The aim of this study was to investigate the composition of microcosm denture plaque biofilms and the susceptibility of Candida spp. within these biofilms to antifungal agents. An in vitro model was employed to grow oral biofilms derived from denture associated stomatitis (DAS) patient samples to assess fungal growth in the presence and absence of antifungal agents. The compositions of genera present in vitro were found to be similar to those exhibited on the mucosa and denture fitting surfaces of DAS samples. Exposure to single agents, e.g., miconazole, fluconazole or chlorhexidine did not inhibit growth of Candida spp. when used in clinically relevant doses. Combinations of miconazole and chlorhexidine, pulsed into the system to mimic patient use, did reduce bacterial and candidal growth for several days. Hence, the use of dual-therapy appeared to be useful in reducing the number of viable organisms within denture plaque grown in vitro although resistance to these agents was also evident.  相似文献

Mussel adhesive proteins (MAPs) attach to all types of inorganic and organic surfaces, even in wet environments. MAP of type 5 (fp-5), in particular, has been considered as a key adhesive material. However, the low availability of fp-5 has hampered its biochemical characterization and practical applications. Here, soluble recombinant fp-5 is mass-produced in Escherichia coli. Tyrosinase-modified recombinant fp-5 showed ~1.11 MPa adhesive shear strength, which is the first report of a bulk-scale adhesive force measurement for purified recombinant of natural MAP type. Surface coatings were also performed through simple dip-coating of various objects. In addition, complex coacervate using recombinant fp-5 and hyaluronic acid was prepared as an efficient adhesive formulation, which greatly improved the bulk adhesive strength. Collectively, it is expected that this work will enhance basic understanding of mussel adhesion and that recombinant fp-5 can be successfully used as a realistic bulk-scale bioadhesive and an efficient surface coating material.  相似文献

Infection of nematodes byDactylaria haptotyla, a nematode-trapping hyphomycete, was studied by electron microscopy. The cytoplasm of the adhesive knob in the fungus contained a number of electron-dense, membrane-bound vesicles, 0.2–0.5 µm in diam. The vesicles were rarely seen in the stalk cell or vegetative cell cytoplasm. When the adhesive knob came into contact with the nematode's cuticle, it secreted an adhesive which was seen in ultrathin sections between the knob and the cuticle as an amorphous mass. At the same time, electron-dense vesicles in the cytoplasm were reduced in number and many small vacuoles developed. Soon after capture of a nematode, the cell wall of the adhesive knob became obscure at the prospective site of penetration, where a vesicle, 0.7 µm in diam, was found in serial thin sections of the knob's cytoplasm. At the site facing the vesicle, the peripheral part of the nematode's cell exhibited a high electron density. The vesicle, which appeared to be derived from smaller electron-dense vesicles coalesced with each other, released its enzymic contents toward the captured nematodes before penetration by the fungus.  相似文献

