Adrenergic and cholinergic mediated glucose oxidation by rat parotid gland acinar cells during aging

A₂-phospholipase from Crotalus Adamanteus venom has been assayed using the fluorescent probe 2-all trans-parinoyllecithin. The hydrolysis of the lecithin in an “albumin-rich” incubation medium is paralleled with a fluorescence hyperpolarization. The mechanism of the hyperpolarization is discussed. The application of the method to assay phospholipases, allows detection of an activity under 1 nanomole/min and continuous monitoring of the hydrolysis.     

Heme complexes of rabbit hemopexin,human hemopexin and human serum albumin: Electron spin resonance and Mőssbauer spectroscopic studies

The electron spin resonance (ESR) spectra of human and rabbit ferriheme-hemopexin complexes at 30^oK show an ESR absorption characterized by g_x = 1.60, g_y = 2.25 and g_z = 2.86, characteristic of low-spin ferriheme-proteins. The low-spin nature of the heme-iron in heme-hemopexin is corroborated by the observation of nuclear hyperfine splitting in M?ssbauer spectra at 4.2^oK. The similarity of the ESR spectra of ferriheme-hemopexin with those of low-spin cytochromes which coordinate heme via two histidine residues points to a similar coordination mechanism in hemopexin. In contrast, the ESR spectra of the 1:1 and 2:1 complexes of heme with human serum albumin display signals at g = 6.0 and g = 2.0, characteristic of high-spin ferrihemeproteins.     

Cyclic AMP potentiates down regulation of epidermal growth factor receptors by platelet-derived growth factor

Pretreatment of $\text{Balb}\text{c}\text{-3T3}$ cells with platelet-derived growth factor (PDGF) has been shown to decrease binding sites for ¹²⁵I-labelled epidermal growth factor (EGF) (1,2,3). Agents which elevate cellular cyclic AMP concentrations enhance this ability, and the change in EGF binding is inversely proportional to the elevation of cyclic AMP. In quiescent density arrested cells, the sensitivity of cells to down regulation of EGF receptors by PDGF is proportional to the cyclic AMP content of the cultures in three different cell lines. Agents which elevate cyclic AMP and which potentiate PDGF mediated heterologous down regulation of EGF receptors are able, like cholera toxin (3), to stimulate cells to synthesize DNA in defined medium in the absence of EGF. Down regulation of EGF receptors by PDGF in combination with agents elevating cyclic AMP effectively mimics the action of EGF.     

Expression profiles of genes involved in apoptosis and selenium metabolism in articular cartilage of patients with Kashin–Beck osteoarthritis

Kashin–Beck disease (KBD) is a special type of endemic osteoarthritis. It has been suggested that alterations in selenium metabolism and apoptosis play a role in KBD. However, the underlying molecular mechanism remains largely unclear. We performed a microarray analysis using RNA isolated from cartilages of KBD patients and healthy controls, through Significance Analysis of Microarray (SAM) software. Functional gene networks and crucial molecules associated with differentially expressed genes were investigated via Ingenuity Pathway Analysis (IPA) and hub gene analysis. Quantitative real-time PCR was used to check the validation of chip test. We identified 52 up-regulated apoptosis-related genes and 26 down-regulated selenium-related genes between KBD and controls, and these genes associated with the “MYC-mediated apoptosis signaling pathway”. We confirmed the results from array studies with quantitative real-time PCR analysis. Our results suggest that abnormal regulation of selenium metabolism and apoptosis through the MYC mediated signaling pathway contributes to the pathogenesis of KBD, but the relationship between apoptosis gene and selenium gene was not found.     

探讨现代医院药剂管理的思路及意义

随着我国医疗体制的改革,医院也面临着新形势下自身管理体系和管理方式的改革和创新。现代医院的管理模式也发生很大变化,从传统经验管理型逐步转变为现代管理型。现代医院管理采用科学的管理方法和管理技术,广泛地运用现代自然科学和社会科学的研究成果使管理手段现代化、科学化。现代医院药剂管理是医院管理工作的重要组成部分,对医疗质量及用药安全起着重要的作用。医院药剂管理的现代化可以保证用药的安全性,提高医疗质量,提高医院的核心竞争力,促进现代市场经济体制下医疗水平的提升和医院的发展。现代医院药剂管理的关键在于如何实现新模式下的管理目标及如何提高新模式下的药剂管理水平。本文结合我院在医疗改革背景下逐渐创新各个环节的管理模式,完善管理体系,突破管理瓶颈而获得的实践体会,进一步探讨现代医院药剂管理的方法及意义,为医院药剂管理工作提供理论指导。     

miRNA-125b在膀胱癌中的表达和意义

目的:探讨膀胱癌组织中miR-125b的表达在膀胱癌发生发展中的作用及其临床意义。方法:采用应用茎环RT-PCR的方法检测66例膀胱尿路上皮癌组织标本的miR-125b的表达,另有16例正常膀胱黏膜组织作对照,并结合临床病理资料进行统计学分析。结果:miR-125b在膀胱癌组织中的表达显著高于非肿瘤的正常膀胱黏膜组织(p0.05),miR-125b的水平还与表达水平与膀胱癌的组织学分级、肿瘤转移、术后复发均明显相关性(P0.05)。结论:miR-125b在膀胱癌组织中表达量升高,且与组织学分级、肿瘤转移、术后复发相关,可能作为膀胱癌诊断和预后指标。     

On the Evaluation of Information Flow in Multivariate Systems by the Directed Transfer Function

The directed transfer function (DTF) has been proposed as a measure of information flow between the components of multivariate time series. In this paper, we discuss the interpretation of the DTF and compare it with other measures for directed relationships. In particular, we show that the DTF does not indicate multivariate or bivariate Granger causality, but that it is closely related to the concept of impulse response function and can be viewed as a spectral measure for the total causal influence from one component to another. Furthermore, we investigate the statistical properties of the DTF and establish a simple significance level for testing for the null hypothesis of no information flow.
