aspects of an activity be it at project, product, company or region level has resulted in the development of various methods and standards. There are several indicator issues to address each aspect of sustainability and it is not easy for decision makers to understand the result due to the use of multiple indicators. In this regard, the paper aims at the identification and combination of indicators allowing to assess the sustainability which is applicable to a carbon fiber recycling sector. Indicators selection were carried out by performing an extensive literature review on existing publications dealing with the different pillars of sustainability and setting a number of selection criteria to prioritize indicators that are relevant to the sector. For the environmental aspect global warming, acidification and human toxicity seem to be the most relevant. The social-economic aspect can be addressed through considering the resource impact assessment by considering the supply risk due to the geological scarcity of a resource and the potential supply disruption due to geopolitical and other social factors. The finding shows that three indicators have been identified enabling the assessment of the environmental pillar. Then the necessity to use extra resources indicators was shown and justified by the need of providing a shorter timeframe perspective as well as to consider the amount of fiber to be recycled in the future and also to determine the potential benefit provided by the creation of this sector to the resource strategy point of view. This will be made possible by using such method as the criticality assessment that enable the consideration of geological and geopolitical supply risk as well as the characterization of the system dependence to a specific resource.Finally, these results lead to the expression of the need to the development of a novel indicator assessing the criticality of carbon fibers as well as the expression of the necessity for further research on the socio-economic perspectives. 相似文献