The localization of specific keratin‐associated beta‐proteins (formerly referred to as beta‐keratins) in the embryonic epidermis of lizards is not known. Two specific keratin‐associated beta‐proteins of the epidermis, one representing the glycine‐rich subfamily (HgG5) and the other the glycine‐cysteine medium‐rich subfamily (HgGC10), have been immunolocalized at the ultrastructural level in the lizard Anolis lineatopus. The periderm and granulated subperiderm are most immunonegative for these proteins. HgG5 is low to absent in theOberhäutchen layer while is present in the forming beta‐layer, and disappears in mesos‐ and alpha‐layers. Instead, HgGC10 is present in the Oberhäutchen, beta‐, and also in the following alpha‐layers, and specifically accumulates in the developing adhesive setae but not in the surrounding cells of the clear layer. Therefore, setae and their terminal spatulae that adhere to surfaces allowing these lizards to walk vertically contain cysteine–glycine rich proteins. The study suggests that, like in adult and regenerating epidermis, the HgGC10 protein is not only accumulated in cells of the beta‐layer but also in those forming the alpha‐layer. This small protein therefore is implicated in resistance, flexibility, and stretching of the epidermal layers. It is also hypothesized that the charges of these proteins may influence adhesion of the setae of pad lamellae. Conversely, glycine‐rich beta‐proteins like HgG5 give rise to the dense, hydrophobic, and chromophobic corneous material of the resistant beta‐layer. This result suggests that the differential accumulation of keratin‐associated beta‐proteins over the alpha‐keratin network determines differences in properties of the stratified layers of the epidermis of lizards. The adsorption behavior of the mussel adhesive protein Mytilus edulis foot protein‐1 (Mefp‐1) has been investigated on a negatively charged polar SiO2 surface and an electrically inert non‐polar CH3‐terminated thiolated gold surface. How the structure of adsorbed Mefp‐1 is changed upon chemically and enzymatically induced cross‐linking using sodium periodate (NaIO4) and catechol oxidase, both of which transform DOPA residues in Mefp‐1 into highly reactive o‐quinones, was also investigated. The results are compared with those resulting from addition of Cu2+ to adsorbed Mefp‐1, which forms complexes with and catalyses oxidation of DOPA residues, previously suggested to participate in the cohesive and adhesive properties of the byssus thread of M. edulis. By combining surface plasmon resonance (SPR) and quartz crystal microbalance/dissipation (QCM‐D) measurements, the effects of these agents were investigated with respect to changes in the amount of coupled water, the viscoelastic properties (rigidity) and the hydrodynamic thickness of the protein adlayers. The layer of Mefp‐1 formed on the bare CH3‐terminated surface was elongated, flexible and coupled hydrodynamically a substantial amount of water, whereas Mefp‐1 formed a rigidly attached adlayer on the SiO2 surface. Upon enzymatically and chemically induced cross‐linking of Mefp‐1 formed on the CH3 surface, the rigidity of the adlayer(s) increased significantly. A similar increase in the rigidity was observed also upon addition of Cu2+, suggesting that the high level of metal ions present in the byssus thread might be essential for the cohesive and adhesive properties of this protein. For the mass‐uptake kinetics of enzymatically induced cross‐linking, three different phases were observed and are interpreted as competition between binding of protein and release of coupled water. For the reaction with NaIO4 and Cu2+, only release of water affected the coupled mass. The importance of this type of information for an improved understanding of the strong adhesion and cohesive properties in marine environments is discussed.  相似文献

Diatoms are a major component of microbial slimes that develop on man-made surfaces placed in the marine environment. Toxic antifouling paints, as well as environmentally friendly, fouling-release coatings, tend to be effective against most fouling organisms, yet fail badly to diatom slimes. Biofouling diatoms have been found to tenaciously adhere to and colonise even the most resistant of artificial surfaces. This review covers the basic biology of fouling marine diatoms, their mechanisms of adhesion and the nature of their adhesives, as well as documenting the various approaches that have been utilised to understand the formation and maintenance of diatom biofouling layers.  相似文献

为研究白蚁饵料成型工艺,比较了9种胶黏剂对白蚁纤维素饵料成型效果、耐水性能以及对白蚁取食的影响。结果表明:20%、40%剂量糊精;50%、100%剂量三聚氰胺甲醛树脂,10%、20%、40%田菁胶、卡拉胶、壳聚糖、明胶;10%、50%、100%聚乙烯醇、硅酸钠对微晶纤维素的成型效果较好,经上述剂量胶黏剂处理后,纤维素饵块的邵氏硬度(HD)极显著高于对照。耐水性能试验至第30天时,50%聚乙烯醇、100%聚乙烯醇、100%三聚氰胺甲醛树脂处理的纤维素饵块的溃散程度指数分别为1.33、1.00、2.00,其余饵块的溃散程度指数均达3级。生测结果显示,在7 d的室内强迫取食试验中,白蚁对50%聚乙烯醇、100%聚乙烯醇、100%三聚氰胺甲醛树脂处理的块状纤维素饵料的取食率均极显著低于对照,说明饵块中添加的上述胶黏剂对白蚁的取食具有一定的影响。综上,50%聚乙烯醇、100%聚乙烯醇、100%三聚氰胺甲醛树脂适用于白蚁纤维素饵料成型,但若想获得白蚁喜食的饵块仍需对配方做进一步的优化。  相似文献

