Bacillus thuringiensis subsp.israelensis andB. sphaericus strains 2362 and 1593 were grown in media based on defatted mustard-seed meal (MSM). The meal contains 40% (w/w) protein, with glutamic acid and arginine as the major amino acids. The toxic potencies of the final bacterial powders towardsCulex pipens quinquefasciatus Say, compared with those of the respective international reference standards, were 46% forB. thuringiensis subsp.israelensis, 62% forB. sphaericus 2362 and 88% forB. sphaericus 1593 when 2% (w/v) MSM was used for growth. With 4% (w/v) MSM,B. thuringiensis subsp.israelensis grew better but had undetectable larvicidal activity, whereas theB. sphaericus strains not only grew better but gave a higher degree of sporulation and toxicity. Roche Susceptibility Test (RST) Medium represents the most completely optimized and convenient fully defined medium described. It requires no post-autoclaving supplementation with vitamins, supports good growth of all common aerobic and anaerobic pathogens and may be used as a broth or agar gel on which the swarming of Proteus spp. is virtually eliminated. The broth as a superior buffering capacity to most complex media and an osmolality and pH close to those of human serum. RST is highly satisfactory for the susceptibility testing of commonly used antibiotics and meets the requirements of the National Committe for Clinical Laboratory Standards of the U.S.A. in almost every respect. 相似文献

Summary The induction and decay of ornithine decarboxylase (ODC) by insulin and asparagine in cultures of H4-II-EC3 (H35) hepatoma
cells was studied in a modified Waymouth medium in the presence of fetal bovine serum (FBS) and in serum-free media. The insulin
response was enhanced by the presence of asparagine although the effect of asparagine was not so much on the initial increase
as it was on a slowing of the decline after the maximum was reached at 6 to 8 h after the supplements were added together
with fresh medium. In all cases the initial ODC activity was zero at zero time for addition of media and supplements, and,
after reaching the maximum, activity declined to near zero by 24 h. Fetal bovine serum gave induction that followed a similar
time course but was inferior to the combintion, of insulin plus asparagine and, in fact, FBS inhibited the latter response.
Putrescine (the product formed from ornithine by ODC), at 10−5
M, markedly inhibited the induction of ODC by insulin or FBS, but the inhibition was less when asparagine was present.

The Δ9-desaturase system in liver microsome from rats treated chronically with ethanol was studied. Stearoyl-CoA desaturase activity decreased by 80% and palmitoyl-CoA desaturase activity was not detectable in microsomes from ethanol-fed rats, while activities of electron transport components such as NADH-cytochrome c and NADH-ferricyanide reductases remained unchanged. However, chronic ethanol administration resulted in an adaptive induction of the activity of NADPH-cytochrome c reductase and the contents of cytochrome b5 and P-450. The activity of the terminal component (cyanide-sensitive factor; CSF) of the desaturase system was greatly depressed by ethanol treatment. The NADH/NAD ratio in microsomes of ethanol-fed rats increased over 2-fold. These results suggest that, during chronic ethanol ingestion, decreased activities of Δ9-desaturases are due mainly to a decreased content of the terminal component of the desaturase system. 相似文献

Summary A series of cell lines unique in insect virus susceptibility pattern have been isolated from the ovaries of the gypsy moth
(Lymantria dispar: Lepidoptera: Lymantriidae) on a synthetic medium with mammalian and avian serum supplementation. Growth curves showed the
poorest growth occurring on peptone-based media with somewhat better growth on amino-acid-based media. The best growth was
obtained with combined media. Serological study distinguished the present cell lines from one another and from cell lines
derived from other insect species grown routinely in the same laboratory. Baculovirus susceptibility among the new lines varied
from no response to a specific complete replication response upon challenge by the homologous (gypsy moth) nuclear polyhedrosis
virus.

