The phycobilisomes and phycobiliproteins of Synechococcus sp. PCC 7002 wild-type strain PR6000 have been isolated and characterized. The hemidiscoidal phycobilisomes of strain PR6000 are composed of eleven different polypeptides: phycocyanin and subunits; allophycocyanin and subunits; subunit of allophycocyanin B; the allophycocyanin -subunit-like polypeptide of Mr 18 000; the linker phycobiliprotein of Mr 99 000; and non-chromophore-carrying linker polypeptides of Mr 33 000, 29 000, 9000, and 8000. Several of these polypeptides were purified to homogeneity and their amino acid compositions and amino-terminal amino acid sequences were determined. Analyses of the phycobiliproteins of Synechococcus sp. PCC 7002 were greatly facilitated by comparative studies performed with a mutant strain, PR6008, constructed to be devoid of the phycocyanin and subunits by recombinant DNA techniques and transformation of strain PR6000. The absence of phycocyanin did not greatly affect the allophycocyanin content of the mutant strain but caused the doubling time to increase 2–7-fold depending upon the light intensity at which the cells were grown. Although intact phycobilisome cores could not be isolated from this mutant, it is probable that functionally intact cores do exist in vivo.Abbreviations used SDS-PAGE

A bacterium tentatively identified as a Pseudomonas sp. was isolated from a laboratory aquifer column in which toluene was degraded under denitrifying conditions. The organism mineralized toluene in pure culture in the absence of molecular oxygen. In carbon balance studies using [ring-UL-14C]toluene, more than 50% of the radioactivity was recovered as 14CO2. Nitrate and nitrous oxide served as electron acceptors for toluene mineralization. The organism was also able to degrade m-xylene, benzoate, benzaldehyde, p-cresol, p-hydroxybenzaldehyde, p-hydroxybenzoate and cyclohexanecarboxylic acid in the absence of molecular oxygen. 相似文献

Polyclonal antiserum specific for ferredoxin-nitrite reductase (EC 1.7.7.1) from the green alga Chlamydomonas reinhardii recognized the nitrite reductase from other green algae, but did not cross-react with the corresponding enzyme from different cyanobacteria or higher plant leaves. An analogous situation was also found for ferredoxin-glutamate synthase (EC 1.4.7.1), using its specific antiserum. Besides, the antibodies raised against C. reinhardii ferredoxin-glutamate synthase were able to inactivate the ferredoxin-dependent activity of nitrite reductase from green algae.These results suggest that there exist similar domains in ferredoxin-nitrite reductases and ferredoxin-glutamate synthases from green algae. In addition, both types of enzymes share common antigenic determinants, probably located at the ferredoxin-binding domain. In spite of their physicochemical resemblances, no apparent antigenic correlation exists between the corresponding enzymes from green algae and those from higher plant leaves or cyanobacteria.Abbreviations Fd

在云南陇川县的蟋蟀Gryllus sp.体上采得一种革螨,系厉螨科Ltelapidae Berlese中未曾描述过的种类,并为之建立新属,订名为滇厉螨属Dianolaelaps gen.nov.,属模标本为蟋蟀滇厉螨Dianolaelaps gryllus sp.nov.。新属的主要鉴别特征是:雌螨螯钳动趾端部分三叉;颚角内侧有沟漕并具齿突;口下片末端,在颚角与下咽间有长圆形结构,其内缘并具透明的尖齿列;涎针有膜状结构包绕;足Ⅰ—Ⅲ基节各具一棘,各基节远端侧具细小毛列。 相似文献