Among the previously established enzyme-linked immunosorbent assays (ELISAs), an ELISA using the full length of a recombinant thrombospondin-related adhesive protein of Babesia gibsoni (rBgTRAPf) is considered as the most sensitive diagnostic method for the detection of an antibody to B. gibsoni in dogs. However, the expression of rBgTRAPf in high concentration is poor and, thus, limits its usefulness as a diagnostic antigen. To improve its expression level, we have truncated BgTRAPf into two fragments having either an N- or a C-terminus (BgTRAPn or BgTRAPc, respectively). The expression of BgTRAPc protein in Escherichia coli yielded adequate recombinant protein. The specificity and sensitivity of ELISAs with the truncated proteins were determined using dog sera experimentally infected with B. gibsoni and specific pathogen-free (SPF) dog sera. A total of 254 field dog sera were examined by the ELISA with rBgTRAPn, rBgTRAPc, and rBgTRAPf as well as by an indirect fluorescent antibody test (IFAT). The specificity of rBgTRAPc was the highest (97.15%), and its kappa value was more (0.8003) than rBgTRAPn (0.7083). With a sufficient level of expression as well as higher specificity and reliable sensitivity, rBgTRAPc appears to be a potential candidate antigen for the serodiagnosis of B. gibsoni infection in dogs.  相似文献

应用扫描电镜观察七星瓢虫Coccinella septempunctata的唇须和附节,以便确认在感应农药残留时起主要作用的化学感受器。研究证明分布在下颚须上的感器类型包括两种锥形感器(兼有机械感受器和化学感受器的功能)和一种钟形感器。同时本文首次报道了对这些感器精确的形态测量,明确了其性二型性。对下颚须的测量表明其左右末节的长度差异与七星瓢虫的性别之间有明显的相关性。文中还记述了下唇须上一种新的钟形感器,其在每个下唇须上的数量大约为12~17个。经测量这些机械感受器的平均直径为2.4 μm。早期研究中所记述的锥形感器被证实是下唇须上仅有的化学感应器,并且首次证明这些锥形感器的数量与性别相关,雄性平均为18个,雌性平均为16个。附节上也存在性别差异: 第2跗节跗垫的宽度在雄性中是从后向前逐渐增大,而在雌性中刚好相反。本文还就这些差异的意义以及新发现感器的功能进行了讨论。  相似文献

The southern green stink bug Nezara viridula L. (Heteroptera, Pentatomidae) is highly polyphagous, preferring apically situated seeds and fruits on more than 150 plant species belonging to over 30 plant families all over the world. This forces them to move over highly variable terrains, including plant stems, leaves, pods and buds, which requires efficient attachment. Stink bugs have long slender legs and feet (tarsi) equipped with paired curved claws, paired soft adhesive pads (pulvilli), and flattened lanceolate hairs (setae), which arise ventrally on the first and second foot segments (tarsomeres). To characterize their attachment abilities on well‐defined test substrates, here we comparatively measured and analyzed the traction forces of bugs walking horizontally and vertically on hydrophilic (water attractive) and hydrophobic (water repellent) glass plates and rods. The latter correspond to the geometry of preferred feeding sites of stink bugs in the field. The results show a clear contribution of tarsal flattened lanceolate hairs to the stink bug's attachment. Higher traction forces are generated on a glass rod than on a glass plate, corresponding to up to individual maximum of 43 times the stink bug's body weight. Substrate hydrophobicity promotes the attachment, while the measured forces are up to eight times lower when tarsal hairs are disabled. The combination of smooth and hairy tarsal pads results in a remarkable attachment ability, which enables N. viridula to climb unstable apical plant parts, and supports their invasive behavior and global dispersion.  相似文献