Summary The air content in three types of propagation media, Jiffy-7 and Jiffy-9 which are Sphagnum peat and Grodan which is rockwool,
were investigated when they were held at moisture tensions of 0,6 and 12 cm measured from the base of the media. At 0 cm tension
the air content (vol. %) was highest in Jiffy-9 and lowest in Jiffy-7. At 12 cm tension the air content was higher in Grodan
than in Jiffy-9 and Jiffy-7.
Oxygen diffusion coefficients (ODC) and oxygen diffusion rates (ODR) were measured at the different air contents. At air contents
below 20 vol. % ODC was about the same for Jiffy-9 and Grodan but at air contents above 20 vol.% it was larger for Jiffy-9
than for Grodan. The oxygen diffusion rate was measured at 0, 4 and 8 cm moisture tension. At all tensions it was approximately
20% higher in Jiffy-9 than in Grodan and Jiffy-7. The ODR in Jiffy-7 and Grodan were affected equally at the same tension,
although Grodan contained more air.

Excitatory amino acid receptor binding parameters were investigated in a spontaneous dog model of chronic hepatic encephalopathy. L-[3H]Glutamate, (+)-[3H]-5-methyl-10,11-dihydro-5H-dibenzo[a,d]cyclohepten-5,10-im ine maleate ([3H]MK-801), [3H]kainate, and alpha-[3H]-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid ([3H]AMPA) binding experiments were performed using crude cerebrocortical synaptosomal membrane preparations from dogs with congenital portosystemic encephalopathy (PSE) and control dogs. There was no change in the affinity or density of L-[3H]-glutamate or [3H]MK-801 binding sites in dogs with congenital PSE compared with control dogs. However, in the PSE dogs there was a significant reduction in the density of [3H]kainate binding sites compared with control dogs and abolition of the low-affinity [3H]AMPA binding site. The relative binding capacity of PSE synaptosomal membranes for [3H]kainate and [3H]AMPA was expressed as the ratio Bmax/KD. There was a significant inverse correlation between the Bmax/KD ratio for [3H]AMPA binding and the worst grade of encephalopathy experienced by each dog. These results suggest that there is a significant perturbation of cerebrocortical non-N-methyl-D-aspartate receptor binding in dogs with congenital PSE which may have relevance to the pathogenesis of hepatic encephalopathy. 相似文献

本文报道了病毒唑及其与干扰素联合对23例慢乙肝近、远期抗病毒效应观察结果,并与12例未经抗病毒治疗的慢乙肝作对照(Ⅲ组),其中病毒唑与干扰素联合组(I组)12例,病毒唑组(Ⅱ组)11例。近期HBeAg、DNAP及HBV·DNA阴转率,I组分别为(41.6%、55.5%及28.s%,Ⅱ组45.4%、100%及33.3%,Ⅲ组8.3%、16.6%及8.3%。远期HBeAg及DNAP阴转率在I组分别为70%、83.7%。Ⅱ组60%、100%,Ⅲ组8.3%,50%。综合抗病毒效应,近期有效率在I、Ⅱ、Ⅲ组分别为50%、72.7%及16.6%。远期则分别为70%、55.5%及8.3%。无一例出现毒副反应。均完成疗程。上述结果提示病毒唑及其与干扰素联合对慢乙肝均有一定抗病毒效应,联合应用并不优于单一用药,但毒副反应也未见加重,而且病毒唑药源较方便,使用较安全,值得进一步研究。 相似文献

通常认为,慢性扁桃体炎主要致病菌是金黄色葡萄球菌和乙型溶血性链球菌。通过50例慢性扁桃体炎的病原微生物培养、组织切片细菌学及电镜等研究,发现慢性扁桃体炎组织中细菌L型也相当常见,L型培养阳性率是88.5%,且组织切片L型感染率与培养阳性率基本一致(P>0.05)。电镜在扁桃体组织间质及上皮细胞、淋巴细胞等多种细胞内均见到细菌L型。提示慢性扁桃体炎与细菌L型感染关系极为密切。并认为,L型侵入组织并在宿主细胞内生长的特性,可能是慢性扁桃体炎反复发作、迁延不愈的重要原因。 相似文献

The inorganic formulations of fourteen common plant tissue culture basal media were examined from the primary literature. Inaccuracies and errors were found for molecular formulae, chemical hydrations, and molar equivalences for iron/EDTA complexation. A comparison with published basal medium formulations from six commercial suppliers uncovered additional inaccuracies, modifications, and errors, thereby emphasizing the need for investigators to examine and describe medium formulations precisely in future publications. 相似文献