Abstract Outer membranes of Shigella species and E. coli K-12 carrying large invasive plasmids and isogenic non-invasive strains without plasmids were analyzed by SDS-PAGE. The immunoblotting analysis of the outer membrane proteins of these bacteria was performed with monoclonal antibody (mAb) made against A and B subunits of Shiga-like toxin (SLT). The SLT was detected in the outer membranes of S. dysenteriae 1 IDBM11, S. sonnei PNS20, S. flexneri M90T, S. dysenteriae 60R, and E. coli K-12 strain AB2463. The two other E. coli K-12 strains, C600 and 933J were included as controls for low and high toxin producers respectively. The outer membrane protein band of molecular weight 70 kDa was common to all bacterial strains studied. The most prominent band of 70 kDa protein was seen to be present in the high toxin producing plasmidless strain of S. dysenteriae 60R and the lysogenic strain of E. coli 933J. The invasive strains of S. dysenteriae 1 and S. flexneri M90T which carry the large invasive plasmids showed the least prominent band of 70 kDa protein.
The immunoblotting analysis of Shiga-toxin partially purified from the S. dysenteriae 60R strain revealed the absence of 70 kDa band on SDS-PAGE, instead the two dissociated subunits were seen. Furthermore, periplasmic Shiga-toxin proteins also showed the complete dissociation into A and B subunits. However, under the same denaturing conditions, the 70 kDa protein band cross-reacting with mAb against A and B subunits was still present in the outer membranes of all different strains. 相似文献

Abstract Perchloroethylene (PCE) was reductively dechlorinated to trichloroethylene in a 10% anaerobic sewage sludge. About 80% of the initially added PCE (300 nmol) was dechlorinated within three weeks. The calculated rates were 250 nM and 445 nM · day−1 during the first and second weeks of incubation, respectively. The depletion of PCE varied in sludges obtained from different sources.
The role of methanogenesis in the dechlorination of PCE was evaluated by inhibiting the methanogens by addition of bromoethane sulfonic acid, a potent methanogenic inhibitor. Dechlorination of PCE was significantly inhibited in sludges amended with the inhibitor. Almost 41–48% less PCE was dechlorinated in sludges containing 5 mM BESA, indicating a relation between the two processes (methanogenesis and dechlorination). Direct proof that methanogens can transform chlorinated aliphatic compounds was obtained using axenic cultures of acetate-cleaving methanogens. Methanosarcina sp , originally isolated from a chlorophenol degrading consortium, showed significantly higher dechlorinating activity as compared to Ms. mazei . Based on these studies and other recently reported observations, it appears that methanogens/methanogenesis play an important role in the anaerobic dechlorination of chlorinated aliphatics such as PCE. 相似文献

Meloidogyne mayaguensis n. sp. is described and illustrated from specimens obtained from galled roots of eggplant, Solanum melongena L., from Puerto Rico. The perineal pattern of females is round to ovoid with fine, widely spaced striae. It has occasional breaks of striation laterally and a circular tail tip area lacking striae. The stylet, 15.8 μm long, has reniform knobs that merge gradually with the stylet shaft. Males have a high, rectangular, smooth head region, not set off from the body contour. The labial disc is continuous with the medial lips which do not slope posteriorly. The styler, 22.9 μm long, has large rounded backward sloping knobs; the shaft is of uneven diameter. Mean body length of second-stage juveniles is 453.6 μm. The truncate head region is not annulated, and the rounded, slightly raised labial disc and the crescentic medial lips form dumbbell-shaped lip structures. The stylet, 11.6 μm long, has rounded, posteriorly sloping knobs. The slender tail, 54.4 μm long, gradually tapers to a bluntly pointed tip. Tomato, tobacco, pepper, and watermelon are good hosts; cotton and peanut are not hosts. M. mayaguensis n. sp. reproduces by mitotic parthenogenesis and has a somatic chromosome number of 2n = 44-45. Summary Effects of the phytotoxic compounds (AAL-toxins) isolated from cell-free culture filtrates of Alternaria alternata f.sp. lycopersici on in vitro pollen development were studied. AAL-toxins inhibited both germination and tube growth of pollen from several Lycopersicon genotypes. Pollen from susceptible genotypes, however, was more sensitive for AAL-toxins than pollen from resistant plants, while pollen of species not belonging to the host range of the fungus was not significantly affected by the tested toxin concentrations. AAL-toxins elicit symptoms in detached leaf bioassays indistinguishable from those observed on leaves of fungal infected tomato plants, and toxins play a major role in the pathogenesis. Apparently, pathogenesis-related processes and mechanisms involved in disease resistance are expressed in both vegetative and generative tissues. This overlap in gene expression between the sporophytic and gametophytic level of a plant may be advantageously utilized in plant breeding programmes.
