U2AF homology motifs (UHM) are protein domains that bind peptidic UHM ligand motifs (ULM) and thus form an intricate network of interactions involved in splicing regulation. Here, we report the backbone assignment of the UHM domain of the splicing factor Puf60 as well as 1H, 15N chemical shifts upon binding of the ULM peptides U2AF65 (85–112), SF1 (1–25), SF3b155 (194–229), SF3b155 (317–357), and Prp16 (201–238).  相似文献

Bacterial thiol peroxidases (Tpxs) are antioxidant enzymes which exist in various bacteria. Tpxs reduce the lipid hydroperoxides to protect the membrane lipid from destruction by reactive oxygen species. Tpxs are essential enzymes for bacterial anaerobic growth. Herein, we report the resonance assignments of 1H, 13C, and 15N atoms in both the reduced and oxidized forms of Bacillus subtilis Tpx.  相似文献

The common thread of evolution runs through all science disciplines, and the concept of evolution enables students to better understand the nature of the universe and our origins. “Science and the Concept of Evolution” is one of two interdisciplinary science Core courses taken by Dowling College undergraduates as part of their General Education requirements. The course examines basic principles and methods of science by following the concept of evolution from the big bang to the origin and evolution of life. Case studies of leading scientists illustrate how their ideas developed and contributed to the evolution of our understanding of the world. Evidences for physical, chemical, and biological evolution are explored, and students learn to view the evolution of matter and of ideas as a natural process of change over space and time.  相似文献

Thermal inactivation kinetic analysis and molecular modeling computation were jointly utilized to illuminate the detailed stabilization mechanism of trypsin caused by methoxypolyethylene glycol (MPEG) modification. First, trypsin was modified by MPEG (molecular mass 350 Da) to enhance its thermal stability. As expected, the modified trypsin was more stable against temperature than the native form. Second, a new kinetic model, which has the ability of taking the thermal denaturation and autolysis effects of proteases into account, was established and used to analyze the thermal inactivation process of the native and modified trypsin. The kinetic analysis showed that the increased thermal stability of MPEG modified trypsin is the joint result of a reduction in autolysis and a decrease in thermal denaturation. Finally, the molecular modeling technique was also employed to calculate some structural information change, i.e. solvent accessible surface, intramolecular hydrogen bond and root mean square fluctuation, between the native and modified trypsin. The results of molecular modeling computation demonstrated that (i) the steric hindrance caused by MPEG chain would result in the decreased rate of autolysis, (ii) the decreased rate of thermal denaturation should be ascribed to the increased number of hydrogen bond, not the result of the increased molecular rigidity.  相似文献

Phyllanthus orbicularis is a medicinal plant, endemic to Cuba, whose aqueous extract has proven antiviral properties. This plant extract is being studied for treatment of viral diseases in animals and humans. Antimutagenic activities of this plant aqueous extract have been investigated as an additional and possible valuable property. Antimutagenesis was assayed against the mutagenic activity of m-phenylenediamine (m-PDA), 2-aminofluorene (2-AF), 1-aminopyrene (1-AP), 2-aminoanthracene (2-AA) and 9-aminophenantrene (9-AP) in Salmonella typhimurium (S. typhimurium) YG1024, in different co-treatment approaches. This plant extract produced a significant decrease of the mutagenesis mediated by these aromatic amines (AA) in the following order: m-PDA>2-AA>2-AF>9-AP>1-AP. Interactions with S9 enzymes and transformation of promutagenic amines and their mutagenic metabolites by chemical reactions to non-mutagenic compounds are proposed as possible mechanisms of antimutagenesis. Mutagenesis mediated by m-PDA was almost completely abolished when S9 mixture was co-incubated with the plant extract during 40 min, previous to the addition of the m-PDA and bacterial cells to the assay. Similar results were found with 2-AA and 1-AP, but the reduction of the mutation rate was not so dramatic. In contrast, the most significant antimutagenic effect against 2-AF and 9-AP was seen when these chemicals were co-incubated with the plant extract, before addition of the S9 mixture and bacterial cells to the assay. Therefore, inhibition or competition for S9 enzymes seems to be the main antimutagenic mechanism of this plant extract against m-PDA, 2-AA and 1-AP, whilst a chemical modification of 2-AF and 9-AP into non-promutagenic derivatives is likely to be the main mechanism of antimutagenesis against both compounds.  相似文献

Forty‐two essential oil samples were isolated from leaves of Xylopia rubescens harvested in three forests of Southern Ivory Coast. All the samples have been submitted to GC‐FID and the retention indices (RIs) of individual components have been measured on two capillary columns of different polarity. In addition, 20 oil samples, selected on the basis of their chromatographic profile, were also analyzed by 13C‐NMR and 24 components (78.0 – 92.4% of the whole compositions) have been identified. The content of the main components varied drastically from sample to sample: furanoguaia‐1,4‐diene (5.7 – 54.1%), furanoguaia‐1,3‐diene (1.1 – 10.5%), (8Z,11Z,14Z)‐heptadeca‐8,11,14‐trien‐2‐one (4.3 – 16.0%), and (E)‐β‐caryophyllene (1.7 – 17.3%). Hierarchical cluster and principal components analysis of the 42 oil compositions allowed the distinction of two well‐differentiated groups of unequal importance within the oil samples. Oil samples of the main group (Group II) contained mainly furanoguaia‐1,4‐diene (mean [M] = 43.1%; standard deviation [SD] = 3.2%) while furanoguaia‐1,3‐diene (M = 8.4%; SD = 0.9%) and (8Z,11Z,14Z)‐heptadeca‐8,11,14‐trien‐2‐one (M = 7.1%; SD = 1.5%) were present at appreciable contents. The composition of Group I was dominated by furanoguaia‐1,4‐diene (M = 17.0%; SD = 8.5%), (8Z,11Z,14Z)‐heptadeca‐8,11,14‐trien‐2‐one (M = 10.2%; SD = 2.4%) and (E)‐β‐caryophyllene (M = 9.5%; SD = 5.3%).  相似文献

The fruit (aril) of the endangered genus Taxus plants is an abandoned herbal resource. Traditionally, people enthusiastically focus on its bark, its renewable, tremendous arils fall into the soil with seeds after they are mature. The present research investigated the fruit of two species from the genus Taxus, Taxus chinensis var. mairei, and Taxus media, with regards to their antioxidant and antihyperglycaemic activities, safety, and bioactive constituents. Results showed that T. chinensis var. mairei and T. media both had certain biological activities with T. chinensis var. mairei better in antioxidant activity and T. media better in antihyperglycaemic activity. Correlation analysis revealed that the differences in bioactivities depended on content of their mainly chemical components. The mice acute oral toxicity test indicated that the methanol extracts of the two biotypes of Taxus were safe. And nineteen compounds were tentatively assigned from the two varieties, via tandem mass spectrometry using a LC‐ESI‐Q‐TOF‐MS instrument, which included phenols, flavonoids, and terpenes. These results indicate a possible application of Taxus fruit extracts in various fields like in food industry, however, this still needs further investigations.  相似文献

The essential oils (EOs) of green seeds from Daucus carota subsp. maximus growing wild in Pantelleria Island (Sicily, Italy) were characterized. EOs were extracted by steam distillation, examined for their inhibitory properties against food‐borne Gram‐positive and Gram‐negative bacteria and analyzed for the chemical composition by gas chromatography (GC) and mass spectrometry (MS). Undiluted EOs showed a large inhibition spectrum against Gram‐positive strains and also vs. Acinetobacter spp. and Stenotrophomonas maltophilia. The minimum inhibition concentration (MIC) was in the range 1.25 – 2.50 μl/ml for the most sensitive strains. The chemical analysis indicated that Dcarota subsp. maximus EOs included 34 compounds (five monoterpene hydrocarbons, six oxygenated monoterpenes, 14 sesquiterpene hydrocarbons, four oxygenated sesquiterpenes, camphorene and four other compounds), accounting for 95.48% of the total oil, and that the major chemicals were carotol, β‐bisabolene, and isoelemicin.  相似文献

为了解华石斛(Dendrobium sinense)的化学成分,采用多种柱色谱技术从其全草乙醇提取液中分离纯化了10个化合物,经波谱分析分别鉴定为:鼓槌石斛素(1)、2′,4′-二羟基查尔酮(2)、2,5,7-三羟基-4-甲氧基-9,10-二氢菲(3)、4,7-二羟基-2,3-二甲氧基-9,10-二氢菲(4)、2,5-二羟基-3,4-二甲氧基-9,10-二氢菲(5)、2,7-二羟基-3,4,6-三甲氧基-9,10-二氢菲(6)、(E)松柏醛(7)、反式对羟基肉桂酸酯(8)、对羟基苯丙酸甲酯(9)和十二元内环酯(10)。所有化合物均为首次从华石斛中分离得到,其中化合物2、6、7和10对乙酰胆碱酯酶具有一定的抑制活性。  相似文献

迄今为止疫苗在人类和动物传染性疾病的控制和预防中依然发挥着其他药物种类难以企及的重要作用。疫苗设计成功的基础在于有效地递送抗原物质以诱发强有力的保护性免疫反应,而疫苗递送载体的合理应用可以加强、改善、甚至改变抗原物质所诱发的免疫应答过程,从而带来优化疫苗接种效果,简化免疫接种程序等有益效果。目前研发中常用的疫苗递送载体可以分为生物载体(如病毒与细菌)与化学载体(如微针与脂质体)两类,在不同递送系统使用方面的重要考虑是有效地利用载体的装载能力和相应特性来达到理想的免疫效果。目前疫苗递送技术的快速进展为现代疫苗的发展提供了有力的技术支撑。  相似文献
